Development of a Vacuum Nozzle Seeder for Large Seeds in Cucurbitaceae

  • This study aimed to develop a seeder for sowing large seeds such as those of fruit vegetables used as rootstocks in grafting. It investigated how the nozzle diameter and the nozzle's vacuum pressure affected the seeding performance. This study found that a nozzle diameter of 1.5 mm was capable of sowing most of the seed sizes used in this study. The highest seeding rates for 'Chambak', 'Tuktozwa', and 'Hukjong' were $97.6\%$, $98.8\%$, and $97.6\%$, respectively. Lineup status after sowing was good. The working performance was 75.6 sheets/hr. An average seeding rate of 1 grain was $97.8\%$.

Characterization and Expression Profile Analysis of a New cDNA Encoding Taxadiene Synthase from Taxus media

  • A full-length cDNA encoding taxadiene synthase (designated as TmTXS), which catalyzes the first committed step in the Taxol biosynthetic pathway, was isolated from young leaves of Taxus media by rapid amplification of cDNA ends (RACE). The full-length cDNA of TmTXS had a 2586 bp open reading frame (ORF) encoding a protein of 862 amino acid residues. The deduced protein had isoelectric point (pI) of 5.32 and a calculated molecular weight of about 98 kDa, similar to previously cloned diterpene cyclases from other Taxus species such as T. brevifolia and T. chinenisis. Sequence comparison analysis showed that TmTXS had high similarity with other members of terpene synthase family of plant origin. Tissue expression pattern analysis revealed that TmTXS expressed strongly in leaves, weak in stems and no expression could be detected in fruits. This is the first report on the mRNA expression profile of genes encoding key enzymes involved in Taxol biosynthetic pathway in different tissues of Taxus plants. Phylogenetic tree analysis showed that TmTXS had closest relationship with taxadiene synthase from T. baccata followed by those from T. chinenisis and T. brevifolia. Expression profiles revealed by RT-PCR under different chemical elicitor treatments such as methyl jasmonate (MJ), silver nitrate (SN) and ammonium ceric sulphate (ACS) were also compared for the first time, and the results revealed that expression of TmTXS was all induced by the tested three treatments and the induction effect by MJ was the strongest, implying that TmTXS was high elicitor responsive.

  • VR(Virtual reality) technologies have given engineers the ability to design, test, and evaluate engineering systems in a virtual environment. The virtual plant is the highlight of the application of the VR technology to plant engineering. Plant design, maintenance, control, management, operation are integrated in the virtual plant. The VR monitoring system including the concept of the virtual plant is developed to replace a current control room that has number of gages and warning lamps in two-dimensional panels which shows the operating status of a plant. The operating status of the plant is displayed in the VR monitoring system through the realistic computer graphics. Sophisticated, realistic and prompt control becomes possible. The VR monitoring system consists of advanced visualization, walk-through simulation and navigation. In the virtual environment, a user can navigate and interact with each component of a plant. In addition, the user can access the information by just clicking interesting component. The VR monitoring system is operated with various modules, such as (1) virtual plant constructed with Graphic Management System (GMS), (2) Touch & Tell System, and (3) Equipment DB System of Part. In order to confirm the usefulness of the VR monitoring system, a pilot gas plant which is currently being used for plant operator training is taken as application. The end of the paper gives an outlook on the future work and a brief conclusion.

Preparation and characterization of SRF(Solid Refuse Fuel) using heavy oil fly ash (중유회를 활용한 고형연료 제조 및 특성)

  • In this study, the characteristics of the SRF (Solid Refuse Fuel) prepared by blending each of the additives (citrus peel, waste wood, coal) in the heavy oil fly ash, evaluating the heavy oil fly ash recyclability. Recycling SRFs were fabricated by pellet extruding method after blending the heavy oil fly ash and additives based on 30% moisture content. As a result, the formability of the SRFs was excellent under condition of blending heavy oil fly ash with coal or citrus peel and the highest calorific value was 4,274 kcal/kg at heavy oil fly ash mixed with coal. Therefore, the formability and calorific value were improved when the heavy oil fly ash was mixed with coal(20 wt%) at 30% moisture content. From these results, the applicability of SRFs with additives was confirmed by using the heavy oil fly ash from J thermal power plant.

  • Lately old LNG carriers increased and ship price is getting down. So Interest for reuse and modification of used LNG carriers is growing. Also the needs for replacement of old power plant is increasing. Additionally eco friendly fuel such as LNG become attractive. Consequently gas power plant is getting much more popular than before. So in this research planning, we consider the floating power plant by modifying LNG carriers. This plant has the various function including storage, power plant and bunkering fuction etc. Through this multifunctional plant, we are ready for the old power plant shutdown and energy crisis in the future when we can supply the urgent mobile floating power plant quickly in time.

  • Supercritical carbon dioxide power system is the next generation electricity technology expected to be highly developed. The power system can improve net efficiency, simplify cycle configuration, and downsize equipment compared to conventional steam power system. In order to dominate the new market in advance, it is required to found Front End Engineering Design (FEED) Framework of the system. Therefore, this study developed the FEED framework including design processes for the supercritical carbon dioxide power system, information elements for each process, and relationships for each element. The developed FEED framework is expected to be able to secure systematic technological capabilities by establishing a common understanding and perspective among multi-field engineers participating in the design.

Transcriptome Profiling and Characterization of Drought-Tolerant Potato Plant (Solanum tuberosum L.)

  • Potato (Solanum tuberosum L.) is the third most important food crop, and breeding drought-tolerant varieties is vital research goal. However, detailed molecular mechanisms in response to drought stress in potatoes are not well known. In this study, we developed EMS-mutagenized potatoes that showed significant tolerance to drought stress compared to the wild-type (WT) 'Desiree' cultivar. In addition, changes to transcripts as a result of drought stress in WT and drought-tolerant (DR) plants were investigated by de novo assembly using the Illumina platform. One-week-old WT and DR plants were treated with -1.8 Mpa polyethylene glycol-8000, and total RNA was prepared from plants harvested at 0, 6, 12, 24, and 48 h for subsequent RNA sequencing. In total, 61,100 transcripts and 5,118 differentially expressed genes (DEGs) displaying up- or down-regulation were identified in pairwise comparisons of WT and DR plants following drought conditions. Transcriptome profiling showed the number of DEGs with up-regulation and down-regulation at 909, 977, 1181, 1225 and 826 between WT and DR plants at 0, 6, 12, 24, and 48 h, respectively. Results of KEGG enrichment showed that the drought tolerance mechanism of the DR plant can mainly be explained by two aspects, the 'photosynthetic-antenna protein' and 'protein processing of the endoplasmic reticulum'. We also divided eight expression patterns in four pairwise comparisons of DR plants (DR0 vs DR6, DR12, DR24, DR48) under PEG treatment. Our comprehensive transcriptome data will further enhance our understanding of the mechanisms regulating drought tolerance in tetraploid potato cultivars.
