Three Possible Mechanisms for Stomatal Opening in Response to Light

  • Environmental factors such as light and low $CO_2$ concentrations trigger events which may result in stomatal opening. Stomatal aperature is largely controlled by the solute contents of guard cells, but not exclusively, by through changes in their content of potassium salts, with $K^+$ balanced either by $Cl^-$ or malate, depending on the species and conditions. However, how these signals are sensed and how they are transduced into driving the ion fluxes that control stomatal movements is not still fully understood. The basic role of stomata is regulating transpiration and photosynthesis. Photosynthesis plays a central role in the physiology of plants and an understanding of its response to light is, therefore, critical to any discussion of how plants sense and respond to light. It had been proposed that the evidences pointed three possible mechanisms for the light response. Firstly, there is a direct response of stomata to light. Secondly. there is an indirect response of stomata to light through the effect of $CO_2$. Lastly, there are some evidences for a third effect of light on stomata. However, attempts to investigate how these three possible mechanisms explained in detail in response to light have not been made. Therefore, this study is examined the differences among these three possible mechanisms.

Formation Constants of 18-Crown-6 Complexes with Substituted Potassium Phenoxides (置換 Potassium phenoxide와 이루는 18-Crown-6 Complex의 形成常數)

  • Recently, the formation of the complexes between macrocyclic polyethers and alkali metal salts have been determined by several methods. It has been suggested that the specificity of the complexation be due to the nature of the surrounding solvent molecules. The formation constant values ($K_f$) of $K^+$ are shown to be the largest among the other alkali metal cations because the ionic diameter of $K^+$ is approximately the same with the hole size of 18-crown-6. In this study the formation constants of the 1 : 1 complexes of 18-crown-6 with potassium p-substituted phenoxide are calculated by the conductance measurement in organic solvents. As a result, the $K_f$ value series among organic solvents are given in the order of $CH_3$OH > DMF > DMSO. It seems that the donor number of the solvent is a main factor in the formation of the complex between $K^+$ metal ion and 18-crown-6 molecules. At the same time, the formation constants increase with increasing the electron-withdrawing power of substituents because the phenoxide ion is stabilized by the charge dispersion.

Evaluation of the Color-change and Stability of Hoecheong (Smalt) Pigments When Exposed to Airborne Environmental Pollutants (회청 안료의 보존 환경에 따른 안정성 평가)

  • Recently, as the climate changes rapidly and the prevalence of airborne fine particulate matter increases, the pattern of pollutants in the atmospheric environment is also changing. Therefore, the importance of studying the stability of pigments used in colored cultural properties is emerging. Hoecheong is an inorganic blue glass pigment called smalt; it is made by using cobalt as a coloring element in potash glass, and was widely used in colored cultural assets, such as murals and paintings. In this study, we collected three other hoecheong pigments to analyze their properties. The percentage of Co and K contained are different according to the manufacturer, and the smalt-3 sample has a lower cobalt content (15.1 wt.%) and higher potassium content (29.6 wt.%). After this analysis, colored specimens were prepared. Prepared specimens were exposed to ultra-violet rays, CO2/NO2, and NaCl, which are known to have the greatest influence on the stability of pigments. We found that factors affecting the color stability were NO2 gas, ultra-violet rays, and water-soluble salts (NaCl). Among them, NO2 has the most severe impact on color change of the pigments. Results of the component analysis showed that the color change depends on the potassium and cobalt content of the hoecheong pigment. Among the specimens, smalt-3 showed the most vulnerability after exposure to NO2 gas and water-soluble salts. Pigment film stability is affected by watersoluble salts, giving rise not only to color change, but also weakening the physical properties of the film. However, there was no significant change in composition and color after exposure to CO2 gas. In conclusion, we found that hoecheong pigments underwent color change and increased instability of the coating film when exposed to any of the atmospheric environmental factors used in this study, except for CO2.

Physicochemical Properties and Microbial Analysis of Korean Solar Salt and Flower of Salt (한국산 꽃소금과 천일염의 이화학적 특성 및 미생물 분석)

  • The present study was conducted to ensure the diversity of domestic solar salt by analyzing the composition and microbiological characteristics of solar salt (from Docho island: DS) and the flower of salt produced in different Korean salt flats (Sinui island: SF, Bigum island: BF, and Docho island: DF). The analyses showed that the moisture content of the three types of flower of salt and solar salt ranged from 10.54~13.82% and NaCl content ranged from 78.81~84.61%. The mineral content of those salts ranged from 3.57~5.51%. The content of insoluble matter in these salts was $0.01{\pm}0.00{\sim}0.05{\pm}0.00%$. The sand content of these salts was $0.01{\pm}0.01{\sim}0.03{\pm}0.01%$. By Hunter's color value analysis, the color of the flower of salt was brighter and whiter than solar salt. The salinity of the flower of salt was a little higher than solar salt as well. The magnesium and potassium ion content of DF was $9,886.72{\pm}104.78mg/kg$ and $2,975.23{\pm}79.73mg/kg$, respectively, which was lower than the content in SF, BF, and DS. The heavy metal content of all salts was acceptable under the Korean Food Sanitation Law. The flower of salt was confirmed to be sweeter and preferable to solar salt. More than 80% of the solar salt crystals were 2~3 mm in size, whereas crystals from the flower of salt were 0.5~2 mm in size. The bacterial diversity of DF and DS were investigated by culture and denaturing gradient gel electrophoresis (DGGE) methods. The number of cultured bacteria in flower of salt was approximately three times more than solar salt. By DGGE analysis, major microbes of DF were Maritimibacter sp., Cupriavidus sp., and unculturable bacteria, and those of DS were Cupriavidus sp., Dunalidella salina and unculturable bacteria. The results of DGGE analysis showed that major microorganisms in solar salts were composed of unidentified and unculturable bacteria and only a few microorganisms were culturable.

