Anti-Asthmatic Effects of Portulaca Oleracea and its Constituents, a Review

  • Objectives: The medicinal plants are believed to enhance the natural resistance of the body to infections. Some of the main constituents of the plant and derived materials such as, proteins, lectins and polysaccharides have anti-inflammatory effects. Portulaca oleracea (P. oleracea) were used traditionally for dietary, food additive, spice and various medicinal purposes. This review article is focus on the anti-asthmatic effects of P. oleracea and its constituents. Methods: Various databases, such as the PubMed, Scopus, and Google Scholar, were searched the keywords including "Portulaca oleracea", "Quercetin", "Anti-inflammatory", "Antioxidant", "Cytokines", "Smooth muscle ", and " Relaxant effects " until the end of Jul 2018. Results: P. oleracea extracts and its constituents increased $IFN-{\gamma}$, IL-2, $IFN{\gamma}/IL-4$ and IL- 10/IL-4 ratio, but decreased secretion of $TNF-{\alpha}$, IL-4 and chemokines in both in vitro and in vivo studies. P. oleracea extracts and quercetin also signifcantly decreased production of NO, stimulated ${\beta}$-adrenoceptor and/or blocking muscarinic receptors in tracheal smooth muscles. Conclusion: P. oleracea extracts and quercetin showed relatively potent anti-asthmatic effects due to decreased production of NO, inflammatory cytokines and chemokines, reduced oxidant while enhanced antioxidant markers, and also showed potent relaxant effects on tracheal smooth muscles via stimulatory on ${\beta}$-adrenoceptor or/and blocking muscarinic receptors.

마치현 에틸아세테이트 분획물의 뇌세포 보호효과 (Neuroprotective Effect of Ethyl Acetate Fraction of Portulaca oleracea L.)

  • Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 마치현의 항산화 작용 (Anti-oxidant Effects of Portulaca oleracea L. on HCl-ethanol Induced Gastritis in Rats)

  • Objectives : The purpose of this study is to evaluate whether or not a pretreatment with Portulaca oleracea has an antioxidant effect in HCl-ethanol induced gastric mucosal damage. Methods : We elucidated the level of reactive oxygen species (ROS), lipid peroxidation, and two important constituents of antioxidant defense such as superoxide dismutase (SOD), glutathione (GSH) in these effects. Results : The oral administration of crude extract from P. oleracea attenuated the gastritic lesion area, submucosal edema and hemorrhage, and mucosal necrosis induced by HCl-ethanol. The MDA levels of control group were higher than those in the rats given the P. oleracea pretreatment. While the GSH levels of control were decreased, the GSH activity on the gastric mucosal layer maintain normal level in rats given the Portulaca oleracea pretreatment before HCl-ethanol induced gastritis significantly increased. However, the SOD activites were not altered by P. oleracea. Conclusions : The administration of Portulaca oleracea have a protective antioxidant effect against the gastric lesion induced by HCl-ethanol and may therefore be a promising drug for gastritis and gastric ulcer.

쇠비름 첨가 설기떡의 품질 특성 및 항산화성 (Antioxidant Activities and Quality Characteristics of Sulgidduk Added with Portulaca oleracea L.)

  • 본 연구는 항산화능이 우수한 것으로 알려진 쇠비름을 선택하여 쇠비름 페이스트를 만든 후 0, 1, 3, 5%를 첨가한 설기떡을 제조하여 품질 특성 및 항산화성을 분석하였다. 쇠비름 첨가량이 증가할수록 산도와 환원당이 증가하였다. 색도는 쇠비름 첨가량이 증가할수록 명도는 감소하고, 적색도와 황색도는 증가하였다. 총페놀 함량은 쇠비름 첨가량이 증가할 수록 증가하였다. DPPH 라디칼 소거능과 superoxide dismutase 유사 활성은 쇠비름 첨가량이 증가할수록 $IC_{50}$ 값과 $EC_{50}$ 값이 낮아져 항산화능이 높았다. 기호도는 쇠비름을 첨가할수록 외관의 기호도는 증가하지만 향미, 맛, 조직감, 전체적 기호도는 3% 쇠비름 첨가군이 가장 높게 나타났고, 5% 쇠비름 첨가군이 감소함을 보였다. 특히 전체적 기호도는 쇠비름 3% 첨가군이 가장 높은 점수를 받았다. 이상의 결과로 보아 쇠비름 첨가가 떡의 항산화능과 기호도를 증가시켜 설기떡의 가치를 높일 수 있을 것으로 보인다. 이 연구를 통해 쇠비름을 첨가한 설기떡의 항산화능이 우수한 것을 볼 수 있었다. 또한, 쇠비름의 향미와 맛을 고려하여 볼 때 쇠비름을 3% 첨가하여 제조하면 외관, 향미, 맛, 전체적 기호도 등 선호도 측면에서 적당할 것으로 생각한다.

