Occurrence of Tulip Fire Caused by Botrytis tulipae in Korea

  • Severe spotted lesions were observed on tulip plants grown in Asan, Yongin, and Icheon areas in Korea during a disease survey in 2000 and 2001. Diseased plants with severe symptoms were blighted and rotted at the late stage of disease development. A total of 15 isolates was obtained from the infected plant parts of tulip. All the isolates were identified as Botrytis tulipae based on their morphological and cultural characteristics. Three isolates of B. tulipae were tested for their pathogenicity to tulip by artificial inoculation. Spotted lesions similar to those observed in the fields were induced on tulip leaves. This is the first report of tulip fore disease caused by B. tulipae in Korea.

  • Plants are inherited spatial and temporal coordination systems in their growth and differentiation processes which are precisely governed by the two interlocked control systems; autogenous and environmental. Looking into the overall course of plant development from molecular to organismal level, it can be comparable to a concerto for plant hormones, environmental stimuli and plant genomic orchestra conducted by an unidentified virtuoso. Some of the recent significant attempts to puzzle out the mystery of the life processes of plant development are briefly reviewed. The revolutionary advances in understanding the mystic processes are contemporarily achieved by the application of various molecular techniques. The characterization of plant genomes is now attained through recombinant DNA approaches, and the sensitive detection of specific gene products during the plant development is perimitted by the immunochemical procedures. However, along with the recognition of underlying molecular events such as developmental changes in gene expression and hormone-receptor interrelation associated with tissue sensitivity to hormones, more emphasis should be placed upon the physiological approaches of organismal level for the understanding the correlative systems of the developmental processes of plants as intact eukaryotic organisms.

배추무사마병의 뿌리혹 형성에 미치는 묘령, 접종원 농도 및 접종방법의 영향 (Effects of Plant Age Inoculum Concentration and Inoculation Method on Root Gall Development of Clubroot Disease of Chinese Cabbage Caused by Planmodiophora brassicae)

  • Effect of inoculum concentration inoculation method and plant age on development of clubroot disease of Chinese cabbage seedling were examined in growth chambers. Root galls were developed at the concentration of 105 resting spore or above per ml of incoulum and as the inoculum concentration became higher rate of development of root galls was faster. In the plants with root gall development fresh weight of above ground parts was reduced to 30-44% of that of healthy plants but root weight increased by 4-10 times. Growth of diseased plants was greatly reduced as compared to healthy plants. Planting in the diseased soil as a inoculation method was most effective for disease development showing uniform infections but time of initial root gall development was delayed by root soaking inoculation. Some plants inoculated by soil drenching method did not develop root galls. However root gall enlargement after its initial formation did not differ greatly among inoculation methods. Nine-day-old seedlings showed poor development of root gall but 16-days-old seedlings was found to be most adequate for inoculation for gall development.

Epigenetic Regulation of Plant Reproductive Development

  • Epigenetics represents a chromatin-mediated transcriptional repression which plays a control role in both animal and plant development. A number of different mechanisms have been described to be involved in the formation of chromatin structure: especially DNA methylation, nucleosomal histone modification, DNA replication timing, and binding of chromatin remodelling proteins. Epigenetic phenomena include genomic imprinting, dosage compensation of X-chromosome linked genes, mutual allelic interactions, paramutation, transvection, silencing of invasive DNA sequences, etc. They are often unstable and inherited in a non-Mendelian way. A number of epigenetic defects has been preferentially described in floral development. Here, epigenetic phenomena in model angiosperm plants and their corresponding mechanisms are reviewed.

Soft Rot of Onion Bulbs Caused by Pseudomonas marginalis Under Low Temperature Storage

  • Soft rot occurred severely in onion bulbs stored under low temperature ($5^{\circ}C$) in storage houses at Changyoung, Kyungnam province, Korea in early 2000. Water-soaking and yellowish-brown lesions initially appeared on the outside scales of diseased onion bulbs, gradually progressing into the inside scales. Among the bacterial isolates obtained from the lesions, K-2 isolate was found to be responsible for the disease, which grew at a temperature range of from $0^{\circ}C$ to $36^{\circ}C$ with optimum temperature of $00^{\circ}$-$33^{\circ}C$. However, it showed strong pathogenicity to onion bulbs at $25^{\circ}C$ and $5^{\circ}C$ at 3 days and 2 months, respectively. The bacterium also caused soft rot on potato and showed hypersensitive reactions to tobacco and potato. The causal bacterium of onion soft rot was identified as Pseudomonas marginalis based on morphological, biochemical, and physiological characteristics including LOPAT, Soft rot in onion under low temperature storage caused by P. marginalis has not been previously reported.

