Effects of The Soy Protein Level on Plasma Glucose, Lipids, and Hormones in Streptozotocin-Diabetic Rats

  • The number of diabetics in Korea is about 3 to 5 percent of the population, and the incidence is increasing yearly due to changes of life style and food intake. Diet is a key element in the management of diabetes, yet the appropriate diet for diabetes remains controversial. We have recently shown that a diet rich in protein of animal origin(casein) seems beneficial to controling plasma glucose and lipids in streptozotocin-induced diabetic rats. It therefore seemed desirable to find out whether the beneficial effect of high casein diet in experimental diabetes could also be reproduced with a vegetable source of protein(soy). The purpose of this study is to compare these results with the results of our previous study. In the present study, non-diabetic and streptozotocin-induced diabetic rats were studied in order to examine the effects of altering the level(20% vs 60%) of dietary soy protein on blood glucose, lipids, and hormones. Results of the present study showed that a high soy protein diet decreased triglyceride concentration in diabetic rats. However, diabetic rats fed a high soy protein diet were not hypocholesterolemic compared to rats fed a control diet. Moreover, diabetic rats fed a high soy protein diet had significantly increased plasma glucose concentration compared to rats fed a control diet. This study was not able to discern a specific effect of dietary protein level on insulin, glucagon, or insulin/glucagon ratio. Except for the hypotriglyceridemic effect, the results were not similar to the findings of our previous study which showed a beneficial effect on streptozotocin-induced diabetic rats fed a high casein diet.

Identification and Characterization of a RecA-like Protein Induced by DNA Damaging Agents in Fluorescent Pseudomonas sp.

  • A RecA-like protein (RecAps) was identified from fluorescent Pseudomonas sp. and the inducible nature of the protein was characterized in detail. It was shown by dose-response and time-course experiments using two DNA damaging agents, nalidixic acid and mitomycin-C, that the cellular level of RecAps protein was increased 3-8 fold compared to that of the control. The most effective doses of nalidixic acid and mitomycin-C for the protein induction were $30{\mu}g/ml$ and $0.3{\mu}g/ml$ at the treatment time point of 150 min, respectively. The enhanced level of RecAps protein was gradually decreased to the control level after 10 hr in normal medium. Interestingly, the cellular level of RecAps protein was increased by the same DNA damaging agents even when cell growth was completely inhibited by treatment with $170{\mu}g/ml$ of chloramphenicol, an inhibitor of protein synthesis, suggesting that new protein synthesis is not required for the induction of RecAps. All these results suggest that a typical S0S repair function driven by RecA-like protein is conserved in Pseudomonas sp. cells as in E, coli.

Statistical Treatment on Amylose and Protein Contents in Rice Variety Germplasm Based on the Data Obtained from Analysis of Near-Infrared Reflectance Spectroscopy (NIRS)

  • The purpose of this study was to statistically analyze amylose and protein content of rice variety resources collected from China (1,542), Japan (1,409), Korea (413), and India (287). The statistical analysis was conducted using ANOVA and DMRT based on the data obtained from NIRS analysis. The average amylose contents were 18.85% in Japanese, 19.99% in Korean, 20.27% in Chinese, and 25.46% in Indian resources. The average protein contents were 7.23% in Korean, 7.73% in Japanese, 8.01% in Chinese, and 8.17% in Indian resources. The amylose and protein content using ANOVA showed significant differences at the level of 0.01. The F-test for amylose content was 158.34, and for protein content was 53.95 compared to critical value 3.78. The amylose and protein content using DMRT (p<0.01) showed significant difference between countries. The value of statistical treatment was divided into three groups such as $China^a$, $Korea^a$, $Japan^b$, $India^c$ in amylose and $China^a$, $India^a$, $Japan^b$, $Korea^c$ in protein. Japanese resources had the lowest level of amylose contents, whereas, the lowest level of protein content was found in Korean resources compared to other origins. Indian resources showed the highest level of amylose and protein contents. It is recommended that these results could be helpful to future breeding experiments.

High-Level Production of Spider Silk Protein by Fed-Batch Cultivation of Recombinant Escherichia coli and Its Purification

  • Silk proteins from Nephila clavipes are fibrous proteins containing repetitive sequences with both crystalline and amorphous domains. In order to obtain high-level production of silk protein, the synthetic genes had 16 contiguous units of the consensus repeat sequence of the silk protein were expressed in Escherichia coli BL21(DE3) under the strong inducible T7 promoter. For production of recombinant silk protein in large amounts, pH-stat fed-batch cultures were carried out. The recombinant silk protein was produced as soluble forms in E. coli, and the recombinant silk protein content was as high as 11% of the total protein. When cells were induced at $OD_{600}$ of 60, the amount of silk protein produced was 6.49 g/L. After simple purification steps, 9.2 mg of silk protein that was more than 80% pure was obtained from a 50 mL culture, and the recovery yield was 26.3%.