  • Objectives : The objective of this study was to examine the effects of P. oleracea into the HCl-ethanol induced gastritis in rats, and to isolate and determine the chemical compounds from P. oleracea. Methods : The rats were orally administered with crude extract or fractions or isolated compounds of P. oleracea 30 mins before the induction of gastric lesion by oral administration of HCl-ethanol. The gastric lesional area was measured using pixel counting software. Then the chemical compounds from P. oleracea was isolated and determined by LC-MS and NMR. Results : The inhibition effect of oral administration of crude extract of P. oleracea at a dose of 500 mg/kg in HCl-ethanol induced gastritis was similar to cimetidine. Then, aqueous fraction at a dose of 240 mg/kg exhibited the effects similar to cimetidine. Then, the aqueous fraction was further separated by MPLC and yielded four sub fractions. Among those sub fractions, agent II at a dose of 40 mg/kg possessed the strongest effect in the HCl-ethanol induced gastritis. The water fraction yielded-Uridine, Adenosine, Guanosine, which were characterized by Mass, 1H-NMR, 13C-NMR. Conclusions : This study suggest that a P. oleracea and its compounds showed potent efficacy on the development of HCl-ethanol induced gastritis. Thus, P. olaracea can be a potential natural resource for the management of gastritis although the mechanism of action involved in the treatment remains to be explored.

Cactus의 광합성과 질소동화작용에 관한 연구 - 한국산 쇠비름(Portulaca oleracea L.) - (A Study on Photosynthesis and Nitrogen Assimilation in Cactus -Portulaca oleracea L.-)

  • Crassulacean acid metaholism (CAM) was investigated in leaves and stems of the succulent $C_4$dicot Portulaca oleracea L. Under 14-hour days, stem tissues showed much greater fluctuation of acidity than leaf tissues. But leaf and stem tissues showed almost same CAM-like pattern of acid fluctuation under 8-hour days. Stem tissues of R oleracea grown under the naturai environment showed high CAM activity, but no CAM activity was seen in leaves of those plants. In the naturally growing plants, the rapid acidification was seen in intact stems at dawn, but defoliated stems showed only a gradual increase. RuBP carlboxylase activity was very high at 2:00 P.M. in both leaves and stems. However, its activity at 1:00 A.M. and 5:30 AM. was hardly detected. particularly, activity of PEP carboxylase in leaves was very high in the early morning, though that in stem tissues was little. These results indicate that $CO_2$ passed through open stomata at dawn may be assimilated by PEP carboxylase in leaves, and then $C_4$ products move to stems. The levels of nitrate concentration and of nitrate reductase were higher in stems than in leaves. The levels were also higher in the light than in the dark. It would be suggested that considerable amount of nitrate absorbed from roots ho assimilated in stems, and nitrate transferred to leaves via stem tissues be reduced there. 마치현 70% 에탄올 추출물의 Heme Oxygenase-1 발현을 통한 산화적 스트레스에 대한 사람각질형성세포 보호 효과 (The Cytoprotective Action of Portulaca oleracea 70% EtOH Extracts via the Heme Oxygenase-1 on Hydrogen Peroxide-induced Oxidative Stress in Human Keratinocyte HaCaT Cells)

  • Keratinocytes are first barrier against outer challenges on skin. However, it is still largely unknown about effective protectors against ultraviolet B (UVB), and oxidative stress in human keratinocyte, HaCaT cells. Inducible heme oxygenase (HO)-1 acts against oxidants that are thought to play a role in the pathogenesis of skin disorders. Therefore, the purpose of this study was to evaluate the effect of Portulaca oleracea 70% EtOH extracts against hydrogen peroxide (H2O2)-induced oxidative stress in human keratinocytes, HaCaT cells. P. oleracea 70% EtOH extracts showed the potent protective effects on H2O2-induced toxicity by induced the expression of HO-1 in human keratinocyte, HaCaT cells. Furthermore, P. oleracea 70 % EtOH extracts caused the nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in human keratinocytes, HaCaT cells. In addition, we found that treatment with c-Jun N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea 70% EtOH extracts-induced HO-1 expression, and JNK inhibitor (SP600125) also inhibited protective effects by P. oleracea 70% EtOH extracts. Therefore, these results suggest that P. oleracea 70 % EtOH extracts increases cellular resistance to H2O2-induced oxidative injury in human keratinocyte, HaCaT cells, presumably through JNK pathway-Nrf2-dependent HO-1 expression.

쇠비름 첨가 설기떡의 저장성 및 노화 특성 (Storage Characteristics and Retrogradation Properties of Sulgidduk Added with Portulaca oleracea L.)

