이 연구의 목적은 대장균 분자 샤페론 GroEL의 시험관 내 단백질 접힘에 있어서 반응온도의 영향과 보조샤페론의 필요 여부를 자발적 재접힘이 가능한 온도와 그렇지 않은 온도조건에서 조사하는 것이다. 여러 조건하에서 GroEL에 의한 두 가지 기질 단백질의 재접힘을 반응속도론적으로 조사하기 위하여 GroEL에 의한 단백질 침전생성억제와 변성된 단백질의 재접힘을 광범위하게 조사하였다. 자발적 재접힘이 가능하지 않은 $37^{\circ}C$에서는 ATP와 완전한 GroEL 시스템이 변성된 폴리펩티드의 재접힘을 위하여 필요하다는 것을 확인하였다. 하지만, 자발적 재접힘이 가능한 낮은 온도에서는 자발적 재접힘과 샤페론 의존적 단백질 재접힘이 서로 경쟁하는 것을 알 수 있었다. 따라서 GroEL은 변성된 폴리펩티드의 자발적 접힘 경로를 더 효율적인 단백질 재접힘 경로인 샤페론 의존적 단백질 재접힘 경로로 유도하는 것으로 보인다.
2020 한국인 영양소 섭취기준 개정에서 단백질은 기본적으로 질소평형을 유지하는데 필요한 단백질 양 0.66 g/kg 체중/일에 이용효율 90%를 적용한 평균필요량 0.73 g/kg 체중/일과 변이계수 12.5%를 적용한 권장섭취량 0.91 g/kg 체중/일을 생애주기별 체위기준치 변화에 따라 제시하였다. 그러나 성장기의 단백질 섭취기준에서는 체중 기준을 2017년에 개정된 소아청소년 성장도표에 근거하고, 일본 DRI 자료를 참고로 단백질 식품 이용효율을 차등 적용하였으며, WHO/FAO/UNU 보고서상의 성장에 필요한 단백질량 오류를 수정·반영하여 기준값을 개정하였다. 그 결과 일부 연령구간에서 섭취기준의 상향조정이 있었으나 한국인의 연령대별 1일 평균 단백질 섭취량은 대부분의 연령대에서 단백질 평균필요량의 100%를 넘는 것으로 나타났고, 남자 75세 이상과 여자 65세 이상을 제외하고는 권장섭취량과 비교했을 때도 많은 것으로 나타나 단백질 섭취기준의 상향 조정은 국민의 평균 단백질 섭취기준을 반영하는 것으로 판단된다. 한편, 75세 이상의 단백질 섭취량은 근감소증 예방, 근력 강화 등을 위해 65-74세 단백질 섭취기준과 동일하게 설정하였다. 이번 개정에서 단백질 에너지 적정비율은 기존의 7-20%를 유지하였으나 향후 단백질의 적정한 섭취수준 및 만성질환예방을 위한 단백질 필요량 설정에 관한 과학적 근거마련을 위해 우리나라 국민을 대상으로 한 다양한 중재연구가 수행되어야 할 것이다.
Porphyromonas gingivalis, a Gram negative, anaerobic, asaccharolytic rod, is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. A 30 kDa (heated 24 kDa) hemin-binding protein whose expression is both hemin and iron regulated has recently been purified and characterized in this oral pathogen. This study has identified a hemin-binding P. gingivalis protein by expression of a P. gingivalis genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. A library of genomic DNA fragments from P. gingivalis was constructed in plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$ , and screened for recombinant clones with hemin-binding activity by plating onto hemin-containing agar. Of approximately 10,000 recombinant E. coli colonies screened on LB-amp-hemin agar, 10 exhibited a clearly pigmented phenotype. Each clone contained various insert DNA. The Hind III fragment transferred to the T7 RNA polymerase/promoter expression vector system produced a sligltly smaller (21 kDa) protein, a precursor form, immunoreactive to the antibody against the 24 kDa protein, suggesting that the cloned DNA fragment probably carried an entire gene for the 24 kDa hemin-binding protein.
