A Genome-Specific PCR Primer Design Program for Open Reading Frames

  • Proper PCR primer design determines the success or failure of Polymerase Chain Reaction (PCR) reactions. In this project, we develop GENE-PRIMER, a genomes specific PCR primer design program that is amenable to a genome-wide scale. To achieve this, we incorporated various parameters with biological significance into our program, namely, primer length, melting temperature of primers Tm, guanine/cytosine (GC) content of primer, homopolymeric runs in primer and self-hybridization tendency of primer. In addition, BLAST algorithm is utilized for the purpose of primer specificity check. In summary, selected primers adhered to both physico-chemical criteria and also display specificity to intended binding site in the genome.

낙동강 상·중·하 수역에서의 질화세균군의 변화 (Changes of Nitrifying Bacteria in the Different Zone (Upper·Mid·Lower Part) of the Nak-Dong River)

  • Nitrifying bacteria were detected by fluorescent in situ hybridization (FISH) method at 6 sampling sites with different eutrophication degree in the Nak-Dong River and their tributaries. And conventional physico-chemical parameters including $NH_4-N$, $NO_3-N$, and TN were determined concurrently. In rainy period (July), there was no noticeable difference between the number of ammonia/nitrite-oxidizing bacteria detected at each site except Sang-Ju and the ratio of nitrifying bacteria to total counts stained by DAPI varied in 6~33%. By contrast, in the dry period (October), both of bacterial population was increased differently and the ratio of nitrifying bacteria to total counts ranged more widely from 6% in heavily polluted water zone, Hwa-Won to 60% in upper tributary with high agricultural land use. Byung-Sung-Chun. In January, the numbers of ammonia-oxidizing bacteria was reduced up to one tenth, while those of nitrite-oxidizing bacteria was apparently increased maybe due to high DO and low DOC.

  • The seeds of Abrus precatorius L. (Family- Fabaceae) constitute the drugs Abrus, Gunja, or Ratti in commerce. In the Indian System of Medicine, the seeds are used for sciatica, paralysis, headache, dysentery, diarrhoea, leprosy, ulcer, nervous disorders, alopecia, as well as anti-inflammatory, antidiabetic, antibacterial, antitumor, sexual stimulant and abortifacient. Seeds are poisonous and therefore are used after mitigation. The protein abrin is responsible for the highly toxic properties of seeds. Quantitative HPTLC analysis of the methanolic extract of seeds determined the presence of 0.4018% gallic acid and 0.4009% glycyrrhizin. The present study was undertaken to develop an HPTLC method, as well as ascertain the physico-chemical, morphological and histological parameters to establish the authenticity of A. precatorius seeds.

  • Physico-chemical characterization of human renal dipeptidase was carried out. It was a glycoprotein with a subunit MW of approximately 47,700 dalton. The pH optimum was at 8 and its stable conformation was retained between pH 5 and 12. The kinetic parameters determined with imipenem, a noval ${\beta}-lactam$ antibiotic, were Vmax, $5.21\;\mu{mol/min/mg}$; km, 4.35 mM ; and Ki with cilastatin, $0.25\;\mu{M}$ Cilastatin demonstrated reversible competitive inhibition.

몇가지 천연 산화방지물의 첨가가 가열 팜유에 미치는 영향 (Influence of some Natural Antioxidants Effect on Thermal Oxidation in Palm Oil)

  • Effectiveness of some natural antioxidants were investigated by measuring the physico-chemical charcteristics and fatty acid composition during thermal oxidation in palm oil. Tocopherol showed most enhanced thermal oxidation stabilities compared to the other natural antioxidants. AR spice was no good AOM stability and changes of acid value but the other parameters were obtained desirable results. The changes of linoleic aicd content was slightly decreased during thermal oxidation. Addition of rosemary and glycyrriza extract increased the stabilities of oil less than tocopherol and AR spice. Order of antioxdative effects was tocopherol, AR spice and others. There was no significiant difference in stability of rosemary and glycyrriza extract.

  • Nowadays the hydrophilic polymeric networks (polymer hydrogels) due to the complex of benefit physico-chemical properties attract a wide attention of specialists working in various fields of science, medicine and technology. The special attention of chemists is aimed on so-called stimuli-sensitive or intelligent hydrogels, which can undergo a volume phase transition in response to change in environmental parameters such as temperature, pH, solvent composition, etc [1]. Scientific group of Kazak State National University, Department of Macromolecular Chemistry works in this field [2-5]. Here we report about our achievements on pH-sensitive hydrogens.

