새로운 생균제 CS61 배양액의 사료 내 급여가 육계의 생산성에 미치는 영향 (Effects of Dietary Supplementation of a New Probiotic CS61 Culture on Performance in Broiler Chickens)

  • 가금류 제품에서 항생제의 잔류와 내성균의 출현으로 인해 항생제 대체물질에 대한 연구가 활발히 진행되고 있으며, 특히 생균제와 활성촉진제 또는 이들을 조합한 신바이오틱의 사용이 권장되고 있다. 본 연구는 새로운 생균제 CS61 배양액의 사료첨가 급여가 육계의 성장 및 사료효율에 미치는 영향과 안전성을 평가하여 항생제를 대체할 수 있는 사료첨가제로서의 개발가능성을 알아보기 위해 수행하였다. CS61 배양액은 0, 0.1 및 1%의 용량으로 28일간 사료에 혼합하여 육계에게 급여하였다. 시험결과, 시험물질 처치군에서 부검 시의 체중과 일당증체량이 대조군에 비해 용량의존적으로 증가하였다. CS61 배양액의 사료 내 첨가는 대조군 동물에 비해 사료효율도 개선하는 것으로 나타났다. 반면, 일반증상과 사망률, 부검소견, 혈액학치 및 혈청생화학치에서는 시험물질의 처치와 관련된 독성소견이 관찰되지 않았다. RAW 264.7 세포를 이용한 일산화질소 시험에서 정제된 CS61 펩타이드는 lipopolysaccharide에 유도된 일산화질소 생성을 용량의존적으로 억제하였다. 본 시험결과는 육계에 CS61 배양액의 사료첨가 급여는 항염증효과를 통해 성장과 사료효율을 개선할 수 있음을 보여주며, 사료첨가제로서 CS61 배양액의 유용성과 개발가능성을 시사해 주고 있다.

  • 곱게 갈아 액화시킨 음식물 찌꺼기를 기질로 생균사료를 생산하기 위한 효모의 호기적 액상발효에 있어서 효모증식에 대한 교반속도의 영향을 연구하였다. screening을 통해 선택한 효모인 Kluyvermyces marxianus를 종균으로 2liter jar fermenter를 이용하여 호기적 조건에서 곱게 분쇄해 10% 고형분 함량을 갖는 기질에 교반속도를 500rpm, 900rpm, 1200rpm를 다르게 조정하여 $35^{\circ}C$에서 증식시켰다. 효모의 증식을 촉진시키기 위하여 고분자물질을 분해하는 효소를 분비하는 곰팡이 Aspergillus oryzae와 혼합발효를 실시하여 단독발효와 비교하였다. 발효결과 교반속도가 증가할수록 생균수도 높아져 400rpm에서보다는 900rpm에서 높은 생균수를 보였으나 1200rpm에서는 오히려 감소하였다. 혼합발효는 amylase의 역가를 높여주어 증식속도를 높여 주나 발효종료단계에서 최고 생균수를 증가시켜주지는 않는 것으로 나타났다.

  • We investigated the effect of dietary supplementation with probiotics on growth performance, feed utilization, innate immunity and disease resistance against Streptococcus iniae in juvenile olive flounder Paralichthys olivaceus. Four experimental diets were made by supplementation with three Bacillus sp. probiotics (B. subtilis, B. pumilus and B. licheniformis) into a basal diet (control) at 0.5% each. Three-hundred fish (average BW, 20 g) were randomly distributed (25 per tank) into twelve 150 L capacity polyvinyl circular tanks with three replicates per dietary treatment. Triplicate groups of fish were fed the experimental diets to apparent satiation (twice per day, 09:00 and 17:00 h) for nine weeks. After the feeding trial, a challenge test was directly conducted against S. iniae. The results indicated that dietary supplementation with the probiotic, Bacillus sp, seemed to improve growth performance of olive flounder. Hematological parameters and innate immunity as determined by nitroblue tetrazolium, myeloperoxidase or superoxide dismutase activities were not significantly affected by the probiotic supplementation. The result from the challenge test suggested that dietary supplementation with probiotics Bacillus sp. can improve the disease resistance of olive flounder against S. iniae.

고온세균을 이용한 남은 음식물의 호기적 액상발효 (Aerobic Liquid Fermentation of Residual Food Waste by Thermophilic Bacteria)

  • 남은 음식물을 기질로 생균사료를 생산하기위하여 고온성 세균에 의한 호기적 액체 발효가 시도되었다. 토양이나 남은 음식물들로부터 11종의 고온성 세균이 $55^{\circ}C$에서의 진탕배양에 의해 분리되었다. 이들 분리균 들의 기질분해능을 평가하기위해 배양액 중에서의 ${\alpha}-amylase$ 와 protease효소역가를 측정 비교하였다. 이 결과 6종의 세균이 선택되었고 남은 음식물 기질 적응력을 높이기 위해 반복 배양을 실시하였다. 발효말기에서의 생균농도는 대부분 $3{\sim}7{\times}10^9/ml$에 이르렀다. 이들 중 B3, B6가 가장 높은 효소역가를 나타냈다. B3, B6, 그리고 분양받은 균주인 Bacillus Stearothermophillus를 2L-jar fermenter를 이용해 혼합발효시킨 결과발효개시 8시간 내에 생균수가 $1.4{\times}10^{10}/ml$에 이르렀다.

