A 10-week feeding trial was conducted to investigate the effects of dietary glucose, dextrin and starch on growth and body composition of juvenile starry flounder. Triplicate groups of fish (average weight, 9.7 g) were fed iso-nitrogenous (53% CP) and iso-caloric (3.8 kcal/g diet) diets containing 20% glucose, 20% dextrin and 5-25% alpha-potato starch with 5-14% lipid levels. Survival was not affected by dietary carbohydrate. Weight gain, feed efficiency and protein efficiency ratio of fish fed the diet containing 20% glucose were the lowest among all groups. The best weight gain was observed in fish fed the diets containing 20% dextrin. Growth and feed efficiency were not affected by dietary -potato starch level. Lipid contents of whole body and liver were not affected by dietary glucose, dextrin and starch at the same level. However, the lipid contents tended to decrease with increasing dietary starch level and those of fish fed the diets containing 5% alpha-potato starch were significantly higher than those receiving 10-25% alpha-potato starch. Liver glycogen content and hepatosomatic index tended to increase with increasing dietary starch level. These results indicate that juvenile starry flounder are able to efficiently utilize dextrin and -potato starch compare to glucose in diets and that alpha-potato starch could be incorporated up to 25% in the diet for optimum growth by juvenile starry flounder.
A 20-week growth trial was conducted in flow-through aquarum system to investigate the practical dietary carbohydrate sources for juvenile abalone (Haliotis discus hannai). Four replicate grops of the abalone averaging 0.125g were fed one of eight diets containing 24.2% wheat flour (WF), 20% dextrin (DEX), 20% sucorse (SUC), 10% $^{\alpha}$-potato starch+10% $^{\beta}$-potato starch (ab-S), 15% $^{\alpha}$-potato starch (a-S15), 20% $^{\alpha}$-potato starch (a-S20), 25% $^{\alpha}$-potato starch (a-S25), or mixture (MIX) with practical ingredients such as soybean meal, corn gluten meal, cotton seed meal and heat flour. In addition, these formulated diets were compare with macroalgae such as dried sea mustard Undaria (D-SM) or dried sea tangle Laminaria(D-ST). Survival rate, weight gain, shell growth and soft body weight of abalone were not significantly affected by the different dietary carbohydrate sources (P>0.05), whereas those fed a-S15 diet were slightly low. These values of abalone fed D-ST were lowest (P<0.05), followed by those fed D-SM. Lipid contents of soft body from abalones fed a-S25, D-ST or D-SM were significantly lower than those of abalone fed other diets (P<0.05). These data indicate that abalone can equally utilize any carbohydrate sources used in this study.
Purple-fleshed potato powder (PFPP) was investigated to determine optimal mixing ratio with milk powder and dextrin to produce a ready-to-eat mashed potato powder. The rheological characteristics, color, and anthocyanin contents were studied at a different concentration of ingredients. The power-law model was applied to explain the mechanical spectra of mashed potatoes which represented the change in structure induced by different mixing ratios. Mixture design was used to obtain the experimental points used to establish the empirical models to describe the effects of each ingredient on the characteristic of the mashed potato. The results of mechanical spectra showed that both storage and loss moduli (G' and G'') were significantly influenced by PFPP and milk powder concentration. The power law parameters n' and n'' showed higher values for the mashed potato with a lower concentration of PFPP and a higher concentration of milk powder, which showed that the gel networks involved in the mashed potato were weaker. The optimum mixing ratio with the highest redness and anthocyanin content, while maintaining the rheological properties similar to the commercial mashed potato, was determined as PFPP:milk powder:dextrin = 90.49:4.86:4.65 (w/w). The proportions of PFPP and milk powder in the formulation significantly changed the characteristics of mashed potato, whereas no significant effect of dextrin was observed in this formulation.
