Cloning and Characterization of the IgA Fc Receptor from Swine

  • The myeloid-specific IgA Fc receptor ($Fc{\alpha}R$) is a cell surface molecule on immunocytes that provides a fundamental connection between humoral and cellular immunity. In this study, the full-length cDNA sequence of swine $Fc{\alpha}RI$ ($swFc{\alpha}RI$) was isolated and characterized and found to contain a 792-base-pair open reading frame, encoding a 264-amino-acid transmembrane glycoprotein with a predicted molecular mass of 29.4 kDa. The $swFc{\alpha}RI$ shares high amino acid sequence homology (>50%) with its counterparts from cattle, seal, and horse. Rosetting analysis confirmed that COS-7 cells transfected with an $swFc{\alpha}RI$ expression plasmid was able to combine with chicken erythrocytes sensitized with porcine IgA, but not IgG.

돼지 Landrace 품종에서 Myostatin 유전자의 유전적 다형성과 성장형질과의 연관성 (Association Between MSTN Gene Polymorphism and Growth Traits in Landrace Pigs)

  • Porcine myostatin(MS1N) gene plays a key role in the differentiation of myoblast and muscle development. Genetic polymorphism was screened by single stranded conformation polymorphism(SSCP) analysis and subsequent DNA sequencing detected a nucleotide substitution(C2150T) in exon 3 of MSIN gene. Phenotypic association of the polymorphism was tested in a Landrace population and positive effects of the allele T for lean growth traits were found in the population. Even though it is not significant, the pigs have IT and TC genotypes were heavier for the body weight at birth and at twenty weeks of age than those containing genotype. Cc. However, the allele T was significantly associated with higher eye muscle area(P < 0.05). As a result of this study, we suggested that the allele T in exon 3 of MSTN gene comes a significant effect for increasing the eye muscle area without decreasing backfat thickness. This polymorphism did not change the amino acid but Taq I -RFLP matched to SSCP band patterns in exon 3 of MSTN gene, which will be an useful molecular marker for breeding of Landrace pigs.

Association of a Pyruvate Kinase M2 (PKM2) Polymorphism with Back Fat Thickness in Berkshire Pigs

  • Pyruvate kinase M2 (PKM2) is a key regulatory enzyme in the glycolytic pathway. It is one of four pyruvate kinase isoenzymes that widely differ in their occurrence according to tissue type. PKM2 is expressed in differentiated tissues, such as fat tissues, lung, as well as normal proliferating cells, embryonic cells, and tumor cells. The objective of this study was to investigate the association of single nucleotide polymorphisms (SNPs) in the PKM2 gene with meat quality traits in Berkshire pigs. We detected a SNP (g.34341 A>G) in the 3'UTR region of the PKM2 gene in 670 Berkshire pigs through DNA sequencing. Three genotypes, AA, AG, and GG, were found for this SNP, but based on an association analysis with meat quality traits, genotype AA was significantly associated with thicker back fat than genotype GG (p=0.027). Therefore, the g.34341 A>G polymorphism in the 3'UTR region of the porcine PKM2 gene could be applied in pig breeding programs to improve back fat thickness.

더덕추출물에 의한 bFGF-유도 시험관내 혈관신생의 억제 (Effects of Codonopsis lanceolata Extracts on bFGF-induced Angiogenesis in vitro)

  • 더덕의 메탄올 추출물(CL-ex)이 혈관신생에 영향을 주는 지의 여부를 알아보기 위하여, 인간태반 정맥내피세포와 돼지 폐동맥내피세포를 이용하여 실험하였다. 실험 결과, 더덕추출물은 FGF에 의해 유도된 시험관 내 혈관신생을 억제하였다. 또한 더덕추출물은 내피세포의 DNA 합성에는 영향을 주지 않았으나, FGF에 의해 증가된 내피세포의 이동을 농도 의존적으로 강하게 억제하였고, MMPs의 분비 역시 감소시켰다. 이러한 결과에 의하면, 더덕에 함유된 혈관신생 억제 물질은 혈관내피세포의 이동 과정에 작용하여 그 효과를 나타내는 것이라 추정된다. 따라서 더덕은 그 발생과 진행이 혈관신생에 의존하고 있는 것으로 알려진 암 등의 질병 예방과 치료를 위한 유용한 식물자원으로 개발될 수 있음을 시사한다.

