Photosensitization Effect and Structure-Activity on Mutagenic Potential by 4-(Nitrobenzyl)Pyridine (4-NBP) Test, of Epoxides, Olefins and Alkylating Agents (4-(Nitrobenzyl)Pyridine에 의한 에폭시드 및 알킬화합물의 변이원성 잠재력에 대한 구조활성 및 광화학효과의 연구)

  • This paper reviews the results of a series of efforts to develop structure-activity models for slow-reacting chemicals and olefins whose toxicity may be enhanced by the ultraviolet radiation. Photoinduced toxicity of 14 compounds was found to be a different result of competing factors of structure, having carbon-carbon double bonds. To mimic the biological consequences of photooxidative damage in mammalian cells, the photochemical mutagenicith of 14 chemicals was tested in the CAS. Simple olefins were photochemically mutagenic or carcinogenic with irradiation, increasing the alkylating activity from zero level to 0.87(abs/gram) for styrene, 0.25 for 1-butene, 0.11 for 1-hexene, respectively, whereas no photochemical mutagenicity was observed with 1-octene in the absence of the CAS. Oxide compounds, however, showed a decreasing trend of photoalkylating activities in the presence or absence of the CAS. We found that the structure-activity relationship was not applicable to our data.

  • The UV action spectra and quantum yields for photodestruction of tryptophan (Trp) in peptide such as N-acetylphenylalanyl-L-tryptophan (NAPT) and 3-methyl indole (scatole) were determined in aerated aqueous and organic solvents. The photodestruction of aqueous NAPT was shown to be initiated by photoionization without requirement of threshold energy, as demonstrated by the similarity of fluence effect curves obtained for the action at various wavelengths and the wavelength dependence of quantum yield comparable to that reported for the photoionization of L-Trp. N-formylkynurenine (NFK)-type photoproduct, which is a photodynamic sensitizer, was not found to be involved in the photodestruction of Trp in NAPT in aqueous solution. In contrast, the action spectra of NAPT and scatole in organic solvents have revealed evidences for the significant role of internal photosensitization by NFK-type photoproduct in photolysis of Trp in peptide.

Blue Light Photosensitization in Mitochondrial Membrane of Plant Cells (식물세포 미토콘드리아막에서 일어나는 청색광 Photosensitization)

  • Plant mitochondria, irradiated with blue-colored $sunlight(350{\sim}500nm)$ under aerobic and anaerobic conditions, were assayed as to the electron transfer activity of respiratory enzyme system, and compared with those irradiated with orange-colored light(white sunlight minus blue-colored light). The respiratory activity of mitochondria was most seriousely inhibited by illumination with blue-colored light under aerobic condition. Deaeration of mitochondrial suspension resulted in substantial decrease of the photoinhibition by blue-colored light. Meanwhile, orange-colored light demonstrated much less effectiveness-almost ineffectiveness-in causing the inhibition of mitochondrial respiration system. The results of enzymatic assay revealed a strong possibility that FMN in NDH and heme group at least in cytochrome c oxidase, but not FAD in SDH, are the photodynamic sensitizers in mitochondrial inner membrane. Also worthwhile to note is the significant difference from the others of SDH in its photoinhibitory response to the light quality of visible light; that the inhibition of SDH by irradiation was not affected by atmospheric condition and that orange-colored light gave rise to considerable extents of inhibition to the enzyme. This observation was tentatively interpreted in terms of photosensitized reaction not involving molecular oxygen possibly catalyzed by Fe-S centers in the enzyme. The superoxide production and the membrane peroxidation of mitochondria under various treatments also indicated that there was blue-light photodynamic reaction in mitochondria involving active oxygens.

A Spectral Comparison Study of PDT Drugs - ALA and ALA-Hexyl ester

  • 5-aminolevulinic acid (ALA) has been used to stimulate endogenous protoporphyrin IX (PpIX) in tumor and then initiate PDT. Recently, ALA-Hexyl ester (He-ALA) was found much effective than ALA on producing PpIX in cancer cells. To clarify the transportation mechanism of ALA and He-ALA, the detection of them is the important step. ALA and its derivatives all don't emit fluorescence, so the Raman spectroscopy was used here for the direct detection of ALA and He-ALA. The results showed that ALA and He-ALA have the common strong Raman peaks at 2930, 2950 CM$\^$-1/, due to the CH$_2$ vibration. The peak 3050 CM$\^$-1/ appeared in ALA spectrum can be attributed to OH vibration, while the peaks of 2860, 2900 CM$\^$-1/ in He-ALA spectrum were assigned as the modes of CH$_3$. This Raman spectral characteristic is consistence with the structure difference of He-ALA and ALA. Thus, Raman spectroscopy provides a new way to detect and distinguish ALA and He-ALA, and could be explored further in biology system.

  • The electrochemical spectral sensitization of $SnO_2$ electrodes with rhodamine B was studied. Photocurrent was measured in the presence of a variety of supersensitizers including ascorbic acid and KI. In the presence of ascorbic acid, the increase in the concentration of ascorbic acid, pH of the solution, or the potential applied to $SnO_2$ up to 0.6 V vs. SCE enhanced the photocurrent. However, ascorbic acid produced considerably high dark current due to its low reduction potential. On the other hand, KI produced low but stable photocurrent. The results, together with the solvent effect on the photocurrent, were taken into account to elucidate the mechanism of photosensitization in the presence of ascorbic acid or KI.

