Control of Agrimony (Agrimonia pilosa) Powdery Mildew Using Cooking Oil and Yolk Mixture (난황유를 이용한 짚신나물 흰가루병의 방제)

  • We've been involved in collection and domestication of Agrimonia pilosa from 2005 to 2007. We observed agrimony powdery mildew caused by Sphaerotheca aphanis. The powdery mildew developed twice a year, in May to June and mid August to September on whole plants of A. pilosa plants, that is, leaves, stem and floral part. We sprayed the Cooking Oil & Yolk mixture (COY) to control the agrimony powdery mildew. When COY was single-sprayed to control agrimony powdery mildew, its control value was 26.9%. However when it was double-sprayed and triple-sprayed at seven days interval, its control value increased up to 80.8 and 82.7%, respectively. Upon the first spray, symptoms of powdery mildew disappeared within three days and suppressive effect of COY was persisted for only seven days. Thereafter, powdery mildew was relapsed to develop. However, on double to triple sprays, powdery mildew was suppressed more than 28 days after final treatment. Therefore, it is concluded that double spray of COY at seven days interval can control effectively agrimony powdery mildew.

Specific Primer Sets for RT-PCR Detection of Major RNA Viruses of Tomato Plants in Korea (국내의 토마토 주요 바이러스 진단을 위한 역전사중합반응법용 프라이머 세트)

  • Major tomato viruses in Korea are Tomato chlorosis virus (ToCV), Tomato spotted wilt virus (TSWV), Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), and Tomato mosaic virus (ToMV). RT-PCR conditions for the viruses were examined, especially in primer set and RT-PCR mixture. Total 46 primer sets from the unique sequence of the viruses were tested for nonspecific background products in a RT-PCR mixture without template. Among them 16 primer sets were applied to healthy tomato RNA, resulting the compatibility between RT-PCR mixture and primer set influenced RT-PCR to reduce nonspecific background products. Based on the combinations among cDNA synthesis parameters and RT-PCR mixtures, two reaction mixtures were finally selected for ToCV detection. The condition allowed to determine more specific primer sets; C029 (ToCV), C072 (TSWV), C070 (CMV), C048 (PepMoV), and C065 (ToMV). These primer sets are expected to be of use to specific detection of the major viruses in tomato plants.

Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants (유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색)

  • This study was designed to investigate antioxidative activity of substances isolated from 25 kinds of useful plants resources toward free radical and human low density lipoprotein(LDL). Methanol extracts of Oenothers odorate had the highest antioxidative activity similar with ${\alpha}$-tocopherol. Methanol extracts of Oenothers odorate was extracted again by the ethylacetate. The ethylacetate soluble acidic fraction obtained from methanol extract of Oenothers odorate showed highest activity toward human LDL. Each fraction was purified through Sepadex LH-20 chromatography by elution of chloroform-methanol mixture (90:10 v/v). Fraction, F-2 obtained from Oenothers odorate showed at highest levels of electron donating activity. Fraction, F-2 was identified as 3,4-dihydroxybenzoic acid and 3-hydroxycinnamic acid.

  • GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Cibotii Rhizoma for quality standardization of a medicinal crude drug GCSB-5. Onitin-4-O-$\beta$-D-glucopyranoside was isolated from Cibotii Rhizoma as the standard compound and identified on the basis of spectroscopic data such as NMR. HPLC analysis method for the determination of onitin-4-O-$\beta$-D-glucopyranoside was established for the quality control of the medicinal plants of Cibotii Rhizoma species, GCSB-5 raw material and GCSB-5 preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline.

Productivity of the Rice Plants at the Abandoned Crop Field Established from the Shattered Grains by Combine Harvesting (Combine 수확시 탈락볍씨의 경련 휴경조건하 자연상태에서의 수량성)

  • The rice plants (Oryza sativa L.) established from the shattered grains by combine harvesting at the previous year showed great variations of yield and yield components from site to site at the abandoned rice crop field. The cultural condition was very similar to direct seeding under no-tillage system but no cultural practices such as application of fertilizer, weed control, irrigation and drainage had been carried out. The highest yield of 188kg/10a was observed at one of the quadrats randomly located on the field, which showed the possibility of exploitation of no-tillage system. The interrelationship between crop growth and coverage of weed species was measured by calculating the correlation coefficients. The investigations of how to establish sufficient number of seedlings per unit area, plant succession on the abandoned crop field, crop mixture with legume crops, and breeding appropriate plant type of rice for the enhancement of competitive ability would be required for the success of no-tillage system.

  • In a rapidly changing fashion design world, creative ideas are always required. Knowledge has been created as an art, exhibiting a new imagination that surpasses reality, while being visualized from the past. The purpose of this study is to derive types of visualization of knowledge for continuous creation of fashion design. The study consists of literature and empirical studies. ATTA, a creativity evaluation method made by Torrance applied for analyzing of Gucci fashion design collections from 2016-2017. Creativity of the Gucci collection by ATTA evaluation items are that first, a vivid idea was revealed through collection history, myths and animal and plants, second, conceptual incongruity is in the composition of the garment, third, provocative questions are the symbolism of the meaning of the text, fourth, different perspectives derive a new formative beauty through the synthesis of twisted elements, fifth, abstraction is a symbolic expression of animals and plants, sixth, synthesis is a mixture of various materials and techniques by plural inspiration, seventh, context, it is developed as a design accompanied by stories of history and myth and eighth, fantasy is fictitious animals and animals and detail. In addition, formativeness of fashion design by visualization of this knowledge was extracted with contamination, symbolism, enjoyment and fabrication. Visualization of the knowledge is expected to be used as a strategy to attract ongoing ideas for creative fashion designs.

