The pharmacokinetic properties of KR-V compounds, recently developed as new anti-HIV agents, were studied after i.v. and p.o. administration in rats. The concentrations of the KR-V series were determined in rat plasma using an high-performance liquid chromatography (HPLC)-UV detection system. Of the 19 KR-V compounds investigated in the present study, only KR-V 3, 10, 14, 16 and 18-1 showed oral bioavailability. The plasma concentration-time data could be adequately described by an one-compartment open model. In the i.v. kinetic study (10mg/kg), the CLt of KR-V 3, 10, 14 and 16 (>4L/hr/kg) were significantly higher than that of KR-V 18-1 (1.1 L/hr/kg). The AUC of KR-V 18-1 was greater ($8.97{\mu}g{\cdot}hr/ml$) than that of the other compounds, but the Vd (0.58 L/kg) was lower. In the p.o. kinetic study (50mg/kg), although the t-1/2 of KR-V 18-1 was shorter than that of the other compounds, the AUC ($3.659{\mu}g{\cdot}hr/ml)$ and $C_{max}(1.891{\mu}g/ml$) were markedly higher. In a seperated in vitro experiment, only KR-V 18-1, of the 5 compounds with bioavailibility, exhibits potent activity against HIV-1 mutant strains. Therefore, KR-V 18-1 is expected to become a new potent anti-AIDS drug candidate/lead compound.

The present study evaluated the effects of isoflavone-rich bean sprout on the lipid metabolism in ethanol-treated rats. Fifty male Sprague-Dawley rats were randomly divided into five groups: normal control, alcohol control, low soybean sprout+ethanol (low SS), high soybean sprout+ethanol (high SS) and isoflavone extract+ethanol (IE). They were fed experimental diets based on Lieber-DeCarli liquid diet for 40 days. Body weight, food intake and feed efficiency ratio (FER) of ethanol-treated groups were significantly suppressed compared with that of the normal control group. Among the ethanol-treated groups, high SS group showed in significant increase in the body weight, food intake and FER. Supplementation of isoflavone-rich soybean sprout powder or isoflavone extract significantly decreased plasma triglyceride (TG), total cholesterol (TC), atherogenic index (AI) and increased the ratio of HDL-cholesterol to TC. Supplements also significantly decreased total lipid, TG and TC in liver tissue compared with that of alcohol control. There was a significant decrease in hepatic lipid peroxidation products in IE group compared with other ethanol-treated groups. This results suggest that supplementation of isoflavone-rich bean sprout powder may exert beneficial effects on lipid metabolism in chronically alcohol-treated animals by improving lipid profiles in plasma and liver tissues.
Objectives : The aim of this study was to investigate the hypoglycemic and hypolipidemic activities and mechanisms of Acanthopanax senticosus (AS) herbal acupuncture. Methods : Anti-diabetic and anti-steatotic activity of the AS herbal acupuncture was investigated on C57BL/6J ob/ob mice. After random grouping at the age of 9 weeks, the herbal acupuncture groups were injected subcutaneously at the left and right Gansu (BL18) corresponding acupuncture points alternately on exactly the same time every day with 0.1ml of either 400 mg/kg or 800 mg/kg of AS (AS400 and AS800) for 8-week period. As a positive control, metformin was administrated at a dose of 300 mg/kg (MT300). Body weights were measured weekly, and on every other week blood was collected for blood glucose analysis. At the end of study, blood was also collected for determination of plasma insulin and lipid levels, after which they were killed and periepidydimal fat, liver, muscle, and pancreas were immediately removed. The removed tissues were instantly soaked in liquid nitrogen and stored at $-70^{\circ}C$ for morphological examination and mRNA analysis. Results : The AS herbal acupuncture significantly prevented weight gain on C57BL/6J ob/ob mice. The AS herbal acupuncture lowered blood glucose and improved glucose tolerance in C57BL/6J ob/ob mice. The increase of insulin response during the OGTT was inhibited by the AS herbal acupuncture. Insulin sensitivity of skeletal tissue was enhanced. Plasma lipid levels were significantly improved in the AS herbal acupuncture groups. The AS herbal acupuncture decreased hepatic lipogenesis and hepatic triglyceride production, and increased fatty acid (FA) transporter that involves in FA uptake. The AS herbal acupuncture inhibited the increase of liver mass by prevention of the accumulation of TG but did not inhibit weight gain of fat tissue on C57BL/6J ob/ob mice. Conclusion : In summary, we have demonstrated several unique properties of the AS herbal acupuncture in decreasing body weight, and reversing insulin resistance and hepatic steatosis in ob/ob mice. This AS herbal acupuncture acts as an insulin sensitizer and specifically decreases circulating glucose and lipids, and suppresses hepatic lipogenesis.
This experiment was conducted to verify whether or not chronic consumption of liquor prepared with Gastrodiae rhizoma exhibits antioxidant activity in brain tissue of rats. In the experiment, male Sprague-Dawley rats were given a liquid diet containing 6% (GA6), 18% (GA18), and 36% (GA36) of total calories as liquor for 6 weeks. The control rats (GAC6, GAC18, GAC36) received an isocaloric diet containing Soju instead of liquor. Caloric intakes and body weight gains of the 36% groups were significantly lower than those of the 6% and 18% groups (p<0.05), whereas alcohol intakes were significantly high in the 36% group (p<0.05). MDA contents of plasma and brain showed a positive correlation with alcohol intakes of the liquor and Soju consuming groups. Especially, contents of MDA were significantly higher in the 18% and 36% groups than the 6% group (p<0.05). SOD activities of serum and brain were significantly highest in the 18% group, and they tended to be higher in the liquor consuming groups compared with the control group. Catalase activity of serum showed no significant changes in the three groups consuming liquor, and catalase activity of brain tissue significantly increased in the GA18 group compared with the control group (p<0.05). In the liquor consuming groups, TAC level of serum tended to be higher in the GA36 group compared with the control group (p<0.05). However, TAC level of brain tissue showed no significant changes between the liquor consuming and control groups. These results show that as alcohol intake increased by chronic administration of liquor and Soju, brain tissue of rats was damaged more due to oxidative stress. However, a liquid diet containing 18% of total calories as liquor instead of Soju is expected to have a protective effect on brain damage induced by oxidative stress.

