Survey on the Changes of Potassium and Magnesium Concentration according to the Retention Time After Rinse of Sucrose and Glucose in Oral (구강 내 자당 및 포도당 잔류 경과 시간에 따른 칼륨과 마그네슘 농도 변화)

  • The purpose of this study was to evaluate the salivary concentration of Potassium and Magnesium cations and their variation before and after sucrose and glucose rinse, and to investigate the relationship between the levels of each compound. Saliva samples were obtained from 40 subjects before and up to 60 min after intake of a 10% sucrose and glucose solution at 1-month intervals. Potassium and Magnesium in human saliva were determined via anion-exchange chromatography with an anion-suppressed conductivity detector using 12 mM sulfuric acid. The concentrations of Potassium and Magnesium before sucrose rinse were 274.3±77.9 mg/ℓ and 4.5±2.5 mg/ℓ, also, the concentrations of Potassium and Magnesium before glucose rinse were 279.2±62.1 mg/ and 4.8±2.0 mg/ℓ, respectively. Potassium and Magnesium concentrations were significantly decreased (p<0.05) after sucrose rinse. The content of potassium and magnesium in saliva before and after rinsing sucrose and glucose is difficult to standardize or classify, as previous research. The reason for the variation between individuals is large, and easily changed by chemical or physiological stimulation. However, this study was experiment for the purpose of accumulating basic data for saliva.

  • Penicillin sensor was manufactured by immobillizing penicillinase with glutaraldehyde on the $H^+$-selective membrane based on PVC-COOH-TDDA. This membrane was not inter fered by $K^+$ ion in Pc-G potassium salt. When enzyme was immobilized with glutaraldehyde, the PVC-COOH matrix was more effective than PVC matrix. Calibration curve calculated from Nernst equation was not linear. But potential was relative to concentration of Pc-G. And maximal potentiometric velocity was also relative to concentration of Pc-G. Therefore, it may be applied to Michaelis-Menten equation. The penicillin sensor was useful for determination of Pc-G at concentration of 0.1~10mM level.

  • The reaction of phenylhydrazine with bromine in sulfuric acid solution has been studied kinetically. The pseudo-second-order rate constant is approximately inversely proportional to hydrogen-ion concentration when the concentration of sulfuric acid is lower than 1M. arom the study of the effect of potassium bromide concentration on the rate constant, it is concluded that both neutral bromine and tribromide ion participate in the reaction, the rate constants in 0.01M $H_2SO_4$ being $5{\times}10^5M^{-1},sec^{-1}\;and\;0. 7{\times}10^5M^{-1},sec^{-1}$, respectively at $20^{\circ}C$. The pseudo-second-order rate constant of 2.4-dinitrophenylhydrazine-bromine reaction is independent of hydrogen ion concentration. From the KBr addition experiment, the rate constants for $Br_2\;and\;Br_3^-$ were obtained as $1.2{\times}10^5M^{-1},sec^{-1}\;and\;2.0{\times}10^4M^{-1},sec^{-1}$, respectively.

Characteristics of Inorganic Ion Absorption of Strawberries Cultivated in Closed Hydroponic System with Different Substrates (고형배지를 이용한 순환식 딸기 수경재배에서 배지 종류별 무기이온 흡수 특성)

  • This experiment was carried out to understand the characteristics of inorganic ion uptake of strawberries to establish a labor saving, environmentally sound closed hydroponic system. Nitrate nitrogen is absorbed in the perlite and granule rockwood with the almost same rate and in the cocopeat with a little bit higher concentration. At the early growth stage, phosphate was absorbed in comparatively high rate, but it is gradually declining to $4me{\cdot}L^{-1}$ in the three treatments except for reused medium. Four treatments showed different absorption rates for potassium, respectively, however, it became $3{\sim}5me{\cdot}L^{-1}$ gradually. But there was few sign to absorb potassium in the reused medium. High concentration of calcium was absorbed in the cocopeat medium and the lowest absorption rate in the granule rockwool, and it turned out to be $2{\sim}3me{\cdot}L^{-1}$ absorption concentration rate in the perlite. All three treatments showed $1me{\cdot}L^{-1}$ at the average absorption concentration rate of magnesium. Iron showed high absorption rate but it showed steadily 1~2 ppm absorption rate. Results of this experiment will be utilized for developing the new substrates for strawberry closed hydroponic system.

