적엽 및 제협처리가 콩의 동화물질 전류에 미치는 영향 (Effects of Leaf and Pod Removal on Assimilate Translocation in Soybean Plants)

  • 콩의 생식생장기인 착협시(R3)에 상부40%와 하부 60%로 구분하여 엽제거와 협제거처리에 의한 엽과 종실의 건물중, 가용성 당, 전분, 단백질 및 기름함량에 미치는 영향을 구명하기 위하여 1992년 고려대학교 자연자원대학 덕소농장에 황금콩을 공시하여 실시한 시험결과를 요약하면 다음과 같다. 1. 상엽하협제거처리의 하부 엽 건물중이 가장 높았고 상부 종실 건물중은 가장 낮았다. 2. 가용성 당합량은 처리간 엽과 종실에서 차이가 없었다. 3. 전분함량은 상엽하협제거의 하부 여에서 가장 높았다. 4. 단백질함량은 하부 엽이 상부 엽보다 높았는데 종실에서는 하엽상협제거의 하부에서 가장 낮았다. 5. 기름합량은 하엽상협제거의 상부 엽과 하부 종실에서 가장 높았다. 6. 콩 생식생장기의 장거리 전류에 있어서 동화물질은 상부에서 하부로 이루어지며, 단백질원은 하부에서 상부로 일어나지만 엽에서의 재이동은 약했다.

Removal of Total Suspended Solids by a Foam Fractionator in a Simulated Seawater Aquaculture System

  • In a simulated seawater aquaculture system, effects of different operating factors like the superficial air velocity (SAY), hydraulic residence time (HRT), protein concentration and foam overflow height on the removal of total suspended solids (TSS) by a foam fractionator, with 20 cm diameter and 120 cm height, were investigated. This experiment was performed on batch and consecutive modes for different combinations of the tested factors, using synthetic wastewater. In 5 consecutive trials, TSS concentration in culture tank water decreased faster, when the foam fractionator was operated at higher SAV and lower HRT. In batch trials, with increasing SAV, TSS removal rate increased, but decreased with increasing HRT. Higher protein concentration in the bulk solution resulted in higher TSS removal rate. TSS concentration in the collected foam condensates increased but the foam overflow rate decreased with increasing foam overflow height. Foam fractionation was effective for removing TSS in seawater aquaculture systems and its performance largely depended on the operating parameters, especially superficial air velocity.

향류 공기 구동식, 고속 폭기식 및 벤튜리식 포말분리기에 의한 양어장수의 단백질 및 부유 고형물의 제거 특성 (Characteristics of Proteins and Total Suspended Solids Removal by Counter Current Air Driven Type, High Speed Aeration Type and Venturi Type Foam Separator in Aquacultural Water)

  • 공기 구동식, 고속 폭기식 및 벤튜리식 포말분리 장치를 동일한 용량으로 제작하여 양어장 순환수 중의 단백질,총 부유 고형물 및 탁도를 제거하는데 있어 각 포말 분리장치의 운전 특성을 알아보고자 하였다. 공기구동식 포말분리기의 유략감소가 거의 없었으나 벤튜리식 포말분리기는 $66.1{\%}$정도의 유량감소가 발생하였으며 고속 폭기식 포말분리기는 $77.2{\%}$의 유량감소가 발생하였다. 공기 구동식, 고속 폭기식 및 벤튜리식 포말분리 장치의 단백질 제거속도는 수력학적 체류시간이 증가함에 따라 지수적으로 감소 하였다. 0.32 분 이상의 수력학적 체류시간에서는 공기 구동식 포말분리장치의 제거속도와 제거율이 가장 높게 나타났으며 그 이하의 수력학적 체류시간에 대해서는 고속 폭기식 포말분리 장치의 제거속도와 제거율이 가장 높은 것으로 나타났다. 수력학적 체류시간의 변화에 따른 총 부유 고형물과 탁도의 제거속도와 제거율의 변화는 단백질 제거속도 및 제거율의 변화와 거의 비슷하였으며 벤튜리식 포말분리기는 짧은 수력학적 체류시간에서는 총 부유고형물의 제거 효율이 다른 두 포말분리기에 비해 떨어지는 것으로 나타났다. 탁도의 제거는 공기 구동식 포말 분리기가 가장 우수한 것으로 나타났다.

