Encephalomyocarditis virus infection in pigs associated with reproductive failure (번식장애를 수반한 돼지의 뇌심근염 바이러스 감염증)

  • Encephalomyocarditis virus infection in pigs, characterized by severe reproductive failure in sows and sudden death in less than 7 day-old-piglets, a period of several months from October to December of 1989, in Korea was occurred. The most outstanding gross lesions at necropsy were found in the heart which were consisted of pale, yellow brown necrotic foci of varying size in myocardium. In some cases, pulmonary edema and liver congestion were observed. Histopathology of the heart revealed the interstitial myocarditis, endocarditis and epicarditis with lymphocytic, plasmacytic and macrophage cell infiltration and, in some cases, calcification of the necrotic myocardial muscle fibers. In some cases of brain, the perivascular cuffing and glial nodules were observed. In the liver and the lacrimal gland varying degrees of multifocal necrosis were seen. The virus was isolated from the heart and the brain in a stillborn piglet and a mummified fetus. This outbreak represents the first case of encephalomyocarditis virus infection of pigs in Korea.

Effect of Feeding Probiotics on Performance and Intestinal Microflora of Laying Hens (생균제의 급여가 산란계의 생산성과 장내 미생물의 변화에 미치는 영향)

  • The objects of this experiment was to investigate the effect of dietary several supplemental probiotics on performance and intestinal microflora of Lohmann brown laying hens from 68 to 80 weeks. Basal diets based on corn and soybean meal contained 18.0% CP and 2,720㎉/kg ME. Bacillus subtilis, Lactobacillus salvarius isolated from piglet(LSP) were fed at the level of 0.1 and 0.2% in a one way design. There were four replicates of 40 hens each per treatment. Egg production, feed intake, feed conversion ratio(FCR), eggshell quality were measured at every four weeks and intestinal microflora were examined at the end of experiment. Egg production of bird fed 0.2% individual probiotics was significantly higher than that of control(P 0.05). Birds fed the diet containing 0.2% LSC and LSP had significantly lower FCR than other treatments(P 0.05). However, egg weight of birds fed control and 0.2% BS diet showed higher than other treatments. Feed intake of 0.2% BS and 0.1% LSP treatment was significantly higher than other treatments, but was not consistency of all treatments(P 0.05). Eggshell breaking strength and thickness of hens fed probiotics tended to increase compared to that of control, but was not significantly different. Intestinal anaerobes, Lactobacillus spp. and yeast of hens fed all tested probiotics were significantly increased compared to those of control. The number of intestinal E. coli of all probiotics treatments except 0.1% LSP tended to decrease. Intestinal Lactobacillus spp. was increased significantly by 0.1% dietary LSC, whereas intestinal yeast showed significant increase in LSP treatments(P〈0.05). The results of this experiment indicated that feeding probiotics to laying hens improved the egg production, FCR and increased beneficial microflora.

Pathological observation on the piglets experimentally infected with Aujeszky′s disease virus isolated in Korea (국내 분리 오제스키병 바이러스 접종자돈의 병리학적 관찰)

  • Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

Studies on enterotoxigenic Escherchia coli isolated from piglets with diarrhea (설사 자돈에서 분리된 병원성 대장균에 관한 연구)

  • This study was carried out to Investigate the biochemical characteristics, antibiotic susceptibility, serogroups and pili producibility test of enterotoxigenic Escherichia coli(ETEC) isolated from piglets with diarrhea in Kangwon province from March to October 1996. 1. Sixty eight E coli strains were isolated from 72 piglets with diarrhea and the biochemical and cultural reaction were compared with the classification criteria of Edwards and Ewing. 2. The serogroups of 26 isolates were classified as 08 : K87 6(8.8%), O20 : K1O1 4(5.9%), O141 : K85 4(5.9%), 09 : K103 : P987 3(4.4%), O45 : K 2(2.9%) 0139 : K82 2(2.9%), O64 : $K^{-}$2(2.9%), O149 : K91 1(1.5%), O157 : K88ac 1(1.5%) and O115 : $K^{-}$1(1.5%), respectively. 3. In antibiotic susceptibility test, the isolates showed high susceptible to Ak, Eno, Na, Gm, Am and Km, whereas resistance to Tc, Sm and Cf. 4. Sixty one strains(89.7%) of 68 I coli Isolates were resistant to one or more drugs. The isolates resistant to 2 and 3 or more drugs were 60.3% and 19.1%, respectively. Amog the 16 multiple resistant patterns, Sm Tc(11.5% ), Cf Sm Tc(11.5% ), Cf Cp Sm Su Tc(9.8% ) and Cf Cp Sm Su Tc(8.2%) patterns were frequently observed. 5. MRHA of guinea pig erythrocytes was detected in 9 out of 26 OK serotype and 9 out of 42 unidentified serotypes. MRHA titers of serotypes showed from 16 to 32 in O141 : K85 and no titers in O139 : K82. 6. By the GM1 ganglioside ELISA, $\beta$-, $\alpha$-, and $\gamma$-hemolysin producing strains was detected as 36, 6, and 5 from heat labile enterotoxin(LT) of 47 ETEC, respectively. The distribution of LT toxin from 112 isolates was showed $\beta$- hemolysin, 2 isolates $\alpha$-hemolysin and 3 isolates $\gamma$-hemol-ysin from 26 OK serotypes.

