Korean Lentinus edodes SR-1 was cultured to multiply the mycelia in the complete broth medium (C/N=13.1) for mushroom, and protein-bound polysaccharides were extracted from the cultured broth containing mycelia (The whole cultured broth was used to increase the yields: 80% of protein-bound polysaccharides were existed at the cell wall of mycelia and 20% of those were secreted extracellularily in this culture). Protein-bound polysaccharides in the cultured broth containing mycelia were extracted by using three different methods: 1) Extraction with hot water, 2) Disintegration of cell wall by glass bead mill treatment before extraction with hot water, and 3) Cellulase treatment before extraction with hot water. The highest yield was obtained (930 mg polysaccharides/100 mL culture broth) when protein-bound polysaccharides were extracted with 2) method. The extracted crude protein-bound polysaccharides were purified using protease, DEAE-cellulose and Sephadex G-100. The growth inhibition activity for $P_{388}$, mouse leukemic cell, increased (53.7, 62.2, 93.7% and 97.4%) as the purification level increased. Protein-bound polysaccharides contained 46.1% of polysaccharides, 7.3% of protein, and trace amounts of minerals. Polysaccharides contained glucose, galactose, xylose and mannose. The content of proline and glycine were high, however, methionine and leucine were not found. The major minerals were Na, K, Zn, and Ca.
Delivery of therapeutic protein drugs is a hot issue in the clinical application, because protein drugs have low side effects and highly therapeutic effects compared with chemical drugs. Despite their prominent advantages, protein drugs have high risk for human therapy such as their easy degradation by proteolytic enzymes, renal filtration and immune response. Over the past few decades, a large number of polysaccharides as vehicles for the protein delivery system have been developed to overcome the problems. This review presents the studies on protein delivery based on polysaccharides used as stabilizer and vehicles comprising nano- or microspheres to overcome inherent limitations of therapeutic proteins.

본 연구는 황색포도상구균의 병원성 인자인 alpha-toxin, casular polysaccharides (CPS)와 fibronectin-binding protein (FnBP)을 이용해 실험동물인 토끼에서의 항체가 형성 능과 공격접종 후 방어능에 관한 연구를 위하여 수행되었으며 향후 젖소 유방염 아단위 항원 을 이용한 백신개발 가능성을 탐색하고자 수행되어졌다. 황색포도상구균의 alpha-toxin, capsu18r polysaccharides (CPS)91 fibronectin-binding protein (FnBP) 항원을 이통재 효소면 역측정법을 통한 혈중 IgG 항체가수준을 측정하였으며 alpha-toxin, capsular polysaccharides (CPS)와 fibronectin-binding protein (FnBP)으로 면역시킨 토끼에서 대조군의 토끼보다 혈 중 항체가 수준이 1차 면역 이후 유의성 있게 높았다 (p<0.05). 백신에 사용된 alpha-toxin, capsular polysaccharides (CPS)와 fibronectin-binding protein (FnBP) 항원중 capsular polysaccharides (CPS)가 다른 alpha toxin과 fibronectin-binding protein (FnBP)의 혈중항 체가 수준과 비교하여 볼 때 비교적 낮은 수준이었다. 세균을 혈중으로 공격접종한 후 대조군 과 백신접종군의 혈중내 세균제거율에 있어 대조군에 비해 백신접종군에서 유의성 있게 낮은 균수를 보였다 (p<0.05). 또한 장기내 균수측정실험결과 대조군의 장기보다 백신접종군에서 유의성 있게 세균수가 낮게 출현하였다 (p<0.05). 결론적으로 본 연구결과 황색포도상구균의 3가지 항훤 alpha-toxin, capsular polysaccharide와 재조합 fibronectin binding protein을 이 퐁한 실험동물에서 안단위 유방염 백신은 방어능이 있다고 생각된다.
The hypoglycemic effects of 2 mushrooms, Pleurotus ostreatus and Lentinus edodes, on streptozotocin(STZ) induced diabetic rats were investigated in this study . Diabets mellitus was induced in male Sprague-Dawley rats by the injection of STZinto the tail vein at a dose of 45mg/kg. Sprague-Dawley male rats(200-250g) were assigned to one control and three STZ-diabetic groups. Diabetic groups were assigned to STZ-control, pleurotus ostreatus and Lenitinus edodes groups. All groups were fed a AIN 76 diet. The two experimental groups were fed with each protein-bound polysaccharide(150mg/kg BW) for 14 days and with carboxymethyl cellulose for STZ-control group. The body weight gain was monitored and the blood levels of glucose and cholesterol were measured . Levels of protein triglyceride, and free fatty acid in plasma were analysed. Serum aminotransferase activity as also measured. The body weight gain was lower in the all diabetic groups than in the of normal group. The weight of spleen was reduced by adminstration of the Lentinus edodes protein-bound polysaccharides. The result suggest that orally administered Lentinus edodes protein-bound polysaccharides exhibited hypoglycemic effect in STZ -induced diabetic rats and that these protein-bound polysaccharides may be useful for the management of diabetes mellitus.
