식품중 Polycyclic Aromatic Hydrocarbons의 위해성평가 (Dietary Risk Assessment for Polycyclic Aromatic Hydrocarbons in Foods)

  • Polycyclic aromatic hydrocarbons(PAHs)의 인체에 대한 위해성 정도를 판단하기 위해 PAHs의 대표독성물질인 benzo(a)pyrene의 독성을 기준으로 하는 TEFs를 적용하여, PAHs의 주요노출식품에 의한 위해성 평가를 수행하였다. 숯불구이쇠고기, 숯불구이돼지고기, 숯불구이닭고기, 햄, 베이컨, 및 식용유지 등을 대상으로 PAHs 오염도를 모니터링 한 결과 숯불구이닭고기에서 총 PAMs농도가 9.3ppb로 가장 높게 검출되었으며 각각의 congener들 모두 다른 식품에서 보다 높게 나타났다. 반면에 숯불구이쇠고기, 베이컨, 숯불구이돼지고기, 햄 및 식용유지의 총 PAHs는 각각 0.2ppb, 0.3ppb, 0.7ppb, 0.8ppb 및 1.2ppb로 비교적 낮은 값을 나타냈다. TEFs값을 적용하여 환산된 오염도 값의 경우에서도 숯불구이닭고기가 가장 높은 값을 보였고, congener들의 농도를 비교했을 때 숯불구이닭고기, 숯불구이돼지고기, 베이컨, 숯불구이쇠고기 및 햄에서 모두 BaP의 농도가 각각 1.88 $\mu\textrm{g}$-TEQ$_{BaP}$/kg, 0.19 $\mu\textrm{g}$-TEQ$_{BaP}$/kg, 0.08 $\mu\textrm{g}$-TEQ$_{BaP}$/kg, 0.04 $\mu\textrm{g}$-TEQ$_{BaP}$/kg 및 0.02 $\mu\textrm{g}$-TEQ$_{BaP}$/kg로 congener들 중 가장 높게 나타났고 식용유지에서는 dibenzo(a,h)anthracene의 농도가 0.21 $\mu\textrm{g}$-TEQ$_{BaP}$/kg으로 가장 높은 값을 나타냈다. 우리나라 사람들의 PHAs 주요노출식품 섭취로 인한 만성1일 인체 노출량은 4.32${\times}$$10^{-4}$ $\mu\textrm{g}$/kg/day으로 산출되었으며 선정된 식품 중에서 숯불구이 닭고기의 노출기여도가 가장 높았다. PAMs의 초과발암 위해도는 만성1일 인체노출량과 BaP의 발암력인 7.3 (mg/kg/day)$^{-1}$을 고려하였을 때 3.44${\times}$$10^{-6}$ , 백만명당 3∼4명 수준이었다. 우리나라 대표식단을 통한 PAMs 노출량을 반영할 수 있는 위해성 평가가 추후 계속되어야 할 것으로 사료된다. 할 것으로 사료된다.

PAH가 CYP1B1 유전자 발현에 미치는 영향 (Effect of PAH on CYP1B1 Gene Expression)

  • Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrachloto-p-dioxin). Recent industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. Acenaphthene, anthracene, benzo(b)fluoranthene, fluorene, fluoranthene, anphthanlene, pyrene, phenanthrene and carbazole were weak responders in MCF-7 cells. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA.

These flavonoids decreased B(k)F infuced luciferase activity at low concentration. But, these flavonoids exhibited stimulatory effect at high concentration.

Biodegradation of Pyrene by the White Rot Fungus, Irpex lacteus

  • Abstract The removal percentage (94%) of 100 ppm of pyrene in a shaken culture of white rot fungus, Irpex lacteus, was much higher than that in a static culture (37.9%). Over 90% of the pyrene disappeared with I. lacteus grown at $15-27^{\circ}C$, yet less than 50% was removed at $37^{\circ}C$. The transformation rates of pyrene ($4.5-5.0{\;}\mu\textrm{g}/ml/day$) were not very different among cultures with 5- 30% inoculum sizes, and over 90% of the 100 ppm pyrene was removed in every case during 20 days of incubation. The biodegradation of pyrene by I. lacteus was confirmed by measuring the $CO_2$ evolved from the mineralization of the added pyrene. The activity of lignin peroxidase (LiP), which is known to be involved in the biodegradation by white rot fungi, was high between 8 to 12 days of incubation. Although manganese peroxidase activity was demonstrated during the same period as LiP, its activity was quite low, and no laccase activity was detected. Even though the activity patterns of ligninolytic enzymes did not coincide with the pyrene removal, this study shows that I. lacteus has a high biodegrading capability and can be a candidate for the bioremediation of polycyclic aromatic hydrocarbon contaminants.inants.

  • Among the AKARI all-sky survey data, the $9{\mu}m$ diffuse map is crucial to study the polycyclic aromatic hydrocarbon (PAH) emission features on large spatial scales, while the $18{\mu}m$ map is useful to trace hot dust emission. To utilize these advantages, we have improved the AKARI mid-infrared (MIR) all-sky survey diffuse maps. For example, we have established special methods to remove the effects of the ionizing radiation in the South Atlantic Anomaly (SAA) and of the scattered light from the moon. Using improved diffuse map data, we study the properties of PAHs and dust in the Galactic center region associated with high-energy phenomena.

