D99 Type I Signal Peptidase Implicated Stabilizing the Protein Structure (Type I 신호펩디드 가수분해효소에 존재하는 D99 아미노산 잔기의 구조적 역할 가능성)

  • Type Ⅰ signal peptidase is an integral membrane protein that functions to cleave signal peptides from secreted and membrane proteins. The enzyme serves as a potential target for the development of novel antibacterial agents due to its unique physiological properties. Despite being one of the best characterized enzymes, the catalysis of Type Ⅰ signal peptidase still remains controversy over the catalytic serine/lysine dyad mechanism. It appears that the dyad proteases are generally less efficient than the prototypical serine/histidine/aspartic acid triad found in most enzymes, although Type Ⅰ signal peptidase is an exception to this rule. In this paper, we have proposed that Type Ⅰ signal peptidase may act as the serine/lysine/aspartic acid triad cataltytic mechanism. Therefore, the aspartic acid 99 residue in the E. coli signal peptidase was chosen and mutated to an alanine to see if there is any possible role of the aspartic acid in the catalytic function. Type Ⅰ signal peptidase D99A protein was inactive in vitro assay using the procoat synthesized by in vitro transcription translation. However, the mutant was active using a highly sensitive in vivo assay. Pulse-chase experiments show that the replacement of aspartic acid 99 with alanine results in a very unstable signal peptidase molecule. Therefore, we conclude that it is unlikely that the residue is directly involved in catalysis, but rather plays an important role in stabilizing the protein structure.

  • Electrocardiogram (ECG) and blood pressure (BP) are main vital signs which are the standards in most medical settings in assessing the most basic body functions. Multi parameters are desired in providing more information for health professionals in order to detect or monitor medical problems of patients more precisely. This study urges us to develop a robust wireless healthcare monitoring system which has multiple physiological signs measurements on real time that applicable to various environments which integrates wireless sensor network technology and code division multiple access (CDMA) network with extended feature of locally standalone diagnosis algorithms that implemented in tell phone. ECG signal and BP parameter of the patients are routinely be monitored, processed and analyzed in details at cell phone locally to produce useful medical information to ease patients for tracking and future reference purposes. Any suspected or unknown patterns of signals will be immediately forwarded to hospital server using cell phone for doctors' evaluation. This feature enables the patients always recognize the importance of self-health checking so that the preventive actions can be taken earlier through this analytic information provided by this monitoring system because "Prevention is better than Cure".

Comparative Studies on The Relationship between Presence and Physiological Arousal in Virtual Reality and Desktop Based Driving Simulator (VR 및 Desktop 기반 운전 시뮬레이터 사이의 실재감과 생리적 각성 반응에 관한 비교연구)

  • 가상현실의 자연스러운 응용분야는 작업 훈련(Task Training) 분야이다. 이러한 작업 훈련 분야에서 가장 중요한 요소 중 하나는 각성 수준(Arousal Level)으로, 적절한 각성 수준은 과제 훈련의 효과를 결정짓는다. 본 연구는 VR 환경과 Desktop 환경의 자동차 운전 시뮬레이터에서 피험자가 실험을 위하여 설계된 2 개의 시나리오를 수행하는 동안 피험자의 생리 신호를 측정하고, 실험 후 각 환경에 대한 주관적인 실재감을 측정하여, 두 실험 환경 사이에 피험자의 주관적인 실재감이 유의한 차이가 있는지 알아보고, 피험자가 수행한 시나리오와 시나리오에 종속된 실험 태스크에 대하여, 각성 수준을 평가할 때 유용한 지표로 활용되는 피부전도도(GSR) 반응이 피험자의 주관적인 실재감의 결과와 어떠한 관계를 보이는지 살펴 보았다. 실험 결과 두 실험 환경 사이의 피험자의 주관적인 실재감은 유의한 차이가 있었고, VR 환경에서 더 높게 나왔다. 그러나 두 실험 환경 사이의 피험자의 생리적 각성 반응은 실재감의 결과처럼 유의한 차이를 보여주지 못했으며, 오히려 시나리오에 종속된 실험 태스크에 따라서 유의한 차이를 보여주었다. 따라서 실재감과 피부전도도 (GSR)측정에 의한 각성도 는 직접적인 관계가 적은 것으로 생각되며, 이는 피부전도도(GSR)를 실재감의 한 척도로 쓰기에는 적합하지 않을 수 있다는 것을 의미한다.

