Effects of Antioxidants on Growth and Betalain Production in Hairy Root Cultures of Phytolacca esculenta van Houtte (자리공(Phytolacca esculenta van Houtte) 모상근의 생장과 Betalain 생산에 미치는 항산화제의 효과)

  • The synthesis of betalain in hairy root cultures of Phytolacca esculenta van Houtte required the light, but the growth of hairy roots was inhibited 5 times under light condition(1500 1X) compared to dark condition. To investigate the growth inhibition of hairy roots under light condition, we surveyed the effects of several antioxidants on growth and betalain production if suspension cultures of hairy roots was increased 1.2-1.4 times under dark condition and 1.3-1.9 times under light condition by the treatment of ascorbic acid, glutathione, $\alpha$-tocopherol, sodium pyrosulfate and propylgallic acid. The betalain production was increased 1.2-2.1 times by antioxidants under light condition. The combination treatment of antioxidants didn't have any significant effect on the growth of hairy roots and betalain synthesis. The antioxidants of the betalain production were more effect under blue light than others. It was discussed that the endogenous oxidants may be produced under blue light.

Effect of Electron Acceptor, Electron Transport Inhibitors and Antioxidants on Growth and Ginsenosides Production in Hairy Root Cultures of Panax ginseng C.A. Meyer (인삼모상근의 생장과 Ginsenosides 생성에 미치는 Electron Acceptor, Electron Transport Inhibitors 및 Antioxidants의 효과)

  • The growth of hairy roots were increased $69\%$ by 0.1 mM DCPIP under light conditions. In these conditions, the contents of seven ginsenosides were none significant variation. The influence of electron inhibitors on growth and ginsenoside contents in ginseng hairy roots was tested. The growth was inhibited $71\%\;and\;22\%$ respectively by CCCP and methylamine. The ginsenoside contents were as decreased above $45\%$ in all treatment tested except triazine treatment. In antioxidants treatment, the growth of hairy roots was increased about $68\%$ by propylgallic acid, about $23\~25\%$ by ascorbic acid or 2,5-dimethylfuran, while the contents of seven ginsenosides were none significant variation. The ginsenoside productivity was high when hairy roots were cultured in $\circledR^{\wedge}MS$ medium for 4 weeks and then transferred to 1/2MS medium with ascorbic acid or 2,5-dimethylfuran for 1 weeks in light conditions. It is suggested that ginsenoside productivity could be accelerated by some antioxidants in hairy root cultures of ginseng.

  • In order to determine the specific active oxygen species directly related to chlorophyll bleaching in the leaf-burning disease, we investigated the effects of singlet oxygen (1O2), superoxide radical (O2-), and hydrogen Peroxide (H2O2) on isolated chloroplast suspension and leaf discs from Panax ginseng C.A. Meyer. When the singlet oxygen was added to the chloroplast suspension, the chlorophyll and carotenoid contents were decreased by more than 809), similar to treatment with high light intensity (100 KLux). We assumed that the conversion of dioxygen (O2) produced either in photolysis or in breakdown of hydrogen peroxide to singlet oxygen resulted from photorespiration. On the basis of these experiments , :he inhibitory effects of active oxygen scavengers propylgallic acid (PGA), 2,5-ditetrabutyl hydroquinon (DBH), sodium pyrosulfate (SPS), and ascorbic acid (ABS) were examined. In chloroplast suspension all four scavengers inhibited chlorophyll bleaching by more than 75fl , and in the leaf discs the inhibition rates of SPS, PGA and ABS were 46%, 51%, and 96% respectively.
