  • The purplish-red pigment was formed on agar plate by superimposed streaking of Streptomyces propurpuratus ATCC 21630 and strain R-89, The strain No.89 was ascribable to the genus Bacillus and designated as Bacillus sp. R-89. Both strain did not produced such pigment when cultivated independently. For hyperpigment production, we selected the mutant S.P-6 from Streptomyces propurpuratus ATCC 21630 by MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) treatment. Maximum purplish-red pigment 1420 mg/$m\ell$ were produced, when the mutant of R-16 and Bacillus sp. R-89 were mixed cultured at 3$0^{\circ}C$ for 72 hr.

  • Optimal conditions of producing red pigment by Monascus anka, Nakazawa, et Sato, IFO 4478 and 6540 were found to be at pH 6.0 and $30^{\circ}C$ for 10 days. When 3.0% steamed rice and 1.0% defatted sesame exfracts were used as substrates, the highest production of red pigment was yielded. Furthermore, mutants such as Monascus anka 4475-27 and 6540-185 induced by N -methyl-N-nitro-N-nitrosoguanidine (MNNG) treatment were found to produce pigment much more than parent strains.

  • Optimization of culture conditions for pigment production of Monascus purpureus P-57 mutant was investigated in liquid culture. The optimum composition of medium for the pigment production was $4\%$ rice power, $0.1\%$ beef extract, $0.03\%$ glutamic acid, $0.1\%\;MgSO_4{\cdot}7H_2O,\;0.25\%\;KH_2PO_4$, the optimum initial pH was 5.0. And the optimum culture conditions was at $30^{\circ}C$ for 8 days under 150 rpm with shaking. M. purpureus P-57 mutant produced the highest pigment as 356.04 units at red pigment and as 268.20 units at yellow pigment, and produced high cell mass as 15.00 g/L in liquid culture under the optimum conditions.

  • In this study, I investigated the applicability of fluorescein-derived xanthene dyes to fluorescent pigment and the controllability of the optical properties of manufactured pigments. Eosin Y (D&C Red No.22) and phloxine B (D&C Red No.28) were mainly used as a dye to prepare the pigment. Dyes dissolved in a solvent were poured into a powder dispersed in the solvent, then dried and pulverized to fabricate the pigments. Optical characteristics related with fluorescence of the prepared pigment were measured. The optical properties of pigments were varied depending on the solvent used, content of the dye in the pigment, and the ratio of dyes when more then two dyes were mixed. According to the experiment result, it seems that some of the dyes attached to the powder showed fluorescence while the rest did not contribute to it. From the result of pigment washing experiment to explore the binding (or interaction) strength and characteristics of the powder-dye system constituting the pigment, it seems that there are two or more different interactions existing in the pigment system, one of which is relatively stronger than the solvent-dye interaction.

Effects of Natural or Synthetic Pigment Supplementation on Egg Production, Egg Quality and Fatty Acid Contents in the Egg Yolk of Laying Hen (산란계 사료에 천연 및 합성착색제 첨가가 산란성적, 난질 및 난황의 지방산 농도에 미치는 영향)

  • Two experiments were conducted to compare the effects of the natural and synthetic commercial pigments on the laying Performance, Pigmentation and fatty acid contents in e99 yolk of laying hens. The experimental diets were formulated to have isocalories and isonitrogen. In experiment I, the diet does not contain the com in which the xanthophylls are free, and in experiment II, the diet contained 54% of corn in which the level of xanthophylls are 19.34 ppm/kg (calculated levels). In the experiment 1, 480 ISA Brown laying hens were divided into eight groups. Each group has 60 birds fed the 0% of corn with 8 types of pigment levels for 4 weeks. in the experiment 2, 600 ISA Brown laying hens were divided into ten groups. Each group has 60 birds fed the 54% of com diet with 10 types of pigment levels for 4 weeks. Feed intake, e99 Production, egg weight and feed efficiency did not have significant difference in experiments I and II. Albumen height and haugh unit did not have significant difference in both experiments. In order to approach the yolk pigmentation to 12∼13 of Roche color fan, addition level of natural red pigment was 25∼30 ppm. In the case of synthetic red pigment, the level was 15∼20 ppm. In this experimental condition, the pigmenting effect of the synthetic pigment was better than that of the natural pigment. In the experiment 2, the pigmenting effect of mixing pigments were investigated between TM2 mixed with natural red pigment, and TM6 mixed with synthesis red pigment. The pigmenting effect of synthetic red pigment was greater than that of the natural red pigment. However, the pigmenting effect of natural pigment was greater than that of synthetic pigments when the levels of synthetic and natural red pigments in diet are higher in TM3 and TM8. The fatty acid content in yolk was not affected by pigment addition.

  • A bacterial strain, JE-34, producing a high concentration of red pigment was isolated from a sediment in East China Sea. It was identified as Zooshikella sp. JE-34 based on the 16S rRNA gene sequence analysis. The red pigment was purified by solvent extraction and HPLC was identified as prodigiosin-like compound. Nutritional and cultural conditions were optimized for the production of prodigiosin-like pigment in the flask level. Optimal culture conditions were at initial medium pH $6.0{\sim}7.0$, $30^{\circ}C$ and 4 days incubation. For carbon and, nitrogen sources were soluble starch and malt extract.

Pigment and Saikosoponin Production Through Bioreactor Culture of Carthamus tinctorius and Bupleurum falcatum

  • Traditional culture technology of medicinal plants mainly depends on the field culture, which has many problems. With progress of modern culture technology, it has become possible to produce valuable secondary metabolites from medicinal plants. In this paper, we discuss about the pigment and saikosaponin production from too medicinal plants, Carthamus tinctorius and Bupleurum falcatum, through bioreactor culture system. A two-stage bioreactor culture system was established for the production of yellow and red pigments and saikosaponins by cell suspension cultures of Carthamus tinctorius and Bupleurum falcatum. In Carthamus tinctorius, balloon type airlift bioreactors and column type airlift bioreactors were employed for the tell culture and for the pigment production, respectively. The greatest pigment production was obtained on White medium supplemented with 4 mg/L kinetin, high levels of sucrose concentration and photosynthetic photon flux. In Bupleurum falcatum, adventitious roots were cultured in balloon type airlift bioreactors and the root growth was greatest on SH medium containing 5 mg/L IBA and 0.2 mg/L kinetin. HPLC analysis showed that the contents of main active saikosaponins a, c, and d in adventitious roots were almost the same as those in field cultured root.

  • Monascus pigment was produced by Monascus species. During Monascus fermentation, citrinin, the mycotoxin was produced with pigments. Citrinin can become a problem for use of monascus pigment as a food colorant. We found adding of S. cerevisiae filtrate during Monascus cultivation could enhance production of red pigment whereas it could reduce citrinin level. When we added the filtrate at 24 hand 48 h, respectively, pigment production increased about 400% and citrinin concentration decreased to 30%. In a glucose medium, there was no special effect by addition of filtrate. On the other hand, the effect was striking in a sucrose medium.

  • In this study various nutritional and environmental parameters such as, initial moisture content, pH. inoculum size, air rate, sample size and nutrient supplement that influence pigment production were evaluated in solid-state cultures. optimum initial moisture content and pH were determined to be 50% and 6.0, respectively. The supplement of the substrate with different carbon, nitrogen, and mineral source reveals a more inhibitive effect as the substrate concentration increase. optimum aeration rate was determined to be 2vvm in flask culture. The maximum amount of red pigment, 3500 OD/g dried fermented rice, was obtained in optimum conditions which is obtained in solid flask culture.

  • Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.
