Study on Antioxidant and Cytotoxic Activities in Ethanol Extract from Prunus mume (오매 에탄올 추출물의 항산화 활성 및 항암 활성 연구)

  • Prunus mume is well known contain many functional materials that play beneficial roles in the human body. Studies have found that many organic acids and polyphenol compounds exist in Prunus mume. In this study, content of total polyphenols and flavonoids, antioxidative activity and cytotoxicity of ethanol extract from Prunus mume were measured Contents of total polyphenolic and total flavonoid compounds in ethanol extract from Prunus mume were 16.92 mg/g and 59.95 mg/g, respectively. The $IC_{50}$ of ethanol extract from Prunus mume were $237.72 {\mu}g$/assay and $239.58 {\mu}g$/assay by DPPH and ABTS radical cation scavenging test, respectively. Additionally, ferric ion reducing antioxidant power (FRAP) value of ethanol extract from Prunus mume was 0.94 mM ($FeSO_4$ eq.) by $800 {\mu}g$/assay. Cytotoxicity of Prunus mume against five kinds of cancer cell lines increased as the extract concentration increased Especially, cytotoxicity of the ethanol extract against A-549 (lung cancer line) was higher than that against any other cancer cell line by both MTT and SRB assay. These results show that ethanol extract of Prunus mume has considerably high antioxidative and cytotoxic activities.

Comparison of Main Compounds and Physiological Activities of Anthriscus sylvestris (L.) Hoffm. Roots and Aerial Parts Extracts (전호 지하부와 지상부 추출물의 주요성분 및 생리활성 비교)

  • The number of people suffering from diabetes have been increased around the world. In this study, we investigated the antidiabetic and antioxidant effects of Anthriscus sylvestris(L.) Hoffm and its main compounds. It was divided into root(R) and aerial part(AP) for comparative analysis. Total polyphenol, total flavonoid content was higher in AP extract, but nodakenin content was higher in R(1169.13 ± 6.00 mg/g) extract. Antioxidant activity was also higher in AP extract. To compare antidiabetic efficacy, we analyzed the effects of R and AP extracts on ɑ-glucosidase inhibition(AGI), dipeptidyl peptidase-4(DPP-4) and protein tyrosine phosphatase(PTP)1B activity. R and AP extracts showed similar effects on AGI and DPP-4 activity in a concentration dependent manner, and there was no effect on PTP1B activation. Glucose uptake(139.51 ± 3.19%) in 3T3-L1 cells was more effective in the AP extract-treated group than the positive control, rosiglitazone, group. Both R and AP extracts were effective in protecting against pancreatic beta cell damage caused by hyperglycemia. These results suggest that Anthriscus sylvestris(L.) Hoffm. could be used as a candidate for diabetes treatment.

Whitening and Anti-oxidative Activities of Chemical Components Extracted from Branches of Sorbus alnifolia

  • In this study were evaluated the whitening and anti-oxidative activities from the extracts of Sorbus alnifolia branches, and identified the chemical structures of the active ingredients. In the whitening tests using α-MSH stimulated B16F10 melanoma cells, the 70% ethanol extract and n-butanol (n-BuOH) fractions concentration-dependently inhibited cellular melanogenesis and intracellular tyrosinase activities without causing cell toxicity. The total polyphenol content of n-BuOH and ethyl acetate (EtOAc) fractions were measured to be respectively 241.1 ± 1.1 and 222.9 ± 2.4 (mg/g GAE), and the total flavonoid content of EtOAc fraction was 75.3 ± 2.0 (mg/g QE). Upon anti-oxidant studies with DPPH and ABTS+ radicals, potent radical scavenging activities were observed in the EtOAc and n-BuOH fractions. Moreover, in the study of cell protection efficacy using HaCaT keratinocytes damaged by H2O2, the EtOAc and n-BuOH fractions showed a very positive results on prevention of oxidative stress. Phytochemical studies for this extract resulted in the isolation of four compounds; 2-oxopomolic acid (1), euscaphic acid (2), epi-catechin (3), prunasin (4). These results suggested that the extract of S. alnifolia branches containing compounds 1-4 as natural ingredients could be used as whitening and anti-oxidant ingredients in cosmetic formulations.

Antioxidant Activity and Quality Characteristics of Pine Needle Cookies (솔잎 쿠키의 항산화활성 및 품질특성)

  • Pine needle powder has medical and functional uses. In this study, cookies were prepared using different 0.5%, 1%, 3%, 5% of pine needle powder. The total phenolic content was measured by the Folin-Ciocalteu method, and antioxidant activity was evaluated by DPPH assay. The antioxidant activity was highly correlated with the total phenolic composition of pine needle cookies (r=0.989). In addition, the quality characteristics of the pine needle cookies were estimated based on the bulk density and pH of the dough, spread factor, loss rate, leavening rate, color, texture profile analysis, and sensory evaluations. The bulk density of the dough and the hardness, total polyphenol contents and DPPH free radical scavenging activity of cookies significantly increased with increasing pine needle powder, while the pH of the dough, spread factor and L values of the cookies decreased with increasing pine needle powder content (p<0.05). Additionally, the consumer acceptability scores for the 3% pine needle cookie groups ranked significantly (p<0.05) higher than those of the other groups in appearance, taste, flavor, texture and overall preference. Taken together, the results of this study suggest that pine needle powder is a good ingredient for increasing the consumer acceptability and the functionality of cookies.

