Changes of plasma progesterone concentrations after induction of estrus in the bitch (발정유기(發情誘起)된 암캐의 혈중(血中) progesterone 농도변화(濃度變化))

  • This study was performed to investigate the patterns of progesterone secretion after induction of estrus in premature, metestrous and anestrous bitches. A total of 22 bitches were used. Of them 18 bitches were treated with hormone to induced estrus and 4 bitches were untreated and served as controls. Estrus was induced with $PGF_{2{\alpha}}$, estrone, estradiol-$17{\beta}$, PMSG and HCG(Treatment A), and with PMSG and HCG(Treatment B). Blood samples were collected via the cephalic vein at 2 to 5 days interval. Blood samples were centrifuged (1,200g, 10min.) within 30 minutes after collection and plasma was stored at $-20^{\circ}C$ until analyzed for the progesterone concentrations. Plamsa progesterone concentrations were measured by radioimmunoassay. The results of estrous induction were determined by estrous signs, ovarian response, egg recovery and progesterone patterns. The results obtained were as follows; 1. All bitches in treatment A showed estrous signs, however the ovarian response and egg recovery were not detectable and the levels of progesterone were nearly same as before. 2. In the treatment B, premature and metestrous bitches showed only estrous signs, however 5 of 7 anestrous bitches (71.4%) showed estrous signs, ovarian response and changes of progesterone levels. In conclusion, clinical estrous behavior can be induced during any phase of the estrous cycle, but ovulation should be induced only if induction occur approximately 4 months or more after the previous estrus.

Patterns of Pulsatile and Surge Modes of Follicle-Stimulating Hormone Treated with Different Progesterone Levels in Ovariectomized Goats (난소제거된 Shiba 염소에서 다른 농도의 Progesterone 처치에 의한 FSH Surge 및 Pulse 분비에 미치는 영향)

  • The objective of the present study was to determine the progesterone levels that effects on the pulsatile and surge modes of FSH secretion. In previous studies we have shown that LH surge occurred in the follicular levels of progesterone, whereas there was no surge mode secretion of LH in either the sub luteal or luteal levels of progesterone. LH pulsatile frequencies were high in two groups such as follicular level and sub luteal level. But in the luteal level of progesterone the pulsatile pattern of LH were strongly suppressed. Namely, sub luteal levels of progesterone, around 1 ng/ml, completely suppressed the LH surge but did not affect the pulsatile frequency of LH secretion. Because of this we hypothesized that the two secretory patterns of FSH are similar to that of LH. Long-term ovariectomized Shiba goats that had received implants of estradiol capsules and three different progesterone silastic packet inducing follicular, subluteal and luteal levels of progesterone were divided into three groups such as non-P, low-P and high-P group. Blood samples were collected daily throughout the experiment for the analysis of gonadal steroid hormone levels and at 10-min intervals for 8 h on Days 0, 3, and 7 (Day 0: just before progesterone treatment) for analysis of the pulsatile frequency of FSH secretion. Then estradiol was infused into the jugular vein of all animals at a rate of 3 ${\mu}/h$ for 16 h on Day 8 to determine whether an FSH surge was induced. Blood samples were collected every 2 h from 4 h before the start of the estradiol infusion until 48 h after the start of the infusion. In each group, the mean ${\pm}$ SEM concentration after progesterone implant treatment was 3.3 ${\pm}$ 0.1 ng/ml for the high P group, 1.1 ${\pm}$ 0.1 ng/ml for the low P group, and < 0.1 ng/ml for the non-P group, concentrations similar to the luteal levels, subluteal levels, and follicular phase levels of the normal estrous cycle, respectively. The FSH pulse frequency was maintained highly in all groups on Day 0, Day 3 and Day 7. An FSH surge was induced in all 4 cases of the Non-P group. In the High P and Low P groups, the plasma concentrations of FSH remained low until 48 h after the start of estradiol infusion, and no occurrence of FSH surge was found in any of the animals. The results of this study not only confirm that the pulsatile patterns of FSH were not inhibited strongly relative to LH, they also suggest that some other mechanism and factor may be controlling the FSH secretion.