돼지 MC1R 유전자변이의 양돈산업 적용 (Commercial Application of Porcine MC1R Gene Polymorphisms to Korean Pork Industry)

  • 본 연구는 한국재래돼지의 MC1R 유전자형을 결정하고 MC1R 유전자의 변이와 육질과의 연관성을 규명하기 위하여 실시하였다. 재래돼지의 MC1R 유전자형을 분석한 결과 중국재래돼지품종이 속하는 $E^{D1}$ 내에서도 0201형에 속하는 것으로 나타났으며 몇몇의 재래돼지에서는 $E^P$ 유전자형이 출현하였는데 이는 Berkshire 품종의 혼입으로 인한 것으로 사료된다. 흑색의 모색과 관련되어 있다고 보고된 Leu102Pro (T1132C) 변이를 재래돼지와 Yorkshire의 양방향 교배로 생산된 F2 집단의 171 두를 대상으로 분석하고 육질형질과의 연관성을 분석하였다. 모든 육질형질에 대하여 유의적인 효과를 나타내지 않았으며 이로써 돼지의 흑색모색과 육질과는 연관성이 없으며 다른 육질관련 유전자에 의해 영향을 받는다는 근거를 제공하였다. 또한 Duroc 품종에서 AA으로 고정되어 있어 다른 품종과의 구분 기준이 되는 Ala243Thr (G1554A) 변이를 이용하여 종돈검정소에서 출하된 Landrace, Yorkshire, Duroc, Berkshire 순종집단에 대한 유전자형 빈도를 조사하였으며 Berkshire를 제외한 나머지 품종에서 이형접합체가 일부 출현하였다.

기관배양을 통한 추간판 재생용 나노파이버 및 온도 감응성 지지체에 대한 검증 (Investigation of Nanofiber and Thermosensitive Scaffold for Intervertebral Disc through Organ Culture)

  • The purpose of this study is to investigate the potential of a novel tissue engineering approach to regenerate intervertebral disc. In this study, thermosensitive scaffold (chitosan-Pluronic hydrogel) and nanofiber were used to replace the nucleus pulposus (NP) and annulus fibrosus of a degenerated intervertebral disc, leading to an eventual regeneration of the disc using the minimally invasive surgical procedure and organ culture. In preliminary study, disc cells were seeded into the scaffolds and cellular responses were assessed by MTT assay and scanning electron microscopy (SEM). Based on these results, we could know that tissue engineered scaffolds might provide favorable environments for the regeneration of tissues. Organ culture was performed in fresh porcine spinal motion segments with endplates on both sides. These spinal motion segments were classified into three groups: control (Intact), injured NP (Defect), and inserting tissue engineered scaffolds (Insert). The specimens were cultivated for 7 days, subsequently structural stability, cell proliferation and morphological changes were evaluated by the relaxation time, quantity of DNA, GAG and histological examination. In these results, inserting group showed higher relaxation time, reduced decrement of DNA contents, and accumulated GAG amount. Consequently, the tissue engineered scaffolds used in this study seen to be a promising base scaffolds for regenerative intervertebral disc due to its capacity to absorb external dynamic loading and the possible ideal environment provided for disc cell growing.

생쥐의 MT Transposon-like Element, Clone MTi7(MTi7) 유전자의 포유류 Homolog 및 Flanking Sequence에 대한 연구 (Studies on Mammalian Homolog and Flanking Sequence of Mouse MT Transposon-like Element, Clone MTi7(MTi7))

  • 본 연구실에서는 이전의 실험에서 suppression subtractive hybridization(SSH)을 통하여 생쥐의 생후 1일자 난소와 5일자 난소에서 차이 나게 발현하는 유전자들의 목록을 얻었고 그 중에서 MT transposon-like element, clone MTi7(MTi7)이 성장하는 난포에서 더 높게 발현한다는 것을 알아냈다(Park et al., 2002). In situ hybridization과 RNA interference를 이용한 연구결과, MTi7은 난자에서 특이 적으로 발현하는 유전자로 특히 난자성숙에 관여하는 것으로 관찰되었다(Park et at., 2003). 그러나 현재까지 MTi7의 염기서열은 생쥐에서만 알려져 있다. 따라서 본 연구는 두 부분으로 나누어서 첫째, MTi7이 다른 포유류에도 존재하는지 알아보기 위해 소,돼지, 흰쥐 그리고 사람 등 각기 다른 네 종의 난소 cDNA를 사용하여 새로운 MTi7을 분리하고자 하였으며, 둘째, 생쥐의 MTi7이 transposon의 특징을 갖고 있어 다른 유전자에 삽입되어 있는지를 알아보기 위해 inverse PCR을 시행하여 MTi7주변의 유전자가 있는지를 조사하였다. 네 종의 난소 cDNA를 사용하여 생쥐의 MTi7과 매우 유사한 염기서열을 갖고 있음을 알았다(87%∼98%). Inverse PCR 결과, 생쥐의 MTi7은 beta-carotene 15, 15'-monooxygenase(Bcdo) 유전자 혹은 serine protease inhibitor, Kunitz type I(Spint 1) 유전자에 삽입되어 있음을 알 수 있었다. 본 연구의 결과로 여러 포유류의 MTi7 sequence를 알아내고, 또한 생쥐의 MTi7이 삽입되어 있는 유전자를 알아냄으로써 머지 않은 장래에 난자형성 및 난포형성과정에 있어서 MTi7의 역할을 밝혀 낼 수 있을 것으로 기대된다.

