Characterization and RT-PCR Detection of Turnip Mosaic Virus Isolated from Chinese Cabbage in Korea (배추에서 분리한 순무 모자이크 바이러스의 특성 및 역전사 중합효소 연쇄반응법(RT-PCR)을 이용한 검정)

  • Turnip mosaic virus)TuMV-Ca) was isolated from a Chinese cabbage showing severe mosaic and black necrotic spots symptoms in Korea. The virus was identified as a strain of TuMV by its host range test, particle morphology, serology, double stranded RNA analysis. For detection of the virus, reverse transcription and polymerase chain reaction(RT-PCR) was performed with a set of 18-mer TuMV-specific primers to amplify a 876 bp DNA fragment The virus was rapidly detected from total nucleic acids of virus infected tissues as well as native viral RNA of purified virion particles by RT-PCR. Detection limit of the viral RNA by RT-PCR was 10 fg.

First Report of Pasteuria nishizawae Sayre, Wergin, & Nishizawa Attcaking Heterodera glycines in Korea (국내 미기록 콩씨스트선충 기생세균, Pasteuria nishizawae Sayre, Wergin & Nishizawa의 보고)

  • Obligate bacterial parasite attacking Heterodera glycines was firstly found from Chungju soybean field in Korea. Diameters of sporangium and central body were 5.6 ${\mu}{\textrm}{m}$ and 1.9 ${\mu}{\textrm}{m}$ under light microscopy (LM), and 3.9 ${\mu}{\textrm}{m}$ and 1.8 ${\mu}{\textrm}{m}$ under transmission electron microscopy (TEM). Endospore showed cup-shaped with smooth-type exosporium without peg-like thickening in polar area under SEM and TEM. Bacteria completed its life cycle in female of soybean cyst nematode after adhering on cuticle of second-stage juvenile. From these results, the Pasteuria found from Chungju was identified with P. nishizawae.

  • The characteristic Ramularia type conidia of Mycosphaerella nawae were formed on naturally infected leaves. Artificial inoculation with the conidia induced typical symptom on leaves, which was not distinguishable from those of ascospore infection, which has been considered as a primary inoculum source in nature. Also the morphology of the conidia produced on PDA was not different from those formed on artificially inoculated leaves or on naturally infected leaves at later stage of symptom development. Accordingly, we report the role of the conidia as a secondary inoculum of the circular leaf spot pathogen of persimmon for the first time.

A Palynological Taxonomy of the Korean Verbenaceae (한국산 마편초과 식물의 화분학적 분류)

  • This research is the palynotaxonomic revision to clarify palynological descriptions and relationships of taxa in Korean Verbenaceae. To do this, morphology, size, sculpture and aperture of pollen grain were investigated with scanning electron microscope. As a result, palynological key as to 5 genera 10 species drawn up. Palynological descriptions of Callicarpa dichotoma, Callicarpa mollis, Callicarpa chejuensis, Vitex chinensis and Caryopteris incana are near completion at first. Vervena officinalis are newly described and descriptions of Callicarpa japonica, Clerodendrum trichotomum, Vitex rotundifolia and Caryopteris divaricata are corrected. Palynological data of Korean Verbenaceae is useful to intrageneric relationships. Namely, as to genus Callicarpa, C. dichotoma and C. japonica are considered to be more advanced than C. mollis and C. chejuensis. As to genus Caryopteris, C. divaricata is more advanced than C. incana.

Isolation of Symbiotic Frankia EuIK1 Strain from Root Nodule of Elaeagnus umbellata (보리수나무 뿌리혹으로부터 Frankia EuIK1 공생균주의 분리)

  • The root nodules of Elaeagnus umbellata were coralloid-shape due to repeated dichotomous branching of nodule meristem. The filamentous endophyte with vesicle cluster ranging from 30 ${\mu}{\textrm}{m}$ to 60 ${\mu}{\textrm}{m}$ in diameter was present only in the cortical cells. The isolated endophytes in vitro culture showed typical Frankia morphology, consisting of highly branched hyphae ranging from 0.8 ${\mu}{\textrm}{m}$ to 1.0 ${\mu}{\textrm}{m}$ in diameter, terminal and intrahyphal sporangia varing in shape and size up to 60 ${\mu}{\textrm}{m}$ in length and laminated vesicles. Its infectivity and effectivity were confirmed by induction of nitrogen-fixing root nodules on the inoculated seedlings of two Elaeagnus species. Consequently, the isolate was confirmed as a true symbiont of Elaeagnus umbellata root nodule and named Frankia EuIK1.