Heteropolysaccharides isolated from liquid cultures of nine Tremella species contained 0.3 to 1.2% protein, 2.7 to 5% ash, 0.9 to 3.4% acetyl groups, 76.5 to 84.2% carbohydrates and trace amounts of starch. The polysaccharides in aqueous solution were slightly acidic (pH 5.1 to 5.6). They consisted of the following monomeric sugars: fucose, ribose, xylose, arabinose, mannose, galactose, glucose and glucuronic acid. The backbones of the polysaccharide structures consisted of $\alpha$-(1$\rightarrow$3)-links while the side chains were $\beta$-linked.
The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of L. edodes were investigated. The use 2% solution of surface active agent, Triton X-100 was effective for extraction of the protein-bound polysaccharide from the mycelium. The extraction of the protein-bound polysaccharides from mycelium with hot water was achieved by 4 hours extraction at 10$0^{\circ}C$. For the separation and partial purification of the protein bound polysaccharides the column chromatography using DEAE-Cellulose, DEAE-Sephadex and Sephadex proved to be effective.
The Basidomycetes fungus Agaricus blazei Murill has been well known as a health food for the prevention of cancer, diabetes, hyperlipidemia, arteriosclerosis and chronic hepatitis. This study was concentrated to investigate the characteristics of crude protein-bound polysaccharides(PBP) compositions extracted from Agaricus blazei Murill. In order to produce crude polysaccharides, culture conditions were examined using YMK media. Total sugars and protein contents of PBP were detected by phenol-sulfuric acid method and Bradford -assay. Hexosamine was found to be involved in the linkage, N-linked and O-linked types. To identify helical conformation existence, wavelength was measured using Congo red after the treatment with alkali solution.
Polysaccharide content in protein-polysaccharides (PPS) extracted from sea mustard, sea tangle and fusiform were 40.61, 38.42 and 52.80% , respectively. 5% of sea tangle PPS showed highest inhibitory activity on 4-nitorquinoline -1-oxide(4-NQO) against Salmonella typhimurium TA100 compared to the other seaweed PPS. 5% of sea mustard PPS showed highest inhibition ration of 62% on 2-nitrofluorene(2-NF)against Salmonella typhimurium TA98. These PPS extracts showed cytotoxic activity against human colon cancer cell (SW-480), and showed mild cytotoxic activity on human stomach cancer cell(SNU-1) and human hepatic cancer cell(HepG 2).
Protein-polysaccharides(PP) extracted from four species of gulfweed, fusiforme, sea-mustard and sea-tangle were examined for antitumor activity toward sarcoma-180 implanted in mice, ICR. Polysaccharides in protein-polysaccharides extracted from gulfweed, fusiforme, sea-mustard and sea-tangle were 61.48, 55.61, 34.06 and 30.28%, respectively. Monosaccharides of four seaweeds consisted of glucose, galactose, mannose, fructose and xylose, and major amino acids consisted of glutamic acid, aspartic acid, cystein, valine and glycine. Antitumor activity of the protein-polysaccharides extracted from sea-mustard showed the highest inhibition ratio of 69.76% when PP was injected to the mice at the dose of 100mg/Kg/day. PP of sea-tangle showed the maximum survival ratio of 25.22% when injected at the dose of 100mg/Kg/day.
Optimum conditions for hydrolysis were investigated to enhance water-solubilities of protein-bound polysaccharides in the basidiocarps of Ganoderma lucidum by treating chymotrypsin. We also attempted with Ganoderma lucidum residue remaining after extracting hot water-soluble compoents in Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chymotrypsin, 2% Gampderma lucidum or 6% Ganoderma lucidum residue, at pH 10 and at $ 40^{\circ}C$), the amounts of total protein and carbohydrate of hydrolysate were measured. Michaelis constant, $K_{m}$, and maximum rate, $V_{max}$, calculated by Lineweaver-Buck plot for the hydrolysis of Ganoderma lucidum were 1.73% and 0.073%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 2.40% and 0.033%/min respectively. The amount of polysaccharide isolated from Ganoderma lucidum (100 g) treated with chymotrypsin was only 3.07 g, but significantly increased amount (14.34 g) of polysaccharides was isolated from Ganoderma lucidum residue (100 g) treated with chymotrypsin. The protein-bound polysaccharide was isolated from the non-hydrolyzed and hydrolyzed sample and molecular weights of the polysaccharide were measured by Sepharose CL-48 gel filtration.
