Genetic Relationships Analysis of Asparagus cochinchinensis $L_{OUR}$ Collections by Random Amplified Polymorphic DNA (RAPD에 의한 천문동 수집종의 유연관계 분석)

  • To analyze the genetic relationships among 23 accessions of Asparagus cochinensis $L_{OUR}$, random amplified polymorphic DNA(RAPD) analysis was performed using artificially synthesized 10 primers. The range of polymorphism was $42.9{\sim}91.7%$ with an average of 72.9% in 85 randomly and specifically amplified DNA fragments. On the. basis of similarity coefficient analysis by unweighed pairgroup method, arithmetic average method(UPGMA), 23 accessions of Asparagus cochinensis $L_{OUR}$ could be classified into 6 groups at the similarity coefficient value of 0.82. Group I contained 5 accessions, Group II contained 6 accessions, Group III contained 6 accessions, GroupIV contained 2 accessions, Group V contained 2 accessions and Group VI contained 2 accessions. The range of total genetic similarity coefficient value of 23 accessions of Asparagus cochinchinensis $L_{OUR}$ was $0.47{\sim}0.92$ and average value was 0.76. To obtain more exact data from PCR, we also tried to develope enhanced RAPD techniques using Bovine Lacto Transfer Technique Optimizer(BLOTTO). In RAPD analysis of Asparagus cochinensis $L_{OUR}$, we could obtain better RAPD results by adding BLOTTO at a final concentration of 1%.

Analysis of Genetic Diversity in Soybean Varieties Using RAPD Markers (사료작물로 이용이 가능한 한국 재배콩의 RAPD 표지인자에 의한 유전적 다양성 분석)

  • Random amplified polymorphic DNA (RAPD) analysis was used to detect the genetic diversity of soybean (Glycine max (L.) Merr.) varieties and field bean (Glycine soza Sieb. and Zucc.) Five soybean varieties and one field bean were analysed with random primers using the polymerase chain reaction (PCR). Nine primers of a total twenty random primer were selected to amplify DNA segments. A total of 74 PCR products were amplified and 67.6% of which were polymorphic. The size of DNA molecule is ranged 0.13~2.0Kb and typically generated four to eight major bands. Specific genetic marker were revealed in primer sequence 5'-CAG GCC CIT C-3', 5'-TGC TCT GCC C-3' and 5'-GTC CAC ACG G-3', respectively. Genetic similarity between each of the varieties were calculated from the pair-wise comparisons of amplification products and a dendrogram was constructed by an unweighted pair-group method with arithmethical means (UPGMA). The results indicate that intervarietal relationships of soybean have a narrow genetic base and between the varieties, Hwanggum-kong and Seckryang-bootkong is more closely related than the rest of varieties, and field bean is related quite distant.

Interspecific Distinguishability of Veiled Lady Mushrooms (Dictyophora spp.) Based on rDNA-ITS Analysis (rDNA-ITS 분석에 의한 망태버섯속균(Dictyophora spp.)의 종간 구분 가능성)

  • To establish the phylogenetic relationships of Dictyophora spp., rDNA-ITS regions of 11 strains of veiled lady mushroom collected from various countries were amplified and sequenced. It was observed that the 11 strains were divided into four groups based on PCR band patterns of each ITS region cleaved by eight different restriction enzymes in cleaved amplified polymorphic sequence analysis (CAPS). The phylogenic relationship of each group by cleaved amplified polymorphic sequence (CAPS) analysis matches well with previously reported morphological phylogeny, such as 5 strains of D. indusiata, 4 strains of D. echinovolvata, and a strain of Phallus rugulosus. Sequence analysis using the cluster V methods showed more detail classification than CAPS analysis. The 5.8S region showed two point nucleotide base exchanges from G to A according to four groups, and four groups were subdivided by sequence variation of ITS I and ITS II regions. But sequence variation of Phallus rugulosus was not showed in full ITS region. This study further delineates the taxonomic level at which ITS sequences, in comparison to ribosomal gene sequence, are most useful in systematics and other mushroom study.

Selection and Agronomic Traits of Radiation-induced Variants in Rice (방사선 이용 벼 돌연변이 계통 선발 및 농경 형질조사)

  • Radiation technique in agriculture was initiated to develop mutant rice. Seeds of Daechungbyeo rice were irradiated with 250 Gy gamma ray for the purpose of inducing and selecting rice variants. Some quantitative traits of the variants in M$_{8}$ generation were evaluated and RAPD analysis was carried out. Variants showed a wider range of agronomic characteristics in both a positive and a negative direction compared with their original variety. The new mutants were characterized by an increased or decreased in plant height, lodging resistance and shorter panicle. RAPD analysis showed that polymorphic bands were presented in most of the primers. In comparison with the original variety, variants were classified into four groups through UPGMA analysis. Among mutants no. 91, 139, 140 and 141 was ranked as salt tolerance and the proline content of these mutants was more increased than that of original variety. The lines of 139, 140 and 141 had the highest genetic distance as compared to original variety in the dendrogram. It is expected that such variants will be useful not only for studying molecular genetics but also for breeding research and genetic analysis.s.