There are some international standards and organization controling the quality characteristics of varieties and seeds such as UPOV, OECD, ISTA, AOSA. The National List system to check variety performance such as quality characteristics of plant varieties and seeds was established by each countries. The UPOV (International Union for the Protection of New varieties of Plant) regulates the requirement of plant variety protection that is distinctness (D), uniformity (U), stability (S) (here in after called DUS), Novelty and Denomination, and some quality characteristics are used to examine the requirement of plant variety protection by each crops. OECD (Organization for Economic Cooperation Development) seed scheme regulates the seed certification for seed trade between countries. ISTA (International Seed Testing Association) and AOSA(Association for Official Seed Analyst) regulates the seed analysis standard and methods. Most of the countries in Europe has the National List system, which check the value for cultivation and use(VCU), that is to say, variety performance such as yield, quality and stress to environment. All the seeds should be enlisted in the National List before sell the seed in their country. All the quality characteristics checked variety performance are for instance, amylose and amylopectin content in rice, molting quality, $\beta$-glucan, protein in barley, protein and sugar content in soybean, sugar and amylopectin content in corn etc. Conditions for the protection of new variety of plant are DUS. Quality characteristics may be the important characteristics and used to check DUS in crop by crop. It is very important to develop a new characteristics and establish standard method fur examine the VCU and DUS test for each crop.

The glutathione S-transferase (GST) system is useful for increasing protein solubility and purifying soluble GST fusion proteins. However, purifying half of the GST fusion proteins is still difficult, because they are virtually insoluble under non-denaturing conditions. To optimize a simple and rapid purification condition for GST-pyruvate kinase muscle 2 (GST-PKM2) protein, we used 1% sarkosyl for lysis and a 1 : 200 ratio of sarkosyl to Triton X-100 (S-T) for purification. We purified the GST-PKM2 protein with a high yield, approximately 5 mg/L culture, which was 33 times higher than that prepared using a conventional method. Notably, the GST-high-temperature requirement A2 (GST-HtrA2) protein, used as a model protein for functional activity, fully maintained its proteolytic activity, even when purified under our S-T condition. This method may be useful to apply to other biologically important proteins that become highly insoluble in the prokaryotic expression system.
The effect of undecorticated sunflower cake (USFC) as critical protein supplement was assessed and compared with deoiled groundnut cake (DGNC) in adult goats and sheep. The animals were fed a basal diet of wheat straw ad libitum and supplemented with either USFC or DGNC to meet their protein requirement for maintenance. Total dry-matter intake by sheep and goats (g/kg $BW^{0.75}$) on USFC was similar to their counterparts on DGNC supplemented group. However, while intake of cake moiety was significantly (p<0.05) higher in USFC, the intake of wheat straw was significantly (p<0.05) higher by animals on DGNC. Digestibility of various nutrients, except lower crude protein digestibility by goats in USFC group, did not differ significantly between animals given DGNC or USFC. DCP and TDN concentration (% DM) was comparable in sheep and goats irrespective of dietary supplement. Similarly, the intake (g/kg $W^{0.75}$) of DCP, DDM, DOM, and TDN was similar between DGNC and USFC in both sheep and goats. It may be concluded that undecorticated sunflower cake is comparable to deoiled groundnut cake as a critical protein supplement to the roughage based diet of small ruminants.
According to the intercombined review of chemical and biological investigation it has been noted that the metabolizable energy per gram dietary protein of mixed diet of daily intake patterned by Korean population has been found 3.4-3.6 Cal., which entails 10-12% level of the protein calorie percentage of total metabolizable energy, the biological value being fallen within the scope 63-73. The structure of dietary protein has revealed that the lysine and isoleucine were primary limiting amino acids and threonine secondary limiting as a general trend, however, it is assumed that the ultimate nutritional effect of dietary protein might be restricted uniformly among regions by the amount of lysine, since the lysine availability has been yielded as low as 72-82% level. As for the net protein utillization NPUst falls in the range of 52-62 and the NPUop 47-58. In either part the mountainous region has demonstrated lowest value and the urban area highest, these trend being obviously associated with the ratio of animal protein relative to the vegetable origin. The net dietary protein calorie percentage (NDpCal %) has been found within the range of 5-7 that may be capable of meeting the requirement for the maintenance of adult, though for the growth it is insufficient. Present level of total caloric intake would not influence on the fate of protein value of prevailing regional diet in terms of caloric restriction, since the present intake of food energy is higher than the lower limit of caloric intake that would impair the biological performance of dietary protein fed ad libitum basis. Based on the protein efficiency, the adequacy of current level of protein intake was analyzed in terms of utilizable protein, and it has been demonstrated that the 37.8g of utilizable protein in the fishery region and 38.2g in the mountainous region were bellow the FAO recommendation. Accordin to the hematological study it may be interpreted that the anemic symptoms of the mountainous region has some possibility of being related to the inferior status of dietary protein in quality as well as in quantity.

