Effects of Inhibitors (DFMO, DFMA) on Polyamine Synthetic Enzymes (ADC, ODC) during Ovarian Development of the Tobacco Budworm, Helicoverpa assulta (담배나방의 난소발생시 폴리아민 합성효소에 미치는 저해제의 효과)

  • Effects of $\alpha$-difluoromethylornithine (DFMO) and $\alpha$-difluoromethylarginine (DFMA), inhibitors of polyamine syntheic enzymes (ornithine decarboxylase, arginine decarboxylase), on ovary were investigated during pupal-adult development of Helicoverpa assulta. Two inhibitors (DFMO, DFMA) showed definite inhibition effects on ovarian development. The inhibition effect on ovaries weight was more marked in DFMA-injection than that observed in DFMO-injection. Two inhibitors (DFMO, DFMA) gave rise to a peculiar decrease in ornithine decarboxylase (ODC) or arginine decarboxylase (ADC) activity in ovaries, 72 hrs (5-day old pupa) post injection. However, DFMO clearly exhibited supression of ODC activity after 96 hrs (6-day old pupa). In addition, two inhibitors (DFMO, DFMA) diminished putrescine content in the ovary. The spermidine level was slightly decreased by each injection of the inhibitors. However, two inhibitors (DFMO, DFMA) raised the spermine content at certain developmental stages in the ovary. Although the effect of DFMA was less severe, two inhibitors (DFMO, DFMA) caused not only an overall delay in ovarian development, but also abnormalities in cellular differentiation. Noted effects in the pupal ovary were the appearance of irregular nurse cells and partial destruction of follicle epidermal cells. Adult ovary showed rapid degradation of nurse cells, a reduction in the number of follicle epidermal cells and immature oocytes that had a low yolk content.

지료조성에 따른 AKD사이징의 효과

  • 1980년대 중반이후 국내에서는 기존의 로진-알람을 이용한 산성 초지 대신 중성 초지 로의 전환이 가속화 되어왔다. 현재 거의 모든 지종이 중성 초지 조건에서 생산되고 있으며 A ASA(alkenyl succinic anhydride)와 함께 대표적 중성 사이즈제로 널리 쓰이고 있는 A AKD(alkyl ketene climer)를 이용한 중성 사이징이 주류를 이루고 있다. 따라서 본 연구에 서는 국내에서 가장 많이 사용되고 있는 AKD를 이용한 중성 사이정의 효과를 파악하기 위 하여 지료 조성에 따른 AKD의 사이즈 발현 특성을 알아보고자 하였다. 공시펼프로는 미세분이 적은 미고해 NBKP와 신문고지를 실험실에서 탈묵한 DIP를 사 용하였으며, 정착제로는 PAM 및 분자량과 전하밀도가 서로 다른 3종의 Polyamine을 사용 하였다. 충전제로는 현장에서 사용되는 공업용 Clay, Talc와 탄산칼습을 사용하였다. 각기 다른 지료 조성에서 AKD 사이즈도의 발현과 효과를 조사한 결과 다음과 같은 결론을 얻었다. 일반적으로 미세분의 제거, 섬유손상의 억제, 적절한 정착제의 사용, 열처리 등의 조건 들이 갖추어 질 때 AKD의 사이정 효율을 보다 높일 수 있었다. 탄산칼습이나 Talc같은 낮 은 표면 에너지를 가지는 충전물들은 AKD 사이정에 긍정적인 효과를 나타내었다. 정착제 로서의 PAM의 첨가는 DIP에서 Polyamine보다 낮은 사이즈도를 보였으나 NBKP에서는 더 효과적이였다. Polyamine은 소량 투입만으로 일정수준의 사이즈도를 나타낼 수 있는 정착제 로 특히 DIP 사이정에 상당히 효과적이였으나 사용량과 충전물의 첨가에 매우 민감하게 반 응하였다.

Cell Biological Studies on Growth and Development Effect of$Ca^{2+}$ and polyamine of $\beta-glucan$synthetase activity in carrot root protoplast (생체생장에 관한 세포생물학적 연구 당근 뿌리의 원형질체에서 polyamine과 $Ca^{2+}$$\beta-glucan$ synthetase 활성도에 미치는 영향)

  • The effect of polyamine, Ca2+ and calmodulin on GS ($\beta$-glucan synthetase) activity was studied in Daucus carota root. The Ca2+ is shown to have no effect on the GS activity whereas the GS II activity is increased in response to increase in concentration of the Ca2+. When the protoplasts are cultured, for 4 days, the GS II activity increases as a tunction of time and reachs a maximum after 3 days at a time when the network of cellulose microfibrils is known to be synthesized. The effect of the Ca2+ and 1mM spermine on the GS II activity turns out to be synergistic, especially more synergistic at lower concentration of the Ca2+. The GS II activity seems to be enhanced by the Ca2+. The GS II activity in the protoplast treated by the calcium channel blocker, verapamil, turns out to be lower than that of the control. Cumulative results suggest that the Ca2+ stimulates the cell wall regeneration via enhancement of the GS II activity responsible for synthesizing the cell wall component throught synergistic effect with spermine.

