Efficient Selection Method for Drought Tolerant Plants Using Osmotic Agents

  • An efficient method to select drought tolerant Korean native plants using in vitro culture system was established in this study. While the plant growths and root inductions of each plant were proportionately affected by concentrations of mannitol on in vitro culturing seven plant species to test tolerance to osmotic stress, growth index (GI) and number of root induction of Chrysanthemi zawadskii var. latilobum and Dianthus chinensis var. semperflorens plantlets were higher than the others in 125mM mannitol. In test with polyethylene glycol (PEG), plantlets of C. zawadskii var. latilobum and D. chinensis var. semperflorens showed higher GI and number of root induction than the others in 33.3mM. On testing whether the well grown plants under osmotic stress are tolerant to virtual drought stress, there were significant differences in the withering rates of C. zawadskii var. latilobum and D. chinensis and those of were Aster yomena and Centaurea cyanus after 12 days without watering. It was found that significantly lower stomata numbers were shown in both drought tolerant plants than the sensitive plants. Averages of the stomata circumferences and the stomata area in the plantlets of the tolerant species were larger than those of the sensitive plants D. chinensis var. semperflorens showed the lowest transpiration level per unit area. The highest stomatal area per unit area was found in C. zawadskii, followed by D. chinensis var. semperflorens, Aster yomena and C. cyanus. In conclusion, C. zawadskii var. latilobum and D. chinensis var. semperflorens were more tolerant to drought than other two species. Furthermore in vitro selection was successfully used to screen drought tolerance species of native plant species.

Effect of Medium Composition on in vitro Propagation and Plantlet Regeneration from Nodal Explants of Cassava (Manihot esculenta Crantz)

  • The Cassava (Manihot esculenta Crantz) is a perennial woody shrub cultivated mainly in the tropics for its starchy tuberous roots. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. Thus, a study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with single node after sterilization, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, single-node stem segments, approximately 1 cm in length, were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate was 55.6%, the shoot number and its length were 1.0/explant and 2.3 cm in the most favorable medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

번식방법(繁殖方法)에 따른 밭고추냉이의 성분비교(成分比較) (Comparison of Chemical Constituents of Upland Wasabia japonica Matsum Grown by Different Propagation Methods)

  • 향신료(香辛料) 및 고소득 작물로 개발가능성이 있는 고추냉이의 번식방법(繁殖方法)에 따른 성분(成分)을 비교(比較)하여 고추냉이 품질평가 및 우수 품종육성의 기초자료(基礎資料)로 제공(提供)하고자 지폐산(脂肺酸), 아미노산 및 유기산 함양(有機酸 含量) 및 조성(組成)을 조사한 결과(結果)를 (요약)要約하면 다음과 같다. 1. 총지방산 함량 및 지방산조성은 번식방법간에 차이가 없었으나 잎의 지방산함량이 다른 부위보다 높았고, 지방산조성은 부위와 관계없이 linolenic acid 함량이 가장 높았고 그 다음이 Palmitic acid, oleic acid, linoleic acid순이었으며 그 외에 lauric acid, myristic acid, palmitoleic acid, stearic acid, arachidonic acid 소량 함유되어 있었다. 2. 총아미노산 함량은 번식방법 비슷하였으나 부위별로는 잎, 근경, 줄기, 화경순으로 높았고, 아미노산조성은 번식방법에 관계없이 각 부위 모두 glutamic acid $(331{\sim}1568mg)$, asparatic acid $(346{\sim}1380mg)$, arginine $(84{\sim}724mg)$, leucine $(49{\sim}1264mg)$의 함량이 많았다. 3. 유기산 조성은 번식방법에 관계없이 비슷하였며 부위에 관계없이 acetic acid $(60.0{\sim}78.2)$가 가장 함량이 많았고 그 다음은 succinic acid $(9.9{\sim}32.0)$, malic acid $(2.9{\sim}7.9%)$로 함량이 많았다.