Changes of the Level of G Protein ${\alpha}-subunit$ mRNA by Withdrawal from Morphine and Butorphanol

  • Morphine or butorphanol was continuously infused into cerebroventricle (i.c.v.) with the rate of $26\;nmol/{\mu}l/h$ for 3 days, and the withdrawal from opioid was rendered 7 hrs after the stopping of infusion. The expression of physical dependence produced by these opioids was evaluated by measuring the naloxone-precipitated withdrawal signs. The withdrawal signs produced in animals dependent on butorphanol (kappa opioid receptor agonist) were similar to those of morphine (mu opioid receptor agonist). Besides the behavioral modifications, opioid withdrawal affected G protein expression in the central nervous system. The G-protein ${\alpha}-subunit$ has been implicated in opioid tolerance and withdrawal. The effects of continuous infusion of morphine or butorphanol on the modulation of G protein ${\alpha}-subunit$ mRNA were investigated by using in situ hybridization study. In situ hybridization showed that the levels of $G\;{\alpha}s$ and $G\;{\alpha}i$ were changed during opioid withdrawal. Specifically, the level of $G\;{\alpha}s$ mRNA was decreased in the cortex and cerebellar granule layer during the morphine and butorphanol withdrawal. The level of $G\;{\alpha}i$ mRNA was decreased in the dentate gyrus and cerebellar granule layer during the morphine withdrawal. However, the level of $G\;{\alpha}i$ mRNA was significantly elevated during the butorphanol withdrawal. These results suggest that region-specific changes of G protein ${\alpha}-subunit$ mRNA were involved in the withdrawal from morphine and butorphanol.

남해도 지역이 임신후반기 임부의 영양실태 조사 (A Nurition Survey of the Latter Half of Pregnancy in Nam-Hae Do)

  • The purpose of this survey was to investigate the nutrient and food intake and haematology of the latter half of pregnant women in Nam Hae Do. The nutrient intake study was performed by Twenty-four hour dietary recall method. As Haematology, RBC, Hb. and Hct were measured. 1. The results of nutritional survey were, 1) The mean nutrient intakes that were below the RDA were Protein, Calorie, Calcium and Iron. 2) The mean nutrient intakes that were above the RDA were Vitamin A, Thiamin, Riboflavin, Ascorbic acid. 3) Most of calorie and other nutrients were obtained from vegetable food sources. 4) Animal protein intake was 33% of total protein intake and most of this value was obtained from fish and shell fishes. 2. The extent of malnutrition was explained in terms of the amount of calorie, protein, calcium and iron. The results were, The predicted percentage of deficiency, in case of Calorie, 53.3% of total subjects. in case of Protein, 52% of total subjects. in case of Calcium, 78.7% of total subjects. in case of Iron, 54.7% of total subjects. 3. The results of Haematology were, 1) The mean level of RBC, Hb and Hct were 3.76$\times$106cm/㎣, 10.47gm%, and 32.56% 2) There were significant correlation between calorie intake and Hct level, protein intake and RBC level, Iron intake and MCHC level.

Influence of Protein and Energy Level in Finishing Diets for Feedlot Hair Lambs: Growth Performance, Dietary Energetics and Carcass Characteristics

  • Forty-eight Pelibuey${\times}$Katahdin male intact lambs ($23.87{\pm}2.84$ kg) were used in an 84-d feeding trial, with six pens per treatment in a $2{\times}2$ factorial design arrangement. The aim of the study was to evaluate the interaction of two dietary energy levels (3.05 and 2.83 Mcal/kg ME) and two dietary protein levels (17.5% and 14.5%) on growth performance, dietary energetics and carcass traits. The dietary treatments used were: i) High protein-high energy (HP-HE); ii) High protein-low energy (HP-LE); iii) Low protein-high energy (LP-HE), and iv) Low protein-low energy (LP-LE). With a high-energy level, dry matter intake (DMI) values were 6.1% lower in the low-protein diets, while with low-energy, the DMI values did not differ between the dietary protein levels. Energy levels did not influence the final weight and average daily gain (ADG), but resulted in lower DMI values and higher gain efficiencies. No effects of protein level were detected on growth performance. The observed dietary net energy (NE) ratio and observed DMI were closer than expected in all treatments and were not affected by the different treatments. There was an interaction (p<0.03) between energy and protein level for kidney-pelvic and heart fat (KPH), KPH was higher in lambs fed high energy and high protein diet but not in high energy and low protein diet. The KPH was increased (20.2%, p = 0.01) in high-energy diets, while fat thickness was increased (21.7%, p = 0.02) in high-protein diets. Therefore, it is concluded that dietary energy levels play a more important role in feed efficiency than protein levels in finishing lambs with a high-energy diet (>2.80 Mcal/kg ME). Providing a level of protein above 14.5% does not improves growth-performance, dietary energetics or carcass dressing percentage.

