Cromakalim (BRL 34915), known as an airway smooth muscle relaxant, inhibited the releases of mediators in the antigen-induced mast cell activation. It has been suggested that cromakalim, in part, inhibited mediator releases by inhibiting the initial increase of 1,2-diacylglycerol (DAG) produced by the activation of the other phospholipase system which is different from phosphatidylcholine-phospholipase D pathway. The aim of this study is to further examine the inhibitory mechanism of cromakalim on the mediator release in the mast cell activation. Guinea pig lung mast cells were purified by using enzyme digestion and percoll density gradient. In purified mast cells prelabeled with $[^3H]PIP_2$, phospholipase C (PLC) activity was assessed by the production of $[^3H]$insitol phosphates. Protein kinase C (PKC) activity was assessed by measuring the protein phosphorylated from mast cells prelabeled with $[{\gamma}-32P]ATP$, and Phospholipase $A_2\;(PLA_2)$ activity by measuring the lyso-phosphatidylcholine produced from mast cell prelabeled with 1-palmitoyl-2-arachidonyl $phosphatidyl-[^{14}C]choline$. Histamine was assayed by fluorometric analyzer, and leukotrienes by radioimmunoassay. The PLC activity was increased by activation of the passively sensitized mast cells. This increased PLC activity was decreased by cromakalim pretreatment. The PKC activity increased by the activation of the passively sensitized mast cells was decreased by calphostin C, staurosporine and cromakalim, respectively. The $PLA_2$ activity was increased in the activated mast cells. The pretreatment of cromakalim did not significantly decrease $PLA_2$ activity. These data show that cromakalim inhibits histamine release by continuously inhibiting signal transduction processes which is mediated via PLC pathway during mast cell activation, but that cromakalim does not affect $PLA_2$ activity related to leukotriene release.
The reduction of carbon dioxide emission is hot issue in the world because we are confronted with serious global warming and climate change. As a part of carbon dioxide reduction efforts, various approaches for increasing filler loading have been carried out in order to decrease the energy consumption in papermaking processes. Effects of the pretreatment of GCC with cationic starch and MFC on the flocculation behaviour of GCC were investigated in this study. Pretreatment of GCC with cationic starch caused the change of electric charge of suspension and flocculation behaviour of GCC. Largest flocculation size was obtained near the isoelectric point in the case of cationic starch treatment. When MFC (30 times grinded) was added after preflocculation of GCC with cationic starch, the flocculation size was increased, but largest flocculation size was obtained at -150 mV of electric charge of suspension in this study. However the addition of highly grinded MFC (60 times grinded) caused smaller flocculation size of GCC than those of MFC (30 times grinded). When GCC and MFC were mixed first, and then cationic starch was added, the characteristics of MFC and the change of electric charge which could be brought by cationic starch did not affect the flocculation size of GCC at all. The flocculation size obtained by the combination of cationic starch and MFC was smaller than those of cationic starch. These results show that flocculation behaviour could be controlled by the change of electric charge of suspension and the combination methods of cationic starch and MFC.
Tetragonia tetragonoides (Aizoaceae) has been known as an anti-cancer agent. The activation of proteinase-activated receptor-2 (PAR-2) by trypsin appears to play a role in inflammation. In the present study, we examined the inhibitory effects of Tetragonia tetragonoides water extract (TTWE) on the production of tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ and tryptase in trypsin-stimulated human leukemic mast cells (HMC-1) expressing PAR-2. HMC-1 cells were stimulated with trypsin in the presence or absence of TTWE (10, 100, and $1000\;{\mu}g/ml$). The level of $TNF-{\alpha}$ secretion from HMC-1 cells was measured by enzyme-linked immunosorbent assay (ELISA). $TNF-{\alpha}$ and tryptase mRNA expression were examined by reverse transcription-PCR. Also, extracellular signal-regulated kinese (ERK) activation was assessed by Western blot analysis. Trypsin activity was measured using the substrate Bz-DL-Arg-p-nitroanilide (BAPNA). It was observed that $TNF-{\alpha}$ secretion, tryptase mRNA and $TNF-{\alpha}$ mRNA expression in trypsin-stimulated HMC-1 cells were inhibited by pretreatment of TTWE ($1000\;{\mu}g/ml$). Furthermore, the pretreatment of TTWE ($1000\;{\mu}g/ml$) resulted in the reduction of ERK phosphorylation and trypsin activity. These results suggest hat TTWE might have the inhibitory effects on the PAR-2-dependent inflammation processes and it is likely to function as PAR-2 antagonist.
