Hypoglycemic effect of a polyherbal aqueous extract in experimentally induced diabetic rats

  • The present study was carried out to investigate the hypoglycemic effect of a polyherbal aqueous extract (Curcuma longa Linn., Emblica officinalis Gaertn., Trigonella foenum-graecum Linn., Enicostemma littorale Blume) in alloxan-induced diabetic rats. Short term experiments showed a decrease in blood glucose levels at $2^{nd}\;hr$ of administration of the aqueous extract in alloxan-induced diabetic rats with increase in serum insulin levels. The extract did not show any effect on blood glucose or serum insulin levels in normoglycaemic rats. Treatment with the extract (1.5 g dry plant equivalent extarct/100 g body weight/day) for 20 days in diabetic rats showed a significant decrease in blood glucose and glycosylated haemoglobin levels and an increase in serum insulin levels. The aqueous extract also showed an enhanced glucose-induced insulin release at 11.1 mM glucose from isolated rat pancreatic islets. The extract did not show any toxicity at the particular dose used.

Allelopathic Effects of Chrysanthemum boreale on Seed Germination and Seedling Growth of the Selected Plants

  • To examine the allelopathic potentials of Chrysanthemum boreale, aqueous extracts and essential oil of the plant were used in these experiments. Seed germination of the receptor species was inhibited by the aqueous extracts and the inhibitory effect was increased in proportion to the concentration of extracts. In contrast, seedling elongation showed varied results. Achyranthes japonica, Bidens bipinnata, Raphanus sativus var. hortensis for. acanthiformis, Plantago asiatica, Pimpinella brachycarpa and Lactuca sativa were inhibited by increasing concentration of the aqueous extract, while Brassica campestris subsp. napus var. pekinensis and Echinochloa crus-galli were stimulated by the extract. Dry weight was also inhibited proportionally by increasing concentration of the aqueous extract, while some species were stimulated by a lower concentration of the extract. The volatile substances of C. boreale did not affect the seed germination of receptor plants, but seedling elongation and dry weight of some species were inhibited dose-dependently. Root hair development of selected plants was inhibited along with the concentration of essential oil. The above mentioned results, therefore, confirmed that the natural substances from C. boreale had allelopathic potentials to other plants.

Anti-oxidative, Acetylcholinesterase Inhibitory Activities and Acute Toxicity Study of Nepeta sibirica L.

  • Nepeta sibirica L. or Siberian catmint is a medicinal plant species used in Mongolian traditional medicine for curing human different disorders and veterinary practices. The previous study of the whole plant concentrated on the determination of its essential oil composition and reported that the major ones are sesquiterpenes, including nepetalactone. The aim of this study was to reveal a new biological activity of the above-ground parts of N. sibirica L. and compare the activity of different extracts correlating with the content of biologically active compounds and evaluate their toxicity. For this purpose, anti-oxidative and acetylcholinesterase inhibitory activities of the above-ground parts of N. sibirica L. aqueous and ethanol (EtOH) (40%, 70%) extracts were assayed spectrophotometrically. The aqueous extract showed positive anti-oxidative activity by both tested DPPH and FRAP assays with IC50 134.24 ± 1.42 mg/mL and FRAP value 1385.15 ± 8.12 µmol/L at 200 ㎍/mL, in contrast to 40% and 70% EtOH extracts. The 70% EtOH extract presented the highest acetylcholinesterase inhibitory activity (IC50 77.29 ± 0.38 mg/mL) followed by 40% EtOH extract (176.72 ± 0.35 mg/mL) and aqueous extract (275.41 ± 0.23 mg/mL). Total phenolics were found to be gallic acid equivalent, % 3.74 ± 0.05 (70% EtOH), 3.94 ± 0.04 (40% EtOH), and 3.79 ± 0.16 (aqueous), whereas the total flavonoids as a rutin equivalent, % as 2.01 ± 0.12, 1.44 ± 0.17 and 1.99 ± 0.02, each. The aqueous extract showed the best anti-oxidative and lowest activity against the acetylcholinesterase; however, the 70% EtOH extract showed the opposite effects than that of the aqueous. No mortality incidence was visible at various doses, indicating that the oral median lethal dose of aqueous and 70% EtOH extracts were considered greater than 5000 mg/kg. N. sibirica L. belongs to the non-toxic category of the OECD 423 classification.

