Quality Characteristics of Chicken Breast Surimi as Affected by Water Washing Time and pH Adjustment

  • This study was carried out to compare the quality properties of chicken breast surimi manufactured by four different procedures/methods. Surimi was made from chicken breast by washing two (T1) or four times (T2) with water as well as by pH adjustments at 3.0 (T3) or 11.0 (T4). The contents of moisture and crude fat were significantly higher in the surimi manufactured from pH-adjusted material than after washing. Again, collagen and yield were significantly higher in chicken breast surimi manufactured from washed than pH-adjusted samples, whereas crude protein was higher in the pH-adjusted than washed surimi samples. There was no significant difference in myofibrillar protein content among the surimi manufactured after different washing times and differences following pH adjustments were found. T4 showed highest myofibrillar protein content rating among the surimi samples. All physical characteristics were higher in pH-adjusted chicken breast surimi than in T1 and T2 washed surimi samples. The pH-adjusted surimi had higher hardness, gumminess and chewiness than washed surimi samples (p<0.05). The chicken breast surimi made by pH adjustments had higher lightness (L*) than when made by washing times, whereas pH 3.0-adjusted surimi samples had lower whiteness (W) then the other surimi samples. Myoglobin content was significantly higher in the surimi manufactured from pH-adjusted chicken breast samples.

  • 새 집으로 이사를 가거나 집, 사무실 등의 벽지나 바닥재를 바꿨을 때, 인테리어 공사를 한 뒤에 나타날 수 있는 코를 찌르는 매캐한 냄새와 눈이 따가워지는 등의 현상이 새집증후군으로 새집을 장만한 기쁨을 누리는 것도 잠시 뿐이다. 새 건축물이나 새 가구에 사용되는 건축자재, 접착제, 벽지, 페인트 등에서 나오는 휘발성 유기화합물들은 거주자들의 건강과 실내 생활의 불쾌감을 유발시킨다. 이 휘발성 유기화합물들은 대표적 물질인 포름알데히드를 비롯하여 벤젠, 톨루엔, 아세톤, 스틸렌 등이 포함되어 있고 이러한 물질들은 장시간에 걸쳐 서서히 방출되어 거주자들에게 급성 또는 만성적인 질환을 야기한다. 유기 휘발성 물질들의 제거 방법으로는 흡착을 이용한 물리적 방법과 휘발성 물질을 다른 물질로 전환시키는 화학적 방법 또는 두 가지가 혼합된 방법이 주로 사용된다. 본 논고에서는 반응물질의 반응속도와 생성물의 방출을 제어하는 방법으로 얻어지는 서방형의 이산화염소 젤팩과 최적화된 홀 경을 가진 제올라이트 흡착제를 혼용하여 공기 중에 부유하는 포름알데히드를 흡착, 분해시켜 제거하는 효과적인 방법에 대하여 제안하는 바이다.

  • 콘크리트의 내구성을 향상시키는 기존의 공법과는 다르게, 시멘트 입자 표면에 아스팔트 막(膜)을 부분적으로 형성시킨 후 카본블랙을 처리(處理)하여 콘크리트의 전반적인 내구성(耐久性)을 현격히 향상시킨 고내구성(高耐久性) 포틀랜드시멘트(ACTPC)의 수화반응(水和反應) 특성(特性)을 X-Ray, SEM, 전도열량계 등을 사용하여 조사한 결과는 다음과 같다. ACTPC가 물과 접촉하면 발수성을 나타내는 아스팔트 막(膜)은 시멘트 입자(粒子)와 물의 접촉 및 시멘트 입자(粒子)로부터 각종 이온들의 용출(溶出)을 방해하여 초기(初期) 수화반응(水和反應)이 억제되지만, 카본블랙은 결정핵 역할을 하므로 일정기간이후의 수화반응(水和反應)을 촉진한다.

Construction of a Bacterial Artificial Chromosome Library Containing Large BamHI Genomic Fragments from Medicago truncatula and Identification of Clones Linked to Hypernodulating Genes