Effects of Bacillus SW1-1 coated diets on innate immunity and disease resistance of olive flounder Paralichthys olivaceus against Edwardsiella tarda infection

  • Bacillus SW1-1 is a probiotic isolated from shrimp intestines. We investigated the effects of Bacillus SW1-1 coated diets on the growth, feed utilization, innate immunity, hematological parameters and resistance to Edwardsiella tarda in olive flounder (Paralichthys olivaceus). A commercial diet was used as the control (AP0) and two other diets were prepared by coating 0.25% (AP25) or 0.50% (AP50) probiotic powder which contains 1.0 × 107 CFU/g Bacillus SW1-1. Four replicate groups of olive flounder (153 ± 2 g) were fed one of the diets for 12 weeks. Growth performance and feed utilization of the fish were not significantly affected by the dietary Bacillus SW1-1. After the challenge with E. tarda, AP50 group showed significantly higher survival than AP0 and AP25 groups. Innate immunity and anti-oxidant capacity of the fish were not significantly affected after the feeding trial. However, after the E. tarda challenge, the innate immune parameters (immunoglobulin, lysozyme and anti-protease) were significantly improved in fish fed AP25 and AP50 diets compared to those in fish fed AP0 diet. After the challenge test, significantly lower glucose level was observed in AP50 group compared to AP0 group. These results indicate that dietary supplementation of Bacillus SW1-1 could increase the disease resistance of olive flounder against E. tarda infection. The optimum coating levels of Bacillus SW1-1 needs to be further elucidated.

Complete genome sequence of probiotic Lactobacillus johnsonii 7409N31 isolated from a healthy Hanwoo calf

  • Lactobacillus johnsonii 7409N31 was isolated from the feces of a healthy 11-day-old Hanwoo calf from a farm in Geochang-gun, Gyeongsangnam-do, Korea. The genome of the strain was completely sequenced using the PacBio RSII sequencing system, and it was confirmed that it was composed of one circular chromosome. The size of the entire genome was 2,198,442 bp, and it had 35.01 mol% guanine + cytosine (G + C) content and 2,222 protein-coding sequences, 24 rRNA, 3 ncRNA, and 112 tRNA genes. Strain 7409N31 possessed genes encoding enzymes involved in the hydrolysis of both fibrous and non-fibrous carbohydrates. These data provide a comprehensive theoretical understanding for developing industrial probiotic feed additives that improve nutrient digestibility.

Comparative Genome Analysis and Evaluation of Probiotic Characteristics of Lactobacillus plantarum Strain JDFM LP11

  • In the current study, the probiotic potential of approximately 250 strains of lactic acid bacteria (LAB) isolated from piglet fecal samples were investigated; among them Lactobacillus plantarum strain JDFM LP11, which possesses significant probiotic potential, with enhanced acid/bile tolerance, attachment to porcine intestinal epithelial cells (IPEC-J2), and antimicrobial activity. The genetic characteristics of strain JDFM LP11 were explored by performing whole genome sequencing (WGS) using a PacBio system. The circular draft genome have a total length of 3,206,883 bp and a total of 3,021 coding sequences were identified. Phylogenetically, three genes, possibly related to survival and metabolic activity in the porcine host, were identified. These genes encode p60, lichenan permease IIC component, and protein TsgA, which are a putative endopeptidase, a component of the phosphotransferase system (PTS), and a major facilitator in the gut environment, respectively. Our findings suggest that understanding the functional and genetic characteristics of L. plantarum strain JDFM LP11, with its candidate genes for gut health, could provide new opportunities and insights into applications in the animal food and feed additive industries.

도축 폐혈액 단백질을 이용한 Probiotics생산에서의 동결건조 조건 (Optimization of Freeze-drying Conditions for Probiotics Production with Animal Blood Proteins Added Medium.)

