  • The polyamine pathway represents a logical target for chemotherapeutic intervention, since depletion of polyamines results in the disruption of a variety of cellular functions, and may in specific cases result in cytotoxicity. Polyamine interaction with DNA has also long been thought to be an important function of the natural polyamines and as more is learned about the specific interactions and the resultant conformational changes which can be influenced by the polyamine binding to DNA the potential for regional and gene-specific changes are becoming more evident. We have prepraed the elaborate polyamines by the reaction of simpler polyamines with polyalkyating agents. Synthesized polyamines were separated and purified by metal complex formation and ion-exchange chromatography. They were characterized by X-ray crystal structure determinations of their metal complexes.

Polyamines and Their Metabolites as Diagnostic Markers of Human Diseases

  • Polyamines, putrescine, spermidine and spermine, are ubiquitous in living cells and are essential for eukaryotic cell growth. These polycations interact with negatively charged molecules such as DNA, RNA, acidic proteins and phospholipids and modulate various cellular functions including macromolecular synthesis. Dysregulation of the polyamine pathway leads to pathological conditions including cancer, inflammation, stroke, renal failure and diabetes. Increase in polyamines and polyamine synthesis enzymes is often associated with tumor growth, and urinary and plasma contents of polyamines and their metabolites have been investigated as diagnostic markers for cancers. Of these, diacetylated derivatives of spermidine and spermine are elevated in the urine of cancer patients and present potential markers for early detection. Enhanced catabolism of cellular polyamines by polyamine oxidases (PAO), spermine oxidase (SMO) or acetylpolyamine oxidase (AcPAO), increases cellular oxidative stress and generates hydrogen peroxide and a reactive toxic metabolite, acrolein, which covalently incorporates into lysine residues of cellular proteins. Levels of protein-conjuagated acrolein (PC-Acro) and polyamine oxidizing enzymes were increased in the locus of brain infarction and in plasma in a mouse model of stroke and also in the plasma of stroke patients. When the combined measurements of PC-Acro, interleukin 6 (IL-6), and C-reactive protein (CRP) were evaluated, even silent brain infarction (SBI) was detected with high sensitivity and specificity. Considering that there are no reliable biochemical markers for early stage of stroke, PC-Acro and PAOs present promising markers. Thus the polyamine metabolites in plasma or urine provide useful tools in early diagnosis of cancer and stroke.

  • We have purified and characterized a deoxyribonuclease from zygotes of the eukaryotic green alga, Chlamydomonas reinhardtii and investigated effects of the polyamines, putrescine, spermidine and spermine on the purifed endonuclease-catalyzed cleavage of plasmid DNA. The enzyme has a molecular weight of about 37 kDa as measured by gel filtration and SDS-polyacrylamide gel electrophoresis. There is no requirement for a divalent cation. The activity is sensitive to ionic strength, as NaCl and KCl result in inhibition. The cleavage of plasmid DNA by the purified endonuclease was effectively inhibited by polyamines. The enzyme activity was inhibited more effectively by spermine than by spermidine. The inhibition by putrescine was lower than the other two polyamines.

당근 현탁배양세포에서 $Ca^{2+}$과 Polyamine이 Callose 함량에 미치는 영향 (Effects of $Ca^{2+}$ and Polyamine on Callose Contents in Carrot Suspension Cultured Cells)

  • The effects of Ca2+ on polyamines on callose contents of carrot suspension cultured cells were studied. The regeneration process of the cell wall of carrot protoplast observed through the electron microscope. Treatment of the carrot suspension cultured cells with Ca2+ and polyamines resulted in considerable increase on callose contents at 0.1 mM of Ca2+ and polyamines, particulary spermidine. Poly-L-lysine and poly-L-ornithine increased about 30% and 100% of callose contents than that of the control respectively, whereas verapamil and flunarizine markedly decreased the callose contents. These effects of Ca2+ of free ion rather than as Ca2+-calmodulin complex. During the cultivation of the protoplast, the regeneration of the cell wall was somewhat observed on the 4th day, however, it was inhibited by verapamil. These results suggested that the promotive action of Ca2+ and polyamines were manifested in the callose contents and the regeneraton of the cell wall.

  • The activity of Daucus carota L. root $\beta$-glucan synthetase II was observed to increase in the presence of polyamines such as putrescine, spermidine and spermine in vitro, whereas the activity of Daucus carota L. root $\beta$-glucan synthetase I was not affected by the polyamine. The activity $\beta$-glucan synthetase II from Daucus carota L. root protoplasts cultured on medium containing 10-6 M polyamines such as putrescine, spermidine and spermine was observed to be higher than that of the control. Daucus carota L. root protoplasts were observed to have the activities of arginine and ornithine decarboxylases and it was noted that they could produce polyamines, which might have an effect on $\beta$-glucan synthetase II activity.

  • An experimental system for the possibility of association between cultured plant cells and a cyanobacterium (N. muscorum) was investigated. The association was established in nitrogen-free medium, and nitrogen source was provided by N. muscorum through its own nitrogen fixation. N. muscorum was distributed on the outer part and intercellular spaces of the cultured plant cells. Polyamines were treated to the nitrogen-free medium to improve the association. Polyamines increased nitrogenase activity and protein amount in the association between N. muscorum and the cultured plant cells, as compared to the association without polyamines. Especially, spermine showed an increasing effect on the nitrogenase activity and the protein content in the association of tobacco and N. muscorum, while spermidine showed similar effect in case of soybean and N. muscorum.