  • Floral structures of 11 korean lauraceous species belonging to six genera have been studied. Machilus is a primitive genus having relatively large bisexual flower, six tepals and staminodes in fourth whorl of stamen. The flowers of Neolitsea are highly reduced state as unisexual flower, six stamens arranged to three whorls and no staminode in the flower, so regared as more advanced group. Intrafamilial classification systems of Nakai (1939) and Pax (1891) are more reasonable but still have some discrepancies with results. The origin of tepals of this group has been a long controversial problem; whether the differentiation of tepals occur. Teratological study of Lindera erythrocarpa, inner tepals of this group must be originated from the staminode and outer tepals from leafy tepals. Although morphological differentiation does not sitll occur, inner and outer whorls of tepals are originated through different way.

  • Ceramium kindoi Yendo demonstrates a Polysiphonia type of life history without diviation in unialgal culture. However, cultures of C. paniculatum Okamura and C. aduncum Nakamura from Kangneung shows considerable phenotypic variations in laboratory. In C. paniculatum, the subulate spines disappear, the transverse cell-rows of corticating bands decrease in number, and the growing direction of corticating cells is changed during the culture. In C. aduncum, a species new to Korea, frond apices become slightly incurved in contrast to strongly rolled ones in the field, and the rows of corticating cells decrease in number and height during the culture. These results suggest that some significant taxonomic characters currently used for identification of Ceramium species are reconsidered.

Variation in Flower Color among Hybrids of Jeoktanshim Hibiscus syriacus L

  • This study was performed to provide basic information of the development and breeding of new cultivars of Hibiscus syriacus L., which have more beautiful and diverse flowers. Morphological characteristics of the flowers and leaves, and genetic variation of the flowers color of two Jeoktanshim-line cultivars, Bulsae and Pyungsung, were crossed each other. The result of the cross between Bulsae and Pyungsung are as follows: Mean flower height and width were 5.35 cm and 7.69 cm, respectively. Mean length and width of petal were 5.43 cm and 3.80 cm respectively, and mean length and width of flower pistil were 4.67 cm and 0.44 cm, respectively. The flower color of all ten individuals was the color of the Jeoktanshim-line, and Pyungsung, and all flower type were I-c type.

  • Ginseng (Panax ginseng C.A. Meyer) hairy root transformed with Agrobacterium rhizogenes (strain A4) was examined cytogenetically and histologically to assess its characteristics. The optimum growth of hairy root obtained in hormone-free MS medium (sucrose 30 g/L, pH5-6) for long period cultures. All hairy root strains (HB1, HB2, HB3) had the 2n diploid number (2n=48) of chromosomes in root tip cells. There were no alteration in chromosome structure except in one cell of HB3 strain. Results of SDS-PAGE showed a few difference in pattern and number of bands between normal and hairy root of ginseng. The root anatomy of normal root and hairy root differed each other. The hairy root had a clearly defined vascular strand, and the morphology of cortical cells were disorganised with large intercellular spaces.

Disaccordance of Genetic Relationship between Randomly Amplyfied Polymorphic DNA and their Morphological Characteristics of Korean Native Aster

  • This study were focused on the genetic relationship using RAPDs and some morphological characteristics among eleven taxa Aster collected in Korea. Twenty random primers were selected, a total 216 DNA bands were generated and 213 bands were shown polymorphism among species. The collected eleven taxa were clustered into five groups at 0.609 similarity index. The first group was A. glehni, A. ageratoides, A. maackii and A. scaber was clustered at 0.713 of genetic distance. The second group was A. tataricus and A. koraiensis and the third group was A. spathulifolius, the forth group was A. yomena, A. hayatae and A. hispidus and the fifth was A. tripolium.
