Hypoxia suffocates histone demethylases to change gene expression: a metabolic control of histone methylation

  • Hypoxia affects various physiological and pathophyological processes. Hypoxia changes the expression of hypoxia-responsive genes through two main pathways. First, hypoxia activates transcription factors (TF) such as Hypoxia-inducible Factor (HIF). Second, hypoxia decreases the activity of Jumonji C domain-containing histone demethylases (JMJDs) that require $O_2$ and ${\alpha}$-Ketoglutarate (${\alpha}$-KG) as substrates. The JMJDs affect gene expression through their regulation of active or repressive histone methylations. Profiling of H3K4me3, H3K9me3, and H3K27me3 under both normoxia and hypoxia identified 75 TFs whose binding motifs were significantly enriched in the methylated regions of the genes. TFs showing similar binding strengths to their target genes might be under the 'metabolic control' which changes histone methylation and gene expression by instant changing catalytic activities of resident histone demethylases.

  • The purpose of this article was to contribute to the knowledge of physiological and pathological changes of aging skeletal muscles, and of therapic method. By aging there were changes of distribution of muscle fibers, the loss of muscle mass, the loss of the number of muscle fibers, the loss of glycolysis capacity, the decrease of the oxidative enzymes and muscle vascularization in the skeletal muscles. And as a pathological change, the exhaustive maximal exercise increased oxidative stress that led to oxidative damage which were shown to be implicated in promoting aging. The property of intensity and duration exercise is important not only in keeping human health and physical fitness from oxidative stress, but also for the maintenance of well-being and quality of life.

Changes in ROS-Scavenging Enzyme Activity in Rice (Oryza sativa L.) Exposed to High Salinity

  • We studied changes in the biochemical and physiological status and ROS-scavenging enzyme (superoxide dismutase, catalase and peroxidase) activity in leaves and roots of rice (Oryza sativa L.) plants exposed to high salinity. Under salt stress, the reduction in RWC (relative water content) in leaves was relatively severe in comparison with that of roots. The proline content was also significantly higher in leaves of rice plants following salt treatment. The activities of CAT and POX in roots increased with increasing NaCl concentration, but the activity of SOD decreased. These results suggest that the increase of endogenous proline is closely associated with the increase of CAT and POX activities, which may play important roles in salt tolerance. Therefore, we conclude that the alleviation of oxidative damage and increased resistance to salinity may result from the presence of efficient antioxidative systems.

틸라피아의 해수수치에 관한 생리학적 연구 III. 호르몬과 혈청성분간의 상관관계 (Physiological Studies on Adaptation of Tilapia(Oreochromis niloticus) in the Various Salinities III. Correlations between Serum Hormones and Components Levels)

  • This study was taken to examine correlationships between ehdocrine changes such as estradiol-17$\beta$, progesterone, T4 and T3 and serum components concentrations of female Oreochromis niloticus living in 0$\textperthousand$, 10$\textperthousand$, 20$\textperthousand$, and 30$\textperthousand$ salt concentrations, respectively. The results obtained in these experiments were summarized as follows. Correlation coefficients of serum albumin and thyroxine were +0.907 and +0.611 in 10$\textperthousand$ and 20$\textperthousand$, respectively. In 30$\textperthousand$ salinity, serum BUN and other 3 kinds of hormones showed all negative correlation coefficients. Correlation coefficients of serum estradiol-17$\beta$ with calcium and cholesterol in 20$\beta$ were +0.624, +0.733, respectively. Correlation coefficient between serum triglycerides and thyroxine in 30$\beta$ was +0.989.

  • 근원성유로 부터 근수축조절단백질들을 추출정제하고 저장중의 변화를 연구하여 다음과 같은 결과를 얻었다. 1. ${\alpha}-actinin$은 그 분자형(分子形)이나 생물활성(生物活性)에 아무런 변화도 일으키지 않았다. 2. 근육저장중에 근원섬유의 troponin-troponin complex의 함량은 감소되고 있으며 troponin-tropomyosin complex의 troponin함유비(含有比)는 낮아지고 있다.

