  • Tumor incidence in wild mammals is reportedly very low. Wild nutria, a large rodent, is known to carry many infectious diseases, but rarely exhibits neoplastic diseases. We necropsied a male wild nutria and found a large nodular mass in the left inguinal region, adjacent to the penis. Histopathologically, the mass was diagnosed as preputial gland adenoma. Spontaneous preputial gland adenomas are extremely rare in all animals. Moreover, reports of tumors in nutrias have been limited to adenocarcinomas of the lungs and uterus, as well as subcutaneous fibromas. Here, we describe preputial gland adenoma in a wild nutria.

  • In order to know the histological changes of rat preputial gland during normal sexual cycle and sex hormone treatment, matured Wistar rats (B.W.about 200g) were used for the experiment. Rats were subcutaneously given $2{\mu}g$ 17-${\beta}$-estradiol (Sigma) and 2.5g progesterone (Nakarai Chem., Japan) daily in 0.5ml propylen glycol for ten days respectively. The results obtained are as follow: At the stage of estrus and metestrus, the eosinophilic crystalloid granules and large vacuoles in the acinar cells appeared numerously, and the excretory ducts were severely extended. The developed connective tissue between the acinus were also found. At the stage of proestrus and diestrus, however, the small vacuoles and a few eosinophilic crystalloid granules appeared in the acinar cell. In the estradiol treatment, on the other hand, severely extended excretory ducts and a small number of the eosinophilic crystalloid granules compared with the progesterone treatment were found. The cyst-like structure was found, and in that the acinus disappeared completely. In the progesterone treatment, remarkably extended excretory ducts and nomerous appearance of oval formed eosinophilic granules in the acinar cells were found. As these findings, it could be suggested that the secretion of rat preputial gland was active at the estrus and metestrus stage, and estrogen may concerned in the secretion, land progesterone in the formation of secretory products.

  • This study investigated that exposure of male mice to estrogen receptor $\alpha$-selective agonist, 4,4',4"-(4-propyl-[1H]-pyrazole-1,3,5-triyl)tris-phenol (PPT) induce morphological changes of accessory genital glands. The male reproductive organs were fixed and processed for light microscopy. The PPT induced decreases of ventral prostates, seminal vesicles and preputial glands weights with experimental time. The glandular lumen of ventral prostate was atrophied compared with control group. Type of epithelial tissues in the prostate was changed from simple columnar epithelium to stratified cuboidal or squamous epithelium. Treated group with the agonist showed that increased connective tissue underlying epithelium in the prostate and seminal vesicle. Especially, the glandular lumen of the seminal vesicle was contracted when PPT-treated animals were compared with control group. Secretion cells of preputial gland were smaller than that of control group. On week 8, PPT treatment caused decrease of epithelial cell height lining the lumen of preputial gland. These results provide information useful in researching the physiological function of estrogen mediated by estrogen receptor $\alpha$ in male accessory genital gland.

Morphological Changes of Accessory Genital Organs Induced by Treatment with Different Concentration of Estrogen Receptor Agonist in the Male Mouse (수컷 생쥐에서 에스트로겐 수용체 촉진제의 농도별 투여에 의한 부속 생식샘의 형태학적 변화)

  • The aim of the present study is to validate the effects of treatment with different concentration of estrogen receptor alpha agonist, propyl pyrazole triol (PPT) on the weight and histological structure in the accessory reproductive organs (ventral prostate, seminal vesicle and preputial gland) of male mouse. Treated groups received different doses of PPT 0.01 mg, 0.1 mg and 1.0 mg per week respectively, for 3, 5, and 8 weeks. In general, the weight of reproductive organs was increased in PPT 0.01 mg and 0.1 mg treatment, however decreased in PPT 1.0 mg treatment. Epithelial tissues in the ventral prostate were changed from simple columnar epithelium to squamous or cuboidal epithelium in the treated groups. On week 3, PPT groups caused decrease of epithelial cell height in the ventral prostate. Lumen of the seminal vesicle was narrowed in the treated group. Epithelial cell height of seminal vesicle was reduced in the PPT treatment. Acinus tissue of preputial gland in PPT 1.0 mg treatment was dramatically atrophied than that of control group. These results are useful as a reference to determine the administration concentration of PPT in experiments for understanding the physiological functions of estrogen in the male.

  • Reductive amination of N-succinyl-chitosan (1) and lactose using sodium cyanoborohydride in 1/15 M phosphate buffer (pH 6.0) for 6 d was suitable for the preparation of lactosaminated N-succinyl-chitosan (2). At 8, 24 and 48 h after i.v. administration of fluorescently labeled 1 (1') or 2 (2'), Peyer's patch, mesenteric lymph nodes, testes, prostate, preputial grand, intestine (small intestine plus cecum), femoral muscle, backbone and peritoneum were taken. Peyer's patch and mesenteric lymph nodes were put together as lymph nodes. Over 10% of dose/g tissue was distributed to the prostate and lymph nodes at 48 h post-administration in both l' and 2'.2' was easily distributed into not only the liver but also prostate, intestine, preputial gland and lymph nodes. Although galactose receptors are known to exist not only on the liver parenchymal cells but also on prostate and testes, the selective distribution of 2' into the prostate and the testes were not observed clearly. This study suggested that 1 and 2 should have possibilities for both the prevention and cure of lymph node metastasis as drug carriers.