  • A rapid and sensitive high performance liquid chromatographic method with UV detection for the analysis of polyamines(putrescine, spermidine, spermine) has been developed. The benzoyl chloride derivatives of putrescine, spermidine and spermine are separated on a mc-Bondapak C18 reverse-phase column with 50% methanol as the mobile phase. This method was applied to the analysis of polyamines in the serum of normal human and uterine cancer patients. The results show that the mean level of polyamines in cancer patients serum is much higher than that in normal human serum.

녹두자엽에서 엽록소 및 단백질함량과 $\delta$-Aminolevulinate Dehydratase활성에 미치는 Polyamine의 영향 (Effects of Polyamines on Chlorophyll and Protein content, and $\delta$-Aminolevulinate Dehydratase Activity in Greening Mung Bean Cotyledons)

  • 녹두 유식물의 자엽에서 엽록소 및 단백질 함량과 $\delta$-aminolevulinate dehydratase(ALAD)활성의 변화에 미치는 polyamine의 영향을 조사하였다. Polyamine은 녹화과정에서 자엽내의 엽록소 생성을 촉진하였으며, 이 효과는 KCl에 의해서 상승되었다. 자엽내의 단백질 함량의 변화 또한 엽록소 함량의 변화와 유사하였다. ALAD활성은 암하에서보다 광선하에서 억제되었으나, 18시간 암처리후의 광조사는 ALAD활성을 증가시켰다. Putre-scine처리에 의한 ALAD활성은 암하에서 촉진효과가 낮았으나 광선하에는 그 활성이 증가되었다. KCl은 암하에서 ALAD활성을 촉진시켰으나 광선하에서는 그 효과가 감소되었다. 또한 polyamine과 KCl의 복합처리에서는 촉진효과가 없었다. 이와같은 결과에서 녹두자엽에서의 색소체발달은 polyamine과 광선에 의해 영향을 받으며, polya- mine은 색소체발달에 중요한 작용을 하는 것으로 사료된다.

전립선암세포에 있어서 폴리아민에 의한 칼슘신호와 세포사멸 (Polyamine Induces Apoptosis Through the Calcium Signaling in Human Prostate Cancer Cells)

  • 폴리아민은 거의 모든 세포에 있어서 성장과 분화에 필수적인 물질이다. 이들 폴리아민의 기작은 상당히 복잡하고 다양하여 그 정확한 작용기전은 아직 확실하지가 않다. 본 논문에서는 폴리아민이 세포 증식을 유도 하기도 하지만 일정농도 이상에서는 오히려 세포사멸을 초래한다는 결과를 밝히고자 한다. 본 실험에 사용된 인간의 전립선 암세포(LNCaP cells)에 있어서, 폴리아민 가운데 putrescien은 세포 증식에 거의 영향을 미치지 않았으나 spermidine과 spermine의 경우 10 ${\mu}M$ 이하에서는 세포 증식을 촉진하였다. 그러나, 20 ${\mu}M$ 이상의 농도에서는 농도와 시간의존적으로 세포사별을 유도 하였다. 폴리아민 처리에 의한 세포사멸의 초기과정인 핵 응축과 염색질 condensation이 Hoechst와 PI 염색에서 뚜렷이 관찰되었다. 또한, 폴리아민 처리시 anti-apoptotic protein으로 알려진 Bcl-2 protein의 발현은 거의 완전히 억제된 반면, pro-apoptotic protein으로 알려진 Bax의 발현은 현저히 증대되었다. 본 연구의 결과에 따르면, 폴리아민에 의해서 유도되는 세포사멸은 세포 내 칼슘농도 변화에 의한 것으로 사료된다. 전립선 암세포에 있어서 폴리아민 처리시 시간과 농도 의존적으로 세포 내 칼슘농도가 증가되었다. 세포막을 통한 칼슘이동을 억제하는 nifedipine과 flufenamic acid 등의 억제제를 처리한 실험 결과 세포내 칼슘증가는 세포 내부의 저장소로부터 칼슘의 유출보다는 주로 세포막상에 있는 비선택적 칼슘통로를 통한 외부의 칼슘 유입에 의한 것으로 판단된다.

Effects of Dietary Fats and Fibers on Modulation of Biomarkers and Tumor Incidence in Rats during 1, 2-Dimethylhydrazine-Induced Colon Carcinogenesis

  • This study investigated the effect of different dietary fats and fibers on colon tumor incidence and cell proliferation, the levels of eicosanoids and polyamines in colonic mucosa of DMH-treated rats. The experiment was conducted on male Sprague Dawley rats using a 2 $\times$3 factorial design with two fats (corn oil and DHA-rich fish oil) and two fibers (cellulose and pectin) and a fiber-free control. The rats were find an experimental diet containing 15% (w/w) dietary fat and 6% (w/w) fiber for 25 weeks. Tumor incidence was Bower in rats fed fish oil as opposed to corn oil. The levels of arachidonic acid (AA) and eicosanoids ($PGE_2, and TXB_2$) in normal colonic mucosa were significantly lower in rats fed fish oil and there was a concomitant increase of docosahexaenoic acid (DHA). The levels of eicosanoids and AA in tumor tissues were significantly higher than those of normal colonic mucosa. The level of polyamines in normal colonic mucosa was not affected by dietary fats but was significantly lower than that in rumor tissues. Dietary fiber did not have a significant effect on rumor incidence and the levels of AA, eicosanoids and polyamines. Overall, fish oil rich in DHA reduced cell prolifiration and thus inhibited colon carcinogenesis through its effect on the distribution of AA and production of eicosanoids in normal colonic mucosa. However, its effect on colon carcinogenesis revealed a lack of consistency depending on the type of dietary fiber in diet.
