Effect of Light Intensity and Nitrogen Source on Hydrogen Production Using Rhodobacter sphaeroldes KD131 (홍색 비유황 광합성 세균 Rhodobacter sphaeroldes KD131의 수소생산에 미치는 빛 세기 및 질소원의 영향)

  • Photobiological hydrogen production using Rhodobacter sphaeroides KD131 was studied on the effect of light intensities and nitrogen sources. Media containing malate and glutamate were shown higher hydrogen production rate than that containing succinate and $(NH_4)_2SO_4$ at the $110\;W/m^2$ illumination by halogen lamp at $30^{\circ}C$. Media lacking glutamate as the nitrogen source exhibited higher hydrogen production than that containing glutamate. Initial cell concentration was optimized to 1.0 at the absorbance of 660 nm. Hydrogen production was increased by increasing the light intensity from 0 to $216\;W/m^2$ but the increasing rate declined over $108\;W/m^2$.

  • A microbiological hydrogen production process was optimized. Anaerobic photosynthetic bacteria like Rhodospirillum rubrum which is known to produce hydrogen from carbon monoxide efficiently and remove sulfur was used. To evaluate the potenital of this microorganism the optimization of media fermentation condition light intensity and light requirement for CO conversionwas tried in batch cultures and the continuous fermenter was also applied for this process. The gas residence time on CO conversion was sought out to get high conversion of carbon monoxide to hydrogen. Through this study the possibility of microbial synthtics gas concersion process was proposed.

  • Chlamydomonas reinhardtii UTEX 90 was cultivated with continuous supply of 2% $CO_2$ using TAP media at $25^\circ{C}$ and produced biomass 1.18 g of dry cell weight/L for 4 days. C. reinhardtii algal biomass(CAB) was concentrated to 20 times by volume and converted into hydrogen and organic acids by anaerobic fermentation using Clostridium butyricum. Organic acids in the fermentate of CAB were consecutively used to produce hydrogen by Rhodobacter sphaeroides KD 131 under the light condition. Approximately 52% of starch in the concentrated CAB which had 4-5.8, 24-26 and 6-7 g/L of starch, protein and fat, respectively was degraded by Cl. butyricum at $37^\circ{C}$. During this process, hydrogen and some organic acids, such as formate, acetate, propionate, and butyrate, respectively were produced. Further conversion of the organic acids in anaerobic fermentate of CAB by Rb. sphaeroides KD131 produced hydrogen from the anaerobic fermentate under the illumination of 8 klux using halogen lamp at $30^\circ{C}$. The result showed that hydrogen was evolved by the anaerobic conversion using Cl. butyricum and then by the photosynthetic fermentation using Rb. sphaeroides KD131. It indicated that the two-step conversion process produced the maximum amount of hydrogen from algal biomass which contained carbohydrate, protein, and fat via organic acids.

  • 유기성 폐기물을 이용하여 생물학적 수소생산 통합화 시스템 연구를 수행하였다. 통합화 시스템은 유기성폐기물의 전처리, 2단계 혐기발효 및 광합성 배양으로 구성된 생물학적 수소생산 공정, 초임계수 가스화 공정, 생산된 가스의 저장, 분리 및 연료전지를 이용한 전력 생산으로 구성되었다. 실험에 사용된 유기성 폐자원은 식품공장 폐수, 과일폐기물, 하수슬러지이며, 전처리는 폐기물에 따라 열처리 및 물리적 처리를 하였으며, 전처리된 시료는 생물학적 수소생산 공정에 직접 적용되었다. Clostridium butyricum 및 메탄 생성조에서 발생하는 하수슬러지중의 미생물 복합체는 수소생산 혐기 발효공정에 사용되었으며, 광합성 수소생산 미생물인 홍색 비유황 세균은 광합성 배양에 사용되었다. 생물학적 공정에서 발생하는 미생물 슬러지는 초임계수 가스화 공정으로 수소를 발생하였으며, 슬러지 중의 COD를 저하시켰다. 생물학적 공정 및 초임계수 가스화 공정에서 발생하는 수소는 가스탱크에 가입상태로 저장한 후, 95%순도로 분리하였으며, 정제된 수소는 연료전지에 연결하여 전력 생산을 하였다.

Utilization of Chitosan-glucan Complex Extracted from Ganoderma Iucidum Wastes as Bioflocculant (생물응집제로서 폐영지박 Chitosan-glucan 복합물의 이용성)

  • For the purpose of development of non-toxic and biodegradable flocculant, chitosan complex was isolated from Ganoderma lucidum wastes. The isolated complex was identified as the expected chitosan-glucan complex by IR specta. The complex was extracted by treatment of 50% NaOH solution at 120$\circ$C for 5 hrs, namely optimal condition and solubilized with 2% acetic acid for fur-ther use as flocculant. Preliminary experiments showed that the solubilized complex had higher flocculation activity of 1.3 fold than commercial chitosan at 400 mg/l concentration in soybean curd wastewater. Also the solubilized complex removed 83% of MLSS and 60% of COD in the soybean curd wastewater treated by photosynthetic bacteria, 50% of turbidity and 21% of MLSS in sugar industry wastewater, and 90% of turbidity and 89% of MLSS in alcohol fermentation wastewater. Bacterial cell flocculation activities of the solubilized chitosan-glucan complex were 89% in Bacillus subtilis broth, 81% in Streptococcus lactis broth, and more than 90% in Escherichia coli broth after standing for 2 days. The results reveal that chitosan-glucan complex from Ganoderma lucidum wastes can substitute for commercial chitosan as non-toxic and biodegradable flocculant.