Ficus bengalensis L.의 알레로파시 효과 (Allelopathic Effects of Extracts from Ficus Bengalensis L.)

  • Well grown trees of ficus bengalensis produce one or more potential inhibitors of seed germination and seedling growth. The aqueous extract of ficus leaf and bark enhanced the shoot length aqueous leaf extract of F. bengalensis. Bark extract of F. bengalensis inhibited the shoot length and root length of the plant at high concentration. Both the bark and leaf extract inhibited the seed germination. The postemergence and preemergence treatment of bark and leaf extract of F. bengalensis reduced the shoot biomass. The result suggest that F. bengalensis may have potential allelochemicals which may be developed as natural herbicides.

Antimicrobial activity and toxicity of Quisqualis indica

  • The plant Quisqualis indica (Compositae) has ehnopharmcological reputation of being used as a healing agent in Bangladesh. In this study, preliminary screenings were conducted to look at the antimicrobial susceptibility and cytotoxicity of the plant extract. The extractives of the plant were subjected to screening for inhibition of microbial growth by the disc diffusion method. The zones of inhibition demonstrated by the n-hexane, carbon tetrachloride, chloroform and aqueous soluble partitionates of the methanolic extract ranged from 8 - 15 mm, 8 - 18 mm, 12 - 20 mm and 10 - 16 mm, respectively at a concentration of 400 ${\mu}g$/disc. All the extractives were also subjected to brine shrimp lethality bioassay for primary cytotoxicity evaluation. Here, the carbon tetrachloride soluble materials demonstrated the highest cytotoxicity with $LC_{50}$ of 0.826 ${\mu}g$/ml, while n-hexane, chloroform and aqueous soluble partitionates of the methanolic extract revealed the $LC_{50}$ of 1.254, 3.866 and 5.366 ${\mu}g$/ml, respectively. This is the first report of the antimicrobial activity and cytotoxicity Q. indica.

약용식물 추출물의 xanthine oxidase 활성 저해 효과 (Inhibitory effect of medicinal plant extracts on xanthine oxidase activity)

  • 약용식물 67종의 생리활성 부위를 대상으로 열수 추출물이 in vitro에서 xanthine oxidase의 활성에 미치는 효과를 확인하였다. 약용식물 중에서 황금, 지각, 하수오, 갈근, 청피, 복분자, 녹차 및 지실 추출물은 xanthine oxidase의 활성을 강력하게 저해하여 항통풍 천연소재로서의 활용 가능성이 높게 나타났다. 하수오 뿌리 추출물은 열과 pH에 대한 안정성이 우수하였고, 농도에 비례하여 xanthine oxidase의 활성을 저해하였다. 하수오 추출물은 2.0 mg/0.1 mL의 농도에서 xanthine oxidase에 대해 73.8%의 저해 효과를 나타내었다.

쌀겨 추출물에 의한 잡초 발아 및 초기생장 억제 효과 (Effect of Aqueous Extracts of Rice Bran on Inhibition of Germination and Early Growth of Weeds)

  • 본 연구는 쌀겨 추출물을 이용하여 일년생 잡초 피와 한련초에 대한 발아 및 초기 생육억제 효과를 알아보기 위하여 수행하였다. 멸균 및 비멸균 쌀겨 추출물에 의한 피의 발아율은 5%(w/v) 처리에서만 무처리에 비하여 처리 후 7일째에 각각 27%와 63% 억제효과를 보였다. 그러나 이 추출물에 대한 피의 초장과 생체중은 멸균 및 비멸균 추출물의 1% 이상 처리에서 유의적인 감소를 보였으며, 5% 처리에서 피의 초장과 생체중은 멸균유무에 관계없이 각각 $66{\sim}68%$$75{\sim}78%$ 억제효과를 보였다. 또한 멸균 및 비멸균 쌀겨추출물에 의한 한련초 발아율은 처리 후 7일에 0.5% 이상 처리부터 유의적인 발아억제를 보였다. 처리 후 7일째에 멸균한 쌀겨 추출물에 대한 한련초의 발아율은 4% 처리에서 비멸균 쌀겨 추출물에서는 5% 처리에서 완전히 억제되었다. 쌀겨 추출물에 대한 한련초의 초장과 생체중에 있어서도 발아율 결과와 유사한 경향을 보였다. 쌀겨 추출물은 멸균한 추출물이 비멸균한 추출물보다 한련초의 발아 및 초기 생장억제에 더 민감한 반응을 나타내었으며 이들 추출물에 대해 피보다는 한련초가 더 민감하게 반응하였다. 따라서 쌀겨는 일부 잡초종자의 발아 및 초기 생육억제 효과에 대한 잠재성을 제시할 수 있었다.