A Korean commercial sweet rice drink "Sikhye" showed sucrose, fructose, glucose, maltose, limit dextrin and various size of maltooligosaccharides in HPLC and TLC analysis. Commercial Sikhye was found to contain 0.09% of limit dextrin and 0.2% of rice residue. Limit dextrin in commercial Sikhye showed both signal of $\alpha$-1,4- and $\alpha$-1,6-glucosidic linkage with its estimation ratio of 15:1 by 1H-NMR analysis. This limit dextrin was hydrolyzed to produce various size of maltooligosaccarides with more longer chain than that of traditional Sikhye by pullulanase. Limit dextrin was digested wit enzymes(30units/ml) of $\alpha$-amylase, $\alpha$-glucosidase and glucoamylase from Aspergillus awamori, sweet potato $\beta$-amylase and human salivary $\alpha$-amylase at 37$^{\circ}C$ for 1 hour, respectively. Hydrolysis rates of these amylases on it were higher than in case of traditional sikhye. $\alpha$-Glucosidase plus human salivary $\alpha$-amylase hydrolyzed it to 61.3%. Hydrolysis rates of these amylases on rice residue were lower than that of traditional Sikye. These results suggest that limit dextrin in commercial Sikhye is less effective than isomaltooligosaccharides in traditional Sikhye as a growth factor for Bifidobacterium while rice residue in commercial Sikhye is more effective than that in traditional Sikhye as dietary fiber.ary fiber.
Sikye was produced from glutinous rice. The glutinous rice Sikhye was found to contain 7.3% of limit dextrin, 10.1% of maltose, 1.3% of maltotriose and 1.75% of rice residue. Limit dextrin in glutinous rice Sikhye was purified by ethanol fractionation followed by gel chromatography on Biogel P-2. The purified limit dextrin showed both signal of $\alpha$-1,4- and $\alpha$-1,6-glucosidic linkage with its estimation ratio of 5:1 by 1H-NMR analysis. Limit dextrin was digested with enzymes(30units/ml) of $\alpha$-amylase, $\alpha$-glucosidase and glucoamylase from Aspergillus awamori, sweet potato $\beta$-amylase and human salivary $\alpha$-amylase at 37$^{\circ}C$ for 1 hour, respectively. The results suggest that glutinous rice is more effective to produce high level of branched maltooligosaccharide compared with rice as raw material for Sikye making.
Cyclodextrin hydrolase from Bacillus stearothermophilus KFCC 21203 was purified and the properties of the purified enzyme were investigated. The enzyme was purified 15 folds with 77 % recovery by ammonium sulfate fractionation, DEAE-cellulose chromatography, and Ultro AcA 34 gel filtration. The specific activity and the molecular weight of the enzyme were 1.30 units/mg protein and about 29,500, respectively, The maximum activity of the enzyme was shown at $55^{\circ}C$ and pH 5.5. However, stable temperature and pH were $40^{\circ}C$ and $5.0{\sim}8.0$, respectively. The Km value for ${\gamma}-cyclodextrin$ was $3.78{\times}10^{-3}$ M. The degradation activity of the enzyme was selectively high for ${\gamma}-cyclodextrin$, and very low for ${\beta}-cyclodextrin$, but not for ${\alpha}-cyclodextrin$. The decomposed products of ${\gamma}-cyclodextrin$ were mainly glucose and maltose, and a little mlatotriose. The activity of the enzyme was very high for amylose, potato starch, corn starch, amylopectin and maltooligomer, and relatively high for glycogen and dextrin. The decomposed products of them were mainly glucose and maltose.
Octenyl succinylated (OSA) potato starch was dextrinized by two methods: ultrasound (at 25, 50, or $70^{\circ}C$ for 1 h; OSA-25UT, OSA-50UT, and OSA-70UT, respectively) and acid hydrolysis (for 1 or 4 h; OSA-AD1H or OSA-AD4H, respectively), and the properties of the resulting starch were analyzed. The melting enthalpy of OSA-70UT decreased the most (from 14.0 to 10.0 mJ/mg), indicating chain degradation. For pasting properties, as ultrasound treatment temperature increased, peak viscosity decreased (2884, 2550, and 1888 cP, respectively), whereas acid hydrolysis increased peak viscosity and decreased pasting temperature. The relative crystallinity of OSA-dextrin produced by ultrasound or acid hydrolysis significantly decreased (from 33.61 to 14.90-26.03 and 19.28-20.05, respectively) as temperature or time increased, yet a B-type crystal pattern was maintained. Regarding emulsifying stability and sensory tests of mayonnaise prepared with OSA potato dextrin, mayonnaise with OSA-70UT was stable for short storage period (1 week), however mayonnaise with OSA-AD1H was the most suitable for long storage periods (from 2 to 4 weeks). In addition, the OSA-70UT was the most acceptable for mayonnaise in the sensory test.
