Ruminal and Intestinal Digestibility of Some Tropical Legume Forages

  • Two experiments were conducted to examine the degradation rates of 4 tropical legume forages in rumen and intestine of Kedah-Kelantan (KK) cattle. Three KK cattle, averaging $173{\pm}17.15kg$ each fitted with a permanent ruminal and a T-shaped duodenal cannulae were used. The cattle were fed a maintenance diet (1% DM of their body weight) composing of 60% oil palm frond (OPF) pellet and 40% of a legume mixture of Arachis pintoi (AP) and Leucaena throughout the study. The overall DM and CP degradabilities in the rumen for Gliricidia sepium (GS) and AP were significantly higher than those for Leucaena leucocephala-Bahru (LB) and Leucaena leucocephala-Rendang (LR). This implies that LB and LR would have higher dietary protein flows into the intestine for the more efficient enzymatic digestion. However, the results of the present study suggested only limited proportions of the ruminal undegraded protein in the Leucaenas were digested in the intestine.

  • An ascorbic acid phosphorylating enzyme, which catalyzes the formation of ascorbic acid-2-phosphate from ascorbic acid and pyrophosphate, was purified 32.7-folds to homogeneity from a cell-free extract of Cellulomonas sp. AP-7. The combination of DEAE- Sephacel ion exchange chromatography and Sephacryl S-200 get filtration was used for their purification. The molecular weight of the native protein was estimated to be 96.lkDa on high performance gel filtration chromatography. The SDS-PAGE analysis indicated that the protein consisted of four identical subunits of 24.6 kDa. The purified enzyme showed the optimal tempeature of 40$\circ$C and optimal pH of 4.5. The Km for ascorbic acid and pyrophosphate were 119 mM and 11.9 mM, respectively. The addition of 5,5'-dithiobis-(2-nitrobenzoic acid) into the reaction mixture resulted in the reduction of the enzyme activity at 51%. The enzyme also had a phosphatase activity at weakly acidic pH and the Km for ascorbic acid-2-phosphate in phosphatase activity was 7.9 mM.

Effect of Glucosinolates of Taramira (Eruca Sativa) Oilcake on Nutrient Utilization and Growth of Crossbred Calves

  • Taramira (Eruca sativa) cake, an unconventional oil cake, replaced 25 and 50 per cent crude protein of mustard cake in the ration of crossbred calves in an experiment of 90 days duration. Total glucosinolate content of the three concentrate mixture was almost similar (18.19, 17.95 and $17.95{\mu}mol/g$ dry matter), however, glucouracin was the major glucosinolate of experimental diets. Similar dry matter Intake, nutrient digestibility (except those of fibre fractions) and nitrogen balances as well as similar serum $T_3$ and $T_4$ levels and growth rate in all the groups indicated that taramira cake can replace 50 per cent crude protein of mustard cake in the diet of crossbred calves.

Zinc Bioavailability in a Semolina/Soy Protein Mixture Was Not Affected by Extrusion Processing

  • Effects of extrusion processing on zinc bioavailability in a product made from 85% semolina and 15% soy protein concentrate were determined using in vitro and in vivo techniques. Soluble and dialyzable zinc contents of the extruded product were 15.3 and 13.0 $\mu$g/g, respectively, compared to 15.6 to 15.0$\mu$g/g in the raw (unprocessed) ingredients. Zinc bioavailability in diets in which all of the zinc (14ppm) was provided either by the extruded product or by its raw ingredients was determined in two groups of male Sprague Dawley rats. No differences were found in concentrations of zinc in plasma, liver and femur between rats fed the two experimental diets. Apparent zinc digestibility was similar for both diets. These results are consistent with the in vitro results showing no effect of extrusion processing on zinc bioavailability.