  • 본 연구는 우리 고유의 우수한 전통음식인 설기떡에 쇠비름을 첨가하여 쌀에 부족하기 쉬운 영양소를 보완할 수 있도록 개발하여 저장 중 노화 특성을 분석하였다. 멥쌀가루에 0, 1, 3 또는 5%의 쇠비름 페이스트를 첨가하여 제조한 후 $15^{\circ}C$에서 3일간 저장하면서 수분, 당도, 경도, 노화 특성 등을 분석하였다. 쇠비름 첨가량이 증가할수록 수분 보유량은 증가하였으며 5% 첨가군의 수분 보유율이 가장 높았다. 쇠비름은 당 함량이 적어 설기떡의 당도에 영향을 주지 못했다. 경도는 쇠비름 첨가량이 증가할수록 감소했고, 저장기간이 증가함에 따라 모든 시료군에서 증가하는 경향을 나타냈다. 탄력성은 쇠비름을 첨가할수록 탄력성이 증가하였다. 응집성은 쇠비름 첨가량의 증가가 설기떡 응집성에는 영향을 주지 않는 것으로 나타나 쇠비름이 설기떡 내부 조직을 치밀하게 하고 결합력을 유지시키는 데는 효과가 미약한 것으로 사료된다. 씹힘성은 5% 쇠비름 첨가군이 가장 낮게 나타났다. 회복탄성력은 쇠비름을 첨가할수록 증가하였고 저장기간이 증가함에 따라 모든 시료군에서 감소하였다. 시차주사 열량계를 측정한 결과 엔탈피는 대조군이 123.47 cal/g으로 가장 크게 나타나고 쇠비름 첨가량이 많을수록 쇠비름 설기떡의 엔탈피가 감소하는 경향을 나타내어, 이를 통해 설기떡 제조 시 쇠비름을 첨가함으로써 노화를 억제하는 데 도움을 주는 것으로 보인다. 관능검사 결과 쇠비름을 첨가할수록 색, 쇠비름향, 쇠비름맛, 촉촉함이 증가하는 경향을 나타냈다. 반면 경도는 쇠비름 첨가군에서 낮아졌다. 다음과 같은 결과로부터 설기떡 제조 시 쇠비름의 첨가는 설기떡의 수분 보유력을 높여 수분 함량의 감소를 억제하고 조직의 변화를 줄여 시간이 경과함에 따른 경도의 단단함을 보완할 수 있을 것으로 보인다. 또한 관능평가를 통해 선호도가 가장 높았던 5% 쇠비름 첨가군이 바람직한 제조방법으로 보인다.

Anxiolytic-like effects of Portulaca oleraceae L. using the elevated plus-maze in mice

  • The purpose of this study was to characterize the putative anxiolytic-like effects of the 70% ethanol extract of Portulaca oleracea (EPO) using an elevated plus maze (EPM) in mice. The EPO was orally administered at 50, 100, 200 or 400 mg/kg to ICR mice, 1 h before the behavioral evaluation in the EPM, respectively. Control mice were treated with an equal volume of 10% tween 80, and positive control mice with diazepam (1 mg/kg). Single treatments of the EPO significantly increased the percentage of time spent and arm entries into the open arms of the EPM versus controls (P < 0.05). Moreover, there were no changes in the locomotor activity and myorelaxant effects in any group compared with the saline controls. In addition, the anxiolytic-like effects of the EPO were blocked by flumazenil (10 mg/kg, i.p), a $GABA_A$ antagonist not by WAY 100635 (0.3 mg/kg, i.p), a 5-$HT_{1A}$ receptor antagonist. These results indicate that P. oleracea is an effective anxiolytic agent, and suggest that the anxiolytic-like effects of P. oleracea is mediated via the GABAergic nervous system.

쇠비름(Portulaca oleracea L.)에 함유(含有)된 생리활성물질(生理活性物質) 탐색(探索) (Identification of Physiologically Active Compounds from Purslane(Portulaca oleracea L.))

  • 밭 및 비농경지(非農耕地)의 우점잡초(優占雜草)인 쇠비름에 함유(含有)되어 있는 생이활성물질(生理活性物質)의 탐색(探索)을 통(通)하여 얻어진 몇가지의 결과(結果)를 요약(要約)하면 다음과 같다. 쇠비름을 건조(乾燥)한 시료(試料)로부터 추출(抽出)한 수용(水溶) 및 methanol 추출액(抽出液)은 상치, 무우 및 비름 등(等)의 식물종자(植物種子) 발아(發芽)를 크게 억제(抑制)시켰다. 쇠비름에 함유(含有)된 phenol 화합물(化合物)은 주(主)로 umbeliferon, p-coumaric, salicylic, vanillic, p-hydroxybenzoic, tannic, gallic산(酸) 등(等)이었다. 쇠비름의 건체(乾體), 생체(生體), 종자(種子) 및 callus 상태(狀態) 시료(試料)에 함유(含有)된 phenol 화합물(化合物)을 비교(比較)하면 건체시료(乾體試料)에서는 gallic산(産) 생체시료(生體試料)에서는 salicylic, vanillic산(酸), 종자시료(種子試料)에서는 ferulic산(酸), callus에서는 p-hydroxybenzoic, p-chlorobenzoic, cinamic산(酸) 등(等)이 많이 함유(含有)되어 있어 시료간(試料間)에 차이(差異)가 있었다. 지방산(脂肪酸)은 건체(乾體), 생체(生體) 및 종자시료(種子試料)에 linolenic산(酸)이 각각(各各) 9.203, 14.186, 1.732 mg/g으로 많이 함유(含有)되어 있었고 callus에는 palmitic산(酸)이 1.601 mg/g으로 많았다. 그리고 유기산(有機酸) 가운데는 oxalic산(酸)이 건체(乾體), 생체(生體) 및 callus 시료(試料)에서, citric산(酸)은 callus 시료(試料)에서 많았다.