  • Irradiation of an acetonitrile solution containing dimethylbis(4-vinylphenylmethyl)silane 1a in the presence of 9,10-dicyanoanthracene leads to formation of the intramolecular photocycloadduct, 2-sila-cyclobuta[2.3]cyclophane(2a). In contrast, prolonged irradiation gave insoluble polymeric material. The photocycloreversion of 2a occurs efficiently (quantum yields exceeds unity) by use of redox-type photosensitization in the presence of magnesium perchlorate. The transient absorption spectra generated by pulse radiolysis and gamma-radiolysis show that the radical cation species generated from 1a is different from that arising from 2a.

  • As sunlight not always optimized for every terrestrial plant in terms of light quality, quantity and duration, some plants suffer detrimental effects of sunlight exposure under certain conditions. Photoinhibition of photosynthesis is a typical phenomenon representing harmful light effects, commonly observed in many photosynthetic organisms. It is generally accepted that functional, structural loss of photosystem II complex(PSII) is the primary event of photoinhibition. Accumulating data also suggest that singlet oxygen($^1$O$_2$) is the main toxic species directly involved in it. There are two different views on the specific site and mechanism of $^1$O$_2$ production in the photosynthetic membrane. One of them favors the PSII reaction center, where the primary charge pairs recombination occurs as a prerequisite for the generation of $^1$O$_2$, and the other inclines to photosensitized $^1$O$_2$ formation by a substance located outside PSII. This article describes how we, as the advocators of the latter concept, have arrived at the conclusion that $^1$O$_2$ immediately involved in PSII photodamage is largely generated from the Rieske center of the cytochrome b$_{6}$/f complex and diffuses into PSII, attacking the reaction center subunits.s.

Photodynamic Therapy for Methicillin-resistant Staphylococcus aureus Using Various Photosensitizer

  • The aim of this study was to evaluate the photodynamic effect of various photosensitizing agents against methicillin-resistant Staphylococcus aureus (MRSA). MRSA was exposed to light from a 632 urn diode laser (15 J/$cm^2$) in the presence of various photosensitizer, such as photofrin, photogem, radachlorine and ALA. In vivo study was performed using ICR mice. Twenty eight mice had a standard wound ($100\;mm^2$) created on the dorsum, and MRSA was inoculated into the wound region. The four groups were classified as follows: (1) the untreated control group (bacteria alone), (2) the bacteria plus light group (15 J/$cm^2$), (3) the bacteria plus photofrin group (kept in the dark), and (4) the photodynamic therapy (PDT) group (bacteria, photofrin, and light). After photofrin (dose 1 mg/kg) injection, the experimental group was irradiated with 632 urn diode laser (15 J/$cm^2$) for 30 minutes after In vitro results of PDT showed the complete killing of MRSA at the photofrin, radachlorine, and photogem However, ALA-PDT was ineffective on MRSA viability. In vivo results showed that photofrin has therapeutic effect on the wound infection. These results demonstrate that selective lethal photosensitization of MRSA can be achieved using phofrin, photogem and radachlorin. Thus, PDT can inactivate MRSA survival.

The Effects of Photodynamic Therapy for Vancomycin-resistant Enterococci

  • The aim of this study was to evaluate the effects of the photosensitizer photogem with light-emitting diode (LED) on vancomycin-resistant enterococci (VRE). Two VRE strains isolated from the feces of patients. that was identificated Enterococcus faecium (vanA) and Enterococcus gallinarum (vanC1) using traditional biochemical tests and confirmed VRE genotyping from using polymerase chain reaction. In addition, three strains were used Enterococcus. faecalis CDC-286 (vanA), E. faecalis CDC-583 (vanB) and E. gallinarum CDC-42 (vanC1). To examine the antimicrobial effect of photogem mediated photodynamic therapy (PDT) against, CFU quantification and Disk diffusion antimicrobial susceptibility test were evaluated. The effects of Photodynamic therapy was not associated with genotype. Photogem mediated PDT perfectly inhibited the colony formation of E. faecalis CDC-286. The number of viable bacteria decreased greatly after PDT application with photogem $50{\mu}g/mL$ and energy density of $15J/cm^2$. The diameter of inhibition zone was increased to after PDT more than before PDT. The case of vancomycin disc on E. faecalis CDC-583 and E. galinanum-Patient were changed from resistant to intermediate resistant, from intermediate resistant to susceptable. These results demonstrate that lethal photosensitization of VRE can be achieved using photogem plus 630 nm LED irradiation.

QUANTUM EFFICIENCY OF PHOTOGENERATION OF SINGLET OXYGEN FROM THE CLUSTER TYPE OF BINUCLEAR IRON-SULFUR CENTER [2Fe-2S]

  • Photosensitization via the singlet oxygen ($^1O_2$) mechanism by the binuclear iron-sulfur center, denoted as [2Fe-2S], was investigated, using a highly purified ferredoxin (Fd) preparation from spinach leaves. Since the apoprotein of Fd contains a good number of amino acid residues that are readily reactive with $^1O_2$ and thus interfere with the detection of $^1O_2$ generated from [2Fe-2S], we attempted to deprive the $^1O_2$-sensitive residues of their $^1O_2$-scavenging capacity as much as possible by treating Fd with rose bengal plus 550 nm monochromatic light and thereby photooxidatively degrading these residues. The photochemically modified Fd was found to keep the structural integrity of its Fe-S group virtually unaffected by the treatment. By employing chemical trap method for measurement and examining the kinetic effects of azide and deuterium oxide on the reactions of $^1O_2$ with various trap compounds, we were able to demonstrate that [2Fe-2S] indeed acts as a photosensitizer via $^1O_2$. Further, the minimum quantum yield of $^1O_2$ production by [2Fe-2S] was estimated to be 0.0047.