  • When at liver homogenates were incubated with 1mM $CCl_4$ for five min, glycogen level was decreased, while treatment with protein fraction $G_4$ increased the glycogen level. In addition $G_4$ inhibited the phosphorylation of 34 KD and 118 KD polypeptides induced by $CCl_4$. These protein were more strongly phosphorylated by $Ca^{2+}$/calmodulin-dependent kinase than by C-kinase. Since 34 KD polypeptide was solely phosphorylated by NaF (50mM), an inhibitor of both glycogen syntheses and phosphoprotein phosphates, it is inferred that 3 KD polypeptide is glycogen synthase-likd protein. Because glycogen synthesis is inhibited by phosphorylation of $Ca^{2+}$-dependent glycogen syntheses, it is suggested that $G_4$ increased liver glycogen level by inhibiting phosphorylation of 34 KD polypeptide which is thought to glycogen syntheses-like protein.

상백피 첨가에 따른 숙육의 연화와 관능적 특성 (Sensory Characteristics and Tenderness of Boiled Beef by Addition of the Barks(Morus alba Linne))

  • Tenderization of beef by the addition of barks(Morus alba Linne) and its sensory properties were observed by shearing test, cooking loss, pH, protein content and sensory evaluation. Shear force was decreased as the addition level of barks (Morus alba Linne) increased. Tenderization effect of beef was increased 8,8% at the addition level of 2.5%, 25% at 5.0%, 4.7% at 7.5%, 58% at 10% barks(Morus alba Linne). Cooking loss was observed 44.50% at the no addition, 45.3% at 2.5%, 45.8% at 5.0%, 47.5% at 7.5%, 50.0% at 10% addition level of barks (Morus alba Linne). As the addition level of barks(Morus alba Linne) increased pH of cooked beef decreased to the range of 5.6 to 5.46. As the addition level of barks (Morus alba Linne) increaed protein content of beef decreased whereas protein content of cooled liquor increased. The addition of barks(Morus alba Linne) improved the sensory quality of cooked beef. Especially, tenderness scored the highest value in 10% added beef and other quality factors color, flavor, juiciness and overall quality were evaluated significantly high in 5% added beef. Therefore 5% addition level of barks(Morus alba Linne) for cooking beef was suggested as the desirable level of addition.

돌둠사료의 적정 단백질 및 지질 함량 (Optimum Dietary Protein and Lipid Levels on Growth in Parrot Fish (Oplegnathus fasciatus))

  • 돌돔 사료의 적정 단백질 및 지질 함량을 구하기 위해 북양어분과 카제인을 단백원으로 하여 사료의 단백질 함량이 30, 40, 50 및 60%가 되도록 하고, 각 단백질 함량에 지질 함량이 8 및 16%가 되는 8종의 실험사료로 평균체중 7 g의 돌돔을 대상으로 8주간 사육 실험하였다. 사료의 단백질 함량에 따른 증체율 및 사료효율은 사료지질 8% 수준에서 사료단백질이 40% 이상인 사료구에서는 사료단백질 함량이 증가함에 따라 유의차 있게 증가하였으며(P<0.05), 사료지질 16% 수준에서는 사료단백질이 증가함에 따라 증가하다가, 사료단백질 50% 이상에서는 유의적인 차이가 없었다(P>0.05). 사료지질 16% 수준에서 broken line model을 이용하여 증체율을 지표로 하여 돌돔의 적정 단백질 요구량을 구한 결과 46%로 추정되었다. 사료의 지질 함량에 따른 증체율은 사료단백질 40 및 50% 수준에서 고지질 사료구(16%)가 유의하게 높았으며, 사료효율은 모든 사료단백질 수준에서 고지질 사료구(16%)가 유의하게 높았다(P<0.05). 단백질 효율 및 단백질 축적율은 모든 사료단백질 수준에서 고지질 사료구가 높았다. 따라서 성장과 사료효율을 기준으로 볼 때 돌돔 사료의 적정 지질 함량은 16% 전후로 추정되었다. 전어체 및 등근육의 성분에서 단백질은 사료의 단백질 함량이 증가함에 따라 증가하다가 사료 단백질 40% 이상에서는 유의차가 없었으며, 지질은 모든 사료단백질 수준에서 고지질 사료구가 높았다. 간 및 내장의 성분에서 단백질은 사료의 단백질 함량에 따라 뚜렷한 경향이 없었지만, 지질은 모든 사료단백질 수준에서 고지질 사료구(16%)가 높았다.
