폐지와 폐목재에서의 잔류성 유기오염물질의 농도 및 배출원 추정 (Levels of Persistent Organic Pollutants in Waste Paper and Waste Lumber and Evaluation of their Sources)

  • 본 연구는 산업 현장에서 나오는 폐지, 폐목재에 함유되어 있는 17종 PCDD/Fs, 12종 Co-PCBs, 16종 PAHs 농도 수준과 이들의 발생원 추적을 위해 수행되었다. 농도 수준은 폐지와 폐목재 순으로 PCDD/Fs는 9.69~176.77 pg/g-dry, 0.14~0.25 pg/g-dry, Co-PCBs는 109.95~4097.25 pg/g-dry, 28.23~59.88 pg/g-dry, PAHs는 9.30~52.18 ng/g-dry, 0.82~1.82 ng/g-dry로 조사되었으며, 모든 대상물질은 폐목재보다 폐지에서 더 높은 농도로 검출되었다. PCDD/Fs 중 OCDD가 가장 높은 농도로 검출되었으며, 소각로 배출가스 중 다이옥신과 유사한 패턴을 보였다. Co-PCBs 이성질체 분포 패턴은 상용 PCB 제품의 이성질체 분포패턴과 유사하게 나타나 이로 인한 오염으로 판단되었고, PAHs의 경우 특정 화합물의 농도비를 이용하여 추정한 결과 폐기물 소각과 같은 연소와 관련이 있음을 알 수 있었다.

  • 고분해능 가스크로마토그래프/고분해능 질량분석기를 이용하여 시화공단 지역 근로자와 일반주민의 혈액에서 다이옥신류를 분석하였다. 평균 다이옥신류의 농도는 근로자가 일반주민보다 높았다. 근로자와 일반주민의 평균 다이옥신 TEQ 농도는 각각 40.3 pg I-TEQ/g lipid, 16.6 pg I-TEQ/g lipid였고, 전체 PCBs 농도는 각각 174.1 ng/g lipid, 151.0 ng/g lipid였다. 주성분분석을 한 결과, 근로자와 일반주민의 다이옥신 이성질체 분포는 구별되었지만, 모두 식품류에서의 분포와 유사하였다. 각 개인의 일일 다이옥신 섭취량을 계산했을 때 40%만이 WHO에서 제시한 1-4 pg I-TEQs/kg bw/day의 허용범주를 만족하였다.

Effects of 3,3′,4,4′,5-pentachloro biphenyl (PCB126) on the Expression the Tight Junction Genes in Cultured Mouse Neonatal Testis

  • In an effort to uncover the spermatogenic impairment by the polychlorinated biphenyls (PCBs), the expression of tight junctions (TJs) genes important for the formation of the blood testis barrier (BTB) were examined following the 3,3',4,4',5-pentachloro biphenyl (PCB126) treatment in cultured neonatal testis in mice. At 4 days (D4) after 10 and 100 nM PCB126 treatment the expression of claudin-11 was significantly increased when compared with vehicle control. In contrast no difference in occludin and claudin-1 expression was found among the experimental group. On D8, 100 nM PCB126 significantly increased the expression of claudin-11 but not occludin and claudin-1. 1 uM PCB126 treatment significantly decreased expressions of occludin and ciaudin -1, suggesting the general toxic effect on the Sertoli cell. Because PCB126 does not alter the proliferative activity of spermatogenic cells and Sertoli cells in neonatal testis, it is likely that increase in the expression of claudin-11 by low dose of PCB126 may attribute to the alteration of the Sertoli cells differentiation in testis. It also emphasized that PCB126 might have differentially affected the transcription of TJ genes in Sertoli cells. In conclusion, this result suggests that the structure of TJ may be targeted by PCB126 in neonatal testis in mice and that co-PCB is potentially harmful to spermatogenesis by alteration of the development of BTB.

한국 연안 이매패류에 잔류하는 PBDEs 및 OCs에 관한 연구 (A Study on Polybrominated Diphenyl Ethers and Organochlorine Compounds Residues Persistent in Korean Coastal Bivalves)