오존산화에 의한 정수장의 Microcystin제거 특성에 관한 연구 (A Study on the Removal Characteristics of Microcystin in the Water Treatement Plant by Ozonation)

  • Microcystin, stable compounds with circular heptapeptides, is presented inside cyanobacterial cell. So far, over 30 types have been known to exist and microcystin-LR, RR among them are the most potent toxin compound. By this reason, a strong oxidant, ozone was used in this study to remove the microcystins produced by cyanobacteria. Removal efficiency of microcystin at M water treatment plant was also evaluated. Microcystin concentration was determined by protein phosphatase inhibition assay. The results showed that dissolved microcystin in raw water detected in the range of 0.011-0.028 ㎍ Microcystin-RR equivalent/l. Above 98% of microcystin was removed through overall treatment system. Therefore, the water treatability of M treatment plant seemed to be excellent. Removal efficiency of microcystin according to unit process varied as characteristics of raw water such as DOC, UV/sub 254/ and turbidity. Removal efficiency of microcystin by ozonation was investigated in laboratory according to contact time and ozone dose. Dissolved microcystin was increased by twice fold according to ozone contact time, but increased by fifth fold according to ozone dose. So, changing of ozone dose more affected microcystin release than changing of ozone contact time. Behavior of microcystin by ozonation was similar to that of DOC, and residual ozone concentration gave influence to removal ratio of microcystin. In conclusion, single ozone treatment wasn't effective on microcystin removal in case of water containing a lot of cells. Therefore, it's more effective to use ozonation process after the removal of cyanobacterial cells in advance.

활어수조에서 넙치 사육시 포말분리장치를 이용한 오염물 제거 (Removal of Waste Generated by Flounder (Paralichthys olivaceus) in Aquarium using a Foam Separator)

  • Removal of waste generated by Paralichthys olivaceus in the seawater aquarium using a foam separator was investigated. Protein concentration without a foam separator continuously increased until 3 days after stocking and reached at 25 mg/L after 5 days stocking, but protein concentration became lower than the initial protein concentration (2.5 mg/L) with a foam separator. The trends of other fish wastes such as ammonia, total suspended solids (TSS) and chemical oxygen demand (COD) were similar to protein. Dissolved oxygen (DO) in the aquarium decreased below 6.0 mg/L without a foam separator, but with a foam separator the average DO in the aquarium was 7.3 mg/L. Foam separation with the increase of superficial air velocity (SAV) was more effective than that with the fixed SAV. This study showed that wastewater. treatment of seawater aquarium using a foam separator is effective method for a fish waste removal and oxygen supply.

미나리 ( Oenanthe stolonifera ) 의 Cd, Zn 제거능과 내성에 관한 연구 (Studies on Cd and Removal Ability and Detoxification of Oenanthe stolonifera)

  • To examine the possibility of biomonitoring of heavy metal removal ability and soil, a study was performed to investigate the heavy metal removal ability and metal-binding protein (MBP) as detoxification process using Oenanthe stolonifera. After O. stolonifera was exposed to individuals (cadmium, zinc) and mixture (cadmium+zinc)for 4 days, removal rate of heavy metal and pH in the treatment medium was measured. MBP was assayed by means of ion exchange column chromatography. The exposure to mixture (Cd:76.8%, Zn:75%) rather than individuals (Cd:82.9%, Zn:90.4%) showed a synergism raising the toxic effect. Initial removal rate was different for each heavy metal : in case of exposure to cadmium it was over 60% on day 1, while for zinc it was 75~90% on day 4. Throughout the experimental period, pH value of treatment medium continuously decreased, since cortex in the roots may secret organic acid to adjust and prevent toxicity of metals. The existence or MBP in the 70~80 fraction and the presence of Zn-enzyme pool was ascertained with the column chromatography. This study demonstrated a possibility that heavy utilized as a biomarker of heavy metal pollution.

Leclercia Adecarboxylata를 이용한 합성폐수의 암모니아성질소 제거특성 및 질소거동 (Removal Characteristic of Ammonia Nitrogen and Behavior of Nitrogen in Synthetic Wastewater Using Leclercia Adecarboxylata)