Computer modeling of crack propagation in concrete retaining walls: A case study

  • Concrete retaining walls are the most common types of geotechnical structures for controlling instable slopes resulting from lateral pressure. In analytical stability, calculation of the concrete retaining walls is regarded as a rigid mass when its safety is required. When cracks in these structures are created, the stability may be enforced and causes to defeat. Therefore, identification, creation and propagation of cracks are among the important steps in control of lacks and stabilization. Using the numerical methods for simulation of crack propagation in concrete retaining walls bodies are among the new aspects of geotechnical analysis. Among the considered analytical methods in geotechnical appraisal, the boundary element method (BEM) for simulation of crack propagation in concrete retaining walls is very convenient. Considered concrete retaining wall of this paper is Pars Power Plant structured in south side in Assalouyeh, SW of Iran. This wall's type is RW6 with 11 m height and 440 m length and endurance of refinery construction lateral forces. To evaluate displacement and stress distributions (${\sigma}_{1,max}/{\sigma}_{3,min}$), the surrounding, especially in tip and its opening crack BEM, is considered an appropriate method. By considering the result of this study, with accurate simulation of crack propagation, it is possible to determine the final status of progressive failure in concrete retaining walls and anticipate the suitable stabilization method.

터빈용 Cr-Mo-V강의 고온 환경변화에 따른 피로거동-고사이클 피로균열의 전파특성- (Fatigue behavior of Cr-Mo-V steel at high temperature for turbines -Propagation characteristics of high cycle fatigue crack-)

  • The rotating bending fatigue tests were performed using the specimens taken from Cr-Mo-V steel, widely sued in thermal power plant turbines, at various temperatures such as room temperature, 300 .deg. C, 425 .deg. C and 550 .deg. C. The characteristics of fatigue crack propagation were examined and analyzed by using fracture mechanics parameter. The plastic replica method was also applied in order to measure the crack length on the basis of serial observation of fatigue crack propagation behavior on the defected specimen surface. The fatigue crack propagation behavior of Cr-Mo-V steel was investigated within the frame work of elastic-plastic fracture mechanics. The propagation law of fatigue crack is obtained uniquely by using the term .sigma. $^{n}$ sub a/where .sigma. $_{a}$ is the service stress, a is the crack length and n is a constant. The values of constant n are nearly equal to 2.48, 2.60 and 8.61 at room temperature, 300 .deg. C and 425 .deg. C.

  • When piping system in a nuclear power plant is subjected to a beyond design seismic condition, it is important to accurately determine possibility of crack initiation and, if initiation occurs, its location and time. From recent experimental works on elbow pipes, it was found that the crack initiation location and crack propagation direction of the SA403 WP316 stainless steel elbow pipe were affected by the pipe thickness. In this paper, the crack initiation location and crack propagation direction for SA403 WP316 stainless steel elbow pipes with different thickness were analyzed via elastic-plastic finite element analysis. Based on FE results, the effect of the pipe thickness on different crack initiation location and crack propagation direction was analyzed using ovality, stress and strain components. It was also confirmed that the presence of internal pressure had no effect on the crack initiation location and crack propagation direction.

Practical Propagation Methods for Production of Prothalli and Sporophytes in Deparia pycnosora (Christ) M. Kato

