벼의 무기양분 흡수에 미치는 pH 의 영향 (The Effect of pH on the Mineral Nutrient Uptake in the Rice Seedlings)

  • Absorption pattern of potassium, calcium, phosphate and nitrate ions, and the pH change during ion-absorption at pH 3.0-11.0 by Oryza sativa L. were studied to investigate indirectly the evidence of H^+-efflux by ATPase. The rice seedlings which were grown either in $L^{\circ} -dark or in L^+-sunlight$, were used both in each ion-absorption to compare with one another. The uptake rate of these ions appeared to favor more in $L^+than in L^{\circ}$, over all range in pH, nearly with the same pattern. The absorption of potassium resulted in bell shape and that of calcium increased linearly to the alkali range. The shape of phosphate-absorption showed nearly the t-distribution curve with high value in acid range and the uptake of nitrate resulted in the dual peaks, but higher in acid range. The pH of the external solution changed from the range of 3.5-11.0 to 3.5-7.0 after lhr-absorptioin, and further acidified after 3hr-absorption. It is suggested that the pH change of the external solution be affected by $ H^+-efflux$ which may be caused by the ATP-hydrolysis.

세포막 활동전압에서 음양(陰陽)의 상호관계 (Interrelation of Yin and Yang in Action Potential of Cell Membrane)

  • This study was undertaken to apply the yin-yang theory in action potential. In order to apply the yin-yang theory in action potential, nature of yin and yang, interrelation of yin and yang and action potential in cell were reviewed. According to the yin-yang theory, inner cellular space corresponds to yin, but outer cellular space corresponds to yang. If we classify ions in intracellular fluid or extracellular fluid by nature of yin and yang, potassium(K+) corresponds to yang within yin(陰中之陽), protein(Pr-) corresponds to yin within yin(陰中之陰) in intracellular fluid, and sodium(Na+) corresponds to yang within yang(陽中之陽), chloride(Cl-) corresponds to yin within yang(陽中之陰) in extracellular fluid. Double donnan equilibrium and equilibrium potential were caused by intracellular anion(Pr-) and extracellular cation(Na+) are related with mutual rooting of yin and yang(陰陽互根) and opposition of yin and yang(陰陽對立). The influx and efflux of ion through cell membrane means waxing and waning of yin and yang(陰陽消長), the change of membrane potential means yin-yang conversion(陰陽轉化) during action potential.

Purification and Characterization of Phocaecin PI80: An Anti-Listerial Bacteriocin Produced by Streptococcus phocae PI80 Isolated from the Gut of Peneaus indicus (Indian White Shrimp)

  • A bacteriocin-producing strain PI80 was isolated from the gut of Penaeus indicus (Indian white shrimp) and identified as Streptococcus phocae PI80. The bacteriocin was purified from a culture supernatant to homogeneity as confirmed by Tricine SDS-PAGE. Reverse-phase HPLC analysis revealed a single active fraction eluted at 12.94 min, and MALDI-TOF mass spectrometry analysis showed the molecular mass to be 9.244 kDa. This molecular mass does not correspond to previously described streptococcal bacteriocins. The purified bacteriocin was named phocaecin PI80 from its producer strain, as this is the first report of bacteriocin production by Streptococcus phocae. The bacteriocin exhibited a broad spectrum of activity and inhibited important pathogens: Listeria monocytogenes, Vibrio parahaemolyticus, and V. fischeri. The antibacterial substance was also sensitive to proteolytic enzymes: trypsin, protease, pepsin, and chymotrypsin, yet insensitive to catalase, peroxidase, and diastase, confirming that the inhibition was due to a proteinaceous molecule (i.e., the bacteriocin), and not due to hydrogen peroxide or diacetyl. Phocaecin PI80 moderately tolerated heat treatment (up to $70^{\circ}C$ for 10 min) and resisted certain solvents (acetone, ethanol, and butanol). A massive leakage of $K^+$ ions from E. coli $DH5\alpha$, L. monocytogenes, and V. parahaemolyticus was induced by phocaecin PI80, as measured by Inductively Coupled Plasma Optical Emission Spectrometry (ICPOES). Therefore, the results of this study show that phocaecin PI80 may be a useful tool for inhibiting L. monocytogenes in seafood products that do not usually undergo adequate heat treatment, whereas the cells of Streptococcus phocae PI80 could be used to control vibriosis in shrimp farming.