Comparative Study of Holmium (III) Selective Sensors Based on Thiacalixarene and Calixarene Derivatives as an Ionophore

  • The two chelates based on calix[4]arene and thiacalix[4]arene have been synthesized and used as neutral ionophores for preparing PVC based membrane sensor selective to $Ho^{3+}$ ion. The addition of potassium tetrakis(4-chlorophenyl)borate (KTpClPB) and various plasticizers, viz., NDPE, o-NPOE, DOP, TEP and DOS have been found to improve significantly the performance of the sensors. The best performance was obtained with the sensor no. 6 having membrane of $L_2$ with composition (w/w) ionophore (2%): KTpClPB (4%): PVC (37%): NDPE (57%). This sensor exhibits Nernatian response with slope $21.10{\pm}0.3mV/decade$ of activity in the concentration range $3.0{\times}10^{-8}-1.0{\times}10^{-2}M\;Ho^{3+}\;ion$, with a detection limit of $1.0{\times}10^{-8}M$. The proposed sensor performs satisfactorily over a wide pH range of 2.8-10, with a fast response time (5 s). The sensor was also found to work successfully in partially non-aqueous media up to 25% (v/v) content of methanol, ethanol and acetonitrile, and can be used for a period of 4 months without any significant drift in potential. The electrode was also used for the determination of $Ho^{3+}$ ions in synthetic mixtures of different ions and the determination of the arsenate ion in different water samples.

Changes in Haemolymph Proteins, Hydrolases, and Inorganic tons of Heliothis assulta Injected with Bacillus thuringiensis (Bacillus thuringiensis의 주입에 따른 담배나방의 혈림프 단백질, 가수분해효소 및 무기이온의 변화)

  • Changes in haemolymph proteins, hydrolases such as esterase(EST), acid phosphatase(ACP) and alkaline phosphatase(ALP) , and inorganic ion(Na+, K+ and Cl- ) contents were induced by the injection of Bacillus thuringiengis into haemocoel of the last instar larva of Heliothis assulta. Protein concentration of haemolymph was increased until 24 hrs after injection, and decreased thereafter. Among the 8 basic protein bands identified through acid - polyacrylamide gel electrophoresis(PAGE), 2 bands(bands a and b) became stronger by the bacterial infection. Activities of EST and ALP increased until 12 hrs after injection and then fell down, whereas ACP activity was decreased continuously with time after injection. Contents of inorganic ions were all increased by the bacterial injection, showing slow rate of increase in the chloride ion, but rapid in the sodium and potassium ions.

Acepromazine inhibits hERG potassium ion channels expressed in human embryonic kidney 293 cells

  • The effects of acepromazine on human ether-$\grave{a}$-go-go-related gene (hERG) potassium channels were investigated using whole-cell voltage-clamp technique in human embryonic kidney (HEK293) cells transfected with hERG. The hERG currents were recorded with or without acepromazine, and the steady-state and peak tail currents were analyzed for the evaluating the drug effects. Acepromazine inhibited the hERG currents in a concentration-dependent manner with an $IC_{50}$ value of $1.5{\mu}M$ and Hill coefficient of 1.1. Acepromazine blocked hERG currents in a voltage-dependent manner between -40 and +10 mV. Before and after application of acepromazine, the half activation potentials of hERG currents changed to hyperpolarizing direction. Acepromazine blocked both the steady-state hERG currents by depolarizing pulse and the peak tail currents by repolarizing pulse; however, the extent of blocking by acepromazine in the repolarizing pulse was more profound than that in the depolarizing pulse, indicating that acepromazine has a high affinity for the open state of the channels, with a relatively lower affinity for the closed state of hERG channels. A fast application of acepromazine during the tail currents inhibited the open state of hERG channels in a concentration-dependent. The steady-state inactivation of hERG currents shifted to the hyperpolarized direction by acepromazine. These results suggest that acepromazine inhibits the hERG channels probably by an open- and inactivated-channel blocking mechanism. Regarding to the fact that the hERG channels are the potential target of drug-induced long QT syndrome, our results suggest that acepromazine can possibly induce a cardiac arrhythmia through the inhibition of hERG channels.