Effect of Goat Grazing on Surface Water Quality of Alpine Grassland

  • The objective of this study was to determine the effect of goat grazing on the surface water quality of the alpine grasslands. Seven sites were selected across the goat farm for water sample collection and analysis. Samples were analyzed for BOD (Biological oxygen demand), total nitrogen, total phosphorous, electrical conductivity and water turbidity. All the above-mentioned parameters remained below the standard limit of Korean government at the end site. Puddles showed higher values, but below standard, as stagnant water has lower physico-chemical properties as of flowing water. The present study clearly showed that goat grazing doesn't affect water quality in grasslands if grazing is according to carrying capacity of grassland and fertilizer application is judicious.

The Investigation of Treatment of Pistachio Processing Industrial Wastewater by Electrochemical Methods in Terms of Chemical Oxygen Demand and Total Phenol Removal

  • This work aims to investigate the efficiency of electrocoagulation (EC) of pistachio processing industrial wastewater (PPIW) using the continuous EC process. The tubular reactor made of stainless steel with an internal diameter of 60 mm was used as a cathode electrode. The effect of some parameters was examined on the removal of chemical oxygen demand (COD) and total phenols (TP) removal efficiency. The influences of the initial pH of wastewater (from 4 to 8), flow rate (from 25 to 125 mL/min), current density (from 7 to 21 mA/cm2), and supporting electrolyte type (NaCl, NaNO3, and Na2SO4), supporting electrolyte concentration (from 10 to 100 mg/L NaCl) on removal efficiency were investigated to determine the best experimental conditions. The examination of the physico-chemical parameters during the EC treatment showed that the best removal efficiency was obtained under conditions where the flow rate was 25 mL/min (20 min reaction time), the pH value was 5.2, and the current density was 21 mA/cm2 has set. Under these experimental conditions, COD and TP removal efficiency were found to be 75% and 97%, respectively, while energy consumption was 18.5 kW h/m3. The study results show that the EC can be applied to PPIW pre-treatment.

Physico-chemical Properties of Giant Embryo Brown Rice (Keunnunbyeo)

  • Brown rice with a giant embryo (GE) was observed on the quality parameters of the enlargement of embryo, nutritional components, and physical properties, in comparison to normal embryo brown rice (NE). Also, the effects of germination on the quality parameters were examined. GE embryo was approximately 2.68 times larger than of NE rice. Total free sugars were significantly higher in GE rice (71.96 vs. 41.17 mg/100 g), and germinated rice increased in fructose, but decreased in sucrose and maltose. No significant difference in mineral contents was found in GE and NE rice and their germinated rice, whereas a significant increment was observed on reducing sugars and gamma-amino butyric acid (GABA) contents in GE rice. The lower water absorption index (WAI) of GE rice resulted in relatively lower pasting viscosity, whereas the increased WSI in germinated rice might be attributable to the significant increment of soluble components in GE rice.

소양호 수중 생태계에서의 세균 생체물질량의 분포 (Distribution of bacterial biomass in the water column of Soyang lake)

  • 소양호에서 측정된 세균 체적, 세균 생물량 및 세균 생산량 등의 미생물학적인 요인의 변화에 미치는 물리화학적 환경요인의 영향을 통계학적 방법으로 분석을 하였다. 상관관계 분석과 중회귀 분석 결과 수온은 대부분의 미생물학적 요인에 폭넓은 영향을 미침을 알 수 있었다. 총 세균수, 세균 체적, 세균 생물 량 및 saprophyte 수외 변화는 엽록소 a와 pheophytin a의 존재와 높은 상관관계를, $^{3}H$-thymidine incorporation rate에 의해 측정된 세균 생산량은 seston의 농도에 큰 영향을 받는 것으로 분석되었다. 소양호 수중생태계에서의 세균 체적 및 세균 생물량의 미생물학적 요인은 미생물 군집에게 탄소와 에너지원으로 작용하는 유기물질의 제공원인 식물성 플랑크톤의 분포와 seston의 농도에 의해 조절되고 있음을 시사하여 준다.