Mass Cultivation and Secondary Metabolite Analysis of Rhodobacter capsulatus PS-2 (광합성세균 Rhodobater capsulatus PS-2의 대량배양 최적화 및 대사산물 분석)

  • Plant growth promoting (PGP) hormones, which are produced in a small quantity by bacteria, affect in plant growth and development. PGPs play an important role on the crop productivity in agricultural field. In this study, a photosynthetic bacterial strain producing the PGP was isolated from paddy soil. Bacterial isolate was gram negative, rod-shaped and motility positive. From the 16s rRNA gene sequence analysis, the isolate was identified as Rhodobacter capsulatus PS-2. The mass cultivation of R. capsulatus PS-2 was optimized by considering of the carbon, nitrogen and inorganic salt sources. Optimal medium composition was determined as Na-succinate 4.5 g, yeast extract 5 g, $K_2HPO_4$ 1 g, $MgSO_4$ 5 g, per liter. From the result of 500 L fermentation for 2 days using the optimal medium, the viable cells were $8.7{\times}10^9cfu/mL$. R. capsulatus PS-2 strain produced the carotenoid and indole-3-acetic acid (IAA). The carotenoid extraction and quantitative analysis were performed by HCl-assisting method. Total carotenoid contents from R. capsulatus PS-2 culture broth were measured as $7.02{\pm}0.04$ and $6.93{\pm}0.05mg/L$ under photoheterotrophic and chemoheterotrophic conditions, respectively. To measure the productivity of IAA, colorimetric method was employed using Salkowski reagent at optical density 535 nm. The results showed that the highest content of IAA was $197.44{\pm}5.92mg/L$ in the optimal medium supplemented with 0.3% tryptophan.

Determination of tyrosinase inhibitory activity and betanin content changes in beetroot (Beta vulgaris) extracts fermented by EM

  • Beet (Beta vulgaris) is a crop similar to sugar beet, chard and leaf beets, and its origin is the Mediterranean coast of southern Europe and Central Asia. Among the components contained in beet, betalain, the main component of the root, has been reported to prevent lipid peroxidation induced by active oxygen and free radicals due to its high radical scavenging ability. Among these, the betalain, betanin (Betanidin 5-O-${\beta}$-glucoside) contains both phenolic and cyclic amine groups, all of which are highly electron-donating and act as antioxidants and has tyrosinase inhibitory activity. Betanin accounts for about 75-95% of the total pigment found in the beet. EM stands for effective microorganisms and is a collection of beneficial microorganisms. EM includes yeast, lactic acid bacteria, mycelia, photosynthetic bacteria, actinomycetes, etc. Human patch test according to CTFA guidelines was observed to be a safe source of no stimulation when 5% (v/v) of the EM fermentation liquid was applied to the human body. In addition, beneficial microorganisms are synergistic in the process of co-existence and cultivation and it has the effect of increasing antioxidant capacity and inhibiting corruption. This study confirms the difference in tyrosinase inhibitory activity and betanin content of beetroot extracts and EM fermented beetroot extracts. Hence, these results confirm that EM fermented beetroot extracts are highly beneficial for the human body.

Effects of Physico-chemical and Microbiological Inhibitors for Odour gas Evolution in the Fermentation of Liverstock Feces (가축(家畜) 분뇨(糞尿) 발효시(醱酵時) 악취(惡臭)가스 생성억제제(生成抑制劑) 시용(施用) 효과에 관(關)한 연구(硏究))

  • A series of laboratory experiment was conducted to find out the effects of physico-chemical, microbiological and commercial inhibitors on the odorous gas evolution in the fermentation of livestock feces. The results obtained were summarized as follows. 1. The rate and amount of evolution of gas were the highest at 7 days after incubation, thereafter gradually decreased until 24 days after incubation. 2. The rate and amount of gases were evolved in order of $CO_2>N_2O>CH_4>NH_3>N_2S$, respectively. 3. The highest amount of methane gas was evolved from the poultry feces, those of carbon dioxide and nitrous oxide were evolved from the pig feces, and that of hydrogen sulfide was dominantly evolved from the cattle feces. 4. Negative correlation were obtained between the total amounts of $NH_3$ and $CH_4$, $CO_2$ and $CH_4$, $N_2O$ and $CH_4$, $N_2O$ and $CH_4$, while positive correlations were obtained between the amounts of $CO_2$ and $N_2O$, $CO_2$ and $NH_3$, and $NH_3$ and $N_2O$, respectively. 5. There was no significantly inhibiting effect obtained that the application of commercial gas inhibitor as VK 88. On the other hand there was significantly inhibiting effect obtained when application of fertile paddy soil and photosynthetic bacteria to the fermentation of livestock feces.
