Streptococcus pneumoniae is one of the major pathogens in community-acquired diseases, and it contains several factors that promote its pathogenesis, including pneumolysin (PLY). PLY is a member of the cholesterol-dependent cytolysin family, which attacks cholesterol-containing membranes, thereby forming ring-shaped pores. Thus, it is a major key target for vaccines against pneumococcal disease. We cloned the PLY gene from S. pneumoniae D39 and inserted it into the pQE-30 vector. Recombinant PLY (rPLY) was overexpressed in Escherichia coli M15 and purified by $Ni^{2+}$ affinity chromatography. Similarly, a PLY-EGFP fusion gene was produced by inserting the EGFP gene at the 3' end of the PLY gene in the same vector, and the recombinant protein was purified. Sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) showed that both recombinant proteins were purified. rPLY exhibited significant hemolytic activity against 1% human red blood cells (RBCs). Complete hemolysis was obtained at 500 ng/ml, and 50% hemolysis was found with a 240 ng/ml concentration. In contrast, rPLY-EGFP did not show hemolytic activity. However, rPLY-EGFP did bind the RBC membrane, indicating that rPLY-EGFP lost hemolytic activity via EGFP fusion, while retaining its membrane-binding ability. These data suggest that PLY's C terminus is important for its hemolytic activity. Therefore, these two recombinant proteins can be extremely useful for investigating the toxin mechanism of PLY and cell damage during pneumonia.
The vibrational characteristics of a radial-ply (155SR13 4PR) and a biased-ply tire (6.15-134PR) were investigated for examining the effects of tires with different structure on the ride characteristics of the vehicle. The natural frequencies at the tread band, mode shapes, and damping factors of two tires at the state of plane vibration were determined experimentally. The test work was performed at four levels of the inflation pressure, ranging from 171.7 kPa to 245.2 kPa, and three levels of the vertical load, deviating by 10% from the standard load designated by the Department of Transportation of the United States of America. The following results were drawn by the analysis of the test results: 1. The first-order natural frequencies of the radial-ply and the biased-ply tires at the tread band were 112 Hz and 159 Hz, respectively, at the state o f the free vibration when the inflation pressure of 196.2 kPa was applied. It was known that the biased-ply tire has higher resonant frequency than the radial-ply tire and the natural frequencies of the both tires move to the high frequency range as t he inflation pressure is increased. 2. The vibration modes of both tires were quite different. No big difference in mode shapes was examined as the inflation pressure was increased. But the natural frequencies of two tires were changed. For the radial-ply tire, no difference in mode shape was found whether the vertical load was applied or not. But a significant difference in mode shape was examined for the biased-ply tire. 3. Any difference was not found in damping factor as the different inflation pressures were applied. 4. When no vertical load was applied, damping factors of the radial-ply and biased-ply tire at the state of the natural vibration ranged from 2.6 to 5.9%, and from 4.1 to 7.8%, respectively. It was estimated that the radial-ply tire would have better cushioning than the biased-ply tire since the vertical spring rate of the radial-ply tire was much less than that of the biased-ply tire, even though the damping effect of the radial-ply tire was smaller than that of the biased-ply tire.
This study was carried out to compare the properties of Com-Ply Boards with those of OSBs. These two kinds of boards were made of same flakes. Com-Ply Boards were covered with three different thickness veneers, such as 4mm, 3mm, and 2.5mm. All manufactured Com-Ply Boards and OSBs were 12mm thick. In MOR, all Com-Ply Boards were higher than OSB, and especially 3mm-veneer and 2.5mm-veneer Com-Ply Board were highest. But OSB showed 420kg/$cm^2$. 3mm-veneer Com-Ply Board showed the highest internal bonding 7.0kg/$cm^2$ and then 2.5mm-veneer showed 6.8kg/$cm^2$, and OSB showed 6.6kg/$cm^2$. Screw holding strength showed the high value 23.9kg/$cm^2$, 25.5kg/$cm^2$, and 29.3kg/$cm^2$ respectively at 3mm-veneer, 2.5mm-veneer Com-Ply Board and OSB. Thickness swelling of Com-Ply Boards was lower than that of OSB, but Water absorption of OSB was lower. Finally it was very encouraging that all properties of OSB without veneer, whose cost is higher than flake and whose properties is better, were not worse than those of Com-Ply Board with veneer. It seems That more researches on OSB will be required.
Urea-formaldehyde (UF) resin was formulated similarly to plywood resin in the laboratory. The synthesized UF resin adhesive was mixed with extender, filler and acid catalyst. The mixture contained 56.1% total solids and 43.9% water. The mixes was used to bond five Com-Ply types using Korean wood species. The Com-Ply made were tested for shear strength and wood failure according to KS F 3101 ordinary plywood as well as for bending strength per KS F 3104 particleboard. The performance test results showed good strength properties for all Com-Ply types made in this study. This result represented that the UF resin adhesive mix was adequate for bonding Com-Ply with domestic wood species.