Chemopreventive Effects of the Extracts from Soybean Fermented with Basidiomycetes (들송이(Agrocybe cylindracea)와 말똥진흙버섯(Phellinus igniarius)을 배양한 대두추출물의 암예방 효과)

  • The inductions of phase II enzymes, such as NAD(P)H : quinone reductase (QR), glutathione S-transferase (GST), glutathione (GSH) level and the inhibition of polyamine metabolism were tested for the chemopreventive potentials of the extracts from the soybean fermented with Agrocybe cylindracea (AC) or Phellinus igniarius (PI). The soybean fermented with AC or PI was potent inducer of QR activity in murine hepatoma Hepa1c1c7 cells. GST activities of the extracts from soybean fermented with AC or PI were higher than that of the extract from soybean not fermented with basidiomycetes. In addition, GSH levels of the extracts from soybean fermented with AC or PI were increased about 1.2 fold or 1.4 fold, respectively. In addition, proliferation of Acanthamoeba castellanii in a broth medium was inhibited by the extracts from soybean fermented with AC or PI at the both concentration of 20 and 40 mg/3 ml. These results suggest that soybean fermented with AC or PI may have chemopreventive potentials by inducing QR activity, increasing GSH and GST levels and inhibiting polyamine metabolism.

Effects of Methylglyoxal-bis (Guanylhydrazone) and Ethylene Synthesis Inhibitor on Adventitious Root formation from Soybean Cotyledon (Methylglyoxal-bis (Guanylhydrazone)와 에틸렌 생합성 저해제가 대두 자엽 부정근 형성에 미치는 영향)

  • The effect of methylglyoxal-bis (guanylhydrazone) (MGBG) and ethylene synthesis inhibitors on adventitious root formation from soybean cotyledon in relation to ethylene production and endogenous polyamine content was investigated. Cotyledon explants cultured on rooting medium formed numerous adventitious rook on the cut surfaces after 2 weeks of culture. However when cultured on rooting medium supplemented with MGBG, the root formation was strongly inhibited, its inhibitory effect was reserved when cultured on medium with MGBG + spermine, MGBG + CoCl$_2$ and MGBG + spermine+CoC1$_2$. A slight reversion of the rooting inhibition was observed in 10$^{-3}$ M MGBG +10$^{-5}$ M spermine treatment, whereas it caused a significant effect in 10$^{-3}$ M MGBG +10$^{-4}$ M treatment .Ethylene production and endogenous polymine content was investgated in 10$^{-3}$ M MGBG , 10$^{-3}$ M MGBG +10$^{-5}$ Mspermine, 10$^{-3}$ M MGBG +10$^{-4}$ M CoCl$_2$and 10$^{-3}$ M MGBG +10$^{-5}$ M spermine +10$^{-4}$ M CoCl$_2$treatments. Ethylene production highest in 10$^{-3}$ M MGBG +10$^{-5}$ M spermine treatment was higher than control. In 10$^{-3}$ M MGBG +10$^{-5}$ M spermine + 10$^{-4}$ M CoCl$_2$ treatment, ethilene production was lowest, whereas polyamine level was highest.

Effect of Amino Acid, Polyamine, and Flavonoid on the Pollen Germination of Peach(Prunus persica SIEB under Low Temperature Conditions (아미노산, Polyamine 및 flavonoid 첨가가 복숭아 화분의 저온 발아에 미치는 영향)

  • The objective of this study was focused on the enhancement of pollen germination frequency in peach (Prunus $persica\;_{SIEB}$) under low temperature conditions. The effect of factors such as amino acid, polyamine, and flavonoid on the pollen germination was investigated, and the results are summarized as follows. When amino acid, polyamine or flavonoid was added to the germination medium at $10^{\circ}C$, pollen germination frequency was strongly promoted. Optimum concentration of each supplement for pollen germination enhancement was $100\;mg{\cdot}L^{-1}\;H_3BO_3$, 10 mM asparagine, 10 mM glutamine, 100 mM spermine, $1000\;{\mu}M$ putrescine, and $1.0\;{\mu}M$ kaemferol. The best combination of factors in pollen germination was $100\;mg{\cdot}L^{-1}\;H_3BO_3+10\;mM$ asparagine, followed by $100\;mg{\cdot}L^{-1}\;H_3BO_310mM$ glutamine, $100\;mg{\cdot}L^{-1}\;H_3BO_3+200mM$ spermine, and 10 mM asparagine. These combinations promoted pollen germination by 18% in 'Nagasawa-Hakuho', and 19% in 'Shuho' compared to their germination percentage on the basal medium.