  • 본 연구에서는 고농도 암모니아성질소로 오염된 고체배지로부터 분리해 낸 Leclercia adecarboxylata를 이용하여 암모니아성질소의 제거특성 및 기작을 파악하여 폐수처리의 적용가능성에 대해 살펴보았다. 질소제거에 있어서 가장 널리 알려진 생물학적 질산화와 후탈질에 의한 질소의 대기로의 방출이 아닌 질소합성균주를 이용한 질소의 체내합성을 이용한 영양물질의 제거 가능성에 대해 접근해 보았다. L. adecarboxylata는 무염분조건에서 암모니아성질소의 제거와 균체중식이 가장 왕성했으나, 염분이 4%를 넘어서게 되면 그 효율은 급격히 저하되었다. 약 80 mg/L의 암모니아성질소는 20시간 이내에 거의 대부분 제거되었으나, 500 mg/L인 시료는 탄소원의 부족으로 인해 50시간 이상 처리후에도 50%의 제거율에도 미치지 못해 탄소원이 많을수록 질소제거율은 높음을 알 수 있었다. 탄소원이 모두 소모되고 난 이후에는 더 이상 질소제거는 이루어지지 않았으나, 탄소원을 추가로 공급했을 때 제거효율은 다시 증가했다. 시료의 pH는 미생물의 증식에 의해 8에서 6.36까지 감소했다. 암모니아성질소가 제거되는 동안 아질산성질소와 질산성질소의 축적은 일어나지 않았고, TKN값은 큰 차이를 보이지 않은 것으로 미루어 볼 때 유기질소로의 합성을 추측할 수 있었다. 유기질소 중 단백질의 농도를 측정해 본 결과 초기시료에서는 불검출 되었으나, 48시간 경과후의 시료에는 193.1 mg/L의 단백질이 검출 되었다. 따라서 L. adecarboxylata는 암모니아성질소를 유기질소로 합성하는 능력이 탁월하여 폐수중의 암모니아성질소의 제거에 이용가치가 클 것으로 판단된다.

  • 휘발성 황화합물 중 생물학적으로 가장 난분해성으로 알려 져 있는 DMS에 대한 분해능이 우수한 신균주를 분리. 동정 하여 Gordonia sihwaniensis PKL-1로 명명하였다. 이 균주는 D DMS 초기농도가 35 mg/L 이상일 때에도 우수한 분해능을 보였고, 회분식 배양에서 최대 비분해속도 $\upsilon_{max}$ 없는 0.0016 mg/(mg-protein).hr이 었고, 최적 의 온도와 pH는 각각 $25^{\circ}C$ 와 pH는 7이였다.

Spirulina platensis를 이용한 축산 폐수처리 및 고단백 사료원의 생산 (The Treatment of Swine Wastes and the Production of High Protein Feedstocks from Photoheterotrophic Growth of Spirulina platensis)

  • Microalga, Spirulina platensis has been cultivated in a pilot scale photo-bioreactor to treat wastewater and to produce high protein feedstocks from swine waste containing medium. 0.31(1/day) of specific growth rate and 0.170 of bioenergeric yield were obtanined from batch cultivation in 30% waster containing medium, compared to 0.71(1/day) and 0.545 from clean culture. An optimal dilution concentration was decided as 20% of working volume, based upon the cell growth and biomass productivity. The removal rate of nitrates in the wastewater was decreased as the adding concentration of wastewater was increased while the decrease of total phosphates was reversed, showing 0.33(1/day) and 0.30(1/day) of rate constants for nitrate removal in 10% addition and for phosphate removal in 30% addition, respectively. The chemical composition and amino acid profile of the biomass were superior to those of commerically available health food product, Spirulina sp.

Developing a Virus-Binding Bacterium Expressing Mx Protein on the Bacterial Surface to Prevent Grouper Nervous Necrosis Virus Infection

  • Grouper nervous necrosis virus (GNNV) infection causes mass grouper mortality, leading to substantial economic loss in Taiwan. Traditional methods of controlling GNNV infections involve the challenge of controlling disinfectant doses; low doses are ineffective, whereas high doses may cause environmental damage. Identifying potential methods to safely control GNNV infection to prevent viral outbreaks is essential. We engineered a virus-binding bacterium expressing a myxovirus resistance (Mx) protein on its surface for GNNV removal from phosphate-buffered saline (PBS), thus increasing the survival of grouper fin (GF-1) cells. We fused the grouper Mx protein (which recognizes and binds to the coat protein of GNNV) to the C-terminus of outer membrane lipoprotein A (lpp-Mx) and to the N-terminus of a bacterial autotransporter adhesin (Mx-AIDA); these constructs were expressed on the surfaces of Escherichia coli BL21 (BL21/lpp-Mx and BL21/Mx-AIDA). We examined bacterial surface expression capacity and GNNV binding activity through enzyme-linked immunosorbent assay; we also evaluated the GNNV removal efficacy of the bacteria and viral cytotoxicity after bacterial adsorption treatment. Although both constructs were successfully expressed, only BL21/lpp-Mx exhibited GNNV binding activity; BL21/lpp-Mx cells removed GNNV and protected GF-1 cells from GNNV infection more efficiently. Moreover, salinity affected the GNNV removal efficacy of BL21/lpp-Mx. Thus, our GNNV-binding bacterium is an efficient microparticle for removing GNNV from 10‰ brackish water and for preventing GNNV infection in groupers.