The Influence of Cortisol Level on Progesterone and Ovulation in the Estrus Dogs (혈액 내 코티졸의 패턴 변화가 발정 암캐의 P4 변화와 배란에 미치는 영향)

  • It is generally accepted that chronic stress impairs female reproduction. It negatively affects ovarian function and the number of ovulated oocytes. Chronic stress lowers the number of retrieved oocytes. Ovarian follicular development is regulated by both pituitary-derived gonadotropins and intraovarian regulatory factors. The main corticosteroids are cortisol, cortisone, 11-deoxycortisol and corticosterone, cortisol being one of the most commonly used welfare and stress physiological indicator. In this study, we investigated the effect of cortisol level on progesterone patterns and ovulation in the dog. Cortisol and progesterone level of serum were analyzed by radioimmunoassay. The day of ovulation was considered as the day when serum progesterone concentration was 6.0~8.0 ng/ml. In vivo dog oocytes were collected by flushing oviducts of mixed-breed bitches at three days after ovulation. We classified dogs as having group 1 (cortisol level, 0 ${\leq}$ or < $2{\mu}g/dl$), group 2 (corisol level, 2 ${\leq}$ or < $4{\mu}g/dl$), group 3 (cortisol level, 4 ${\leq}$ or < $6{\mu}g/dl$) and group 4 (cortisol level, $6{\mu}g/dl$ ${\leq}$). The patterns of progesterone were not different in four cortisol groups. The average numbers of retrieved oocytes was not different in four cortisol groups. These results suggest that different cortisol levels on estrus dogs do not affect ovulation, number of ovulated oocytes and progesterone changes.

  • The effect of lanthanum ion (La3 +), which is associated with the mobilization of internal calcium, on the regulation of oocyte maturation was investigated with Rana dybowskii follicles. Follicular oocytes matured (germinal vesicle breakdown, GVBD) dose dependently when they were exposed to La3+ (O.O1-1.O mM) and the maturation occurred in 9-12 hours after the la3+(0.33 mM) stimulation. lanthanum also accelerated the onset of maturation of the lollicular oocytes exhibiting spontaneous maturation. Three hours of exposure to La3+ was enough to induce the maturation. The La3 + -induced maturation was not associated with progesterone production by follicle cells, and the maturation was inhibited by forskolin (9 $\mu$ M), and cyclobeximide (0.01 - 1.0 - $\mu$g/2 ml) in the medium. The La3+ and hormone stimulated maturation showed the same patterns of protein phosphorylation and dephosphorylation during the maturation. The data suggest that the oocyte maturation by La3+ stimulation is very similar to that by progesterone. Thus, it seems that internal mobilization of Ca2+ plays a key role in the initiation of oocyte maturation in amphibia.

The Use of Radioimmunoassay to Moniter Reproductive Status of Cheju Native Cattle and the Effect of Su, pp.ementary Feeding on Reproduction 1. Body Weight Changes, Breeding Performances and Progesterone Levels from Weaning until First Calving

  • Studies were conducted to investigate the relationship between hormone leveles and nutritional levels for improving performance of Cheju native cattle. In June 1984 a trial was initiated using 8 Cheju native calves after weaning, fed at two su, pp.ementary feeding levels (NRC 100% and 70%). The body weight, breedng performence, change in progesterone level during pregnancy and estrus cycle were evaluated. Mean body weight at 6 months of age was 155kg when fed 100% NRC ration but it was only 137kg when heifers received the 70% NRC ration. At 10, 15 and 20 months of age the body weight was 66, 160 and 115kg, respectively, showing that heifers fed the standard ration gained weight rapidly (P<0.01). Average size of the lefe ovary in the standard group was 2.1${\times}$1.6cm and right ovary was 2.6${\times}$1.8cm. However in the restricted feeding group the ovaries were found to be smaller. Diameter of graffian follicles showed a similar tendency to ovarian size in the two groups. The first oestrus in the standard feeding group a, pp.ared at 14.6 months when body weight was 265kg. Age at first calving was on average 28.9 months at a body weight of 436kg. On the other hand when heifers were fed the restricted ration the first oestrus a, pp.ared at 23.0 months at a body weight of 250kg. Average age at first calving was 38.9 months which was 10 months later than the average in the standard feeding group (P<0.01). In standard feeding group the progesterone level was 2.0ng/ml at two weeks after pregnancy and gradully increased up to 4 weeks and peaked at 18 weeks. This peak (6.4-6.5ng/ml) was maintained up to 24 weeks when progesterone level decreased until it reached 2.1ng/ml at the end of pregnancy. In the restricted group progesterone level up to 16 weeks followed a similar pattern to the standard group but there was a tendency in the restricted group to have lower progesterone levels(P<0.01). The standard and srstricted groups showed similar patterns of progesterone concentration during the oestrus cylce. There were no statistically significant differences in progesterone levels between standard and restricted groups but there was variation between induvidual animals.