Total Polyphenol Content and Antioxidative Activities of Mistletoe(Viscum album) Extracts by Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 겨우살이(Mistletoe, Viscum album) 추출물의 폴리페놀 함량 및 항산화 활성)

  • This study was performed to investigate antioxidant activities of Mistletoe (MS) extracts by measuring electron donating ability (EDA), super oxide dismutase (SOD)-like activity and total polyphenol content (TPC). The extracts were obtained using supercritical carbon dioxide, hot-water and ethanol extraction. EDAs was the higher in ethanol extract (70.53%) than water (36.73%) or supercritical carbon dioxide extract (15.73% at 400 bar, $30^{\circ}C$) at concentration of 10 mg/mL. However, SOD-like activities and TPCs were the highest in the supercritical carbon dioxide extract. At concentration of 1 mg/mL, supercritical carbon dioxide extract (22.54% at 200 bar, $40^{\circ}C$) was higher SOD activity than water (7.85%) and ethanol extract (6.12%). Regarding TPCs of MS extracts, the content was the highest in supercritical carbon dioxide extract (17.57 wt% catechin equivalent at 200 bar, $50^{\circ}C$) followed by ethanol and water extract. These results suggest that it can be useful to develop functional foods using antioxidative active compounds of MS with high polyphenol contents.

Optimization of Hot-water Extraction Conditions of Bioactive Compounds from Coffee Residue Extracts (커피박으로부터 생리활성물질 생산 증대를 위한 열수추출 공정 개발)

  • In this study, the optimization of extraction parameters (solvent, temperature, time, solvent concentration) for the maximization of polyphenol extraction was performed to produce value-added food and cosmetic additives using a byproduct of coffee extraction process (coffee residue). All of the extraction parameters evaluated in this experiment had significant effects on polyphenol extraction and the results showed the effect of NaOH concentration on the polyphenol production was most significant among tested parameters. Especially, hot water extraction using acid or base was effective rather than hot-water extraction and the addition of 0.1 mol of NaOH increased 1.5 times extraction concentration compared with hot-water extraction using distilled water. It was found that hot-water extraction with NaOH was more effective than hot-water extraction, and 36.5 mg GAE/g DM was obtained under optimum condition of $100^{\circ}C$, 2 mol of NaOH and 30 min. This result was 2.9 times higher than that of 12.5 mg GAE/g DM obtained from the hot-water extraction before optimization. Thus, coffee residue could be used for food and cosmetic industry as a high-value additive such as antioxidant.

Secondary Metabolites from Enzymatic Oxidation of Caffeic Acid with Pancreatic Lipase Inhibitory Activity (카페인산의 효소적 산화반응으로부터 췌장 지방분해효소 저해 물질의 분리)

  • Pancreatic lipase is a potential therapeutic target for the treatment of diet-induced obesity in humans. As part of our continuing search for novel bioactive compounds, the convenient enzymatic transformation of caffeic acid into neolignans as well as related oxidized-products enhanced pancreatic lipase inhibitory activity. Enzymatic transformation of caffeic acid (1) using polyphenol oxidase originating from Korean pear yielded four oxidized metabolites, which were identified by different spectroscopic techniques ($^1H$,$^{13}C$ NMR, DEP/T, $^1H-^1H$ COSY, HMBC, HMQC, and NOESY). The anti-obesity efficacy of caffeic acid reactant was tested by in vitro porcine pancreatic lipase assay. All tested samples showed dose-dependent pancreatic lipase inhibitory activities. Four oxidative products including phellinsin A (2), caffeicinic acid (3), isocaffeicinic acid (4), and 7,8-erythro-caffeicin (5) were isolated and identified. The major metabolites (2~5) were evaluated for their pancreatic lipase inhibitory activity, and oxidized-products (2~3) improved potency against pancreatic lipase when compared to original caffeic acid. This result suggested that the neolignans isolated from oxidative transformation of caffeic acid might be beneficial in the treatment of obesity and relevant diseases, and the convenient enzymatic transformation by polyphenol oxidase may be a valuable method for structural modification and enhancement of activity.