Relationship between Morphological and Physiological Characters of White Clover Cultivars with Different Leaf Size (잎의 크기가 다른 White Clover 품종들의 몇가지 형태 및 생리적 특성간의 관계)

  • Several benefits provided by white clover (Tnifoliurn repens L.) can be elevated as relationship between its morphological and physiological characteristics is clarified. The experiment was done to analyze the relationship between them in the clover with different leaf size. Individual plants of Osceola (large leaf), Grasslands Huia (medium leaf) and Aberystwyth S184 (small leaf) were grown in 15m plastic pot containing a 1:2:1 soil:sand:Promix mixture for 55 days, and then clipped to remove all fully expanded leaves every 7, 14 or 28 days. To measure the cultivar response, plants were sampled for morphological and physiological parameters on the date (0) after final harvest and 1, 3, 7, 14 and 28 days after the final harvest and then their relationship was analyzed. Osceola had greater leaf area per trifoliolate but less no. of leaves per plant to result in similar total leaf area per plant of all cltivars. Stolon length and no. of growing tips per plant declined with smaller leaf size although the result in biomass was reverse. Stolon of all cultivars showed the greatest fluctuation in total nonstructud carbohydrate during the regrowing period but nitrogen concentrations of all their fractions and dinitrogen fixation did similar patterns. Stolon and root of Osceola, moreover, were the highest ones. Biomass, stolon length and total leaf area per plant of all cultivars were positively correlated to carbohydrate concentration of all frauions and dinitrogen fixation. In Osceola relationship between nitrogen concentration of stolon and the characters showed in Osceola was positively done.

100 MWe Oxyfuel Power Plant Boiler System Process Design and Operation Parameters Sensitivity Analysis (100 MWe급 순산소연소 발전소 보일러계통 공정설계 및 운전변수 민감도 예측)

  • The oxy-fuel combustion is $CO_2$ capture technology that uses mixture of pure $O_2$ and recirculated exhaust as oxidizer. Currently some Oxy-fuel power plants demonstration project is underway in worldwide. Meanwhile research project for converting 125 MWe Young-Dong power plant to 100 MWe oxy-fuel power plants is progress. In this paper, 1 D process analytical approach was applied for conducting process design and operating parameters sensitivity analysis for oxy-fuel combustion of Young-Dong power plant. As a result, appropriate gas recirculation rates was 74.3% that in order to maintain normal rating superheater, reheater steam temperature and boiler heat transfer patterns. And boiler efficiency 85.0%, CPU inlet $CO_2$ mole concentration 71.34% was predicted for retrofitted boiler. The oxygen concentration in the secondary recycle gas is predicted as 27.1%. Meanwhile the oxygen concentration 22.4% and moisture concentration 5.3% predicted for primary recycle gas. As the primary and secondary gas recirculation increases, then heat absorption of the reheater is tends to increases whereas superheater side is decreased, and also the efficiency is tends to decrease, according to results of sensitivity analysis for operating parameters. In addition, the ambient air ingression have a tendency to lead to decline of efficiency for boiler as well as decline of $CO_2$ purity of CPU inlet.

Fiber-optic humidity sensor system for the monitoring and detection of coolant leakage in nuclear power plants

  • In this study, we developed a fiber-optic humidity sensor (FOHS) system for the monitoring and detection of coolant leakage in nuclear power plants. The FOHS system includes an FOHS, a spectrometer, a halogen white-light source, and a Y-coupler. The FOHS is composed of a humidity-sensing material, a metal tube, a multi-mode plastic optical fiber, and a subminiature version A (SMA) fiber-optic connector. The humidity-sensing material is synthesized from a mixture of polyvinylidene fluoride (PVDF) in dimethyl sulfoxide (DMSO) and hydroxyethyl cellulose (HEC) in distilled water. We measured the optical intensity of the light signals reflected from the FOHS placed inside the humidity chamber with relative humidity (RH) variation from 40 to 95%. We found that the optical intensity of the sensing probe increased linearly with the RH. The reversibility and reproducibility of the FOHS were also evaluated.

Agrobacterium tumefaciens Mediated Genetic Transformation of Pigeonpea [Cajanus cajan (L.) Millsp.]

  • Optimal protocol for efficient genetic transformation has been defined to aid future strategies of genetic engineering in pigeon pea with agronomically important genes. Transgenic pigeonpea plants were successfully produced through Agrobacterium tumefaciens-mediated genetic transformation method using cotyledonary node explants by employing defined culture media. The explants were co-cultivated with A. tumefaciens strain C-58 harboring the binary plasmid, pCAMBIA-1301 [con-ferring $\beta$-glucuronidase(GUS) activity and resistance to hygromycin] and cultured on selection medium (regeneration medium supplemented with hygromycin) to select putatively transformed shoots. The shoots were then rooted on root induction medium and transferred to pots containing sand and soil mixture in the ratio of 1:1. About 22 putative TO transgenic plants have been produced. Stable expression and integration of the transgenes in the putative transgenics were confirmed by GUS assay, PCR and Southern blot hybridization with a transformation efficiency of over 45%. Stable integration and expression of the marker gene has been confirmed in the TO and T1 transgenics through PCR, and Southern hybridization.
