Studies on the Interspecific and Intergeneric Hybridization in Herbage Grasses II. Morphological physiological charateristics of Italian ryegrass X tall fescue hybrid (화본과목초의 종.속간 잡종에 관한 연구 II. Italian ryegrass X tall fescue F1 잡종의 형태 및 생리적 특성)

  • Tall fescue(Festuca amndinacea Schreb.) is a productive pasture grass that is widely adapted to the soil and climate of Korea. It is regarded, however, as relatively low in palatability and nutritive value. On the other hand, Italian ryegrass(lo1ium rnultz$om Lam.) is highly nutritious and palatable, but is essentially weak in cold tolerance. Numerous investigators have succeeded in producing hybrids between Lolium spp. and Festuca spp. but no improved varieties have resulted. Therefore, intergeneric hybridization of Italian ryegrass with tall fescue is employed in 1984 to improve the adaptive and cold resistance quality of Italian ryegrass. In the mean time, F1-hybrid plants obtained by crossing Italian ryegrass x tall fescue were observed in open field. The results are summarized as follows: 1. Leaf-size and silica cell hair were intermediate between the types of parents but the shape of auricle tended to show Italian ryegrass and auricle-cilia to show tall fescue. 2. Roots of $F_1$-hybrid plants were mainly distributed at top soil and did not produced florescence material. 3. Head type of hybrids was intermediate between the types of parents but ray length was somewhat less than the length of tall fescue ray. Typical Italian ryegrass or tall fescue-like phenotypes were not observed. 4. Anthers of $F_1$-hybrid plants were formed but pollen were not formed. Even though pollen were formed, they were all empty. 5. Hybrid plants were more winterhardy and summer persistency than Italian ryegrass. They showed a equivalent growth to Italian ryegrass in spring and to tall fescue in summer. 6. Chromosome of $F_1$-hybrids was allopolyploid(2n=4~=28) and all $F_1$-hybrid plants were sterile.

Comparison of IgE induction in mice by pollens from three pine tree species

  • Background: Over the years, pine pollens have been excluded as an allergen due to its relatively large size, low protein content, and waxy hydrophobic layer, despite their abundance. However, recent studies suggest the possibilities of pine pollens being allergens, and it has been reported that allergy symptoms were highly prevalent in areas with considerably large pine forests and high possibility of exposure to the pollen. Therefore, we conducted a comparative analysis of the allergenicities of the pollens from the dominant species of Korean pines, red pine (Pinus densiflora), black pine (Pinus thunbergii), and pitch pine (Pinus rigida), in mice. Methods: The protein composition of the pollens from the three pine species was compared via sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The pine pollens and proteins extracted from the pollens were introduced to BALB/c mice by nasal inhalation and application to exposed skin and the IgE produced by the mice were extracted from blood and analyzed via ELISA. Results: SDS-PAGE showed differing protein compositions of the pollens of the three pine species. Analysis of blood IgE compositions showed a similar amount of IgE produced when pollens were applied to skin. In contrast, when mice inhaled the pollens, P. densiflora was shown to induce significantly more IgE production than those of the other two species. Conclusions: The experimental results demonstrate that the pollens of all three South Korean pine species induce IgE production, and this production was more pronounced when the pollens were inhaled than when they were applied to the skin. Of the three species, the pollen of P. densiflora was found to induce the highest level of IgE production.

A Systematic Study of Elsholtzia Willd. (Lamiaceae) in Korea (한국산 향유속(Elsholtzia Willd., 꿀풀과)의 분류학적 연구)

  • Examined were the vegetative and reproductive characters such as external morphology, anatomy (leaf petiole, blade and stem), pollen and nutlet for the taxonomic revision of Elsholtzia in Korea. The inflorescence of all studied taxa of Elsholtzia in Korea is secund and 0.5-10 cm long. Leaves are ovate or nearly and $0.2-10{\times}0.2-6.0cm$. The shapes of inflorescence and leaves are very useful taxonomic characters. The stomata types are anomocytic and diacytic. The morphology of nutlet is ovoid and pitted in surface pattern of exocarp. All studied taxa are myxocarpy. The pollen grains are of medium size and subprolate in shape, and have typical bi-reticulate exine surface. Four species are recognized for Korean Elsholtzia by the identification key and descriptions in this study.