  • The etiological survey on porcine epidemic diarrhea virus(PEDV) by immunofluorescence antibody test(IFA) showed the positive rusult from the intestines of piglet died from acute diarrhea. The viral agent of PED was also isolated from intestine, which showed positive reaction by immunofluorescence test. After passage in Vero cell, the viral agent was further cloned by plaque purification and designated as KPEDV-9. The immunoblotting analysis using hyperimmune sera and porcine sera revealed the presence of several polypetide bands with molecular weight(M.W.) of 88K, 74K, 70K, 58~54, 54~46K, 44~40K and 33~32K, respectively.

Effects of loose farrowing facilities on reproductive performance in primiparous sows

  • The present study investigated the effects of loose farrowing type during gestation and after farrowing on reproductive performance and of lactating sows. A total of 22 primiparous sows (Landrace; average initial body weights [BW], 228.54 ± 12.79 kg) were allotted to one of two treatments on the basis of body weight. Sows were divided into two experimental groups, conventional farrowing crates (CON), and loosed-farrowing pens (LFP). The experiment duration was around 38 days ranging from 10 days before parturition to 28 days after parturition. Gestating sows at the age of 105 d were placed in gestational stalls (group housing). All the sows were fed a common diet according to the National Research Council requirements for lactation. Cross-fostering was performed within 1 day of parturition. From 1 day after weaning, estrus detection was performed twice-daily (0900 and 1730 h) for 10 min by boar exposure. There were significant effects of LFP housing type on the farrowing duration, and farrowing interval. At the farrowing time, none of the litter parameters including total born, stillborn, mummy, born alive piglets and total litter weight and piglet weight were affected. There were no effects of housing type on the mortality of piglets at d 1, 3, 7, 21, and 28. In conclusion, the result of this study showed that there is no performance difference between the crated or LFP sows, which indicate that the LFP housing has the potential to be used as an alternative to the crated house without any detrimental effects in reproduction performance of lactating sows.

Studies on the pathogenic Escherichia coli isolated from piglets with diarrhea in Chonnam (전남 지방의 설사 자돈에서 분리된 병원성 대장균에 관한 연구)

  • Porcine E coli infection is a disease caused by Enterotoxin produced by Enterotoxigenic Escherichia coli(ETEC). Enteric colibacillosis has become an economically important disease in pigs as a result of increasing intensification of farrowing management. The present study undertaken to obtain the antibiotic sensitivity and distribution of serogroups and pili producibility test of ETEC from E. coli isolates in Chonnam. The results obtained were as follows. 1. A total of 71 isolates identified as E, coli employing IMViC system from rectal specimens of 54 piglets with diarrhea. 2. In antibiotic sensitivity test, isolates showed high sensitivity to AN, CM, Fox, GM, but resistance to EM, NA TC. 3. The distribution of 25 Isolates of serogroups were 0141:K85(11.3%), 08:K87(8.5%), 064:K (5.6%), 0138:K8l (4.2%), 0139 :K82(2.8%), 0157:K88ac(1.4%) and 0149:K9l (1.4%). 4. MRHA of guinea pig erythrocytes was detected in 8 out of 25OK serotypes and 9 out of 46 unidentified serotypes. MRHA titers of serotypes showed from 64 to 128 in 0141: K85, 2 in 0138:K8l and no titers in 0139:K82. 5. The production of heat labile enterotoxin of ETEC was detect 39 out of 52 isolates showed $\beta$-hemolysin, 7 out of 52 isolates showed ${\gamma}$-hemolysin and 6 out of 52 isolates showed ${\gamma}$-hemolysin by $GM_1$ganglioside ELISA. The distribution of LT toxin were in 12 isolate showed $\beta$-hemolysin, 2 isolates showed ${\alpha}$-hemolysin and 3 isolates showed ${\gamma}$-hemolysin in 25 OK serotypes.