A QUALITY CHECK OF THE AKARI MID-INFRARED ALL-SKY DIFFUSE MAP TOWARD THE MASSIVE STAR-FORMING REGIONS NGC 6334 AND NGC 6357

  • We present a comparative study of CO and polycyclic aromatic hydrocarbon (PAH) emission toward a region including the massive star-forming regions of NGC 6334 and NGC 6357. We use the NANTEN $^{12}CO(J=1-0)$ data and the AKARI $9{\mu}m$ All-Sky diffuse map in order to evaluate the calibration accuracy of the AKARI data. We confirm that the overall CO distribution shows a good spatial correspondence with the PAH emission, and their intensities exhibit a good power-law correlation with a spatial resolution down to 4' over the region of $10^{\circ}{\times}10^{\circ}$. We also reveal poorer correlation for small scale structures between the two quantities toward NGC 6357, due to strong UV radiation from local sources. Larger scatter in the correlation toward NGC 6357 indicates higher ionization degree and/or PAH excitation than that of NGC 6334.

소나무잎을 Passive Air Sampler로 이용하여 지역별 대기 중 다환방향족 탄화수소의 농도 및 발생원 추정 (Estimation of Atmospheric PAH Concentrations and Sources at Several Sites Using Pine Needles as a Passive Air Sampler)

  • Objectives: This study was carried out in order to estimate atmospheric polycyclic aromatic hydrocarbon (PAH) concentrations and sources using pine needles as a passive air sampler (PAS) in urban (Pyeongtaek), semirural (Anseong) and rural (Jincheon) sites. Methods: One-year-old pine needles were collected for analysis of their PAH concentrations ($C_{p,n}g/g$ dry) at the end of December. PAHs concentrations in the ambient air ($C_a$, $ng/m^3$) were calculated with a $Log(C_p/C_a)-LogK_{oa}$ correlational equation. Results: PAHs concentrations in ambient air ($C_a$) were high, in the order of urban ($114.03ng/m^3$), semirural ($105.17ng/m^3$) and rural ($61.91ng/m^3$) sites. However, distributions of PAH isomer concentrations were very similar. PAHs of which molecular weight is smaller than 228.30 (AcPy, Acp, Flu, Phen, Ant, Flt, Pyr, BaA, Chry) made up most of the PAHs in the ambient air (96.6-98.5%). Conclusion: At urban, semirural and rural sites, it was concluded that the main source of PAHs in the ambient air ratio of each PAH isomer concentration was cars, especially diesel vehicles.

  • In mammalian, cytochrome P4501A1 (CYP1A1) is very important for metabolism of xenobiotics such as PAHs(Polycyclic aromatic hydrocarbon) and heterocyclic amine, and it is induced by environmental contaminants such as PAHs, TCDD(2,3,7,8-tetrchlorodibenzo-p-dioxin) and 3-MC (3-methylcholanthrene). In fish, like mammalian, when it is exposed to environmental contaminants, they cause specific and sensitive induction of CYP1A. Therefore, induction of CYP1A in fish and mammalian is widely used as a biomarker for exposure of environmental contaminants. In this study, to compare the function of Cyp1a1 in fish with it in mammalian, we have used rainbow trout(Oncorhynchys mykiss) hepatoma cells (RTH-149) and mouse hepatocyte (Hepa-I). in order to examine induction of Cyp1a1 by TCDD, we have used the bioassay system. We examined effects of TCDD on the Cyp1a1-luciferase reporter gene activity, 7-ethoxyresorufin O-deethylase(EROD) activity and Cypa mRNA level.

Fluorescence Characteristic Spectra of Domestic Fuel Products through Laser Induced Fluorescence Detection

  • Traditional investigation procedures of soil and groundwater contamination are followed by soil gas sampling, soil sampling, groundwater sampling, establishment of monitoring wells, and groundwater monitoring. It often takes several weeks to obtain the analysis reports, and sometimes, it needs supplemental sampling and analysis to delineate the polluted area. Laser induced fluorescence (LIF) system is designed for the detection of free-phase petroleum pollutants, and it is suitable for on-site real-time site investigation when coupling with a direct push testing tool. Petroleum products always contain polycyclic aromatic hydrocarbon (PAH) compounds possessing fluorescence characteristics that make them detectable through LIF detection. In this study, LIF spectroscopy of 5 major fuel products was conducted to establish the databank of LIF fluorescence characteristic spectra, including gasoline, diesel, jet fuel, marine fuel and low-sulfur fuel. Multivariate statistical tools were also applied to distinguish LIF fluorescence characteristic spectra among the mixtures of selected fuel products. This study successfully demonstrated the feasibility of identifying fuel species based on LIF characteristic fluorescence spectra, also LIF seemed to be uncovered its powerful ability of tracing underground petroleum leakages.

자동차에서 배출되는 가스상 유해대기오염물질 (HAPs) 배출량 추정 (Estimation of Gaseous Hazardous Air Pollutants Emission from Vehicles)

  • Hazardous Air Pollutants (HAPs) are difficult to measure, analyze and assess for risk because of low ambient concentrations and varieties. Types of HAPs are Volatile organic compounds (VOCs), Polycyclic aromatic hydrocarbon (PAHs) and Aldehydes. HAP emissions from vehicles are a contributor to serious adverse health effects in urban areas. In this study, hazardous air pollutant emissions from road transport vehicles by Non-methane volatile organic compounds (NMVOC) weight fraction and PAHs emission factors are estimated in 2008. The top-five-most hazardous air pollutant emissions were estimated to toluene 864.3 ton/yr, acrolein 690.6 ton/yr, acetaldehyde 554.5 ton/yr, formaldehyde 498.7 ton/yr, propionaldehyde 421.6 ton/yr in 2008. The results for a cancer and non-cancer risk assessment of HAPs emissions show that the major cancer driver is formaldehyde and the non-cancer driver is acrolein.