  • Bivalve samples from 30 locations Korean coastal waters were analyzed to characterize the contamination with and distribution of polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs). The congeners of PBDE in 30 bivalve samples were analyzed by high resolution gas chromatography with a high resolution mass detector. The levels of PBDEs in bivalve samples ranged from 0.092 to 13.5 ng/g wet weight. These values for PBDEs concentrations in bivalves from Korea were higher than or comparable to available values for bivalves from other countries. In the congener profiles, BDE-47 (mean contribution=42.2%) was predominant, followed by BDE-99 (23.1%), BDE-100 (11.9%) and BDE-154 (3.5%). Among the OCs analyzed, $\Sigma$PCBs and $\Sigma$DDTs levels were highest, followed by $\Sigma$HCHs, $\Sigma$CHLs and HCB. The wet weight based $\Sigma$PCBs and $\Sigma$DDTs ranged from 0.28 ng/g to 19.4 ng/g, and from 0.19 ng/g to 4.86 ng/g, respectively. The $\Sigma$PCBs and $\Sigma$DDT slevels in bivalves along the entire Korean coast were lower than those of the USA, and below the Food and Drug Administration (USA. FDA) limits for fish and shell fish. However, the human health criteria and wild life criteria of the USA. Environmental Protection Agency (USA. EPA) were exceeded for DDE and $\Sigma$HCHs at most of the sampling points.

아산만해역 어류체내와 퇴적물중의 PCB와 유기염소계농약 분포 (On the Distribution of PCBs and Organochlorine Pesticides in Fish and Sediment of the Asan Bay)

  • Distribution of Polychlorinated biphenyls (PCBs) and organochlorine pesticides in sediments, seawater and fish tissues were examined. Concentrations of ${\Sigma}PCBs$ and ${\Sigma}DDTs$ in the intertidal sediments of Asan Bay ranged from $1.90{\sim}3.27\;ng\;g^{-1}$ dw (dry weight) and $0.51{\sim}1.26\;ng\;g^{-1}$ dw, respectively. ${\Sigma}PCBs$ concentrations in the tissue of greenling (Hexagrammos otakii), olive flounder (Paralichthys olivaceus) and finespotted flounder (Pleuronichthys cornutus) were $1.27{\pm}0.55$, $1.30{\pm}0.71$ and $1.19{\pm}0.43\;ng\;g^{-1}$ ww (wet weight), respectively. The profile of PCBs in the sediments and the 3 species of fish differed as witnessed by the dominance of tetra- to penta-chlorinated congeners in sediments while penta- to hexa-chlorinated congeners dominated in fish. ${\Sigma}DDTs$ concentrations were $0.67{\pm}0.52$, $0.79{\pm}0.61$, $1.58{\pm}1.05\;ng\;g^{-1}$ ww respectively in the tissue of greenling, olive flounder and finespotted flounder. The p,p'-DDE appeared to be the most important metabolite of DDT in studied fishes. The concentrations of PCBs and organochlorine pesticides measured in this study for fish were generally lower than those of previous studies. Compared to the pollutant concentration quality guidelines in marine fish, the environment of Asan Bay appears to be in a healthy state.

Thermal Destruction of Waste Insulating Oil Containing PCBs under High Temperature and Pressurized Conditions

  • This experimental study was performed to obtain thermal energy from the combustion of synthetic gas, produced by the pyrolysis of insulating oil containing polychlorinated biphenyls (PCBs) in a high temperature and high pressure reactor. The average synthetic gas generated was $59.67Am^3/hr$ via the steady state gasification of insulating oil waste (20 kg/hr) with average concentrations (standard deviation) of $CO_2$, CO, and $H_2$ in the synthetic gas of $38.63{\pm}3.11%$, $35.18{\pm}1.93%$, and $28.42{\pm}1.68%$, respectively. The concentrations of the PCBs in the transformer insulating oil and synthetic gas after its gasification, and the concentrations of the dioxins that could be produced from the incomplete degradation of PCBs were measured. It was revealed that the PCBs in the insulating oil were composed of the series from tetrachlorobiphenyl to octachlorobiphenyl. However, only the #49, #44, #52, and #47/75/48 congeners were detected from the synthetic gas after gasification of the insulating oil and in the flue gas from the combustor. In conclusion, the experimental conditions suggested in this study were very useful for the appropriate treatment of insulating oil containing PCBs. Also, fuel gas containing CO and $H_2$ can be obtained from the pyrolysis of insulating oil containing PCBs.

수원근교(水原近郊) 관개수중(灌漑水中)의 PCBs 및 유기염소계(有機鹽素系) 살충제(殺蟲劑)의 잔류평가(殘留評價) (Evaluation of Polychlorinated Biphenyls(PCBs) and Organochlorine Insecticide Residues in Irrigation Waters in the Periphery of Suwon)

  • 1982년 11월부터 1983년 10월까지 서호를 포함한 수인근교의 저수지 및 한강의 하천수질 시료를 수집, PCBs 및 유기염소계살충제 잔류수준을 평가한 결과는 다음과 같다. 1) PCBs는 거의 모든 수질시료에서 검출되었다. 2) PCBs의 펑균잔류수준은 0.009${\sim}$0.5ppb였고 유기 염소계 살충제의 평균잔류수준은 불검출${\sim}$0.008ppb 였다. 3) PCBs와 총 DDT의 평균잔류수준비는 시료채취 지점에 따라 차이가 있었으며, 이 비는 한강수질시료에서는 500으로 가장 높았고, 원천 수질에서 가장 낮았다. 4) 관개수중 PCBs는 공업 및 도시폐수에서 유래된 것으로 보였다.