Seroprevalence of Mycoplasma hyopneumoniae infection in Korean swine herds (우리나라 양돈장에서 Mycoplasma hyopneumoniae 유병율 조사)

  • Serum samples of 1,175 pigs from 148 Korean swine farms not using Mycoplasma hyopneumoniae (M. hyo) vaccines were collected for seroepidemiological study of M. hyo infection by indirect ELISA method. Informations of each farm were provided about province where the farm was located and season when blood samples were collected. Then, the selected farms were divided into farm units which had 5 serum samples according to production stages : sow, suckling piglet (<30 days old), nursery pig (30-70 days old), and growing pig (>70 days old). Seroprevalence of M. hyo infection according to production stages, province, and season was investigated by using ELISA-positve rate of the selected samples for each study. This study showed that 85.34% (78.94-91.78%, 95% CI) of farms were positive to M. hyo infection and 34.81% (32.09-37.53%, 95% CI) among pigs were sero-positive to M. hyo infection in Korean swine farms. In the study of seroprevalence by production stage, most farms had sows and growing pigs which were sero-positive to M. hyo infection (sow: 83.05%, growing pigs: 87.72%) and most pigs seemed to be naturally infected by M. hyo at 8-10 weeks of age. Also, M. hyo infection showed seasonal pattern that most pigs were infected in late fall to early winter. However, in the study of seroprevalence by province, there was no significant correlation between province and M. hyo sero-positive rate.

Comparison of Plasma Proteome Expression between the Young and Mature Adult Pigs

  • Here, we present an approach of blood plasma proteome profiling and their comparisons between the young and the adult pigs as prerequisite for the identification of bio-markers related to the health conditions, growth performance and meat quality. To profile the proteome in porcine plasma, blood samples were collected from 19 young piglets and 20 adult male barrows and the plasma was retrieved. Then, protein profiling was initiated using one and two-dimensional electrophoresis. Proteins were spotted and then identified by MALDI-TOF-TOF and LC-MS-MS. In the results, more than thirty-six and twenty eight protein spots were selected in young piglets and adult pigs, respectively and twenty three proteins were identified. The proteome profile images were compared between those ones using Image Master Version 7.0. The image of expressed proteome showed that most of proteins from plasma of young piglet separated clearly and concentrated in 2DE display compared to ones from adult. Image analysis in detail was carried out to look for the specific proteins related to age progression. It demonstrated that the characteristics of proteome expression could be distinct to their age stages. Further investigations needed to proceed to understand the age dependent change of protein conformation and biological meaning of those differences in proteome expression between young and mature adult pigs.

Development of a Small Animal CT using a Linear Detector Array and Small-Scale Slip Rings

  • We have developed a small bore x-ray CT for small animal imaging with a linear x-ray detector array and small-scale slip rings. The linear x-ray detector array consists of 1024 elements of 400□m×400□m with a gadolinium oxysulfide (GOS) scintillator on top of them. To avoid use of expensive large diameter slip rings for projection data transmission from the X­ray detector to the image reconstruction system, we used the wireless LAN technology. The projection data are temporally stored in the data acquisition system residing on the rotating gantry during the scan and they are transmitted to the image reconstruction system after the scan. With the wireless LAN technology, we only needed to use small-scale slip rings to deliver the AC electric power to the X-ray generator and the power supply on the rotating gantry. The performances of the small animal CT system, such as SNR, contrast, and spatial resolution, have been evaluated through experiments using various phantoms. It has been experimentally found that the SNR is almost linearly proportional to the tube current and tube voltage, and the minimum resolvable contrast is less than 30 CT numbers at 40kVp/3.0㎃. The spatial resolution of the small animal CT system has been found to be about 0.9Ip/㎜. Postmortem images of a piglet is also presented.
