Phytol, SSADH Inhibitory Diterpenoid of Lactuca sativa

  • The succinic semialdehyde dehydrogenase (SSADH) inhibitory component was isolated from the EtOAc fraction of Lactuca sativa through repeated column chromatography; then, it was identified as phytol, a diterpenoid, based on the interpretation of several spectral data. Incubation of SSADH with the phytol results in a time-dependent loss of enzymatic activity, suggesting that enzyme modification is irreversible. The inactivation followed pseudo-first-order kinetics with the second-rate order constant of $6.15{\times}10^{-2}mM^{-1}min^{-1}.$ Complete protection from inactivation was afforded by the coenzyme $NAD^{+}$, whereas substrate succinic semialdehyde failed to prevent the inactivation of the enzyme; therefore, it seems likely that phytol covalently binds at or near the active site of the enzyme. It is postulated that the phytol is able to elevate the neurotransmitter GABA levels in central nervous system through its inhibitory action on one of the GABA degradative enzymes, SSADH.

Anticancer Properties of Icariside II in Human Oral Squamous Cell Carcinoma Cells

  • OSCC is currently the most common malignancy of the head and neck, affecting tens of thousands of patients per year worldwide. Natural flavonoids from plants are potential sources for novel anti-cancer drugs. Icariin is the active ingredient of flavonol glycoside, which is derived from the medical plant Herba Epimedii. A metabolite of icariin, icariside II exhibits a variety of pharmacological actions, including anti-rheumatic, anti-depressant, cardiovascular protective, and immunomodulatory functions. However, the exact mechanism causing the apoptosis-inducing effect of icariside II in OSCC is still not fully understood. In the present study, we assessed the anti-cancer effect of icariside II in OSCC cell lines by measuring its effect on cell viability, cell proliferation, and mitochondria membrane potential (MMP). Icariside II treatment of OSCC cells resulted in a dose- and time-dependent decrease in cell viability. Hoechst staining indicated apoptosis in icariside II-treated HSC cells. Icariside II inhibited cell proliferation and induced apoptosis in HSC cells, with significant increases in all present parameters in HSC-4 cells. The results clearly suggested that icariside II induced apoptosis via activation of intrinsic pathways and caspase cascades in HSC-4 cell lines. The collective findings of the study suggested that Icariside II is a potential treatment for OSCC; in addition, the data could provide a basis for the development of a novel anti-cancer strategy.

Wood Anatomy and Phylogeny of Laurus ( Lauraceae )

  • Wood and bark structure of Laurus, which is considered as an important spice resources was characterized by several transitional and advanced features. Annual rings are distinct: vessel elements are moderately short and narrow, both simple and sealariform perforation plates. alternate intervascular pitting; irregularly septate fiber are present; parenchyma strands are scanty paratracheal: rays are both homogeneous and heterogeneous with multiseriate: phloem rays are dilated. Comparisons with other genera of Lauraceae suggest that Laurus is transitional and advanced state in evolutionary trends in the family.

한국산 사초과 식물 잎의 표피형에 대하여(6) (A Study of Epidermal Patterns of the Leaf Blades on Korean Sedges, Eriophorum, Fuirena, Kobresia, Rhynchospora and Scirpus(6))

  • Author has studied and reported on taxonomy of Korean sedges, using gross morphology, anatomy and epidermal patterns of the leaf blades(1969, 1971, 1973, 1974). This paper is the 6th report of epidermal patterns of leaf blade on sedges and includes 5 genera, Eriophorum, Fuirena, Kobresia, Rhynchospora and Scirpus. The author proposed to find epidermal patterns of leaf blades as an important taxonomic characteristic of sedges classification. The result of this study, the elements of leaf epidermis, subsidal cells, silica body, cell wall of long cell, prickles, and arrangement of the elements are considered to be significant characteristics for the identification and classification of sedge.

Leaf Anatomy of Parkia clappertoniana Keay(Mimosaceae)

  • The bipinnate-compound leaf of Parkia clappertoniana has prominent, long petiole with a pulvinal base. The pulvinus has wood, periderm, lenticels and extensive cortical layer. Its vascular bundles are elongated with alternating rays. The vascular bundles of the petiole, rachis and veins are oval, with conspicuous bundle caps. Stomatal complex is predominantly paracytic with occasional occurrence of anomocytic stomata near the midrib. The oval stomata have conspicuous stomatal ledges. Trichomes are unicellular, acicular and restricted to the rachis, petiole and pinnule mid-ribs and margin. Ecological and taxonomic significance of features is discussed.

깽깽이풀의 해부학적 연구 (Anatomical Study on the "Ggaenggaengipul(Jeffersonia dubia $B_{ENTH}$)")

  • Korean folk medicine "Ggaenggaengipul" has beenused to dlear heat and treat chronic childfood mutitional impairment, diarrhea, jaundice, haemorrhoid, inflammation, anepithymia, nausea and egestion .The crude drug often used as a supstitute for the more expensive "Huang Lian " (황연) in Korea and China. With regard to the botainicla origic of " Ggaenggaenigipul" , it has never been studied pharmacognostically. To clarify botanical origin of " Ggaenggaegipul" , the morphological and anatomicla characteristics of the leaves, petiole, rhizoma, and radix of Jeffersonia dubia BENTH were studied.

