Preparation and Investigation on Swelling and Drug Delivery Properties of a Novel Silver/Salep-g-Poly(Acrylic Acid) Nanocomposite Hydrogel

  • Novel silver/salep-g-poly(acrylic acid) nanocomposite hydrogel were prepared in aqueous solution using poly(acrylic acid) grafted onto salep as a biopolymer based material. FT-IR spectra confirmed that poly(acrylic acid) (PAA) had been grafted onto salep in graft copolymerization reaction. TEM observations showed that silver nanoparticles have been uniformly dispersed in polymeric matrix. Effects of pH, acrylic acid (AA) amount and silver ion concentration on swelling capabilities were investigated. Results indicate that modifying AA and silver ion can improve swelling properties of the resultant nanocomposite hydrogel. pH response of this nanocomposite hydrogel in acidic and neutral pH made it suitable for drug delivery applications.

  • Polymer hydrogel has attracted considerable interest as a soft material which is finding expanding applications in pharmaceutics and various biomedical fields. In this work, modified PNVP hydrogels were synthesized by crosslinking polymerization of NVP monomer in the presence of PEG macromer with a methoxy end. The effect of the tethered PEG chain on the properties of the hydrogel was investigated in terms of its swelling capacity, compression gel strength, and the morphology of the resulting hydrogels. These PEG-modified PNVP hydrogels possessed good biocompatibility and a decreased protein (fibrinogen) adsorption, thereby indicating their potential as novel drug delivery matrices and scaffold for tissue engineering.

  • Hyaluronic acid and chitosan-based poly(ethylene oxide) (HA-PEO and Chitosan-PEO) hydrogels have been employed as unique biomedical polymeric materials with properties such as bioactivity from polysaccharide, biocompatibility of HA and chitosan as well as PEO and control release of bioactive molecules from the hydrogel itself. We here examine in situ hydrogels based on hyaluronic acid and chitosan in terms of their synthesis, mechanical properties, morphologies and in vitro cellular interactions on their surface and inside. In vivo bone regeneration of HA-PEO and Chitosan-PEO hydrogels was compared with in mouse model.

  • In tissue engineering, scaffolds play an important role in the growth of cells to 3-D organs or tissues. For the success of tissue engineering, they should be mimicked to meet the requirements of natural extracellular matrix (ECM) in the body, such as mechanical properties, adhesiveness, porosity, biodegradability, and growth factor release, etc. Contrary to other materials, polymeric materials are adequate to engineer scaffolds for tissue engineering because controlling the structure and the ratio of components and designing various shapes and size are possible. In this review, the importance, major characteristics, processes, and recent examples of polymeric scaffolds for tissue engineering applications are discussed.

  • Radiation technology is an effective way for regulating polymeric materials to physicochemical and mechanical properties. New polymeric hydrogels based on vinyl ethers have been synthesized by the $\gamma$-initiated polymerization method. In this paper, we have studied the effect of radiation on mechanical and electrochemical properties of new rubber-like polymeric composite materials based on polyvinylether of ethyleneglycol (PVEEG).

  • New polymeric hydrogels based on vinyl ethers have been synthesized by the ${\gamma}$-initiated polymerization method. Their physical chemistry and physical mechanical properities have been studied. It has been shown that structure and swelling behavior of the hydrogels can be regulated by the changing of synthesis conditions nature of monomers. Novel stimuli-sensitive polymers have been synthesized by the varying of macrochains hydrophilic-hydrophobic balance. The some biomedical aspects of application of hydrogels in capacity of drain aging polymeric materials in ophthalmology surgery, implants in plastic surgery as well as drug delivery systems have been investigated.

히드록시프로필셀룰로오스와 카르보폴을 이용한 부유성 히드로겔 매트릭스의 제조 및 in Vitro 평가 (Preparation and in vitro Evaluation of a Buoyant Hydrogel Matrix with Hydroxypropylcellulose and Carbopol)

  • The study was carried out for the preparation and evaluation of a buoyant hydrogel matrix (BHM), which is buoyant in a neutral or in pH 2.0 buffer solution, by the aspects of buoyancy, swelling, and drug release. Physical mixtures of HPC and CP in various molar ratio were employed as a mucoadhesive polymer which swells and controls the rate of drug release. Anhydrous citric acid and sodium bicarbonate in the molar ratio of 1:3 were employed as effervescing agents which provide a buoyancy for the mucoadhesive polymeric matrix. The buoyancy in vitro was expressed as both floating time$(T_{fl})$ and surfing time$(T_{sf})$, which are the time required for floating from the bottom to the surface of the medium and the time to keep the floated state at the surface of medium during release studies, respectively. A close relationship was observed between the buoyancy and the amount of effervescing agent added. $T_{fl}$ of the buoyant hydrogel matrices were decreased to about 10 seconds linearly with increasing the amount of effervescing agent in the range of 5 to 15%. $T_{sf}$ of the buoyant hydrogel matrices were varied from 1 to 3 hr depending on the amount of effervescing agent. The swelling was observed by changes in diameter of the buoyant hydrogel matrices in distilled water or acidic buffer solution, resulted in dependences on pH and the amount of effervescing agents. The release of hydrochlorothiazide from the buoyant hydrogel matrices were followed by apparent zero-order kinetics, while the buoyant hydrogel matrices were floated at the surface and maintaining their swollen shapes.