Proline-Rich Acidic Protein 1 (PRAP1) is a Target of ARID1A and PGR in the Murine Uterus

  • ARID1A and PGR plays an important role in embryo implantation and decidualization during early pregnancy. Uterine specific Arid1a knockout ($Pgr^{cre/+}Arid1a^{f/f}$) mice exhibit in non-receptive endometrium at day 3.5 of gestation (GD 3.5). In previous studies, using transcriptomic analysis in the uterus of $Pgr^{cre/+}Arid1a^{f/f}$ mice, we identified proline-rich acidic protein 1 (PRAP1) as one of the down-regulated genes by ARID1A in the uterus. In the present study, we performed RT-qPCR and immunohistochemistry analysis to investigate the regulation of PRAP1 by ARID1A and determine expression patterns of PRAP1 in the uterus during early pregnancy. During early pregnancy, PRAP1 expression was strong at day 0.5 of gestation (GD 0.5) and then decreased at GD 3.5 in the epithelium and stroma. After implantation, PRAP1 expression was remarkably reduced in the uterus. However, the expression of PRAP1 at GD 3.5 was remarkably increased in the $Pgr^{cre/+}Arid1a^{f/f}$ mice. To determine the ovarian steroid hormone regulation of PRAP1, we examined the expression of PRAP1 in ovariectomized control, $Pgr^{cre/+}Arid1a^{f/f}$, and progesterone receptor knock-out (PRKO) mice treated with progesterone. While PRAP1 proteins were strongly expressed in the luminal and glandular epithelium of control mice treated with vehicle, progesterone treatment suppressed the expression of PRAP1. However, PRAP1 was not suppressed in both the $Pgr^{cre/+}Arid1a^{f/f}$ and PRKO mice compared to controls. Our results identified PRAP1 as a novel target of ARID1A and PGR in the murine uterus.

Studies on the Effects of Steroids on DNA Synthesis of Chromosmoes in Synchronized Human Cells (同時分裂促進된 사람의 培養細胞에 있어서 染色體의 DNA 合成에 미치는 Steroids의 영향)

  • The frequencies of chromosomal aberrations, unmerical variations at various time intervals and DNA synthetic patterns after the treatment with steroids in synchronized human kidney cells treated with 5-AU were investigated in the present experiment. 1. In 5-AU treated group, the frequency of chromosomal aberrations per cell was 0.131, 3 times of control group. In 5-AU + progesterone and 5-AU + testosterone groups, the frequency of chromosomal aberrations per cell was 0.340 and 0.452 respectively. 2. In 5-AU treated group, the frequency of cells with abnormal chromosome number was 0.8%, which was distributed throughout the time regardless of time interval. In 5-AU + progesterone and 5-AU + testosterone groups, the frequencies of cells with abnormal chromosome number were 2.2% and 4.3% respectively and they increased with the time. In 5-AU + progesterone group, the frequency of chromosomal aberrations exhiited the peaks at 12 and 18 hour stage after the treatment with steroids and, in 5-AU + testosterone group, it decreased with the time and in 5-AU treated group no significant difference was observed 3. The increase of labeled metaphases and labelling intensities in 5-AU treated cells are the result of the accumulation of cells at S stage by 5-AU. The decrease of labeled metaphases, labelling intensities and the delay of DNA synthetic time were observed in steroid groups. DNA synthetic pattern of sex chromosomes differs according to the step of cell cycle and DNA synthetic time is irregular because of double treatments with 5-AU and steroids.