Comparison with in Vivo Pollen Development of Domestic Cultivars in Brassica Napus L. (국내육성 유채품종의 생체 내 화분발육 비교)

  • This study was showed into the pollen development with in vivo by bud size and genotype. Microspores of buds from 2.0 mm to 2.5 mm of all genotypes were composed of mainly tetrad cells and early uninucleate stage cells. Microspores derived from buds of 2.5-3.0 mm were exposed cells of early uninucleate, middle uninucleate, and late uninucleate. Microspores from buds of 3.0-3.5 mm contained mostly late uninucleate stage cells and showed some early binucleate stage cells. Microspores of buds with 3.5-4.0 mm in length were composed of mainly binucleate stage cells and decreased late uninucleate stage cells. Microspore with more than 4.0 mm were entered into binucleate stage cells of divided generative nucleus and vegetative nucleus. In 'Tamlayuchae', microspores derived from buds of 3.5-4.0 mm were observed cells of late uninucleate stage and early binucleate stage because of late microspore development. In MS-maintainer, the spring type, microspore derived from buds of 2.5-3.0 mm were observed tetrad stage cells.

Characteristics of Self-compatible Variety from Native Lilium tigrinum Thunberg (우리나라 자생 참나리에서 선발된 자가결실성 2배체 품종의 특성(特性))

  • This study was carried out to breed a self-compatible variety of Tiger Lily from the seedlings originated from Chinju city, Gyeongsangnam-do. The morphological characteristics, ploidy, and the resistance of seedlings to virus infection were investigated. A progeny test was also conducted to examine whether the propagated progenies had the same characteristics as the mother plant. The self-compatible diploid lily variety developed in this study showed a tall type like native triploid lily, Lilium tigrinum, and bulbils were formed on both lily varieties. The morphological characteristics of the flowers and leaves were not much different between the varieties, while the size of the bulbils and bulbs showed significant differences. The percentage of seed generation by self-pollination was 72.6% for the self-compatible variety, while there was no seed generated for native triploid lily. The number of chromosome was 2n = 26 (x = 13, diploid) for the self-compatible variety, while 2n = 39 (x = 13, triploid) in native lily variety. The progenies of the self-compatible diploid lily variety showed the same characteristics as those of its mother plant in morphology, seed germination, and polyploidy. The mother plant of the self-compatible diploid lily variety showed 58% pollen germination and the 2-year-old and 3-year-old progenies showed similar germination percentages. The pollen grains of Korean native triploid lily, however, never germinated.

Genetic Gain and Diversity in a Clonal Seed Orchard of Pinus Koraiensis Under Various Thinning Intensities (잣나무 클론 채종원에서 간벌 강도에 따른 개량효과와 유전다양성)

  • Estimates of genetic gain (in volume growth) and diversity (expressed as status number, $N_s$) were determined in a clonal seed orchard of Pinus koraiensis. The genetic thinning was based on clonal breeding values (represented by general combining ability) obtained from progeny tests, clonal fertility estimated by strobilus production, and clonal size variation determined by the ramet numbers per clone. Parental GCA values for volume growth were calculated, based on height and diameter at breast height measured from field trials. Clonal fertility was estimated from the assessments of strobilus production over twelve years from 1991 to 2003, and used for the calculation of status number. There are 179 clones and 5,268 ramets in 12ha area of P. koraiensis clonal seed orchard. Genetic gain and diversity estimates were determined under assumptions of 30% pollen contamination and inferior genetic value of contaminating pollen. Genetic gain increased as thinning rates were set from 10% to 60%. However, for the higher thinning intensities, the increase of genetic gain was not remarkable. Genetic thinning by means of truncation selection resulted in a greater genetic gain but a large decrease in status number. Status number was represented around 40 clones for 10% through 60% thinning intensities, but for the higher thinning intensities, it was a bit fluctuated. Based on the present results, it could be concluded that thinning rate should not be stronger than 60% to optimize genetic gain while conserving genetic diversity. Consequently 50% or 60% thinning rate might be appropriate for genetic thinning in the clonal seed orchard of P. koraiensis. The effect of pollen contamination on the genetic gain and the consequence of genetic thinning for seed production in the clonal seed orchard, and seed orchard management scheme were also discussed.

