수화 겔의 유변성질에 있어서 합성 조건과 수화 물 양의 영향이 연구되었다. cone-plate 레오메타를 사용하여 polyacrylamide 수화 겔의 비 뉴톤 유동 곡선을 얻었다. 이렇게 얻은 유동곡선에 유동에 대한 비뉴톤식을 적용하여 유변 파라메타를 계산하여 얻었다. polyacrylamide 수화 겔은 전단속도가 증가함에 따라 구조가 약해지는 흐름변성 현상을 나타낸다. 이들 유동성질은 유동단위의 특성과 유동 분절사이의 상호 관계에 의해서 나타나는 물질적인 성질이다.
We have investigated the rheological behaviours of emulsions containing polymers. The polymers used in this study were polyacrylamide and carboxy vinyl polymer. The emulsion with polyacrylamide shows better prior preference than that with carboxy vinyl polymer on the skin texture. In the toxicity test, such as, LLNA cell toxicity, skin organ culture and human patch test, polyacrylamide was the appropriate materials for the cosmetics. The viscosity of polyacrylamide shows maximum value in neutral pH and good stability against temperature. The decreasing effect of viscosity against salt was not good enough but showed a better result than that of carboxy vinyl polymer. Two times the amount of polyacrylamide should be used for the formation of a similar viscosity of carboxy vinyl polymer. Yield value of polyacrylamide was low and the effect for the stabilizing emulsions was also low.
Two polyacrylamide degrading bacterial strains, No. 2 and No. 11, were isolated from soil, and identified as Bacillus sphaericus No.2 and Acinetobacter sp. No. 11, respectively. Both strains grew on medium containing polyacrylamide as sole carbon and nitrogen sources. B. sphaericus No. 2 and A. sp. No. 11 reduced by 16% and 19%of the initial polyacrylamide concentration, respectively. Optimal pH and temperature in growth of Acinetobacter sp. No. 11 were 8.0 and $37^{\circ}C$, respectively. After 14-day cultivation of A. sp. No. 11, the average molecular weight of polyacrylamide has been shifted from $2.3{\times}10^6\;to\;0.5{\time}106$.
Diffusion and partition of progesterone into the polyacrylamide gel was examined. Diffusion coefficient of progesterone decreased down to an asymptotic value as the concentration of the organic solvents in the diffusing medium increased. However the partition coefficient diminished steadily. Crosslinking density in the gel didn't affected the diffusion coefficient considerably but lowered the partition coefficient due to the contraction of pore volume of the gel. Progesterone showed higher diffusion coefficient as well as partition coefficient in the polyurethane than in the polyacrylamide gel, which seems to be ascribed to the difference in hydrophobicity, pore volume and pore size of the polymer matrix.
본 실험은 보리품종의 구분에 가장 적합한 전기영동법과 효소를 구명하고자 6개 보리품종(알찬보리, 수원 216호, 조풍보리, 사천 6호, 향맥, 히노데하다가)의 종실 단백질을 7.5% polyacrylamide slab gel, 2-30% polyacrylamide porosity gradient tube gel, isoelectricfocusing(pH 4-9)과 starch gel을 사용 전기영동 후 protein band pattern과 esterase, acid phosphatase, malate dehydrogenase, glutamate dehydrogenase 및 leucine aminopeptidase의 동위효소 pattern을 관찰하였던 결과는 다음과 같았다. 1. 7.5% polyacrylamide slab gel에서 단백질 pattern과 2-30% polyacrylamide porosity gradient tube gel에서의 단백질과 esterase의 band pattern이 품종간 뚜렷한 차이를 보여 보리품종구분에 가장 적합하였다. 2. Acid phosphatase, malate dehydrogenase, glutamate dehydrogenase 및 leucine aminopeptidase 등의 동위효소 pattern은 모든 실험한 품종이 동일한 형태를 보여 품종구분에 이용하기에는 적합하지 않았다.