  • In the model legume Medicago truncatula, two mutants, sickle and sunn, exhibit morphologically and genetically distinct hypernodulation phenotypes. However, efforts to isolate the single recessive and single semidominant genes for sickle and sunn, respectively, by map-based cloning have so far been unsuccessful, partly due to the absence of clones that enable walks from linked marker positions. To help resolve these difficulties, a new bacterial artificial chromosome (BAC) library was constructed using BamHI-digested genomic fragments. A total of 23,808 clones were collected from ligation mixtures prepared with double-size-selected high-molecular-weight DNA. The average insert size was 116 kb based on an analysis of 88 randomly selected clones using NotI digestion and pulsed-field gel electrophoresis. About 18.5% of the library clones lacked inserts. The frequency of the BAC clones carrying chloroplast or mitochondrial DNA was 0.98% and 0.03%, respectively. The library represented approximately 4.9 haploid M. truncatula genomes. Hybridization of the BAC clone filters with a $C_{0}t-l$ DNA probe revealed that approximately 37% of the clones likely carried repetitive sequence-enriched DNA. An ordered array of pooled BAC DNA was screened by polymerase chain reactions using 13 sequence-characterized molecular markers that belonged to the eight linkage groups. Except for two markers, one to five positive BAC clones were obtained per marker. Accordingly, the sickle- and sunn-linked BAC clones identified herein will be useful for the isolation of these biotechnologically important genes. The new library will also provide clones that fill the gaps between preexisting BAC contigs, facilitating the physical mapping and genome sequencing of M. truncatula.

Characterization of by-products from organosolv pretreatments of yellow poplar wood (Liriodendron tulipifera) in the presence of acid and alkali catalysts

  • Organic by-products derived from cellulose and lignin during organosolv pretreatments of yellow poplar wood (Liriodendron tulipifera) in the presence of $H_2SO_4$ and NaOH as catalysts, respectively, were subjected to various analyses to elucidate their effects on further performance of biological ethanol fermentation and provide preliminary data for the structure and utilization of organosolv lignin. Monomeric sugars amounted to ca. 2.2-7.7% in the organosoluble fraction of the organosolv pretreatment with $H_2SO_4$, while significantly low amount of sugars (0.2-0.3%) were determined in that of the organosolv pretreatment with NaOH. In case of addition of $H_2SO_4$ during organosolv pretreatment of biomass, a fermentation of the organosoluble fraction could be considered as an essential process to increase an efficiency of biomass utilization as well as yield of bioethanol. Precipitates, insoluble by-products in the solvent mixture, were also cficiency oed by diverse analytical methods and revealed that these were typically composed of a lignin moiety regardless of catalyst. According to the results of nuclear magnetic resonance (NMR), Fourier Tcinsform Infrared Spectroscopy (FT-IR) and Gel permeation chromatograp r (GPC), the main components of precipitates seem to be lignin polymers. However, their structures could be slightly modified during pretreatment and mixed with some carbohydrates by chemical bonds and/or physical associations.

소나무 수피의 총합적 이용(제6보) -미분쇄 수피의 알칼리성 아황산염-안트라퀴논 증해와 증해폐액의 특성- (Utilization of Pine Bark(Part 6) -Alkaline Sulfite-Anthraquinone Cooking of Underground Bark and Characterization of the Spent Liquor-)

  • Alkaline sulfite-anthraquinone(AS-AQ) cooking process has been applied to unground pine bark. The properties of the spent liquor such as molecular weight distributions, sulfonic equivalent weights, degree of sulfonation, phenolic hydroxyl groups and kaolin dispersing ability have been investigated to evaluate the possibility of using this liquor as concrete additives or binders. In the case of ground bark meal, more than 90% delignification was achieved at the optimal cooking conditions. However, applying these conditions on unground bark the delignification was slightly retarded. The decrease in the degree of delignification may be attributed to less penetration and diffusion of chemicals during the cooking of the bark. Increasing the cooking temperature only by $5^{\circ}C$ improved the delignification of the bark and about 90% delignification can be achieved. These results indicate that bark can be used successfully during AS-AQ cooking without any mechanical or physical pretreatment. The properties of lignin or polyphenol sulfonates in the spent liquor after AS-AQ cooking of the bark were compared with Sunflo-R, which is commercial lignosulfonate(CLS) prepared from wood. The weight average molecular weights(Mws) estimated by gel-filteration chromatography was found to be ranging from 1,200~1,800. These are considerably lower than those in CLS. Lignin or polyphenol sulfonates in the spent liquor of bark and CLS have similar degree of sulfonation, but the phenolic hydroxyl group was 1.8 times higher than CLS. Moreover, the dispersing abilities of the spent liquors were better than that of CLS, especially after 0.1% addition to kaolin suspension.

Development, Structure, and Diversity of Microbial Lotic Calcareous Mat Communities

  • The phylogenetic diversity of microbial communities in calcareous mats from Spearfish Creek, a freshwater stream located in the Black Hills of South Dakota, was examined using PCR-based 16S rDNA sequence analysis. In this study, two types of calcareous mats were compared: mature mats formed on the natural substrate of rock surfaces and developing mats on an artificial substrate of glass slides. Among 63 unique isolates from a clone library of 16S rRNA genes from mature mat samples, there were 8 phyla of Bacteria represented. The predominant phylum was Proteobacteria (48%), with the $\beta$ subclass being the largest group. Denaturing gradient gel electrophoresis (DGGE) analysis of 16S rRNA genes from slide samples collected at intervals for four months showed considerable diversity of the microbial community from the earliest stages of community development. Amplicons isolated from DGGE gels and sequenced indicated that community succession has occurred without increasing microbial diversity. However, light microscopic analysis revealed a significant increase in microbial cell density throughout the community development. Scanning electron microscopy of mat samples provides evidence that diatoms are also important members of calcareous mat communities.