Changes in Steroid Hormones Levels of Olive Flounder, Paralichthys olivaceus Exposed to Phenanthrene

  • Phenanthrene, one of Polycyclic aromatic hydrocarbons with three aromatic rings, is a ubiquitous contaminant in the environment. Phenanthrene has been identified in ambient air, drinking water and sediment. We examined the effect of phenanthrene on steroid hormones level of olive flounder, Paralichthys olivaceus. Plasma testosterone level was increased significantly in fish exposed to phenanthrene($\geq1.0 \mu M$) at 4th week. However, there was no significant changes of estradiol-17$\beta$ concentration in fish exposed to phenanthrene. The physiological variation in phenanthrene exposed fish was a dramatic increase in plasma cortisol level. It is concluded that chronic exposure of phenanthrene can induce increase of plasma testosterone levels and elevate the plasma cortisol level in flounder, Paralichthys olivaceus.

  • The cells that make up the body continuously undergo cell division to maintain life, but stress and carcinogens, and physiological stimuli suppress immunological functions in the body and change cells to cancerous cells. The only way to overcome cancer is by treatment through early detection and proper diagnosis. However, because early stage of cancer shows subjective symptoms that are similar to those of other common illnesses, even doctors have difficulty separating cancer from common illnesses in its early stage. However, the Ki(bioenergy) present in our body recognizes and changes to the most minute changes in the body. This method of detecting the change in bioenergy to discover a disease in the early stage to raise treatment success is called the OBET: O-Ring, Bioenergy Test. This method, which is based on the theories of oriental medicine, focuses on tumors, especially malignant tumors, and has shown value in diagnosis and treatment, as witnessed in the clinical applications. The diagnosis method and cases of OBET: O-Ring, Bioenergy Test are presented.

유지식품의 조리 중 기능성분의 변화 (Changes of Functional Components Present in Lipid Foods during Cooking)

  • Lipid-based functional components present in foods undergo chemical changes during cooking. Useful n-3 and n-6 fatty acids, phytosterols, tocopherols, and carotenoids are degraded by thermal cooking such as frying, resulting in loss of their physiological functions. However, conjugated linoleic acid and diacylglycerols can be formed during heating, which would be beneficial to the health. Degree of degradation and formation of the functional components depends on the cooking method, cooking temperature and time, lipid matrix containing the components, and the presence of other materials. Although it is clear that the content of each functional component varies during long-heating in a model system consisting of small numbers of components, the real foods cooked in a small scale for a limited cooking time do not show highly significant differences in the functional components contents from raw food materials.

Changes in Skin Temperature and Physiological Reactions in Murrah Buffalo During Solar Exposure in Summer

  • Six adult female Murrah buffaloes of about 12 years were exposed to solar radiation during summer when minimum and maximum ambient temperatures were 27.1 and $44.1^{\circ}C$, respectively. The skin surface temperature at forehead, middle pinna, neck, rump, foreleg, hind legs were recorded using non-contact temperature measuring instrument and respiration rate and rectal temperature were measured throughout the 24 hours starting from 6:30 AM. The diurnal fluctuations and temperature gradients have been reported for buffaloes. During summer when ambient temperature and solar radiation was maximum, adult buffaloes were not able to maintain their thermal balance even after increasing the pulmonary frequency 5 - 6 times. The changes in skin temperature at various sites indicate that the temperature of skin surface not only varies in relation to exposure but also due to water diffusion and evaporation.

Vasoactive Intestinal Peptide(VIP)의 백서신장기능(白鼠腎臟機能)에 미치는 영향(影響) (Effect of Vasoactive Intestinal Peptide on Renal Function in Rats)

  • Vasoactive intestinal peptide (VIP) found in duodenal mucosa originally has been suggested as a neurotransmitter. Its localization, however, now known, is not limited to the gastrointestinal tract, but scattered at many different kinds of tissues, smooth muscles, endocrine gland and exocrine gland as well as central and peripheral neural tissues. To investigate the effect of VIP on renal function, an experiment has been done in anesthetized male rats. The results obtained were: 1) Urinary output and creatinine clearance decreased significantly during the period of infusion of VIP, 2.0ug/rat/7minutes. 2) Urinary excretion of sodium, potassium and chloride decreased but without significance by infusion of VIP. 3) Blood pressure, systolic and diastolic, decreased by VIP administered intravenously in the period of infusion. 4) Changes of urinary output, sodium and chloride excretion was correlated with changes of creatinine clearance. The above data suggest that VIP administered intravenously can suppress the renal hemodynamics indirectly, and also decrease electrolyte excretion through its renal hemodynamic change.