Effect of Several Kinds of Composts on Growth Status of Aerial Parts in Ginseng Seedlings (유기질비료가 묘삼 지상부 생육에 미치는 영향)

  • This experiment were conducted to select cheaper and more favorable compost among several kinds of composts which were produced by companies, farmers etc. instead of Yakto. Yakto was similar to Standard Yakto in the content of inorganic salts, but PL and HJ composts contained more P$_2$O$\_$5/, K$_2$O, CaO and MgO content compared with Standard Yakto and EC in Yakto was also higher than Standard Yakto did, but the phosphorus, magnesium content and EC of Mushroom compost was higher than that of Standard Yakto. On the other hands, Rice straw compost contained higher contents of phosphorus, potassium and magnesium than Yakto did. Germination rate and early growth of lettuce and C/N ratio were tested for checking decomposed degree. The compost of HJ, Mushroom and Rice straw revealed higher C/N ratio, lower germination rate and shorter shoot length than Yakto showed. It was guessed that these composts were not decomposed enough. Emergence and survival rate of ginseng seeds in Yakto were the highest, but its germination in the composts of Mushroom, PL, HJ and Rice straw, in order, was decreased, and its survival rate in the composts of PL, Mushroom, HJ and Rice straw, in order, was decreased. It was concluded that fully decomposed manure should be used and it needs to Investigate perfect decomposed composts of PL and Mushroom instead of Yakto.

Characterization and Anti-Gastric Ulcer Activity of Bamboo Salt (죽염의 특성 분석과 항위궤양효과)

  • Bamboo salt has been used as a traditional remedy for gastric ulcer and gastro-intestinal disorders. It is produced by baking the salt packed in bamboo cylinder nine times under the fire of pine tree. Three of commercially available bamboo salt products (bamboo salt A, B, and C) were characterized by qualitative and quantitative analyses using inductively coupled plasma (ICP) spectrometer, ion chromatograph (IC), X-ray diffractometer (XRD), and electron microscope (EM). Compared with crude salt, the contents of iron, silicon, potassium, and phosphate in the bamboo salt products were higher whereas the sulfate content was lower. Water-insoluble fraction of bamboo salts contained the following compounds; MgO, $SiO_2,\;Mg_2Si0_4,\;and\;CaMgSi0_4$. The study on the microscopic structures of the bamboo salts were shown to have smooth surface and fused shape compared with crude salt. Among the three bamboo salt products, product A was used to test a possible inhibitory effect on gastric acid secretion. Each test material (bamboo salt A, crude salt, and reagent-grade NaCl) was given orally to Sprague-Dawley rats at doses of 0.2, 1.0, and 2.0 g/kg for 28 days before pyrolus ligation. Twenty four hours after the last administration of the test materials, volume, pH, total acidity, and pepsin activity of gastric juice were measured by the Shay-ligation method. No significant differences were observed in the secretion of gastric acid between treated groups (bamboo salt-, crude salt- and reagent-grade NaCI-treated groups) and control group (distilled water-treated group). This result demonstrated that bamboo salt did not exert anti-ulcer activities in experimental animals used in the present study.

Korean solar salts reduce obesity and alter its related markers in diet-induced obese mice