This study aims to accurately predict the first ply failure loads of laminated composite hypar shell roofs with different boundary conditions. The geometrically nonlinear finite element method (FEM) is used to analyse different symmetric and anti-symmetric, cross and angle ply shells. The first ply failure loads are obtained through different well-established failure criteria including Puck's criterion along with the serviceability criterion of deflection. The close agreement of the published and present results for different validation problems proves the correctness of the finite element model used in the present study. The effects of edge conditions on first ply failure behavior are discussed critically from practical engineering point of view. Factor of safety values and failure zones are also reported to suggest design and non-destructive monitoring guidelines to practicing engineers. Apart from these, the present study indicates the rank wise relative performances of different shell options. The study establishes that the angle ply laminates in general perform better than the cross ply ones. Among the stacking sequences considered here, three layered symmetric angle ply laminates offer the highest first ply failure load. The probable failure zones on the different shell surfaces, identified in this paper, are the areas where non-destructive health monitoring may be restricted to. The contributions made through this paper are expected to serve as important design aids to engineers engaged in composite hypar shell design and construction.
This research was carried out to investigate the effects of cyclic humidification on the dimensional stability of particleboard manufactured according the three specific gravity levels and com-ply fabricated with a core of particleboard and veneer or plywood as the face and back materials. Both the particleboard and com-ply were subjected to four cycles of 50-90-50% relative humidity. The results are summarized as follows: Particleboard and com-ply followed a clear pattern of increasing dimensions and weight on the adsorption cycle and decreasing dimensions and weight on the humidification cycle. After the fourth cycle, the dimensions and weight of both particleboard and com-ply were greater than those originally measured. The greater part of increase in occurred during the first humidification cycle. The specific gravity of particleboard has a significant effect thickness and volume change. The dimensional stability of com-ply was better than that of particleboard. In addition, the dimensional stability of com-ply bonded with plywood was better than that of com-ply bonded with veneer.

The aim of this research is to evaluate the relationship between the total effective moment $(M^E)$ and Bemoulli-Euler bending moment (M) when the ply orientations of UD CFRP in Piezoelectric Zirconate Titanate Composite Actuator (PZTCA) are changed. The obtained results as follows. Firstly, as the performance test results by the CFRP ply orientation, the performance of [0] and [90] were stable. However, while the performance of [+45] was suddenly decreased after 5 hours. Secondly, the change of $M^E$ by the CFRP ply orientation was evaluated. As the CFRP ply orientation was increased from [0] to [+60], the $M^E$ were gradually decreased. However, they became a little bit increased from [+60] to [90]. Finally, after the change of M by the CFRP ply orientation was evaluated, it was found that $M^E=2.2M$ was valid for just [0] and that there was a relationship between $M^E$ and M according to the ply orientation.
In this study, a block shear strength test was conducted to compare and analyze the strength and failure mode on the glued laminated timber, CLT, and Ply-lam CLT, which are mainly used for the construction of wood construction as engineering wood. Through this, the Ply-lam CLT manufacturing conditions for optimum production, such as the type of lamina, plywood, adhesive, and layer composition, were investigated. The results are as follow. Through block shear strength test, it showed high strength in the order of glued laminated timber, Ply-lam CLT and CLT. In particular, the shear strength of Ply-lam CLT, which is made of a composite structure of larch plywood and larch lamina, passed 7.1 N/㎟, which is a Korean industrial standards for block shear strength of structural glued laminated timber. In addition, in this study, there was no different in shear strength according to the adhesive type used for glulam, CLT, and Ply-lam CLT adhesion. However, in the case of Ply-lam CLT, the difference in shear strength of Ply-lam CLT was shown according to the type of lamina and plywood. The results showed high strength in the order of Larix kaempferi > Mixed light hardwood ≒ Pinus densiflora, sieb, et, Zucc plywood. The optimal configuration of Ply-lam CLT is when larch plywood and larch lamina are used, and it is decided that the adhesive can be used by selecting PRF and PUR according to the application. The results of block shear strength failure mode by type of wood based materials were analyzed. The failure mode showed shear parallel-to-grain for glulam, rolling shear for CLT, and shear parallel-to-grain and rolling for ply-lam CLT. This is closely related to shear strength results and is decided to indicate higher shear strength in Ply-lam CLT than in CLT due to rolling shear.

In this research, the effects of fiber stacking sequence on damage behaviors of FML(Fiber Metal Laminates) subject to indentation loading. SOP (Singly Oriented Ply) FML and angle ply FML were fabricated to study fiber orientation effects and angle ply effects. FML were fabricated by using 1050 aluminum laminate and carbon/epoxy prepreg. To increase adhesive bonding strength, Al laminate was etched using FPL methods. The static indentation test were conducted by using UTM under the 2side clamped conditions. During the tests, load and displacement curve and crack initiation and propagation behaviors were investigated. As fiber orientation angle increases, the crack initiation load of SOP FML increases because the stiffness induced by fiber orientation is increased. The penetration load of SOP FML is influenced by the deformation tendency and boundary conditions. However, the macro-crack of angle ply FML was initiated by fiber breakage of lower ply because angle plies in Angle ply FML prevents the crack growth and consolidation. The Angle ply FML has a critical cross-angle which prevent crack growth and consolidation. Damage behavior of Angle ply FML is changed around the critical cross-angle.