한국 약식동원 식물자원의 항산화 활성 비교 (The Screening of Antioxidant Activity in some Korean Medicinal Plants)

  • This study was conducted to find the powerful free radical scavenging effect in methanol extracts from 35 kinds of Korean medicinal plants obtained from the Plant Extract Bank as natural plant products. Antioxidant activity of the plant extracts was determined by measuring the scavenging effect of 1,1-diphenyl-2-picryl hydrazyl radical (DPPH). In the screening of plant extracts tested, at 200 ppm level, almost all plantex tracts showed significantly high antionidant activity. In the reinvestigation of 15 kinds of Korean medicinal plants at 50 ppm level selected as good sources with good free radical scavenging effect at 200 ppm level, the DPPH scavenging effects were similar to those of L-ascorbic acid in whole plant extract of Fragaria yezoensis (90.4%), in the plant stems and roots extract of Gingko biloba, and the leaves extract of Rhododendron mucronulatum var. ciliatum (90%). The fruits extract of Cornus officinalis showed the strongest antioxidant activity (95%). These results suggest that the methanol extracts from Korean medicinal plants can be a potential source of antioxidants that can be used to control oxidative stress in the body, But further study is needed to measure their safety for consumption

Shoot induction and regeneration using internodal transverse thin cell layer culture in Sesamum indicum L.

  • An efficient protocol for shoot regeneration was developed for sesame (Sesamum indicum L.) internodes using the transverse thin cell layer (tTCL) culture method. The frequency of shoot regeneration and the number of adventitious buds produced from regenerated shoots depend significantly on explant age, thickness of the tTCL sections, and the phytohormones supplemented to the culture medium. A combination of 6-benzyladenine (2.0 $mg\;l^{-1}$) and a-naphthaleneacetic acid (0.5 $mg\;l^{-1}$) was found to be the best phytohormone combination for shoot bud induction, with the maximum number of shoots obtained when the tTCL sections were 0.5-1.0 mm thick and derived from 4- to 6-week-old seedlings of sesame. Well-developed shoots were rooted on MS medium without phytohormones, and 80% of the regenerated plantlets were successfully established in soil.

Changes in the Composition and Microbial Community of the Pepper Rhizosphere in Field with Bacterial Wilt Disease

  • Bacterial wilt caused by Ralstonia solanacearum is considered one of the most harmful diseases of pepper plants. Recently, research on plant disease control through the rhizosphere microbiome has been actively conducted. In this study, the relationship with disease occurrence between the neighboring plant confirmed by analyzing the physicochemical properties of the rhizosphere soil and changes in the microbial community. The results confirmed that the microbial community changes significantly depending on the organic matters, P2O5, and clay in the soil. Despite significant differences in microbial communities according to soil composition, Actinobacteriota at the phylum level was higher in healthy plant rhizosphere (mean of relative abundance, D: 8.05 ± 1.13; H: 10.06 ± 1.59). These results suggest that Actinobacteriota may be associated with bacterial wilt disease. In this study, we present basic information for constructing of healthy soil in the future by presenting the major microbial groups that can suppress bacterial wilt.

  • Potassium ($K^+$) is one of the most abundant cations in higher plant. It comprises about 10% of plant dry weight and it plays roles in numerous functions such as osmo- and turgor regulation, charge balance of plasma membrane and control of stomata and organ movement. Several potassium transporters and potassium channels regulate $K^+$ homeostasis in response to $K^+$ uptake systems. In this review, we describe the biological, biochemical and physiological characteristics of shaker like potassium channels in higher plant. Especially, we searched the rice genome databases and analysized expressed genes, genome structures and protein domain characteristics of shaker like potassium channels.

Effects of LED Light Quality of Urban Agricultural Plant Factories on the Growth of Daughter Plants of 'Seolhyang' Strawberry

  • This study was conducted to examine the influence of Light-Emitting Diode (LED) light quality in urban agricultural plant factories on the growth and development of Seolhyang strawberry daughter plants in order to improve the efficiency of daughter plant growth and urban agriculture. LED light quality by demonstrated that above-ground growth and development were greatest for daughter plant 2. Daughter plant 1 showed the next highest growth and development, followed by daughter plant 3. Among the different qualities of LED light, the stem was thickest and growth rate of leaves was highest for R + B III (LED quality: red 660 nm + blue 450 nm/photosynthetic photon flux density (PPFD): $241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R (red $660nm/115-117{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). Plant height, leaf width, petiole length, and the leaf growth rate were highest for W (white fluorescent lamp/$241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R + B I (red 660nm+blue 450nm/$80-82{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). For above-ground growth and development, as the plants surpassed the seedling age, mixed light (red + blue), rather than monochromatic light (red or blue), and higher PPFD values tended to increase development. Regarding the quality of the LED light, daughter plant 2 showed the highest chlorophyll content, followed by daughter plant 1, and daughter plant 3 showed the least chlorophyll content. When the wavelength was monochromatic, chlorophyll content increased, compared to that when PPFD values were increased. Mixed light vitality was highest in daughter plant 2, followed by 1, and 3, showed increased photosynthesis when PPFD values were high with mixed light, in contrast to the results observed for chlorophyll content.
