Trends in antimicrobial sales for livestock and fisheries in Korea during 2003-2012 (국내 가축 및 수산에서 항생제 판매 추이(2003-2012))

  • In this study, trends in the sales of antimicrobials for use in livestock facilities and fisheries from 2003 to 2012 were investigated with regard to antimicrobial group, antimicrobial usage, and animal species. The overall amount of antimicrobials sold each year from 2003 to 2007 was 1,500 tons, after which they decreased, with the lowest sales being 936 tons in 2012. The total volume of antimicrobials used for feed additives decreased markedly by 94% from 2003 to 2012, which was mainly attributed to banning of feed additives. However, antimicrobial consumption through self prescription by farmers for disease prevention and treatment increased by 25% from 2003 to 2012. The largest volume of antimicrobials sold was for use in pigs (48~57%), followed by poultry (18~24%), fisheries (11~25%), and cattle (5~8%). Tetracycline was the highest selling antimicrobial, followed by penicillins and sulfonamides, although the overall sale of all three antimicrobials gradually decreased over the study period. This study demonstrated that the total consumption of antimicrobials has gradually decreased since 2008. Nevertheless, usage by nonprofessionals increased, which can ultimately cause emergence and spread of antimicrobial resistance. Thus, early establishment of veterinary prescription guidelines for prudent use of antimicrobials is urgently needed in Korea.

Prevalence state of canine brucellosis in South Korea during 2015 and 2016

  • We investigated the prevalence of canine brucellosis in companion and stray dogs between March 2015 and December 2016 and determined the disease characteristics based on the geographic distribution, sex and age of the dogs in South Korea. We conducted a large-scale survey using serological and bacteriological tests. Samples were collected from 2,394 dogs (1,825 companions and 569 strays). Thirty (1.3%) samples were positive for Brucella canis antibodies including 16 (0.9%) from companion dogs and 14 (2.5%) from stray dogs. Two (1.0%) of the 196 samples cultured from the stray dogs were positive. When compared with male dogs, the female companion and stray dogs had a significantly higher prevalence of brucellosis. Moreover, the prevalence of canine brucellosis was significantly higher in stray dogs older than 6 years and the prevalence of the disease in companion dogs was highest in Incheon (2.1%) and Jeolla (2.1%) provinces. Stray dogs from the Daejeon metropolitan area had the highest prevalence of brucellosis (7.9%). National control measures for canine brucellosis have not previously been implemented. Our findings suggest that appropriate screening tests and control measures are necessary to improve the health of dogs and to protect public health in Korea, particularly with the rapid growth of the companion animal industry.

Development of new analytical methods using high performance liquid chromatography for animal hormones; gonadorelin, progesterone, oxytocin and estradiol (고속액체크로마토그래피를 이용한 가축용 호르몬제(고나도렐린, 프로게스테론, 옥시토신, 에스트라디올) 분석방법 개발)

  • The aim of this study is to develop an optimal analytical method for gonadorelin, progesterone, oxytocin and estradiol, the major components of hormones. A relatively simple and reproducible method using high performance liquid chromatography was developed and as a result of the measurement of specificity, linearity, repeatability, accuracy and intermediated precision, the validity of the developed method was verified with the result of meeting the verification criteria of analytical method validation. Using this newly developed method, 12 post-market veterinary products were tested and the ingredient content were 91.9~116.4%, which satisfied the 90~120% condition of the administrative measure standard. Therefore, if the newly developed method is used for the collection examination of hormone in veterinary medicine, it can be useful as an approved test method.

Navel Orangeworm (Lepidoptera: Pyralidae) Intercepted on Fresh Oranges from the USA at the Korean Port of Entry (미국산 오렌지 생과실에서 검출된 Navel Orangeworm (나비목: 명나방과))

  • Navel orangeworm, Amyelois transitella (Walker) was intercepted at the Korean port of entry. It was detected by an inspection for a consignment of fresh oranges from the USA by the Busan Newport District Office, Yeongnam regional office, Animal Plant and Fisheries Quarantine and Inspection Agency on January 31, 2012. This is the first time that the larvae of this pest species has been detected on fresh oranges, although it has been occasionally detected on walnuts. This report aims to ensure biosecurity by providing diagnostic characters to identify the caterpillars and conducting an imported risk analysis on fresh oranges from the USA.

Infection of Foot and Mouth Disease of Serotype A in Farm-raised Deer in Korea (국내 사슴에서 구제역 A형 감염증례)

  • This report describes a case of foot and mouth disease (FMD) in farm-raised Sika deer. FMD serotype A occurred in six cattle farms in January 2010 since the outbreak of pig cases 2002 in South Korea. In addition to the six cattle farms where the disease occurred during January, positive reactions to FMD antibodies were found in two heads of deer on March at a deer farm in serological tests intended to lift the movement restriction of the susceptible animals imposed within risk zone. In the specimens collected from 12 heads for confirmatory tests in the same farm, no virus was detected in antigen tests and neutralising antibody titers in all raising deer in the farm were relatively high. So it seems likely that the animals had been infected with FMD at least one month earlier when they were found.