To develop the classification method of Agricultural by-productions for biogas production

  • The objective of this study was to develop the classification method of various organic wastes. Specifically, the effects of proximate composition on the biogas production and degradation rates of agricultural by-production was investigated and a new standards for mixture of various organic wastes based on proximate composition combination was developed. Agricultural by-products (ABPs) with medium total carbohydrate, medium crude protein and low fat contents demonstrated the single step digestion process. ABPs with low total carbohydrate, high crude protein and high fat contents demonstrated the two step digestion process of Diauxic growth. The single ABP (Class No. 15) and the mixed ABPs (Class No. 12+18, 6+12+22, 9+12+18) after 10days showed the similar biogas yield pattern. We can use the classification method for the more ABPs and organic wastes from factory and municipal waste treatment plant for the high efficient biogas production.

  • Objectives : This research was investigated the effect of the Ginseng Radix plus Chaenomelis Fructus on Alzheimer's disease. Methods : Specifically, the effects of the Ginseng Radix plus Chaenomelis Fructus extract on $IL-1{\beta}$, $TNF-{\alpha}$ of BV2 microglia cell line treated with lipopolysacchride. Results : The Ginseng Radix plus Chaenomclis Fructus extract suppressed the over-expression of $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, and CD68/CD11b, in the mice with Alzheimer's disease induced by ${\beta}$ amyloid peptide. Conclusion: These results suggest that the Ginseng Radix plus Chaenomelis Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Ginseng Radix plus Chaenomelis Fructus extract for Alzheimer's disease is suggested for future research.

Efficient Generation of Human IgG1 Light Kappa Constant Region Knock-in Mouse by CRISPR/Cas9 System

  • Mice with specific modified genes are useful means of studying development and disease. The CRISPR/Cas9 system is a very powerful and effective tool for generating genetically modified mice in a simple and fast manner. To generate human IgG light kappa constant knock-in mice, we tested by microinjection of a mixture of Cas9 protein, single-guide RNA and target homologous recombinant donor DNA into zygotes. We found that the injection of 10 ng/μL of Cas9 protein and crRNA/tracrRNA, rather than single guide RNA, induced the production of knock-in mice more effectively. Thus, our study provides valuable information that will help to improve the production of knock-in mice and contribute the successful generation of humanized Ab-producing mice in Korea.

Improved Phosphotyrosine Analysis by TLC and HPLC

  • We describe here the conditions of thin layer chromatography (TLC) and high pressures liquid chromatography (HPLC) to improve the analytical method of phosphotyrosine (p-Tyr) in biological sample. TLC was performed on silica plate with the mixture of propanol and water (2.1 : 1 v/v) as a mobile phase and $R_1$ values were 0.42, 0.39 and 0.33 for phosphotyrosine, phosphothreonine and phosphoserine, respectively. HPLC was performed on $NH_2$ column with a mobile phase of potassium biphosphate solution by UV deterction at 192 nm. The optimum condition of HPLC was obtained at 0.01 M, pH 4.5 with a clear separation within 12 min. These procedures have been applied to the analysis of phosphotyrosine obtained from tyrosine-phosphorylated enolase. Both TLC and HPLC methods were suitable to analyze tyrosine-phosphorylated protein without being affected by contaminants from hydrolysates.

Potentiometric Homogeneous Enzyme-Linked Binding Assays for Riboflavin and Riboflavin Binding Protein

  • Adenosine deaminase (ADA) has been utilized as the label in devising a potentiometric homogeneous assay for riboflavin and riboflavin binding protein (RBP). The proposed homogeneous assay method employs an ADA-biotin conjugate as the signal generator and an avidin-riboflavin conjugate as the signal modulator in the solution phase. The catalytic activity of the ADA-biotin conjugate is inhibited in the presence of an excess amount of the avidin-riboflavin conjugate, and the observed inhibition is reversed in an amount proportional to the concentration of RBP added. When the analyte riboflavin is added to this mixture of ADA-biotin, avidin-riboflavin and RBP, the activity of the enzyme conjugate is re-inhibited in an amount proportional to the concentration of riboflavin. Since the enzyme label used in this system is ADA, an ammonia-producing enzyme, a potentiometric rather than photometric detection scheme is used to monitor the enzymatic activity in the assay.

  • The 3rd order nonlinear optical susceptibility was measured through the self-induced ellipse rotation. The phase separation temperature increases with the increase of salt and it decreases with the increase of glycerol.