Characterization of the pcbD Gene Encoding 2-Hydroxy-6-Ox0-6-Phenylgexa-2,4-Dienoate Hydrolase from Pseudomonas sp. P20

  • 2-Hydroxy-6-oxo-6phenylhexa-2,4-dienoate (HOPDA) hydrolase catalyzes the hydrolytic cleavage of HOPDA to bemzpate and 2-hydroxypenta-2, 4-dienoate (HPD) during microbial catabolism of biphenyl and polychlorinated biphenyls. A HOPDA hydrolase gene (pcbD) was isolated from the genomic library of Pseudomonas sp. P20 and designated as pCNUO1201; a 7.5-kb XbaI DNA fragment from Pseudomonas sp. P20 was inserted into the pBluescript SK(+) XbaI site. E. coli HB101 harboring pCNU1201 exhibited HOPDA hydrolase activity. The open reading frame (ORF) corresponding to the pcbD gene consisted of 855 base pairs with an ATG initiation codon and a TGA termination codon. The ORF was preceded by a rebosome-binding sequence of 5'-TGGAGC-3' and its G+C content was 55 mol%. The pcbD gene of Pseudomonas sp. P20 was located immedeately downstream of the pcbC gene encoding 2,3- dihydroxybiphenyl 1,2-dioxygenase, and approximately 4-kb upstream of the pcbE gene encoding HPD hydratase. The pcbK gene was able to encode a polypeptide with a molecular weight of 31,732 containing 284 amino acid residues. The deduced amino acid sequence of the HOPDA hydrolase of Pseudomonas sp. P20 exhibited high identity (62%) with those of the HOPDA hydrolases of P. putida KF715, P. pseudoalcaligenes KF707, and Burkholderia cepacia LB400, and also significant homology with those of other hydrolytic enzymes including esterase, transferase, and peptidase.

Effect of PCB118 on expression of COX-2 and cPLA2 in rat testes

  • Polychlorinated biphenyls (PCBs) are synthetic organic compounds with two benzene rings and well known environmental pollutants. This study examined the effect of persistent exposure to 2,$3^{\prime}$,4,$4^{\prime}$,5-pentachlorobiphenyl (PCB118) on the proinflammatory and proapoptotic factors in male rats. Male Sprague Dawley rats were administered weekly intraperitoneal injections of either PCB118 (20 mg/kg) dissolved in corn oil or corn oil alone. One week after 2 and 5 administrations, the rats were sacrificed by a pentobarbital injection. The effect of PCB118 on the expression of cyclooxygenase 2 (COX-2), cytosolic phospholipase A2 (cPLA2), peroxisome proliferator-activated receptor gamma, Bcl and Bcl-2-associated X protein (BAX) was investigated. The level of COX-2 and cPLA2 expression was higher in the PCB118-treated rats than the control. These results suggest that PCB118 has a proinflammatory effect in rats.

Pseudomonas sp. DJ-12의 pcbCD 유전자의 클로닝과 Escherichia coli에서의 발현 (Cloning and Expression of pcbCD Genes in Escherichia coli from Pseudomonas sp. DJ-12)

  • Polychlorinaed biphenyls(PCBs) 와 biphenyl을 분해하는 Pseudomonas sp. DJ-12에서는 그 초기 분해과정에 pcb ABCD 유전자들이 관여하고 있음이 밝혀졌다. 그 중 pcbCD와 pcdD 유전자를 E. coli XL1-Blue에 클로닝하여 E. coli CU103 과 CU105 균주를 각각 제조하였다. E. coli CU103은 2,3-dehydroxybuphenyl dioxygenase(2,3-DHBP)와 meta-cleavage compound(MCP) hydrolase를 생성하여 2,3-dihydroxybiphenyl을 benzoate로 변환시켜 주었다. E. coli CU1 과 CU103 에서 pcbC 유전자의 산물인 2,3-DHBP dioxygense의 활성도는 Pseudomonas sp. DJ-12에서 보다 약 17배 높았으며, E. coli CU105에서 pcbD의 산물인 MCP hydrolase는 약 3배 더 높게 나타났다.