Anti-diabetic activity of Thespesia lampas Dalz & Gibs on alloxan induced rats

  • Anti-diabetic effect was observed with Thespesia lampas Dalz & Gibs (Family: Malvaceae) when given as a root extract in normal as well as alloxan induced diabetic rats. The effects, however, were more pronounced in diabetic animals in which administration of plant extract for 15 days after alloxan induced diabetes, significantly reduced blood glucose levels. After alloxan induced diabetes it was observed that both standard drug (glibenclamide) and aqueous extract of Thespesia lampas were significantly superior to control in reducing blood sugar on long term treatment (15 days). The aqueous extract of T. lampas (300 and 600 mg/kg) reduced the blood glucose levels from $349.2{\pm}7.2$ to $120.7{\pm}4.6$ and $346.3{\pm}3.4$ to $101.8{\pm}6.3$, respectively. The data suggested that T. lampas could be of beneficial in diabetes mellitus in controlling blood sugar. The present investigation established pharmacological evidence to support the folklore claim as an anti-diabetic.

목련잎 추출액을 이용한 자성 나노입자의 합성 (Synthesis of Magnetic Nanoparticles Using Magnolia kobus Leaf Extract)

  • When iron oxide magnetic nanoparticles were synthesized by co-precipitation method using aqueous ammonia as reducing agent, the synthesized particles were aggregated and thus precipitation occurred. Using Magnolia kobus leaf extract as reducing agent, spherical nanoparticles of 50~200 nm were synthesized with low yield. By using both Magnolia kobus leaf extract and aqueous ammonia as reducing and stabilizing agents, smaller nanoparticles of 40~120 nm could be synthesized with various shapes. The synthesized magnetic nanoparticles were characterized with field emission transmission electron microscopy (FE-TEM) and scanning electron microscopy (SEM). TEM and SEM images showed that the magnetic nanoparticles are a mixture of triangles, tetragons, rods and spherical structures.

Antifilarial potential of the root extracts of Mirabilis jalapa Linn. (Nyctaginaceae) on cattle filarial parasite Setaria cervi

  • Effect of aqueous and alcoholic extracts of the roots of Mirabilis jalapa Linn. Four Oclock plant, on the spontaneous movements of both the whole worm and the nerve-muscle (n.m.) preparation of Setaria cervi and on the survival of microfilariae in vitro was studied. Alcoholic extract caused the inhibition of spontaneous movements of the whole worm and the n.m. preparation of S. cervi, whereas aqueous extract caused inhibition of spontaneous movements of the n.m. preparation. The initial stimulatory effect was not observed by aqueous and alcoholic extracts on n.m. preparation while effect of alcoholic extract on the whole worm was characterized by an increase in the amplitude of contractions followed by reversible paralysis. The concentrations required to inhibit the movements of the whole worm and n.m. preparation for alcoholic extract of root were $270\;{\mu}g/mL$ and $40\;{\mu}g/mL$, respectively whereas an aqueous extract caused inhibition of n.m. preparation at $30\;{\mu}g/mL$ suggesting a cuticular permeability barrier. Alcoholic extract of the roots of M. jalapa caused concentration related effect on the survival of microfilariae of S. cervi. The $LC_{50}$ and $LC_{90}$ for alcoholic extract as observed after 6 hrs. were found to be 10 ng/mL and 18 ng/mL., respectively.