The pharmacokinetics of recombinant human granulocyte colony stimulating factor (rhG-CSF) following intravenous (i.v.), intramuscular (i.m.) and subcutaneous (s.c.) administration of HM1041l-lyo and HM10411-liq (lyophilized and liquid formulations of rhG-CSF, recently under development by Hanmi Pharmaceutical Company) were studied in rats, and compared with that of Filgrastim (conventional formulation of rhG-CSF on market). The plasma concentration of rhG-CSF was quantified using a specific ELISA. The pharmacokinetic parameters of rhG-CSF, after i.v., i.m. and s.c. administration of Filgrastim, HM1041l-lyo and HM1041l-liq to rats at a rhG-CSF dose of $10\;{\mu}g/kg$, were almost identical among the three formulations. No dose-dependency was observed in the pharmacokinetic parameters of rhG-CSF following i.v. administration in the dose range of $5{\sim}100\;{\mu}g/kg$. rhG-CSF, after i.v. administration of the three preparations at a dose of $10\;{\mu}g/kg$ to rats, was detected at low levels in all of the body tissues with highest tissue/plasma ratio of $0.46{\sim}0.51$ for the kidney at 30 min after the administration. The pharmacokinetics of rhG-CSF, after i.v. administration to mice at a dose of $10\;{\mu}g/kg$, were comparable among the three formulations. In conclusion, HM10411-lyo and HM10411-liq exhibited similar pharmacokinetics for rhG-CSF with Filgrastim regandless of animal species. Considering the fact that HM10411 series, contrary to Filgrastim, are proteins lacking a methionine residue, the methionine moiety in rhG-CSF molecule does not appear to influence the pharmacokinetics of the protein significantly.
A study on the oral bioavailability and pharmacokinetics of an anticoccidal agent, toltrazuril, was conducted in broilers following a single oral doses of 10 mg/kg body weight (BW) or 40 mg/kg BW. The concentrations of toltrazuril in plasma were determined by a high-performance liquid chromatography/mass spectrometry. Plasma concentration-time data after single oral administration were analyzed by a non-compartmental analysis. Toltrazuril was very well-absorbed through the gastrointestinal tract with $C_{max}$ of $18.04{\pm}5.80{\mu}g/mL$ and $47.15{\pm}9.40{\mu]g/mL$ at $4.33{\pm}1.51h$ and $3.67{\pm}1.15h$ after oral dose of 10 mg/kg and 40 mg/kg in broilers, respectively. A comparison between 10 mg/kg and 40 mg/kg dose groups showed that $t_{max}$ were similar while $C_{max}$ and area under curve (AUC) increased with increasing dose.