Comparison of Antioxidant Activities by Different Extraction Temperatures of Some Commercially Available Cultivars of Rice Bran in Korea (추출온도에 따른 국내 벼 품종 미강의 항산화 활성 비교)

  • This study was conducted to determine antioxidant compounds and antioxidant activities of rice bran at temperatures of $10^{\circ}C$, $25^{\circ}C$, and $60^{\circ}C$ using 70% ethanol solution. DPPH, ABTS radical scavenging activities and reducing power have been used to investigate the relative antioxidant activities of extracts. The contents of total polyphenol and total flavonoids in the 70% ethanol extracts were measured by spectrophotometer. The total polyphenol content in the rice bran extracts were 790.39~930.07, 782.71~1,039.57, and 848.22~1,052.96 mg gallic acid eq./100 g in $10^{\circ}C$, $25^{\circ}C$ and $60^{\circ}C$, respectively. Antioxidant activities, such as DPPH, ABTS radical scavenging activity and reducing power, in all cultivars and temperatures was the highest in Cheonghaejinmi rice bran extracted at $25^{\circ}C$. The results obtained in the in vitro models suggest that extracts from rice bran, especially Cheonghaejinmi, has significant health-promoting effects, as it exhibits antioxidant activities.

  • 아로니아(Aronia czarna)는 anthocyanin, polyphenol, flavonoid, cathechine, chlorogenic acid와 같은 생리활성물질이 풍부하게 존재하며 항산화, 항암, 항균, 피부건강개선, 노화방지 등 다양한 생리활성에 대한 효능이 있는 것으로 알려져 있어 건강 및 기능성식품, 화장품 등의 원료 소재로 각광받고 있다. 생물전환(Bioconversion)은 미생물 또는 효소의 생물학적 촉매 반응을 활용하여 기존 소재의 성분을 변환시키는 기술이다, 최근 생물전환을 활용한 천연소재의 생리활성 물질 기능성, 생체이용률, 안전성을 증대시키기 위한 방안으로 많은 연구가 진행되고 있으며 식품, 의약품, 화장품 등 다양한 분야에서 활성화 되고 있다. 본 연구는 젓갈로부터 분리한 균주를 유전학적 특성을 확인하기 위하여 16S rDNA 염기서열을 분석한 뒤 그중 유산균을 발효공정에 활용하였다. 전북 순창에서 수확된 아로니아 분말과 발효공정을 수행하였으며 아로니아 최적 추출조건 선정, 발효공정 전 후 추출물의 기능성 평가를 진행하기 위하여 DPPH radical scavenging activity, Total polyphenol 함량을 확인하여 항산화 효능 및 유효성분 함량을 평가하였다. 또한 대식세포인 Raw 264.7을 사용하여 MTT assay, Nitric oxide (NO) 생성 억제 효능을 확인하여 세포독성 및 항염증 활성을 평가하였다. 실험결과, 젓갈류 발효물로부터 16종의 다양한 균주를 확보하였으며, 그중 L. rhamnosus, L. plantarum, P. pentosaceus 균을 발효 공정에 활용하여 유용 균주를 선정 결과 P. pentosaceus 종 유산균 처리군에서 무처리군 대비 DPPH radical 소거능 및 polyphenol 함량이 증가됨을 확인하였다. 발효공정 후 항산화 활성은 무처리군 대비 약 119%, polyphenol의 함량은 무처리군 대비 약 119%로 증가됨을 확인되었다. 또한 Raw 264.7 세포실험 결과 발효공정 후 독성활성이 감소되는 경향을 확인되었으며, 항염증 활성이 월등히 증가됨을 확인하였다.

Polyphenol Profiling and Comparison of Extraction Solvents on Useful Component Content and Antioxidant Activity of Rhododendron micranthum Shoot (꼬리진달래 신초의 폴리페놀 프로파일링과 추출용매에 따른 유용성분 함량 및 항산화 활성 비교)

  • This study was conducted to provide information on the possibility of using Rhododendron micranthum as resource plant. Through polyphenol profiling of R. micranthum shoot extract, useful components were selected and the content of useful components and antioxidant activity by extraction solvent were compared. As a result, 37 polyphenol compounds were identified in R. micranthum shoot extract through LC-ESI-MS analysis. Contents by extraction solvent were compared of chlorogenic acid, astragalin, myricetin, and afzelin. Chlorogenic acid was highest at 6.57±0.12 mg/g in the ethanol extract, astragalin was 2.29±0.02 mg/g in the ethanol extract, myricetin was 4.77±0.06 and 4.52±0.27 mg/g in the methanol, ethanol extracts, and afzelin was highest at 0.10±0.01 mg/g in the methanol extract. Total polyphenol content of the R. micranthum shoot extract according to the extraction solvent was highest in the ethanol extract at 80.01±2.36 mg/g, and total flavonoid content was also highest in the ethanol extract at 78.08±3.44 mg/g. The IC50 value of DPPH radical scavenging activity according to the extraction solvent of R. micranthum shoot was lowest in methanol extract at 943.57±10.68 mg/L, and the IC50 value of ABTS radical scavenging activity was also lowest in methanol extract at 641.60±7.58 mg/L.