섬유성 기질을 물 가수분해하여 생긴 당을 최대한 회수하는 여러 회수방법을 조사하였다. 사용한 전처리 조건은 반응온도가 $170\~180^{\circ}C$, 반응시간이 1시간이었다. 반응 온도가 170에서 $180^{\circ}C$로 증가함에 따라 xylan의 용해율은 증가하지만 $180^{\circ}C$ 이상에서는 상당양의 당이 분해됨을 알 수 있었다. 가수분해 결과로부터 남은 고체기질과 당화액에서 얻은 당의 물질수지를 조사하였는데 glucan의 경우는 비교적 잘 맞으나, 헤미셀룰로오스는 처리되지 않은 기질에 포함된 초기양과 비교할 때 상당한 차이가 났다. 그래서 여러가지 회수방법을 고안하여 당의 물질수지를 조사한 결과 회수기간동안 열을 가하거나 초음파를 조사하는 방법과 같은 물리적인 자극을 주면 액상에서 헤미셀룰로오스 회수율이 증가되는 것이 확인되었다. 이런 사실로부터 당 회수시 기질이 크고 전처리 후 침출용매를 많이 사용할 수 없는 상업적인 공정의 경우 상당양의 올리고당이 고체기공내 갇혀 액상으로 회수되지 못할 것으로 생각된다.
안정동위원소 분석기법(SIA)은 생태학 및 환경학적 측면에서 유기물의 기원 추적 및 생지화학적인 문제를 해결함에 있어서 유용하기 때문에 그 사용이 꾸준히 증가되고 있다. C, N, S, H, O 등의 원소는 모두 하나 이상의 동위원소를 가지고 있으며, 동위원소 조성비는 질량분석기를 이용하여 매우 정밀하게 측정할 수 있다. 이러한 동위원소 조성비는 물리적, 화학적, 생물학적 반응을 통한 분별작용으로 인하여 예측 가능한 방식으로 변경된다. 더 나아가 안정동위원소 기법은 생태와 환경 학문에서 유기물의 기원을 밝히고, 생태계내 먹이망 구조를 밝히고, 영양염 등의 질산염 순환을 이해하고, 동물의 이동경로를 밝히고, 오염원을 추적하는 등 매우 강력한 도구로서 사용할 수 있다. 그러므로 생태학 및 환경학 분야에서 안정동위원소의 활용이 매우 유용하다.
Reactive oxygen species (ROS) and nitrogen species (RNS) are implicated in cellular signaling processes and as a cause of oxidative stress. Recent studies indicate that ROS and RNS are important signaling molecules involved in nociceptive transmission. Xanthine oxidase (XO) system is a well-known system for superoxide anions ($O{_2}^{{\cdot}_-}$) generation, and sodium nitroprusside (SNP) is a representative nitric oxide (NO) donor. Patch clamp recording in spinal slices was used to investigate the role of $O{_2}^{{\cdot}_-}$ and NO on substantia gelatinosa (SG) neuronal excitability. Application of xanthine and xanthine oxidase (X/XO) compound induced membrane depolarization. Low concentration SNP ($10{\mu}M$) induced depolarization of the membrane, whereas high concentration SNP (1 mM) evoked membrane hyperpolarization. These responses were significantly decreased by pretreatment with phenyl N-tert-butylnitrone (PBN; nonspecific ROS and RNS scavenger). Addition of thapsigargin to an external calcium free solution for blocking synaptic transmission, led to significantly decreased X/XO-induced responses. Additionally, X/XO and SNP-induced responses were unchanged in the presence of intracellular applied PBN, indicative of the involvement of presynaptic action. Inclusion of GDP-${\beta}$-S or suramin (G protein inhibitors) in the patch pipette decreased SNP-induced responses, whereas it failed to decrease X/XO-induced responses. Pretreatment with n-ethylmaleimide (NEM; thiol-alkylating agent) decreased the effects of SNP, suggesting that these responses were mediated by direct oxidation of channel protein, whereas X/XO-induced responses were unchanged. These data suggested that ROS and RNS play distinct roles in the regulation of the membrane excitability of SG neurons related to the pain transmission.