  • Mesenchymal stem cells (MSCs) are useful candidates for tissue engineering and cell therapy. Physiological cell environment not only connects cells to each other, but also connects cells to the extracellular matrix that provide mechanical support, thus exposing the entire cell surface and activating signaling pathways. Hydrogel is a polymeric material that swells in water and maintains a distinct 3-dimensional (3D) network structure by cross linking. In this study, we investigated the optimized cellular function for canine adipose tissue-derived MSCs (cAD-MSCs) using hydrogel. We observed that the expression levels of Ki67 and proliferating cell nuclear antigen, which are involved in cell proliferation and stemness, were increased in transwell-hydrogel (3D-TN) compared to the transwell-normal (TN). Also, transforming growth factor-${\beta}1$ and SOX9, which are typical bone morphogenesis-inducing factors, were increased in 3D-TN compared to the TN. Collagen type II alpha 1, which is a chondrocyte-specific marker, was increased in 3D-TN compared to the TN. Osteocalcin, which is a osteocyte-specific marker, was increased in 3D-TN compared to the TN. Collectively, preconditioning cAD-MSCs via 3D culture systems can enhance inherent secretory properties that may improve the potency and efficacy of MSCs-based therapies for bone regeneration process.

Enhancing the Physical Properties and Lifespan of Bacterial Quorum Quenching Media through Combination of Ionic Cross-Linking and Dehydration

  • Quorum quenching (QQ) bacteria entrapped in a polymeric composite hydrogel (QQ medium) have been successfully applied in membrane bioreactors (MBRs) for effective biofouling control. However, in order to bring QQ technology closer to practice, the physical strength and lifetime of QQ media should be improved. In this study, enforcement of physical strength, as well as an extension of the lifetime of a previously reported QQ bacteria entrapping hollow cylinder (QQ-HC), was sought by adding a dehydration procedure following the cross-linking of the polymeric hydrogel by inorganic compounds like $Ca^{2+}$ and boric acid. Such prepared medium demonstrated enhanced physical strength possibly through an increased degree of physical cross-linking. As a result, a longer lifetime of QQ-HCs was confirmed, which led to improved biofouling mitigation performance of QQ-HC in an MBR. Furthermore, QQ-HCs stored under dehydrated condition showed higher QQ activity when the storage time lasted more than 90 days owing to enhanced cell viability. In addition, the dormant QQ activity after the dehydration step could be easily restored through reactivation with real wastewater, and the reduced weight of the dehydrated media is expected to make handling and transportation of QQ media highly convenient and economical in practice.

Histopathological Effects of Tissue Adhesives on Experimental Peripheral Nerve Transection Model in Rats

  • Objective : Our aim was to evaluate the histopathological effects of tissue adhesives on peripheral nerve regeneration after experimental sciatic nerve transection in rats and to search whether these tissue adhesives may possess a therapeutic potential in peripheral nerve injuries. Methods : This experimental study was performed using 42 female Wistar-Albino rats distributed in 6 groups subsequent to transection of right sciatic nerves. Group I underwent external circumferential neurolysis; Group II received suture repair; Group III had local polymeric hydrogel based tissue adhesive administration; Group IV received suture repair and polymeric hydrogel based tissue adhesive application together; Group V had gelatin based tissue adhesive application and Group VI had suture repair and gelatin based tissue adhesive together. After a 6-week follow-up period, biopsies were obtained from site of neural injury and groups were compared with respect to histopathological scoring based on inflammatory, degenerative, necrotic and fibrotic changes. Results : There were remarkable differences between control group and study groups with respect to inflammation (p=0.001), degeneration (p=0.002), necrosis (p=0.007), fibrosis (p<0.001) and vascularity (p=0.001). Histopathological scores were similar between study groups and the only noteworthy difference was that Group V displayed a lower score for necrosis and higher score in terms of vascularization. Conclusion : Our results imply that tissue adhesives can be useful in repair of peripheral nerve injuries by decreasing the surgical trauma and shortening the duration of intervention. Results with gelatin based tissue adhesive are especially promising since more intense vascularity was observed in tissue after application. However, trials on larger series with longer durations of follow-up are essential for reaching more reliable conclusions.