  • 본 연구는 도축 폐기물인 가축혈액의 이용을 위해 사료첨가제로서 혈분 성분이 함유된 유산균 probiotics의 생산을 목적으로 하여 수행되었다. 혈액으로부터 얻은 혈장을 질소원으로 이용한 BBPB 배지에, 시판 중인 미생물 사료첨가제로부터 분리된 probiotics용 Lactobacillus sp.를 배양하여 대조구인 MRS배지의 74%에 이르는 높은 생균수를 얻을 수 있었다. 얻어진 배양액의 동결 건조는 배양시간, 배양액의 pH보정, 배양액의 농축, 급속동결 등의 조건에 의해 균체의 생존율이 영향을 받았으며, 24시간 배양한 배양액을 pH 6.4로 보정한 뒤 10배로 농축하여 -5$0^{\circ}C$에서 3시간 동안 동결한 후 건조하는 조건으로 최적화되었다. BBPB 배지에 잔여하는 혈장 단백질 성분은 유산균의 동결건조 안정화 효과도 가지고 있는 것으로 나타났고 그 수준은 MRS와 유사하였다. 제제화를 위한 동결건조 안정제로는 Sucrose가 높은 효과를 나타내었고 10%첨가시 48.3%의 생존율로 $3.0{\times}^{10}$ CFU/g의 유산균 제제를 얻을 수 있었다. 본 연구에서의 동결건조조건을 이용하면 도축 폐혈액을 이용한 problotics의 산업적 생산이 가능할 것으로 사료된다.

Intestinal Colonization Characteristics of Lactobacillus spp. Isolated from Chicken Cecum and Competitive Inhibition Against Salmonella typhimurium

  • Probiotics are live microbial feed supplements which beneficially affect the host animal by improving its intestinal microflora. To select the best Lactobacillus spp. as a chicken probiotic, probiotic characteristics of 10 selected Lactobacillus strains isolated from chicken cecum or obtained from KCTC were investigated. The strains were examined for resistance to pH 2.0 and 0.3% oxgall, and adhesion to cecal mucus and cecal epithelial cells. All strains grew in MRS containing 0.3% oxgall. However, Lb. plantarum AYM-10, Lb. fermentum YL-3, AYM-3, and Lb. paracasei YL-6 showed relatively high resistance to 0.3% oxgall. Lb. fermentum YL-3, YM-5, AYM-3, and Lb. paracasei YL-6 survived 4 hours of incubation at pH 2.0. Lb. fermentum YL-3, KCTC 3112, and Lb. plantarum AYL-5 were strongly adhesive to cecal mucus, while the rest showed moderate or low adhesion. Lb. plantarum AYM-10, AYL-1, and AYL-5 had good adhering properties to cecal epithelial cells (30.7$\pm$10.82, 40.2$\pm$20.90, and 14.5$\pm$4.22, respectively). Lb. fermentum YL-3, AYM-3, and KCTC 3547 showed Intermediate adhesion ability, and Lb. plantarum showed better adhesion ability to cecal epithelial cells than Lb. fermentum. Attached Lb. fermentum YL-3 to cecum after 60 min incubation was confirmed using CLSM. Lb. fermentum YL-3 attached to a matrix which was composed of a mucus layer adjacent to intracrypts and pericryptal region. Some Lb. fermentum YL-3 bound to mucosal epithelial cells. From these results, Lb. fermentum YL-3 was selected as a chicken probiotic. In vivo trials of chicks inoculated with Lb. fermentum YL-3 had decreased Salmonella population in cecal contents and livers (p<0.5).

Effects of Dietary Enterococcus faecium SF68 on Growth Performance, Nutrient Digestibility, Blood Characteristics and Faecal Noxious Gas Content in Finishing Pigs

  • The objective of this study was to investigate the effects of feeding probiotic (Enterococcus faecium SF68, EF) on growth performance, nutrient digestibility, blood characteristics and faecal noxious gas content in finishing pigs. A total of eighty [($Landrace{\times}Yorkshire$)${\times}Duroc$] pigs with an initial BW of $50.47{\pm}2.13kg$ were used in this 8-week experiment. Pigs were allotted to four treatments (4 replicates per treatment and 5 pigs per pen) according to a randomized complete block design. Dietary treatments were: 1) CON (control; basal diet), 2) CTC (control diet+0.1% antibiotic, chlortetracycline), 3) EF1 (control diet+0.1% probiotic, EF) and 4) EF2 (control diet+0.2% probiotic, EF). During weeks 0-4, ADG was not affected by the addition of antibiotic or EF (p>0.05). In weeks 4-8, ADG tended to increase in CTC and EF treatments compared to CON treatment (p<0.10). ADFI and gain/feed were not affected in each 4-week period and the entire experimental period (p>0.05). Digestibilities of DM and N were higher in EF supplemented treatments than in CON and CTC treatments (p<0.05). Blood characteristics of WBC, RBC and lymphocyte were not affected in pigs given diets containing EF (p>0.05). Supplementation of EF in the diet decreased faecal ammonia nitrogen ($NH_3$-N) and hydrogen sulphide ($H_2S$) concentrations (p<0.05). Faecal acetic acid concentration tended to decrease (p<0.10) while propionic acid and butyric acid concentrations were significantly lower on diets with EF supplementation than on the diet containing antibiotic (p<0.05). In conclusion, dietary supplementation of EF can increase nutrient digestibility and decrease faecal $NH_3$-N, $H_2S$ and volatile fatty acid (VFA) concentrations in finishing pigs.