Pigment concentrates with violet-red color and sweet taste were obtained from purple-fleshed sweet potato(PFSP) using ethyl alcohol and water. Extract from general potato(GP) were used as a control. The relative stability of PFSP pigment concentrate(PFSPPC) in a storage test over 15 days was confirmed in the order of dark > fluorescence > sun-light irradiation. The relative stability of GP pigment concentrate(GPPC) in a storage test over 15 days was confirmed in the order of sun-light > fluorescence > dark storage. The RRP of PFSPPC was higher than that of GPPC, but the color strength of GPPC was 1/2 that of PFSPPC. Treatment of PFSPPC with aluminum potassium sulfate(0.2~0.3%, w/w) best improved its stability. The improved RRPs of PFSPPC were 45.16~47.31% in sun light irradiation, 55.91~60.22% in fluorescence irradiation, and 76.34~75.97% in dark storage conditions. In substituting aluminum potassium sulfate for chitosan, an amount of 0.2~0.3%(w/w) was suitable, giving similar results in improving pigment stability for all concentrates tested. Also, freeze-dried PFSPPC powder was manufactured as a substitute for dextrin, and also as a substitute for chitosan to the extent of 0.25%(w/w). The results of storage stabilite for freeze-dried PFSPPC and GPPC powder over 15 days, irradiation were, PRRs of 74.47~89.36% and 61.54~76.92%, respectively. The stability improving effect of freeze dried PFSPPC powder was confirmed by the results of storage experiments at various conditions. The use of freeze-dried PFSPPC powder was therefore confirmed to be an effective treatment for general foods.

The present study was undertaken to evaluate the effect of indigestible dextrin on bowel function and serum lipid. Recently developed indigestible dextrin is water soluble dietary fiber obtained by heating and enzyme-treatment of potato starch with low viscosity. SD male rats weighing 80~85g were divided into three groups; control diet(IC), control +5% pectin(IP), control +5% indigestible dextin(ID). Experimental rats were fed one of the experimental diets ad libitum for six weeks. Weight gain, food intake, and food efficiency ratio were not significantly different among other groups. Those of ID tended to be higher than in IP and IC. Weights of liver, spleen and kidney were not significantly different among groups. Epididymal fat pads weight in W and ID tended to be lower than in IC. The weight of cecum was sinificantly higher in m than in other groups(p<0.05). The weight and length of large intestine tended to be higher in IP and ID than in IC. Total feces excretion of ID and If tended to be more than that of IC. Transit time was shorter in IP than in other groups. Serum total lipid, total cholesterol and TG levels were higher in IC group than other groups. The fecal excretion of lipid was significantly higher in IP than in other groups(p<0.05). That of ID tended to be more than that of IC, so fat apparent absorption of ID was lower than that of IC. Ca absorption were decreased more in IP than in other groups(p<0.05).
A factorial ($4{\times}2{\times}2$) experimental design was employed to determine apparent digestibilities of dry matter (DM), protein, lipid, energy and nitrogen-free extract (NFE) of the test diets containing either ${\alpha}$-potato starch (A-PS), ${\beta}$-potato starch (B-PS), ${\beta}$-corn starch (B-CS) or dextrin (DEX) as dietary carbohydrate energy for juvenile (average weight 30 g) and grower (average weight 300 g) rockfish reared at 13$^{\circ}C$ and 20$^{\circ}C$. Chromic oxide was used as an inert marker. Feces were collected by fecal collectors attached to rearing tanks from triplicate groups of juvenile and grower rockfish. Digestibilities of DM, energy and NFE of the test diets were significantly affected by dietary carbohydrate and water temperature (p<0.01), but not by fish size. DM digestibility of the fish fed the A-PS diet was significantly higher than that of fish fed other treatments, except for the DEX diet at 20$^{\circ}C$. DM digestibility of rockfish fed the B-CS diet was significantly lower than that of other diets. A similar pattern was observed in apparent digestibility of energy. NFE digestibility of fish fed the test diets was significantly affected by carbohydrate and significantly correlated to DM (r = 0.97, p<0.01) and energy (r = 0.99, p<0.01) digestibilities, regardless of water temperature and fish size. NFE digestibility of the fish fed the ${\beta}$-starch diets was relatively lower compared to that of the ${\alpha}$-starch diets, and ranged from 35 to 43% and 20 to 27% for B-PS and B-CS, respectively. The present findings indicate that carbohydrate and water temperature significantly affected digestibilities of dry matter, energy and nitrogen-free extract of rockfish. Among dietary carbohydrates, ${\alpha}$-potato starch could be effectively used as dietary carbohydrate energy for rockfish at 13$^{\circ}C$ and 20$^{\circ}C$.
