Characterization of Transgenic Tall Fescue Plants Overexpressing NDP Kinase Gene in Response to Cold Stress (NDP Kinase 유전자를 과발현시킨 형질전환 톨 페스큐 식물체의 저온 스트레스에 대한 내성 특성)

  • Oxidative stress is the main limiting factor in crop productivity. To solve global environmental problems using the plant biotechnology, we have developed on the oxidative stress-tolerant transgenic tall fescue plants via Agrobacterium-mediated genetic transformation method. In order to develop transgenic tall fescue (Festuca arundinacea Schreb.) plants with enhanced tolerance to multiple environmental stresses, nucleotide diphosphate kinase gene under the control of CaMV35S promoter were introduced into genome of tall fescue plants. Proteomic analysis revealed that transgenic tall fescue not only accumulated NDP kinase 2 protein in their cells, but also induced several other antioxindative enzyme-related proteins. When leaf discs of transgenic plants were subjected to cold stress, they showed approximately 30% less damage than wild-type plants. In addition, transgenic tall fescue plants showed normal growth when transgenic plants were subjected to $4^{\circ}C$ for 3 days treatments. These results suggest that transgene is important in ROS scavenging by induction of antioxidative proteins, and could improve abiotic stress tolerance in transgenic tall fescue plants.

Growth Characteristics and Economic Efficiency of Nursery Plants Production According to Transplanting Container for Acclimatization of Mass Propagated Plantlets via Bioreactor Culture of Ever-bearing Strawberry 'Goha' (생물반응기를 통해 대량증식된 사계성 딸기 '고하' 소식물체의 순화용기에 따른 생육특성과 묘생산 경제성)

  • This study was conducted to find out the optimum container for increasing acclimatization rate of in vitro mass propagated plantlets of Ever-bearing strawberry (Fragaria ${\times}$ ananassa Duch.) via bioreactor. Four types of containers were used such as transparent plastic container (TPC), plug tray (PT), I-pot (IP), and black vinyl pot (BVP). Number of date maintaining soil water content above 10% was five days in TPC, three to four days in BVP, two days in PT, and one day in IP. Survival rate of plantlets was 80% in BVP, 70% in TPC, 55% in IP, and 15% in PT. In TPC, growth increment of plantlets was the greatest among all the tested containers and the lowest in IP. Numbers of runner per plant were 3.3 in BVP, 2.9 in TPC, 1.6 in PT, and 1.2 in IP. Total cost was 44,405,300 won/10 a in BVP, resulting in reducing more 6,659,400 won/10 a than IP's (51,064,700 won/10 a). Around 102,718 plants/10 a were produced by using BVP, suggesting that 30,265.1 plants/10 a more could be produced than IP (72,452.9 plants/10 a). Production cost per plant was 432.3 won in BVP, resulting in reducing 272.5 won than IP's (704.8 won). As a result, BVP was appropriate for acclimatization of in vitro plantlets through bioreactor system.

  • Environmental stress is the major limiting factor in plant productivity. As an effort to solve the global food and environmental problems using the plant biotechnology, we have developed transgenic tall fescue (Festuca arundinacea Schreb.) plants via Agrobacterium-mediated gene transfer method. To develop transgenic tall fescue plants with enhanced tolerance to the environmental stresses, both CuZn superoxide dismutase (CuZnSOD) and ascorbate peroxidase (APX) genes were incorporated in a pIG121 binary vector and the both of the genes were controlled separately by an oxidative stress-inducible sweet potato peroxidase 2 (SWPA2) premoter expressed in chloroplasts. Leaf discs of transgenic plants showed 10-30% less damage compared to the wild-type when they exposed to a wide range of environmental stresses including methyl viologen (MV), $H_2O_2$ and heavy metals. In addition, when $200{\mu}M$ MV was sprayed onto the whole plants, transgenic plants showed a significant reduction of visible damage compared to wild-type plants that were almost damaged. These results suggest that over expression of CuZnSOD and APX genes in transgenic plants might be a useful strategy to protect the crops against a wide range of environmental stresses.

Effect ofMunicipal Sludge on contents of Cadmium and Zinc in Crop Plants (도시하수(都市下水) Sludge 의 경작지(耕作地) 처리(處理)가 식물체(植物體)중 Cd 및 Zn 함량(含量)에 끼치는 영향(影響))