Comparative Study on the Runoff Process of Granite Drainage Basins in Korea and Mongolia

  • Dissected erosional surfaces are widely distributed in the western part of Korea (e.g. Icheon, Chungju, Jecheon, Seosan). The deposits with thickness of less than 2m occur on the smooth bedrock surface are composed of poorly sorted subangular gravels with less than 20cm diameter. However, only weathered mantle of granites without the gravel layer are observed at some outcrops. The results of grain size analysis of deposits of Icheon district revealed that the characteristic of the gain size distribution is very similar with the results of sheetflood deposits presented by Blair (1999) in the Death Valley. Loess layer with buried soil layers of MIS7 covers the sheetflood deposits. The loess layer implies that the sheetflood deposits occurred before MIS7 based on the typical Loess sequences presented by Naruse et al.(2003). On the other hand, the climate of Korean Peninsula in MIS2 was very dry and cold (Yoon and Hwnag, 2003) by pollen analysis. This is because Yellow Sea was completely emerged during the MIS2(e.g. Sau\ito, 1998). So, it is thought that the climate in Korean Peninsula of not only MIS2 but also other glacial ages such as MIS8 was similar with present Mongolian climates. Tanaka et al.(2005) pointed out that Hortonian overlandflow occurs in grass vegetated granite basin in Mongolia. Therefore, dissected piedmont gentle slopes in the western Korea were possibly formed by sheetflood erosion during probably MIS8 as pediment widely distributed in Mongolia.

A Palynological Taxonomy of the Korean Verbenaceae (한국산 마편초과 식물의 화분학적 분류)

  • This research is the palynotaxonomic revision to clarify palynological descriptions and relationships of taxa in Korean Verbenaceae. To do this, morphology, size, sculpture and aperture of pollen grain were investigated with scanning electron microscope. As a result, palynological key as to 5 genera 10 species drawn up. Palynological descriptions of Callicarpa dichotoma, Callicarpa mollis, Callicarpa chejuensis, Vitex chinensis and Caryopteris incana are near completion at first. Vervena officinalis are newly described and descriptions of Callicarpa japonica, Clerodendrum trichotomum, Vitex rotundifolia and Caryopteris divaricata are corrected. Palynological data of Korean Verbenaceae is useful to intrageneric relationships. Namely, as to genus Callicarpa, C. dichotoma and C. japonica are considered to be more advanced than C. mollis and C. chejuensis. As to genus Caryopteris, C. divaricata is more advanced than C. incana.

Analysis of Deposited Dust Particles using SEM/EDX (SEM/EDX 분석법을 이용한 침착먼지에 대한 분석사례)

  • In this study, the composition and morphology of deposited dust particles with size ranging from a few to tens ㎛ were investigated using SEM/EDX (scanning electron microscopy with energy dispersive x-ray spectrometer). Then deposited dust particles were classified into 8 groups: quartz, aluminosilicates, ca-rich, Fe/Ti oxide, carbon-rich, industrial particle, Fe-rich, and biogenic particle. The sources of deposited dust were high in the order of aluminosilicates 41% > biogenic 18% > Fe-rich 11% > quartz and C-rich 8% > industrial 7% > Fe/Ti oxide 5% > Ca-rich 1%. In particular, the ratio of biogenic particles was relatively high due to influence of pollen. The ratio of carbon-rich was 11% at YM site, 10% at MD site, and 4% at MO site, and the site close to the large emission source was high.

Cleaved Amplified Polymorphic Sequence and Amplified Fragment Length Polymorphism Markers Linked to the Fertility Restorer Gene in Chili Pepper (Capsicum annuum L.)

  • Cytoplasmic male sterility (CMS) in plants, which is due to failure to produce functional pollen, is a maternally inherited trait. Specific nuclear genes that suppress CMS, termed fertility restorer (Rf) genes, have been identified in several plants. In this study, Rfl-inked molecular markers in pepper (Capsicum annuum L.) were detected by bulked segregant analysis of eight amplified fragment length polymorphisms (AFLPs). Only AFRF8 was successfully converted to a cleaved amplified polymorphic sequence (CAPS) marker. This was named AFRF8CAPS and genotype determination using it agreed with that obtained with the original AFRF8. A linkage map with a total size of 54.1 cM was constructed with AFRF8CAPS and the seven AFLP markers using the Kosambi function. The AFRF8CAPS marker was shown to be closest to Rf with a genetic distance of 1.8 cM. These markers will be useful for fast and reliable detection of restorer lines during $F_1$ hybrid seed production and breeding programs in pepper.