Polytetrafluoroethylene (PTFE) membrane has high resistance to chlorine, which is a great advantages in chemical cleaning to recover water flux during membrane processes in drinking water systems. A humic kaolin water with approximately 4 mg/L of DOC and 10 NTU of turbidity was prepared as a feed water. Coagulation pretreatment with or without settling was applied. The coagulation with settling showed the greatest water production. The reduced flux was effectively recovered by NaOCl cleaning, i.e., 21% recovery by 50 mg/L of NaOCl cleaning and 49% recovery by 500 mg/L NaOCl cleaning. The images of SEM and AFM analyses were corresponded to the water flux variation. However, when the floc was accumulated on the membrane surfaces, the efficiency of NaOCl cleaning was substantially limited. In addition, dynamic contact angle became greater after cleaning, which indicates changes in characteristics of fouling layer such as surface hydrophobicity. Proper cleaning technologies during enhanced backwash using NaOCl would expand application of PTFE membranes in drinking water systems.
이산화탄소 저감을 위한 바이오에너지의 개발이 활발히 진행되고 있는 가운데 본 연구에서는 산당화과정을 이용하여 볏짚으로부터 셀룰로스 에탄올의 제조공정을 해석하고자 하였다. 전처리 과정으로는 초음파에너지를 이용한 수화과정과 10~30 wt%의 황산을 이용한 산당화과정을 진행하였으며, 발효과정에서는 10~50 wt%의 효모를 이용하여 3~6 일간 발효시켜 셀룰로스 에탄올 수율을 결정하였다. 최적 전처리조건으로는 375W의 초음파세기로 30 min 간 수화시킨 후 20 wt%의 황산을 이용하여 산당화과정을 2 h 동안 진행하는 것을 추천할 수 있으며, 30 wt%의 효모를 이용하여 3일간 발효하는 것이 가장 높은 셀룰로스 에탄올 수율을 얻을 수 있었다.
자동차의 유리를 폴리카보네이트로 대체하기 위하여 폴리카보네이트 판 위에 하드 코팅을 시도하였다. 본 연구에서는 tetraethyl orthosilicate (TEOS), methyltriethoxysilane (MTES)으로부터 졸-겔 과정을 이용하여 코팅 막을 형성하였는데 이 때 가장 우수한 물성을 갖는 배합 조건을 찾아보았다. 폴리카보네이트 판의 전처리, TEOS와 MTES의 비율, 용매의 선택, aging 시간, 산 촉매의 양, 코팅의 횟수 등을 변화시켜가면서 형성된 코팅 막의 물성을 측정하였다. 측정 결과 폴리메칠메타크릴레이트를 이용한 전처리가 접착력 향상에 매우 효과적이었으며 이러한 졸-겔 과장으로부터 연필경도 2 H 정도의 아주 매끄러운 코팅 막이 형성되었다.
다양한 하 폐수 처리공정 중 혐기성 소화공정은 이산화탄소 배출을 감소시키고 생성되는 메탄을 에너지로서 사용할 수 있는 장점을 가지고 있다. 본 논문에서는 이러한 혐기성 소화공정의 문제점과 보완점에 대해 살펴보았다. 가수분해과정은 혐기성 소화과정 중 율속단계에 해당하여 소화공정과정을 촉진시키고 바이오가스의 생산을 증가시키기 위하여 다양한 전처리 방법이 개발되어왔다. 현재 혐기성 소화공정을 위한 전처리 방법 중 열처리 방법, 초음파 처리, 기계적 처리방법, 화학적 처리방법 등이 상업적으로 이용되고 있으며, 이들 공정은 슬러지 플록 또는 세포의 파괴를 통해 세포분획물이 생물학적으로 분해될 수 있는 형태로 전환시키는 것을 목적으로 한다. 이러한 과정들 모두는 특정 상황에 따른 장점과 단점을 모두 지니고 있으므로 각 공정과정에 대한 이해와 이를 통한 적용을 통해 특정 슬러지에 적합한 최적의 전처리 공정을 도출해 낼 필요가 있다. 또한 혐기성 소화공정의 효율증대와 경제성 확대를 위한 혐기성 소화공정 개발이 필요하다고 할 수 있다.