  • Deparia pycnosora (Christ) M. Kato is a fern used as ornamental plant. In addition, it is called "Teol-go-sa-ri" in Korean name. The aim of this study was to develop a practical propagation method of D. pycnosora using tissue culture technique. Prothallus obtained from spore germination was the used as experiment materials. The prothalli (300 mg) used in all experiments were sub-cultured for 8-week intervals. The most suitable media for prothallus propagation were identified by culturing 300 mg of prothalli in $1/4{\times}$, $1/2{\times}$, $1{\times}$, $2{\times}$ MS medium and in Knop medium for 8 weeks. Also, the prothalli were cultured by chopping with a scalpel. In addition, sucrose, activated charcoal, and total nitrogen source were added in different concentrations based on the culture medium selected. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\times}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark) in in vitro. The results showed that optimum was achieved prothallus fresh weight and development in $1{\times}$ MS medium. When other components were added to the basic $1{\times}$ MS medium, prothallus propagation was maximized in $1{\times}$ MS medium supplemented with 2% sucrose, 0.2% activated charcoal, and 60 mM total nitrogen. To select a suitable soil mixture for sporophyte formation, 1.0 g of prothallus was blended with distilled water, spread on five combinations of different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and cultivated for 12 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). As a results, horticultural substrate alone, 2:1 (v:v) mixtures of horticultural substrate and perlite, and 2:1 mixtures of horticultural substrate and decomposed granite induced 208.0, 201.3 and 248.8 sporophytes per pot, respectively. Effect of plant growth regulators and antioxidants on in vitro plant regeneration and callus induction from leaf explants of purple passion fruit (Passiflora edulis Sims)

  • Purple passion fruit (Passiflora edulis Sims) is one of the introduced tropical plants, an increasing interest has arisen due to its distinctive taste and attractive flavor. It is expected that passion fruit production and planted area will increase gradually in the years ahead because of high profitability and consumer's demands of healthful ingredients. So we tried to investigate the effect of plant growth regulators and antioxidants on in vitro plant regeneration and callus induction from leaf explants of passion fruit for an establishment of optimal mass propagation system. Young leaf explants of purple passion fruit were cultured in Murashige and Skoog (MS) medium containing different growth regulators and antioxidant additives to induce the shoot organogenesis. After 8 weeks, the highest embryogenic callus formation rate was obtained in MS medium supplemented with $1mg{\cdot}L^{-1}$ 6-benzylaminopurine (BAP) and $2mg{\cdot}L^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), furthermore, the shoot development via organogenesis was also observed. Silver nitrate ($AgNO_3$), which was added into the medium to minimize the adverse effects of leached phenolics, was effective for reduction of medium browning and sudden explant death. In the medium supplemented with $1mg{\cdot}L^{-1}$ BAP and $1mg{\cdot}L^{-1}$ gibberellic acid ($GA_3$), shoots were most vigorously regenerated and elongated. Most shoots rooted successfully in half strength medium with $1mg{\cdot}L^{-1}$ indol-3 acetic acid (IAA), and more than 90% of plantlets survived after 4-month acclimatization period.

Regeneration of plants from alginate-encapsulated axenic nodal segments of Paederia foetida L. - A medicinally important and vulnerable plant species

  • Paederia foetida L. is an important medicinal plant that has been used for the treatment of various gastrointestinal related ailments by different tribal communities in India. This plant is also known for its use as a food. Due to overexploitation, P. foetida has been classified as a vulnerable plant in some states of India. The propagation of P. foetida by conventional methods is easy but very slow. Synthetic seed technology offers incredible potential for in vitro propagation of threatened and commercially valuable plants, and can also facilitate the storage and exchange of axenic plant material between laboratories. However, synthetic seed production for P. foetida has not yet been reported. Thus, to the best of our knowledge, the present study is the first attempt to produce synthetic seeds of P. foetida by calcium alginate encapsulation of in vitro regenerated axenic nodal segments. Sodium alginate (3%) and CaCl2 (100 mM) were found to be the optimal materials for the preparation of ideal synthetic seeds, both in terms of morphology and germination ability. The synthetic seeds showed the best germination (formation of both shoot as well as root; 83.3%) on ½ MS medium augmented with 0.5 mg/L indole-3-acetic acid. The plantlets obtained from these synthetic seeds could be successfully acclimatized under field conditions. We also studied the storage of these synthetic seeds at low temperature and their subsequent sprouting/germination. The seeds showed a germination rate of 63.3% even after 21 days of storage at 4 ℃; thus, they could be useful for transfer and exchange of P. foetida germplasm.