Review of Rice Quality under Various Growth and Storage Conditions and its Evaluation using Spectroscopic Technology

  • Purpose: Grain quality is a general concept that covers many characteristics, ranging from physical to biochemical and physiochemical properties. Rice aging during storage is currently a challenge in the rice industry, and is a complicated process involving changes in all of the above properties. Spectroscopic techniques can be used to obtain information on the quality of rice samples in a non-destructive manner. Methods: The objective of this review was to highlight the factors that contribute to rice quality and aging, and to describe various spectroscopic modalities, particularly vibrational and hyperspectral imaging, for the assessment of rice quality. Results: Starch and protein are the main components of the rice endosperm, and are therefore key factors contributing to eating and cooking quality. While the overall starch, protein, and lipid content in the rice grain remains essentially unchanged during storage, structural changes do occur. These changes affect pasting and gel properties, and ultimately the flavor of cooked rice. In addition, grain quality is significantly affected by growing and environmental conditions, such as water availability, temperature, fertilizer application, and salinity stress. These properties can be evaluated using spectroscopic techniques, and rice samples can be discriminated by using multivariate statistical analysis methods. Conclusion: Hyperspectral imaging and vibrational spectroscopy techniques have good potential for determining rice quality properties in a non-invasive manner, i.e., not requiring the introduction of instruments into the rice grain.

저지방 마요네즈의 물성과 관능적 특성 (Sensory and Physical Properties of Low-fat Mayonnaise Made with Starch-based Fat Replacers)

  • 맛, 조직감, 안정성 등에 영향을 미치지 않는 지방대체물질을 이용한 저열량, 저지방 마요네즈를 제조하기 위하여 4종류의 지방대체물질에 대한 점도를 측정하고, 지방대체물질의 식용유에 대한 유지대체비율을 달리하여 제조한 저지방 마요네즈에 대해 물성검사와 관능 검사를 시행하였다. 지방대체물질 paste는 모두 의가소성유체의 특성을 보였고, 의가소성과 항응복력은 Neutral DR(DR), N-Lite L(NL), Sta Slim 150(SS), Stellar 100X(SX)순으로 증가하였다. 지방대체물질이 첨가된 마요네즈의 경도와 부착성도 위와 같은 순으로 증가하였고, 지방대체물질의 첨가비율이 높은 마요네즈일수록 경도와 부착성이 낮았다. DR이 첨가된 마요네즈의 spreadability는 DR paste의 점조도지수를 잘 반영해준다. 지방대체물질의 이용은 마요네즈의 oily odor와 greasy aftertaste를 감소시켰다.

게 페이스트 첨가 패티의 제조 및 특성 (Preparation and Characteristics of Patty with Red-Tanner Crab (Chionoecetes japonicus) Paste)

  • New types of patties were prepared with surimi and crab paste, and then characterized. With an increase of additional ratios of crab paste, the moisture (64.8 to $61.5\%$) of the patties slightly decreased while the ash contents (2.3 to $3.7\%$) increased. The Hunter's color values of the patties decreased in lightness (L value, 58.79 to 54.27) but increased in redness (a value, -2.77 to 3.54). Thus, the patties that had added crab paste appeared slightly more reddish. According to the increasing of additional ratios of crab paste, sensory scores on color and flavor increased whereas the texture score decreased. From the above results, based on physical properties and sensory evaluation, the desirable additional ratio of crab paste for preparing patty was $15\%$. The calcium and phosphorus contents of the patties with the addition of $15\%$ crab paste were 148.8 mg/100 g and 139.6 mg/100 g, respectively, and their ratio (approximately 1:1) was the optimal range in body absorption efficiency. The calcium absorption rate with the $15\%$ additions was 4.9 times higher than that of the patty with no additions. Total amino acid content (16,302 mg/100 g) of the $15\%$ additions showed no difference to the patty without additions, and the major amino acids were aspartic acid, glutamic acid, leucine and lysine. Exceptionally, the cysteine content of the patty with the $15\%$ crab paste additions was 2 times higher than that found in the patty without crab paste. The major fatty acids were 16:0, 18:1n-9 and 18:2n-6 of the total lipids, and there was no difference between the patties with and without crab paste. The above results showed that crab paste can be used as a crab surimi gel source.