Olfactory receptors in non-chemosensory tissues

  • Olfactory receptors (ORs) detect volatile chemicals that lead to the initial perception of smell in the brain. The olfactory receptor (OR) is the first protein that recognizes odorants in the olfactory signal pathway and it is present in over 1,000 genes in mice. It is also the largest member of the G protein-coupled receptors (GPCRs). Most ORs are extensively expressed in the nasal olfactory epithelium where they perform the appropriate physiological functions that fit their location. However, recent whole-genome sequencing shows that ORs have been found outside of the olfactory system, suggesting that ORs may play an important role in the ectopic expression of non-chemosensory tissues. The ectopic expressions of ORs and their physiological functions have attracted more attention recently since MOR23 and testicular hOR17-4 have been found to be involved in skeletal muscle development, regeneration, and human sperm chemotaxis, respectively. When identifying additional expression profiles and functions of ORs in non-olfactory tissues, there are limitations posed by the small number of antibodies available for similar OR genes. This review presents the results of a research series that identifies ectopic expressions and functions of ORs in non-chemosensory tissues to provide insight into future research directions.

Molecular Cloning and Sequencing of the Bacillus subtilis cdd Gene Encoding Dooxycytindine-Cytidine Deaminase

  • The cdd gene of Bacillus subtilis, encoding the deoxycytidinecytidine deaminase of pyrimidine nucleotide biosynthesis has been cloned into the EcoRl site of pBR322. The recombinant plasmid, pSol, promoted the synthesis of 100-140 fold elevated levels of the enzyme. A comparison of the polypeptides encoded by cdd complementing and non-complementing plasmids in the mini cell showed the gene product to have a molecular mass of approximately 14 kDa. The nucleotide sequence of the gene and 460 base pairs upstream from the coding region was determined. An open-reading frame, encoding a protein with a calculated molecular mass of 14337 Da, was deduced to be the coding region for cdd. However, the enzyme has an apparent molecular mass of 54 kDa as determined by gel filteration, whereas sucrose density gradient centrifugation shows 58 kDa. It means that the enzyme could be forming a tetramer in a physiological state. About 28 amino acids of the N-tetramer in a physiological state. About 28 amino acids of the N-terminal presumably form a signal for membrane translocation and six cystein residues are contained in the structure. S1 nuclease mapping indicated that transcription of cdd is initiated 17 base pairs upstream from the translational start. The structural characterization of the odd gene was performed.

Effects of Intrarenal Arterial Infusion of Pro-Atrial Natriuretic Peptides on Renal Function in Unanesthetized Rabbits (가토 신장기능에 미치는 Pro-Atrial Natriuretic Peptide의 영향)

  • It is well known that the atrial natriuretic peptide (ANP) has a prepro-hormone of 151 amino-acids which loses their hydrophobic signal peptide to form 126 amino acid prohormone. The whole prohormone is released and then cleaved by proteases into more than one circulating forms. Recently, Winters et al. (1988a, b) reported that high concentrations of N-terminal fragments of prepro-ANP $(26{\sim}55),\;(56{\sim}92)\;and\;(104{\sim}123)$ were detected in human plasma. However, their physiological roles have not been established. The present study was conducted to determine whether the N-terminal fragments of pro-ANP have any effect on the renal function and to compare the effect with those of G-terminal fragments of pro-ANP The results indicate that intrarenal arterial infusions of prepro-ANP $(26{\sim}41),\;(26{\sim}55),\;(56{\sim}92)\;and\;(104{\sim}123)$ induced no significant changes in renal function. Whereas ${\alpha}-human$ ANP $(prepro-ANP,\;124{\sim}151)$ and pro-ANP caused a significant increase in urine volume, renal plasma flow, glomerular filtration rate, urinary excretions of sodium, chloride and potassium, and fractional excretion of sodium. These results suggest that the N-terminal fragments of pro-ANP are ineffective, while the C-terminal fragments retain the natriuretic and diuretic activities.