Genetic Diversity and Population Genetic Structure of Exochorda serratifolia in South Korea (가침박달 집단의 유전다양성 및 유전구조 분석)

  • Genetic diversity and population genetic structure were estimated in nine natural populations of Exochorda serratifolia in South Korea using ISSR marker system. Average of polymorphic loci per primer was 5.8 (S.D.=2.32) and percentage of polymorphic loci per population was 78.7% with total 35 loci from 6 ISSR primers. In AMOVA, 27.8% of total genetic variation came from genetic difference among populations and 72.2% was resulted from difference among individual trees within populations. Genetic differentiations by Bayesian inference were 0.249 of ${\theta}^{11}$ and 0.227 of $G_{ST}$. Inbreeding coefficient for total populations was 0.412. There was significant correlation between genetic distance and geographic distance among populations. On the results of Bayesian cluster analysis, nine populations were assigned into three groups. The first group included 5 populations, and the second and the third had two populations per group, respectively. These three regions could explain 10.0% of total genetic variation from hierarchical AMOVA, and the levels of among-population and among-individual were explained 19.7% and 70.3%, respectively. The geographic distribution of populations following the three Bayesian clusters could be explained with mountain range as Baekdudaegan which is the main chain of mountains in Korea. The mountains as the physical barrier might hamper gene flow in the pearlbush. So when protected areas are designated for conservation of this species, we should consider those three regions into considerations and would better to choose at least one population per region.

The classification and comparison of genetic diversity of genus Malus using RAPD (RAPD를 이용한 능금속 식물종의 계통관계와 유전적 다양성)

  • Cenus Malus is a long-lived woody species primarily distributed throughout Asia. Many species of this genus are regarded as agriculturally and ecologically important. The phynetics and genetic diversity among eight species of genus Malus were reconstructed using the random amplified polymorphic DNA (RAPD) markers. In a simple measure of intraspecies variability by the percentage of polymorphic bands, the M. micromalus exhibited the lowest variation (34.7%). The M. pumila showed the highest (50.0%). Mean number of alleles per locus (A) ranged from 1.347 to 1.500 with a mean of 1.437. The phenotypic frequency of each band was calculated and used in estimating genetic diversify (H) within species. The mean of H was 0.190 across species, varying from 0.155 to 0.220. In particular, two cultivated species, M. pumila and M. asiatica, had high expected diversity, 0.314 and 0.307, respectively. On a per locus basis, the proportion of total genetic variation due to differences among species ranged from 0.388 to 0.472 with a mean of 0.423, indicating that 42.3% of the total variation was found among species. The phylogenetic tree showed three distinct elates. One includes M. sieversii, M. pumila, and M. asiatica. Another includes three M. baccata taxa. The other includes M. sieboldii, M. floribunsa, and M. micromalus. One variety and one form of M. sieboldii were well separated each other. RAPD markers are useful in germ-plasm classification of genus Malus and evolutionary studies.

RAPD-SCAR Markers Linked to Medium-Leaf Zoysiagrass Ecotypes (한국잔디 중지 변이개체와 연관된 RAPD-SCAR 마커)

  • Two medium-leaf ecotypes (CY6069, CY6097) belonging to one species (Zoysia japonica) of Korean lawngrasses were selected in sod production fields in Jang Seong, Korea. They were reported to have distinct morphological and growth rate characteristics different from the preferred medium-leaf type zoysiagrass in Korea. This study was conducted to define further the genotypic difference at the molecular level and to develop DNA marker based on the specific DNA fragment. Polymorphic DNA fragments were first explored by using randomly amplified polymorphic DNA (RAPD) primers, which were then converted into PCR-based sequence characterized amplified region (SCAR) markers. The CY6069-specific primer set amplified about 550 bp successfully, while the CY6097 marker produced the expected 690 bp band, by which those markers were nominated by CY6069_550 and CY6069_690 SCARs, respectively. Together with the reported morphological and other phenotypic features, the SCAR markers confirmed in this study will be useful to identify those medium-leaf zoysiagrass genotypes when they are cultivated with other vegetatively propagated warm-season turfgrasses in sod farms.