This study evaluated the dietary lysine requirement by measuring the plasma free lysine concentrations in rainbow trout, Oncorhynchus mykiss after dorsal aorta cannulation. A basal diet containing 36.6 % crude protein (29.6 % crystalline amino acids mixture, 5 % casein and 2 % gelatin) was formulated to one of the seven L-amino acid based diets containing graded levels of lysine (0.72, 1.12, 1.52, 1.92, 2.32, 2.72 or 3.52 % dry diet). A total of 35 fish averaging $512{\pm}6.8g$ ($mean{\pm}SD$) were randomly distributed into seven groups with five fish in each group. After 48 h of feed deprivation, each group of fish was fed one of the experimental diets by intubation at 1 % body weight. Blood samples were taken at 0, 5 and 24 h after intubation. Post-prandial plasma free lysine concentrations (PPlys, 5 h after intubation) of fish fed diets containing ${\geq}2.32%$ lysine were higher than those of fish fed diets containing ${\leq}1.92%$ lysine. Post-absorptive free lysine concentrations (PAlys, 24 h after intubation) of fish fed diets containing 2.32 and 3.52 % lysine were higher than those of fish fed diets containing ${\leq}1.52%$ lysine. The brokenline regression analysis on the basis of PPlys and PAlys indicated that the lysine requirement of rainbow trout could be 2.34 and 2.20 % in diet. Therefore, these results strongly suggested that the dietary lysine requirement based on the broken-line model analyses of PPlys and PAlys could be greater than 2.20 % but less than 2.34 % (corresponding to be $6.01%{\leq},but{\leq}6.39%$ in dietary protein basis, respectively) in rainbow trout. Also, these results shown that the quantitative estimation of lysine requirement by using PPlys and PAlys could be an acceptable method in fish.
Objectives: This study aimed to evaluate dietary protein intake and its adequacy among Korean adults during recent 10 years. Methods: Based on the 2010 ~ 2019 Korea National Health and Nutrition Examination Survey (KNHANES) data, a total of 51,296 adults aged 19 years old or more who participated in a one-day 24-hr dietary recall were included. Dietary protein intake was estimated as percentages of total energy (% of energy) and grams per body weight (g/kg/day) and compared with the 2020 Dietary Reference Intakes for Koreans to evaluate the adequacy of protein intake. In addition, proportions of people whose protein intakes were less than the estimated average requirement (EAR) and above the upper limit of the acceptable macronutrient distribution range (AMDR) (> 20% of energy) were calculated according to sociodemographic characteristics. Results: Protein intake was increased from 14.7% of energy in 2010 to 15.6% of energy in 2019 among Korean adults. However, there was no increase in protein intake relative to the recommended nutrient intake (% RNI) during the recent 10 years. Protein intake relative to the RNI was decreased from 130.2% in 2010 to 121.1% in 2019 (P for trend < 0.0001) among total participants, and a significant decreasing trend was observed in all age groups except for over 65 years old. However, protein intake relative to the RNI was lowest in the elderly (98.6%). Proportions of low protein intake (< EAR) and high protein intake (> AMDR) increased in the past 10 years (P for trend < 0.0001 for all), and these were associated with socioeconomic statuses, such as education and household income levels. Conclusions: These findings suggest that protein adequacy in Korean adults has not been improved over the past decade compared with recommended levels. Nutritional education and intervention programs should consider different intake levels according to sociodemographic characteristics.
Purpose: Recently, high-protein diets have become highly popular, and the market for protein products has steadily increased in Korea together with the development of various types of such products. However, there is limited information on the consumption of protein supplements (PS) or protein-fortified foods (PF). Thus, this study aimed to evaluate the use of PS/PF among young adults in Jeju. Methods: A total of 350 adults (140 men and 210 women) aged 19-39 years voluntarily participated in this study from June 2022 to May 2023. PS/PF use was measured using a questionnaire. Dietary intake was assessed using a 24-hour dietary recall. Results: Approximately 31.4% of the participants (n = 110) had consumed PS/PF for more than 2 weeks during the past year and 71.8% of them (n = 79) were still consuming these products (PS/PF consumers). The PS/PF consumers tended to be male and physically active (p < 0.05 for all). The most frequent reason for PS/PF use was muscle gain (59.5%), followed by protein supplementation (19.0%) and body fat loss (13.9%), and the most frequent type of PS/PF consumed was powders (70.6%), followed by drinks (17.7%) and bars (8.8%). The PS/PF consumers tended to consume a high-protein low-carbohydrate diet compared to the non-consumers. The prevalence of consuming dietary protein less than the estimated average requirement (EAR) was significantly lower in PS/PF consumers (13.9%) compared to non-consumers (25.4%; p = 0.0316). Conclusion: These findings indicate that the necessity of protein supplementation should be determined based on the current dietary protein intake and individual requirements. The study also provides the basic information for establishing guidelines for appropriate protein intake.