  • Four crop plants, such as green onion(Allium ascalonicum L.), radish(Raphanus sativus L.), potato(Solanum tubersum L.) and chinese cabbage(Brassica pekinensis Rupr), were grown in greenhouses to determine the accumulations of Cd and Zn absorbed by the plants grown on soils treated with municipal sewage sludge of different levels of 0, 22.5, 45.0 and 90.0 ton $ha^{-1}$ as soil dry weight. The result was analyzed to be comparable to the heavy metal content in the plant tissues among the crop species and the statistical characteristics were investigated to determin a possible relationship between causes of increased contents of Cd and Zn in plants and in soils. The results were as follows : 1. The Cd and Zn content in experimental crop plants increased with increase in quantity of treatment sludge. 2. The Cd and Zn content in roots of all experimental plants increased with sludge treatment amount in soil, in the following order : green onion>potato>chinese cabbage>radish. 3. The Cd content in leaves of crop plants were high in this order : green onion>radish>potato>chinese cabbage, and the Zn content in leaves of crop plants was high in this order. : potato>radish>chinese cabbage>green onion. 4. Concentrations of Cd in potato tubers, radish roots and chinese cabbage leaves ranged from 0.03 to 0.12 ppm. 5. Concentrations of Cd and Zn in plant tissues grown in sludge treated soil of 90.0 ton $ha^{-1}$ were 1.2 to 10 times and 0.4 to 2.3 times higher as compared with those absorbed from sludge-free soil, respectively. 6. The accumulation rates of Cd and Zn in sludge-treated plant tissue were almost constant except for green onion roots.

Biological Control of Mulberry Root Knot Nematode Meloidogyne incognita by Trichoderma harzianum

  • Trichoderma harzianum-THN1 parasitising the egg masses of root knot nematode Meloidogyne incognita was isolated from galled mulberry roots and evaluated for its potential to control root knot disease. In pot experiments root galling was reduced and leaf yield increased significantly following soil treatment with T. harzianum-THN1. The extracts obtained from the soils inoculated with T. harzianum-THN1 drastically inhibited the hatching of nematode eggs and the effect was irreversible even after the eggs were transferred to fresh water. The fungus was equally effective in controlling the disease in nematode infested mulberry garden under field conditions which was significant over the most commonly used egg parasitic fungus Paecilomyces lilacinus. The disease reduction recorded with T. harzianum was on par with the plants treated with the nematicide Carbofuran. The results suggest that T. harzianum- THN1 could be used as a potent ecofriendly biocontrol agent against M. incognita in mulberry without any residual toxicity to silkworms. T. harzianum- THN1 can form an important component of integrated disease management package in mulberry cultivation.

Effect of Bradyrhizobium japonicum on Chlorophyll Content, Nodulation, and Plant Growth in Soybean

  • Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.

  • Twenty kinds of minerel nutrient solution were prepared and supplied to the tomatoes planted in Norwegian quartz pots. These plants were cultured for nine weeks and several physiogical phenomena were observed during the growing period, and after harvest, the alkaloid contents were determined. The highest growth potential was in NS 3 group while the highest alkaloid content wasein KMg 18 group.

  • For improvement of water quality, $20m^2$ of artificial floating plant islands planted with Iris pseudoacorus, were installed in small pond on March, 1999. Small pond has surface area $1,000m^2$ and mean depth 1.5 m. The density of plants was 16 per $m^2$ by using jute pot. Environmental parameters such as COD, SS, T-N, T-P and planktons were biweekly measured from 29 March to 28 September. Because of the small portion of floating island, the effect for water quality improvement was not sufficient. But considering the data of plant growth and nitrogen and phosphorus uptake capacity of plant, about 40% of coverage by artificial floating island was needed for elimination of whole nutrients from inflow.

Role of Siderophores in Biocontrol of Fusarium solani and Enhanced Growth Response of Bean by Pseudomonas fluorescens GL20

  • Plant growth-promoting Psudomonas fluorescens GL20 was isolated from a ginseng rhizosphere on chrome azurol Sagar. P. fluorescens GL20 produced a large amount of hydoxamate siderophore in an iron-deficient medium. The siderophore showed significantly high specific activity of 20.2 unit. Using an in vitro antifungal test, P. fluorescens GL20 considerably suppressed growth of phytopathogenic fungus Fusarium solani, inhibiting spore germination and germ tube elongation. In pot trials of kidney beans with P. fluorescens GL20, disease incidence was remarkably reduced up to $68{\%}$ compared with that of F. solani alone, and plant growth was also increased nearly 1.6 fold as compared to that of the untreated control, promoting elongation and development of the roots. These results indicate that the plant growth-promoting activity of P. fluorescens GL20 can play an important role in biological control of soil-borne plant disease in a rhizosphere, enhancing the growth of plants.

Serratia plymuthica Strain A2l-4: A Potential Biocontrol Agent Against Phytophthora Blight of Pepper

  • A promising biocontrol agent, A2l-4, against Phytophthora blight of pepper was selected from 351 bacterial isolates collected from rhizosphere soils and roots of onion (Allium fistulosum L.). The isolate A21-4 was identified as Serratia plymuthica based on its 16S rRNA sequence and key characteristics as compared with that of an authentic culture of S. plymuthica (ATCC No. 6109D01). The isolate readily colonized on roots of various crops including pepper when inoculated on seed and not. Strain A2l-4 showed narrow spectrum of antibiotic activity, as revealed in its strong inhibitory activity to the genera Pythium and Phytophthora, but not to Fuasrium and Rhizoctonia. In pot experiments, none of the pepper seedlings treated with A2l-4 were infected by Phytophthora capsici, while 86% of the control plants were killed by the pathogen.