  • BACKGROUND/OBJECTIVE: The aim of this experiments was to show anti-obesity effects of Korean solar salt from different salt fields in diet-induced obese mice. MATERIALS/METHODS: Diet-induced obesity (DIO) was induced by a high-fat diet (HFD; 45% cal from fat) in C57BL/6J mice for eight weeks. The mice were fed with the designated diets (chow diet for Normal, HFD for Control, 0.47%-salt-mixed HFD for purified salt (PS), Guerande solar salt from France (SS-G), solar salt from Y salt field (SS-Y), solar salts from T salt field (SS-T) and S salt field (SS-S)) for another eight weeks. We checked body weight, food efficiency ratio (FER) and tissue weights (liver and epididymal adipose tissue (EAT)), and observed serum concentrations of triacylglycerol (TG), total cholesterol (TC), leptin and insulin. We also evaluated gene expressions of adipogenic / lipogenic mRNAs of $C/EBP{\alpha}$, $PPAR{\gamma}$ and FAS and beta-oxidation-related factors ($PPAR{\alpha}$ and CPT-1) in liver and EAT. The mineral composition of salt samples were analyzed using inductively coupled plasma optical emission spectrometry (ICP-OES). RESULTS: SS-T and SS-S significantly reduced body weight gain, FER, and weight of EAT compared to control and other samples (P < 0.05). SS-T and SS-S also significantly decreased serum levels of TG, TC, leptin and insulin (P < 0.05). SS-T and SS-S suppressed expressions of adipogenic / lipogenic mRNAs in liver and EAT, while promoting expression of beta-oxidation-related factors. The lowest sodium concentration was observed in SS-T ($30.30{\pm}0.59%$), and the lowest sodium-to-potassium (Na/K) ratio was found in SS-S (17.81). CONCLUSIONS: Our study shows that well-processed Korean solar salt may have anti-obesity effects in vivo, probably owing to its differences in mineral composition and other components, presumably resulting from the manufacturing processes. Further research is needed into the mechanism and to explore optimal manufacturing processes.

Soil Environmental Investigation of Plastic Film House in Chonnam Area (전남지역 시설재배 토양환경 실태조사)

  • The investigation and the analysis of soil chemical properties, microbial flora and cultural practices in plastic house of 60 farms (pepper 20, cucumber 20, oriental melon 20) were conducted in Chonnam area film. An amount of fertilizer application of farms was more than that of standard fertilizer application. That was phosphate at two and a half times, potassium at twice and manure at three times the amount of standard application. It was inferred that excessive fertilizer application would lead to salts accumulation in soil. Continuous cropping injury would be incurred by excesive salts accumulation in soil. Even if inorganic elements in soil culturing peppers, cucumbers and oriental melons were greater than optimum level, base-saturation degree of soil culturing peppers, cucumbers and oriental melons was 40:18:12, 55:16:14 and 40:27:11 respectively. That was lower than the optimum levels (60:15:5). Therefore, crops would be affected by uptake inhibition by nutrition imbalance. Density of Fusarium is related to plant decease in soil i.e. pepper $26.2{\times}10^2\;cfu\;g^{-1}$, cucumber $75.8{\times}10^2\;cfu\;g^{-1}$ and oriental melon $6.5{\times}10^2\;cfu\;g^{-1}$. B/F of soil planted with pepper, cucumber and oriental melon was 91.2, 80.4 and 18.8, respectively.

Synthesis and Characterization of Pyridinium Dinitramide Salt (피리디니움 디나이트라아마이드염의 합성과 특성연구)

  • A new solid oxidizer, pyridinium dinitramide (Py-DN) is a low toxic energetic material which can be utilized as a HPGP (high performance green propellant). In this work, Py-DN was synthesized using various starting materials including potassium sulfamate, pyridine hydrochloride, strong nitric acid and sulfuric acid. Physical and chemical properties of the Py-DN were characterized using UV-Vis, FT-IR and a thermal analyzer and their properties were compared to those of previously prepared salts including ammonium dinitramide[ADN, $NH_4N(NO_2)_2$] and guanidine dinitramide[GDN, $NH_2C(NH_2)NH_2N(NO_2)_2$] in our lab. Endothermic and exothermic decomposition temperatures of Py-DN were $77.4^{\circ}C$ and $144.7^{\circ}C$, respectively. The combustion caloric value was 1739 J/g, which is thermally more sensitive than that of conventional dinitramides. It may enable to lower the decomposition temperature, which can reduce preheating temperature required for satellite thruster applications.

Study of the Effects of Potassium Chloride and Calcium Chloride on the Development of Drosophila melanogaster (염화칼륨과 염화칼슘의 초파리 발생에 미치는 영향에 대하여)

  • In order to see the single effect and interaction of potassium chloride(KCI) and calcium the chloride ($CaCl_2$) on the development of Drosophila melanogaster, the emergence rates of the five strains of the flies were examined from the media containing 0.0M, 0.5M, 0.1M, 0.2M, of $CaCl_2$ or KCI plus various concentrations of KCI or $CaCl_2$, and the results are presented as follows: 1. The emergence rate of D. melanogaster is not significantly different among strains but strikingly different among concentrations of KCI or $CaCl_2$; the emergence rate decreases as concentration of KCI or $CaCl_2$ increases. 2. The inhibitory effect of $CaCl_2$ on the emergence rates is greater than that of KCI, implying that the resistibility of the flies to KCI is greater than to $CaCl_2$. 3. The addition of small amount of $CaCl_2$ or KCI to KCI or $CaCl_2$ media counteracts the inhibitory effect of KCI or $CaCl_2$, but the addition of a little greater amounts of $CaCl_2$ or KCI to the KCI or $CaCl_2$ media enhance the inhibitory effect of the salts. 4. Thus, there may be some interaction of $K^+$ and $Ca^++$ ions in the inhibitory effect on the development(emergence rates) of Drosophila melanogaster.