Genetical and Physiological Mechanisms of Adult Diapause in Insects

  • Adult diapause in insects is characterized by suppression of reproductive development. It is induced by environmental cues such as photoperiod, temperature, food availability, and other conditions Diapause-inducing environment is recognized and analyzed by the brain of the insects. The interpreted information is conveyed via endocrine system to target tissues such as ovaries, fat body, and other tissues. From this signal hierarchy of a brain-endocrine-target tissue axis, several factors are involved to express a diapause trait in a quantitative mode, even though the insects show a binomial phenotye between being in diapause or not. Recent works estimated that the number of the factors is relatively small by a series of crossing trials between high and low diapause lines. Heritability of the diapause is quite high (ca. 70%) in some species. Epistasis, sex-linkage, pleiotropism, and other nongenetic components also affect diapause inheritance. Most physiological studies have been focused on control mechanisms of the juvenile hormone (JH) synthesis in corpora allata (CA) because JH level in hemolymph of teneral adults is critical to decide a later developmental mode. Allatostatin, an antagonizer of JH synthesis, has been believed to be a potent brain message to CA for adult diapause induction.

The Effect of Cold Air Stimulation on Electroencephalogram and Electrocardiogram during the Driver's Drowsiness (운전자 졸음시 냉풍 자극이 뇌파 및 심전도 반응에 미치는 영향)

  • The purpose of this study was to analyze physiological changes via a cold air reaction experiment to generate basic data that are useful for the development of an automobile active air conditioning system to prevent drowsiness. The $CO_2$ concentration causing drowsiness in vehicle operation was kept below a certain level. Air was blown to the driver's face by using an indoor air cooling apparatus. Sleepiness and the arousal state of the driver in cold wind were measured by physiological signals. It was evident in the EEG that alpha waves decreased and beta waves increased, caused by cold air stimulation. The ${\alpha}/{\beta}$ ratio was reduced by about 52.9% and an alert state confirmed. In the electrocardiogram analysis, the efficiency of cold air stimulation was confirmed by the mean heart rate interval change. The R-R interval had a delay time of about one minute compared to the EEG response. The findings confirmed an arousal effect from sleepiness due to cold air stimulation.

Ginseng; Recent Advances and Trends (인삼연구의 최근 발전과 경향)

  • Ginseng, the root of Panax ginseng C.A. Meyer, is well-known oriental herbal medicine. The number of paper reporting the effects on its physiological, pharmacological, and behavioral effects has been increased every year, since ginsenosides isolated from ginseng are known to be biologically active components. This brief review summarizes some of new findings from recently published papers on ginsenosides or ginseng saponins. Therefore, this paper includes the various effects of ginsenosides on neuronal cell growths, on behavior of experimental animals, on enzyme activities, on the release and uptake of neurotransmitters, on neuronal cell excitability, on the motility of intestine, on antitumor activity, on cardiovascular system and metabolism. In spite of various effects of ginsenosides on various cells or organs, it is still to date impossible for one to clearly explain the exact mechanism on the action of ginsenosides. However, in this article I will discuss several papers providing possible explanations on the physiological and pharmacological actions including signal transudation pathway of ginsenosides. The elucidation of the exact mechanism of ginsenosides on cellular or molecular level will not only give us a chance to explain why people have used ginseng as an elixir of life for several thousands of year but also give us a crucial chance to apply ginseng to modern medicine.

A Study on Visual and Auditory Emotion under Color and Music Stimuli (색과 음악 자극에 의한 시청각 감성지표에 관한 연구)

  • The purpose of this study is to estimate human emotion quantitatively under colors and music stimuli and to examine the for correlation between color and music sensibility. Physiological signals(electroencephalogram, electrocardiogram, Galvanic skin conductivity and respiration rate) were measured to compare color with music sensibilities. The personality of the subject were investigated using factor analysis and semantic differential method of 20 items(7 interval scaled). The results showed that red, yellow and violets color provoked active and exciting senses mainly as dance, rock and blues music. While blue, cyan and pink colors ware involved in tranquil and resting emotions deeply as classic and ballade music.