Objectives : This study was conducted to carry out Purification of Melittin and other peptide components from Bee Venom using gel filtration chromatography and propionic acid/urea polyacrylamide gel electrophoresis Methods : Melittin and other peptide components were separated from bee venom by using gel filtration chromatography on Sephadex G-50 column in 0.05M ammonium acetate buffer. Results : Melittin and other peptide components were separated from bee venom by using gel filtration chromatography on Sephadex G-50 column in 0.05M ammonium acetate buffer. The fractions obtained from gel filtration chromatography was analyzed by using SDS-PAGE and propionic acid/urea polyacrylamide gel electrophoresis. The melittin obtained from the gel filtration contained residual amount of phospholipase $A_2$ and a protein with molecular weight of 6,000. The contaminating proteins were removed by the second gel filtration chromatography. Conclusion : Gel filtration chromatography and propionic acid/urea polyacrylamide gel electrophoresis are useful to separate peptide components including melittin from bee venom.
연근별로 인삼 단백질의 생화학적 특성을 체계적으로 규명하기 위하여 polyacrylamide gel 전기영동, SDS-PAGE, gel filteration 및 아미노산 자동분석기에 의한 실험결과는 다음과 같다. 인삼의 수용성 단백질을 Sephadex G-200으로 분획하여 얻은 2개의 peak 중주 단백질은 SDS-polyacrylamide gel 전기영동에 의해 분자량은 43,000으로 추정되었다. Polyacrylamide gel 전기영동 및 SDS-PAGE에 의한 band의 수는 연근에 따라 별 차이 없이 각각 8 및 $7{\sim}11$개 확인되었다. 인삼의 수용성 단백질과 그의 주 단백질을 아미노산 분석한 결과 arginine이 각각 45.17과 57.36%로 최대의 함량을 나타내었고 methionine과 cystine의 함량은 매우 낮았다.

Polyacrylamide의 농도에 따른 2개 Band의 log mobility의 plot는 charge isomer를 가리키는 평행선을 나타내었다. 4. 두 효소단백질 Band의 작용 pattern을 알기 위하여 2개의 Band를 각각 분리하여 추출하고 soluble starch에 작용시켜 생성된 oligosaccharide의 pattern을 paper chromatography로 확인한 바 2개의 효소단백질 Band는 동일한 작용 pattern을 나타내었다. 5. Soluble starch로부터 생성되는 유일한 초기 가수분해산물은 maltohexaose이었다.
This experiment was carried out to improve the quality of high absorbent polymer which has excellent absorptivity and water retention compared to pulp and absorption sheet, through absorption characteristics-absorptive power, water retention, absorption rate, gel strength, pH, particle size, and moisture content. - of six polymers, namely, anionic polyacrylamide, cationic polyacrylamide, polyacrylic, acid polyvinyle alcohol 500, and 1500, and a-cellulose. and to examine the possibility of substitution of amide groups for carboxyl group and/or hydroxyl group which were commercial high absorbent polymer by comparing the absorption characteristics of the polymers. Polyacrylamide has high absorptive power and water retention, but has low gel strength and poor absorption rate. The rest of polymers were similiar to ${\alpha}$-cellulose in every respect. Thus, polyacrylamides could be replaced with polyacrylic acid and polyvinyl alcohol which are presently a high absorbent polymers. In comparing the absorption characteristics and the absorptive power of the polymers-anionic polyacrylamide, cationic polyacrylamide, polyacrylic acid, polyvinyle alcohol. a-cellulose-the absorptive power was in inverse proportion to the gel strength and absorption rates, affected by the particle size and pH change.
This experiment was executed to investigate the possibility of the application of taxonomic method through the isoelectric focusing with polyacrylamide gel and sodium dodecyl sulfate-polyacrylamide gel electrophoresis with seeds in the identification of turfgrasses. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis was used to investigate the pattern of seed proteins which were extracted from 18 cultivars of cool season turfgrass and 4 cultivars of warm season turfgrass. The isoelectric focusing with polyacrylarnide gel was used to investigate the activity of the three isozymes of esterase, peroxidase and phosphoglucose isomerase which were extracted from 18 cultivars of cool season turfgrass and 4 cultivars of warm season turfgrass. The results were summarized as follows. 1. The difference of the patterns of seed proteins was observed with sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The identification of intra-genus was easily detected. 2. The three isozymes of esterase, peroxidase and phosphoglucose isomerase were investigated through isoelectric focusing with polyacrylamide gel. As a result, esterase was most effective among three isozymes in the identification of turfgrass cultivars 3. In the past cultivar identification was primarily based on visual morphological characters, but there was a lot of difficulty. If we should use electrophoresis, we will be able to identifvturfgrass cultivars more effectively.