The Inhibition of Oxidative Stress by Chios Gum Mastic is Associated with Autophagy

  • Chios Gum Mastic (CGM) is a natural resin extracted from the leaves of Pistacia lentiscus, a plant endemic to the Greek island of Chios. It has been used by traditional healers, and it has antibacterial, antifungal properties, and therapeutic benefits for the skin. The CGM reduces the formation of dental plaque and bacterial growth in oral saliva, and recent studies have demonstrated the role of antioxidant activity of CGM. Although CGM has been widely investigated, its protective effect against oxidative-damage to keratinocytes, as well as the relationship between CGM and autophagy, has not been investigated. The aim of this study was to assess the protective effect of CGM against $H_2O_2$-induced oxidative stress and to evaluate the autophagic features induced by CGM in human keratinocytes. The pretreatment with CGM significantly reduced apoptosis in $H_2O_2$-exposed HaCaT cells. It promoted the degradation of caspase-3, caspase-8, and caspase-9; and it induced the formation of the processed PARP. The treatment with CGM caused an increase in vesicle formation compared to control group. The level of p62 was reduced and the conversion of LC3-I to LC3-II was increased in CGM treated HaCaT cells. Also, the treatment with CGM increased cleavage of ATG5-ATG12 complex. In summary, CGM helps the cells to survive under stressful conditions by preventing apoptosis and enhancing autophagy. Besides, the present investigation provides evidence to support the antioxidant potential of CGM in vitro and opens up a new horizon for future experiments.

Fruit Wall Anatomy of Ocotea (Lauraceae)

  • 종간 유연관계 및 속의 이해를 돕기 위하여 Ocotea속의 과실 해부형태의 연구가 14종에 대하여 수행되었다. 조사된 종들간의 과실해부형태는 거의 비슷했으나 종들간 차이는 중과피내에 후벽세포의 유무에 있었고, 만일 중과피내에 후벽세포가 존재한다면 어느부위에, 또는 어떠한 형태로 존재하는가에 달려있다. 조사된 종들을 가지고 비교한 결과, Ocotea속은 적어도 5개의 group으로 나눌 수 있었다. 즉, 1) 후벽세포가 발달하지 않은 종 O. atrriensis, O. cujumari, O. helicterifolia, O. rubra and O. schomburgkiana; 2) 후벽세포가 외하피에만 발달한 종 O. aciphylla, O. javitensis, and O. sp. [Werff et al. 12676]; 3) 후벽세포가 내하피에만 발달한 종 O. tonuzii; 4) 후벽세포가 외하피와 내하피에 모두 발달한 종 O. foerens, O. quixos, and O. veraguensis; 과 5) 후벽세포가 중과피의 여기저기에 발달한 종 O. floribunda and O. nitida으로 나눌 수 있었다. 아울러, Ocotea속에 있어서 과피 해부형태의 다양성이 논의되었으며, 속에 대한 분류학적 재인식이 요구되었다. 한편, 과실의 해부형태 형질의 진화경향은 중과피내에 후벽세포가 발달하지 않은 종으로부터 후벽세포가 발달한 종으로 진화했을거스로 추정되었다.

Krigia속의 과피의 해부학적 구조와 분류학적 의미 (Fruit wall anatomy of the genus Krigia (Asteraceae, Lactuceae) and their taxonomic implications)

  • Krigia속 7종 모두와 근연속인 Nothocalais cuspidata 1종에 대해 과피 해부학적 형질을 조사하여 분류학적 유연관계를 추론하였다. 과피 해부학적 형질 중 과피의 모양, 늑의 수는 Krigia속 모든 종이 같은 형질을 공유하고 있으나, costa 의 모양, costa와 intercosta에서 중과피를 이루는 libriform 섬유세포와 fiber-sclereid 세포층의 수와 이들 세포층의 발달정도가 종간에서 차이점이 나타났다. Krigia biflora, K. cespitosa, K. occidentalis와 K. wrightii는 costa와 intercosta에서 libriform 섬유세포가 잘 발달되어있는 반면에 K. dandelion, K. montana와 K. virginica는 잘 발달되어있지 않고, 대부분 fiber-sclereid 세포로 이루어져있다. 이전 분류체계에서 Krigia 절에 속했던 K. biflora는 과피 해부학적 형질상의 차이로 볼 때 Cymbia절에 더 많은 유사성을 가지고 있다.

Inhibitory effects of some medicinal plant extracts on the tyrosinase promoter activity on B16 mouse melanoma cells

  • Melanin is specifically produced in melanocytes. The pathway for melanin biosynthesis is mainly controlled by tyrosinase. To estimate the inhibitory effect of melanin biosynthesis from 31 medicinal plants extracts, we tested the inhibitory effects of the tyrosinase promoter on B16 mouse melanoma cells. The result of this study demonstrated that Mori Radicis Cortex and Castena Fractus extracts only in tested medicinal plant extracts have high inhibitory effects on tyrosinase promoters with very low cytotoxicity on B16 mouse melanoma cells. Therefore, extracts of Mori Radicis Cortex and Castena Fractus were evaluated as very effective negative regulators of tyrosinase gene expression.