Genetic variations and relationships of Phragmites japonica and P. communis according to water environment change (수환경변화에 따른 갈대와 달뿌리풀의 유전적 변이 및 유연관계)

  • We performed RAPD analysis to investigate the inter-/intraspecific relationships and regional variations of Phragmites japonica and P. communis according to the environmental change. One hundred and fourty nine genetic effective polymorphic bands between 300 bp and 1,900 bp were marked from RAPD PCR with nine oligoprimers. From the RAPD analysis by Nei-Li's genetic distance, the dissimilarity indices among the populations of Phragmites japonica were relatively low from 0.012 to 0.061, and Phragmites communis were also low from 0.033 to 0.095. It showed the close genetic relationships among the same species populations, and both species were distinctly independent with relatively high level of dissimilarity indices (0.043 - 0.132). The obvious genetic markers to distinguish two species were confirmed and those profiles were suggested. From the UPGMA phenogram by RAPD analysis, both species showed the water environment related cluster patterns by distributional regions. RAPD analysis was useful to delimit two species taxonomically and to investigate the genetic relationships among inter-/intraspecific populations.

Variations in Mutant Plants by chemical mutagen treatments of Dianthus superbus L. (술패랭이에서 화학돌연변이물질 처리에 따른 돌연변이 유기 및 변이 분석)

  • This study was conducted to determine the effects and optimum concentrantion of chemical mutagens, colchicine, EMS (ethyl methan sulfonate), MNU (1-methyl-3-1­-nitrosoguanidinenitro), sodium azide $(NaN_3)$ for induction of mutant plants. In order to induce the mutants of Dianthus superbus L, immature seed were pre-soaked in the warter adding each mutagens and concentration of EMS, colchicine, MNU, and sodium azide $(NaN_3)$. Comparision of morphological characteristic and seed germination in each mutant plants differed depending on mutagen sources and their concentrations. When 0.2% EMS were treated on seed, germination decreased to 12% while untreated control was germinated 76.6% for twenty days. Treatments of colchicine appeared higher germination than other mutagen but not survived. The survival rate was extremely decreased in MNU treatment at 0.5mM and chlorophyll-mutant plantlets were obtained by sodium azide treatment at 0.2mM. Chlorophyll mutants were produced by pre-soaking the immature seed of Dianthus superbus L. with mutagen, sodium azide. The control plants appeared normal green leaf color, while mutant plant after mutagenic treatment of immature seed results in yellow­-green stripes and albino in normal green leaf tissue. RAPD was carried out to check the genetic modification of regenerated plants by mutagen treatments at 0.2mM sodium azide. Three polymorphic DNA fragments out of thirty-seven obtained by RAPDs were observed in regenerated plants using five decamer primers.

부착성 요각류 Tigriopus japonicus의 nauplius 생산을 위한 중형규조류의 먹이효율

  • 본 연구는 부착규조류에 따른 부착성 요각류 Tigriopus japonicus의 nauplius 생산력을 알아보기 위하여 실시하였다. 실험에 사용된 T. japonicus는 부산 동백섬 부경대학교 수산과학연구소 부근에 있는 tidal pool에서 동물성부유생물망으로 채집하였다. 먹이생물로는 부경대학교 한국해양 미세조류 은행에서 보관중인 부착규조류 중 중형종인 Caloneis schroder를 대표종으로 이와 크기가(14.8~27.5$\mu\textrm{m}$) 유사한 종들을 대상으로 형태를 현미경 하에서 관찰하고, 지역과 분리일자 등을 고려하여 유사종을 선별한 후 최종적으로 종의 유전적 유사성을 밝히기 위하여 RAPD-PCR (random amplified polymorphic DNA polymerase chain reaction)을 실시하였다. 이 중 Caloneis schroder와 유전적으로 유사성이 낮은 Navicula spp. (KMCC B-245, 393, 394, 581) 4종을 선별하여 T. japonicus의 먹이로 사용하여 포란한 암컷의 nauplius 생산력을 3반복 조사하였다. Genomic DNA는 대부분의 종에서 성공적으로 검출되었으며, 종에 따라 PCR 증폭산물이 나타나지 않은 경우도 있었으므로, PCR 산물이 나타난 종에 대해서만 분석하였고, 증폭된 DNA band는 대부분 크기 0.5~2.0kb 범위에서 나타났다. 실험에 사용된 부착규조류 간의 유사성을 알아보기 위하여 similarity matrix를 분석한 결과 F값의 범위는 0.00에서 1.00까지 였으며, Caloneis schroder와 유사성이 낮은 종들에 비하여 유사성이 높은 종들이 더 많이 나타났다. 이들을 먹이로 하여 포란한 T. japonicus의 실험구별 nauplius 평균 개체수를 살펴보면, KMCC B-394가 255.7마리로 가장 높았던 반면 KMCC B-581가 29.7마리로 가장 낮았다. 그 외 KMCC B-245가 120.0마리, KMCC B-393가 76.0마리, Caloneis schroder가 32.3마리 각각 나타났다. 이와같은 결과를 볼 때 T. japonicus의 nauplius 생산력은 규조 종에 따라 큰 차이가 있는 것으로 판단된다.