cDNA Cloning and Polymorphism of the Porcine Carbonic Anhydrase III (CA3) Gene

  • Carbonic anhydrase III (CA3) is a member of a multigene family that encode carbonic anhydrase isozymes. In this study, a complete coding sequence of the pig CA3 gene which encodes a 260 amino-acid protein was determined. The amino acid comparison showed high sequence similarities with previously identified human (86.5%) CA3 gene and mouse (91.5%) Car3 gene. The partial genomic DNA sequences were also investigated. The length of intron 1 was 727 bp. Comparative sequencing of three pig breeds revealed that there was a T${\rightarrow}$C substitution at position 363 within intron 1. The substitution was situated within a NcoI recognition site and was developed as a PCR-restriction fragment length polymorphism (RFLP) marker for further use in population variation investigations and association analysis. Two alleles (A and B) were identified, and 617 bp fragments were observed for the AA genotype and 236 bp and 381 bp fragments for the BB genotype. The polymorphism of CA3 was detected in 8 pig breeds. Allele B was predominant in the Western pig breeds. In addition, association studies of the CA3 polymorphism with carcass traits in 140 $Yorkshire{\times}Meishan$ $F_2$ offspring showed that the NcoI PCR- RFLP genotype may be associated with variation in several carcass traits of interest for pig breeding. Allele B was associated with increases in lean meat percentage, loin eye height and loin eye area. Statistically significant association with backfat thickness was also found; pigs with the AB genotype had much less backfat thickness than AA or BB genotypes.

오제스키병의 생체 조기진단을 위한 면역세포화학, In situ hybridization 및 전자현미경적 연구 (Immunocytochemistry, In situ hybridization and electron microscopy for early diagnosis of Aujeszky's in living pigs)

  • The purpose of this study was to establish early diagnostic methods for the detection of Aujeszky's disease viral antigens and nucleic acid in nasal cells, and buffy coats from experimentally infected living pigs by a combination of immunocytochemistry, in situ hybridization with digoxigenin(DIG)-labled probe and electron microscopy. Forty days old piglets were inoculated intranasally with $10^{7.0}TCID_{50}$ of Aujeszky's disease virus (ADV, NYJ-1-87 strain). The viral antigens and nucleic acid of ADV were detected in nasal cells, and buffy coat for 20 days after inoculation by immunocytochemistry, in situ hybridization with DIG-labeled probe and electron microscopical method. The results were compared with conventional methods such as a porcine Aujeszky's disease serodiagnostic(PAD) kit, neutralization test(NT) and virus isolation. 1. The viral antigens, nucleic acids and capsids of ADV were detected in nasal cells, buffy coats from 3 days to 20 days after inoculation by immunocytochemistry, in situ hybridization with DIG-labeled probe and electron microscopy, respectively. 2. When viral antigens were detected by the immunocytochemical technique, a diffuse brown deposit was observed in the nucleus and cytoplasm of nasal cells, buffy coats and PK-15 cells under a microscope. 3. DIG-labeled DNA probe was prepared by amplification of conserved sequence of recombinant ADV-gp50 clone with polymerase chain reacction. When ADV-DNA was detected by ISH with DIG-labeled probe, purplish blue pigmentation were observed in the nuclei and cytoplasms of ADV-infected cells under a microscope. Positive signals were observed in nasal cells and in the buffy coat and PK-15 cells at the first day after inoculation. 4. Where ADV-capsids were detected by transmission electron microscopical method, aggregation of capsids was observed in the nuclei and cytoplasms of nasal cells, buffy coats and PK-15 cells. The results suggested that these methods were considered as the highly sensitive and reliable tools for rapid and confirmative diagnosis of Aujeszky's disease in living pigs.

  • 4개의 후보 유전자를 분석해본 결과, 돼지의 주요 염색체 부위 및 유전자들이 주요 경제성 요인들과 관계가 있는 것으로 확인됐다. 양돈업계에서 DNA 기술을 이용한 염색체 정보를 활용하기 위해 본 연구에서는 4개의 후보 유전자에서 생성된 중합효소연쇄반응(PCR) 생성물을 비교 재 서열 함으로써 단일염기변이(SNP) 표지들을 개발했다. 또한 이들 4개의 SNP에 대해 PCR 제한효소 절편길이 다형 성(RFLP)분석을 전개한 후, 이를 대한민국 내 버크셔 종 돼지 개체군의 유전자형을 분석하는데 활용했다. 본 연구는 유용한 단일염기변이를 식별하고 돼지개체군 내 경제적으로 중요한 특성들과 SNP의 연관성을 확인하는 데 그 목적이 있다.