Validation of Korean Meat Products and Processed Cheese for the Detection of GMO using p35S and tNOS Primers

  • In this study, 543 samples of press hams, sausages, processed ground meat and processed cheese acquired from retail markets in Seoul and Gyeonggi province in Korea from 2005 to 2010 were monitored using a one-step multiplex polymerase chain reaction (PCR) method that involves the amplification of specific soya or maize endogenous genes and the amplification of 35S promoter (p35S) and nopaline synthase terminator (tNOS) for GMO detection. Among the 543 samples, 477 samples were amplified for maize and/or soybean endogenous genes. Although one sausage sample collected in 2008 showed amplification of tNOS, the result was assumed to be false positive based on the results from further tests of other sausage samples of the same brand. Our results demonstrate the absence of GM soya and/or maze of livestock products in the Korean market during 2005-2010. In addition, the one-step multiplex PCR using previously constructed primer sets appears to be useful as a screening method for the detection of GMOs in processed livestock products. However, more specific methods should be established and employed to detect the event-specific GM gene for positive reaction samples by screening tests in processed livestock products.

Intestinal T cell lymphoma in a cat, Korea (고양이 장에서 발생한 T 세포 림프종)

  • An 11 year-old male Korean short-haired cat was presented to local animal hospital due to weight loss, vomiting, and intestinal hypomotility. After the cat was euthanized by poor clinical outcomes, necropsy was performed at Animal and Plant Quarantine Agency. At necropsy, the stomach was enlarged and had some nearly complete pellet food and the yellow mucous contents. The lumen of the middle and lower parts of the jejunum became narrow. Histopathologically, medium-sized lymphoid cells with hyperchromatic nuclei enclosed by scant cytoplasm were diffusely proliferated from mucosa to serosa of the small intestine. These findings were mainly observed in the jejunum and slightly in the duodenum and ileum. The monomorphous lymphocytes were 1 to 1.5 times larger than red blood cells and had few mitotic figures. Metastasis of the tumor cells to other organs was not observed. In the result of immunohistochemical analysis for identifying the origin of tumor cells, CD3 was expressed, but $CD79{\alpha}$ was not detected in the infiltrated cells. This case was diagnosed as T cell intestinal lymphoma in a Korean short-haired cat based on the clinical signs, gross findings, histopathology, and immunohistochemistry.

Bilateral renal cystadenocarcinoma with lung metastasis in German sheperd dog (독일세퍼드견에서의 폐전이 소견의 양측성 신장 낭선암종)

  • Primary renal tumors are uncommon in dogs with prevalence rate of approximately 1%. Renal carcinoma originating from epithelium of proximal convoluted tubules are more likely to be affected to Middle-aged dogs (average age, 8y), males about twice as often as bitches. A 10-year-old, female, German Shepherd dog with history of anorexia, vomitting and hematuria was referred to the Animal Disease Diagnostic Division in Animal and Plant Quarantine Agency. The dog was necropsied and several organs were collected, fixed in 10% phosphate-buffered formalin, embedded in paraffin wax and sectioned for histopathology. Grossly, the kidneys were bilaterally enlarged ($18{\times}12{\times}8cm$; left, $18{\times}10{\times}8cm$; right). The numerous cysts varying sizes from 3 to 6 cm in diameter were protruding from the surface of both kidney. A large nodule ($10{\times}6{\times}6cm$) was discovered between cardiac and diaphragmatic lobe in the right lung. Immunohistochemical examination revealed strong positive reaction to cytokeratin and ki-67 in the nuclei of the epithelial tumor cells. But showed negative reactions to vimentin and CD10. Based on the pathological and immunohistochemical examination, we diagnosed as the bilateral renal cystadenocarcinoma in German shepherd dog.

Polioencephalomalacia diagnosed in necropsied cattle in Korea

  • Polioencephalomalacia (PEM) is a neurologic disease of ruminants diagnosed by pathological approach. There is very little information available to understand bovine PEM in Korea. In this study, we investigated the diagnostic rate and pathological features of PEM in 334 necropsied cases of cattle submitted to the Animal and Plant Quarantine Agency in Korea from 2015~2017. PEM was diagnosed only in 13 (3.9%) Hanwoo, the Korean native cattle, and sporadically occurred in our country. The disease was the most diagnosed in fall season, and the age of the cows ranged from 1 month to 3 year. In all PEM cases, softening, and yellow discoloration of the gray matter in the cerebrum and fluorescence at the sites using ultraviolet illumination were grossly observed. Microscopically abundance of glial cells with vacuolar large cytoplasm and neuronal necrosis were commonly observed. This study suggests that future studies are necessary to identify the cause and pathogenesis for the control of PEM in our country.

Isolation, characterization, and evaluation of Bacillus thuringiensis isolated from cow milk

  • Probiotics colonize the intestines and exert an antibacterial effect on pathogens. Therefore, probiotics could be used as a preventive agent against lethal infections. To isolate probiotic microorganisms, 116 bacterial strains were isolated from healthy cow's milk and were subjected to Gram-stain, morphology and biochemical analyses, Vitek analysis, and 16S rRNA analysis. One of the strains identified as Bacillus (B.) thuringiensis 87 was found to grow very well at pH 4.0~7.0 and to be resistant to high concentrations of bile salts (0.3~0.9% w/v). B. thuringiensis was susceptible to the antibiotics used in the treatment of bovine mastitis, yet it exhibited an antimicrobial effect against Staphylococcus (S.) aureus 305. Moreover, it protected mice from experimental lethal infections of E. coli O55, Salmonella typhimurium 01D, and S. aureus 305 through a significant induction of interferon-${\gamma}$, even at four-week post-administration of B. thuringiensis. Although oral administration of B. thuringiensis 87 did not provide significant protection against these lethal challenges, these results suggest that B. thuringiensis 87 could be a feasible candidate as a probiotic strain.