Two experiments were conducted to evaluate the effects of the level of corn dry distillers grains with solubles (CDDGS) supplementation on growing performance, blood metabolites, digestion characteristics and ruminal fermentation patterns in steers grazing dormant forage. In Exp. 1, of growth performance, 120 steers ($204{\pm}5kg$ initial body weight [BW]) were distributed randomly into 3 groups (each of 40 steers), which were provided with the following levels of CDDGS supplement: 0%, 0.25%, or 0.50% BW. All groups of steers were grazed for 30 days in each of 3 grazing periods (March, April, and May). Approximately 1,000 ha of the land was divided with electric fencing into 3 equally sized pastures (333 ha in size). Blood samples were collected monthly from 20 steers in each grazing group for analysis of glucose (G), urea-nitrogen (UN) and non-esterified fatty acids. Final BW, average daily gain (ADG) and supplement conversion (CDDGS-C) increased with increasing levels of CDDGS supplementation (p<0.05).The CDDGS supplementation also increased the plasma G and UN concentrations (p<0.05). In Exp. 2, of digestive metabolism, 9 ruminally cannulated steers ($BW=350{\pm}3kg$) were distributed, following a completely randomized design, into groups of three in each pasture. The ruminally cannulated steers were provided the same levels of CDDGS supplementation as in the growing performance study (0%, 0.25%, and 0.50% BW), and they grazed along with the other 40 steers throughout the grazing periods. The dry matter intake, crude protein intake, neutral detergent fiber intake (NDFI), apparent digestibility of dry matter (ADDM), crude protein (ADCP) and neutral detergent fiber (ADNDF) increased with increasing levels of CDDGS supplementation (p<0.05). The ruminal degradation rates of CP (kdCP), NDF (kdNDF) and passage rate (kp) also increased with increasing levels of CDDGS supplementation (p<0.05). Ruminal ammonia nitrogen ($NH_3$-N) and propionate concentrations also increased with increasing levels of CDDGS supplementation (p<0.05). However, acetate concentrations decreased with increasing levels of CDDGS supplementation (p<0.05). Liquid dilution rate increased with increasing levels of CDDGS supplementation but ruminal liquid volume decreased (p<0.05). On the basis of these findings, we can conclude that CDDGS supplementation enhanced the productive performance of cattle grazing native rangeland without negatively affecting forage intake, glucose and urea-nitrogen blood concentrations, ruminal degradation and ruminal fermentation patterns.

The FPDs(Flat Panel Displays) such as LCD(Liquid Crystal Display) and PDP(Plasma Display Panel) and OLED(Organic Light Emitting Diode), recently, have been substituted for CRT(Cathode Ray Tube) displays because they have a convex surface, small volume, light weight and lower electric power consumption. The productivity of FPDs is greatly dependent on the area of thin glass panel with 0.6 - 0.8mm thickness because FPDs are manufactured by cutting a large-scaled thin glass panel with patterns to the required product dimensions. So FPD's industries are trying to increase the area of thin glass panel. For example, the thin glass panel size of the 8th generation is 2,200mm in width, 2,600mm in length and 0.7mm in thickness. The air flows both in the thin glass panel and in the porous PE-plate surface were modeled and analyzed, from which a working condition was estimated. The thin glass panel on the porous PE-plate surface with self-lubricating characteristics was investigated and compared with that on the square duct floating bar surface with many holes of 1mm diameter when the thin glass panel contacts the floating bar surface due to malfunction of electric power supply.
This paper reports the amount of $^{222}Rn$ and $^{238}U$ in 18 sites of ground water and 30 sites of surface water. The instrument used to count $^{222}Rn$ activity was the liquid scintillation counter (LSC) which could resolute ${\alpha}$ and ${\beta}$ radiations. And $^{238}U$ was analyzed by the inductively coupled plasma (ICP). Radon and Uranium were not detected in raw and treated water which were sampled in a water treatment plant. However, radon ($^{222}Rn$) was high concentration in ground water from Jeon-la, Gang-won. So was uranium ($^{238}U$) in case of ground water from Gang-won, Choong-chung. Radon ($^{222}Rn$) activities were detected less than 15 pCi/L at 5 sampling points, 15~300 pCi/L at 7 sampling points, 300~4000 pCi/L at 6 sampling points. However, Radon ($^{222}Rn$) activities of all ground water samples were less than 4,000 pCi/L, which was bellow American Alternative Maximum Contamination Level (AMCL). Uranium ($^{238}U$) concentrations were less than $0.1{\mu}g/L$ at 5 sampling points, from $0.1{\mu}g/L$ to $20{\mu}g/L$ at 13 sampling points. Uranium was not detected in about 30% of the whole samples, but the concentration ranged from relatively low to high concentrations depending on the sampling point. The minimum detectable activity (MDA) of radon was 15 pCi/L. and the detection limit of uranium was $0.1{\mu}g/L$.

Mo isotope, one of highly redox-sensitive isotopes, has been shown to be useful tracers of geochemical processes. Many studies for Mo isotope have documented with the help of recently developed analysis tools, but it has not yet been documented in the Korea. In this study, we introduce two-stage column separation method of Mo using column tube (BioRad PolyPrep(R) column, 10 ml) and anion exchange resin (BioRad Resin AG(R) 1-X8, 200-400 mesh). Mo isotope ratios in the solid SRMs (BHVO-2, SDO-1, PACS-2) and liquid SRM (IAPSO) were measured on MC-ICP-MS (Multi-collector Inductively Coupled Plasma Mass Spectrometer) and then compared with reference Mo isotope ratios. Mo isotope ratios in our study overlap with reference Mo isotope ratios within analytical error.