EFFECTS OF POLYPHENOLS OF Cocos Nucifera HUSK FIBRE ON SELECTED KIDNEY FUNCTION INDICES IN MICE

  • Decoction of Cocos nucifera husk fibre is used indigenously in Nigeria for malaria treatment. Polyphenols have been identified as the phytochemicals responsible for the antimalarial activity of Cocos nucifera husk fibre, though their toxicity has not been evaluated. The polyphenols of Cocos nucifera husk fibre were therefore evaluated for their effects on selected kidney function indices in mice. Fifty mice were randomly divided into five groups (A-E) of ten mice each. Mice in group A were orally administered 5% DMSO solution while those in groups B, C, D and E were orally administered 31.25, 62.5, 125 and 250 mg/Kg body weight of the polyphenols respectively for seven days. Serum urea, creatinine and uric acid concentrations were determined. Serum levels of sodium, potassium, chloride and calcium ions and kidney alkaline phosphatase (ALP), glutamate dehydrogenase (GDH) and gamma-glutamyltransferase (GGT) activities were also determined. The results showed that the polyphenols significantly reduced (p<0.05) urea concentration at 250 mg/Kg body weight and creatinine concentration at all doses compared to controls. The polyphenols caused no significant alteration (p>0.05) in serum uric acid concentration and kidney ALP, GGT and GDH activities compared to controls. There was significant increase (p<0.05) in serum sodium ion concentration at 31.25, 125 and 250 mg/Kg body weight of polyphenols whereas significant increase (p<0.05) in serum potassium and chloride ions was observed at 62.5 and 250 mg/Kg body weight compared to controls. Thus, polyphenols of Cocos nucifera husk fibre may adversely affect some osmoregulatory functions of the kidney, especially at higher concentrations.

  • The removal of calcium ions using zeolite to solve problems of the CaCO3 manufacturing process using cement kiln dust was investigated. To do so, a modified zeolite was employed and experiments were conducted to select the optimal zeolite type considered the binding cation and structure, evaluate the removal performance of calcium ions, the influence of the type and concentration of the modifying solution, and the removal selectivity when K coexists. Among five zeolites, 13X zeolite was found to have the best calcium ion removal performance, and it was confirmed that the removal performance was enhanced when KCl was used as a modifying solution instead of NaCl. This study is expected to be the basis for the solution of carbonation process and high concentration of KCl recovery technology.

Aspects of Nutrient Transportation after Animal Manure Application in Jeju Field Soil (제주 밭토양에서 가축분 퇴비의 시용에 따른 양분의 이동양상)

  • Recently, saw dust manure has been widely used in horticultural crop production in Korea. Animal manure is produced by decaying of livestock manure mixed with saw dust, and contained higher nutrients and ion concentration than the traditional manure made from rice straw and grasses. Therefore, a continuous application of the animal manure disregarding to soil characteristics may be ruined the soil conditions. This study was conducted to investigate the transportation of the nitrogen, phosphate, potassium, and ion concentration of the animal manure applied to volcanic and non-volcanic soils in Jeju islana Soil chemical analysis were done before and 180 days after animal manure application. After animal manure application, $NO_3$-N moved up to 90cm in volcanic soil, while the movement was limited to 60 cm in non-volcanic soil. Phosphate concentration was high up to 30 cm, where crop roots are mainly distributed, in volcanic soil, however, the phosphate moved up to 60 cm in volcanic soil. Exchangeable potassium moved up to 90 cm in volcanic soil, but the movement wns limited up to 60 cm in non-volcanic soil. For both soil types, no significant different in ion concentrations was observed up to 60 cm in soil depth, though the concentrations were higher in volcanic ash soils as compared to the non-volcanic ash soils.