Effects of Prostaglandin $F_{2{\alpha}}$ on the Concentrations of Ovarian and Pituitary Hormones in Korean Native Goats (한국재래산양(韓國在來山羊)에 있어서 Prostaglandin $F_{2{\alpha}}$의 투여(投與)가 난소(卵巢) 및 뇌하수체(腦下垂體) Hormone의 함량(含量)에 미치는 영향(影響))

  • This experiment was conducted to examine the $PGF{2{\alpha}}$-induced changes in concentrations of ovarian and pituitary hormones of Korean native goats. Each goats received two injections of $PGF{2{\alpha}}$ (5mg each ; 3 hours apart) on day 10 of the estrous cycle. Jugular venous blood samples were collected at 0, 3, 6, 12, 24, 48 and 72 hours postinjection for quantification of LH, FSH, prolactin, progesterone and estradiol-$17{\beta}$. The results were summarized as follows ; The blood serum concentration of progesterone was decreased from pretreatment level of $4.15{\pm}1.8ng/ml$ to $2.52{\pm}1.2ng/ml$ (about 60%) within 3 hours and to $0.81{\pm}0.3ng/ml$ at 12 hours of the $PGF{2{\alpha}}$ injection. After 12 hours, the concentrations of progesterone were less than 1.02ng/ml by 72 hours postinjection. The concentrations of estradiol-$17{\beta}$ following treatment increased (p < 0.05) over the 72 hours. Initial concentration of LH was $3.0{\pm}0.3{\mu}IU/ml$. After treatment with $PGF{2{\alpha}}$, concentrations of LH increased within 12 hours but declined 12 and 72 hours from $4.1{\mu}IU/ml$ to $2.5{\mu}IU/ml$. Prior to administration of $PGF{2{\alpha}}$, mean concentration of FSH was $3.5{\pm}0.5{\mu}IU/ml$. Concentrations of FSH declined over time in goats treated with $PGF{2{\alpha}}$ on day 10 postestrus. The mean prolactin concentrations in the blood serum after $PGF{2{\alpha}}$ treatment were not significantly different from those of the pretreatment. It is concluded that the initial increase in LH is dependent on a decrease in serum progesterone and differences in patterns of secretion of gonadotropins might be caused by differences in progesterone or progesterone-estradiol ratio when luteal regression is induced on day 10 of the estrous cycle.

  • Objective: Progesterone receptor (PGR) and AT-rich interactive domain 1A (ARID1A) have important roles in the establishment and maintenance of pregnancy in the uterus. In present studies, we examined the expression of mitochondrial tumor suppressor 1 (MTUS1) in the murine uterus during early pregnancy as well as in response to ovarian steroid hormone treatment. Methods: We performed quantitative reverse transcription polymerase chain reaction and immunohistochemistry analysis to investigate the regulation of MTUS1 by ARID1A and determined expression patterns of MTUS1 in the uterus during early pregnancy. Results: The expression of MTUS1 was detected on day 0.5 of gestation (GD 0.5) and then gradually increased until GD 3.5 in the luminal and glandular epithelium. However, the expression of MTUS1 was significantly reduced in the uterine epithelial cells of $Pgr^{cre/+}Arid1a^{f/f}$ and Pgr knockout (PRKO) mice at GD 3.5. Furthermore, MTUS1 expression was remarkably induced after P4 treatment in the luminal and glandular epithelium of the wild-type mice. However, the induction of MTUS1 expression was not detected in uteri of $Pgr^{cre/+}Arid1a^{f/f}$ or PRKO mice treated with P4. Conclusion: These results suggest that MTUS1 is a novel target gene by ARID1A and PGR in the uterine epithelial cells.

The Reproductive Patterns and Clinical Application of Endangered Common Chimpanzees by Monitoring the Steroid Hormone Measurements in Fecal Samples (분변내 스테로이드 호르몬의 측정을 통한 멸종 위기 침팬지의 번식형태와 임상적용)

  • To monitor the reproductive patterns of endangered common chimpanzee including annual reproductive cycle, amenorrhea, breeding season, and pregnancy diagnosis, Time-Resolved Fluorescence Immuno Assay (TR-FIA) was used to trace MRH (estradiol, progesterone, testosterone) and human chorionic gonadotropin (HCG). In result of this research, age was not the important factor in determining the reproduction capability in common chimpanzee; it was rather greatly influenced by the combination of various factors such as individual's fertilizing ability, presence of obstetrical diseases, the pattern of mating behavior, and the mental communication under the introduction of a mating partner. This research will play an important role in operating conservation project for common chimpanzees and can be extended also for shedding new light on understanding human menopause and obstetrical diseases.
