Changes of Lipid Composition of Korean Black Soybean before and after Soaking (수침과정 전후의 한국산 검정콩의 지질성분 변화에 관한 연구)

  • Total lipids of Korean black soybean (Glycine man Merr) during soaking in water were extracted, purified and fractionated into three lipid classes, and then lipid contents and their fatty acid compositions were investigated. The lipids of the beans consisted of 89.1% neutral lipids, 1.5% glycolipids and 9.4% phospholipids, and these fractions did not change significantly during the soaking period. The neutral lipid fraction of the beans contained 92.1% triglyceride, 3.0% sterol esters and hydrocarbons, 2.8% diglyceride, 1.5% free fatty acids, 0.3% free sterols and 0.3% monoglyceride, and no significant changes were found in the composition of neutral lipid fraction from the soaked beans. Major components of the glycolipid fraction were esterified steryl glycosides (43.6%), steryl glycosides (26.6%) and digalactosyl diglycerides (14.5%), and these fractions did not change significantly during the soaking period. On the other hand, phosphatidyl choline (41.6%) and phosphatidyl ethanolamine (39.5%) were most abundant components found in the phospholipid fraction, and the contents of phospholipids changed a little during the soaking period. Linoleic acid, oleic acid and palmitic acid were the major fatty acids found in total lipids, neutral lipids, glycolipids and phospholipids. A few changes in the major fatty acid compositions of phospholipids were observed during the soaking period.

Studies on the Lipid Composition of Bush Clover (Lespedeza bicolor) Seed (싸리종자(Lespedeza bicolor)의 지질성분(脂質成分)에 관한 연구(硏究))

  • Lipids in Bush Clover (Lespedza bicolor) seed were extracted with the mix ture of chloro-form-methanol (2 : 1, v/v) and then fractionated into neutral lipids, glycolipids and phospholipids by silicic acid column chromatography. Components and fatty acid composition of each fraction were determined by thin layer and gas chromatographies. The results were summarized as follows. In Bush Clover seed, the contents of neutral lipids, glycolipids and phospholipids were 71.75%, 23.26% and 4.99% respectively. Triglycerides(61.90%) and free fatty acids(22.04%) were the major components among the neutral lipids. Esterified sterols, free sterols, diglycerides and monoglycerides were the minor components. The major components of glycolipids were monogalactosyl diglycerides(38.19%) the others were esterified steryl glycosides, cerebrosides and digalactosyl diglycerides. The major components of the phospholipids were phosphatidyl cholines(36.46%), phosphatidyl inositols(21.52%) and phosphatidyl ethanolamines(17.29%). The major fatty acid of total lipid, neutral lipids and glycolipids were linoleic acid, linolenic acid, oleic acid and palmitic acid. On the other hand, predominate fatty acid of phospholipids were linoleic acid, palmitic acid, linolenic acid and stearic acid.

Roles of Phospholipids in Flavor Stability of Soybean Oil (대두유 향미안정성에 있어서 인지방질의 역할)

  • The effects of phosphatidyl choline (PC), phosphatidyl ethanolamine (PE), phosphatidyl inositol (PI), phosphatidic acid (PA), phosphatidyl glycerol (PG), and cardiolipin (CL) on the flavor stability of purified soybean oil were studied. Purified soybean oil obtained from soybean oil by silicic acid chromatography does not contain measurable iron, tocopherols and phospholipids. Three hundred ppm of PC, PE, PI, PA, PG, or CL was added to the purified soybean oil, with and without 1ppm ferrous iron added. The flavor stability of sample, which was stored at $60^{\circ}C$ for 10 days in dark oven, was determined by a combination of volatile compounds formation and molecular oxygen disappearence in the headspace of air-tightly sealed serum bottle every 48 hrs. Results showed that, in general, phospholipids worked as prooxidant in the pufified soybean oil without ferrous iron added, and worked as antioxidant in the oil, when added 1ppm ferrous iron. The results also suggest that phospholipids work as prooxidant by increasing the solubility of oxygen on the surface of oil, and work as antioxidant in the oil containing 1 ppm ferrous iron by chelating iron. The results showed that PE and PA are better antioxidants than PC and PG. CL and PI showed the lest antioxidant activities in the oil will 1ppm ferrous iron added.

Effect of 2,4-Dichlorophenoxyacetic Acid (2,4-D) and Naphthoxyacetic Acid (NOA) on the Biochemical Changes in the Fat Body and Haemolymph of the Silkworm, Bombix mori L.

  • The effect of topical application with 200,400 and 600 $\mu$g/ml 2,4-Dichlorophenoxyacetic Acid (2,4-D) and Naphthoxyacetic Acid (NOA) to the fifth stadium larvae of the silkworm, B. mori on fat body glycogen, protein, total lipids, phospholipids, neutral lipids and haemolymph trehalose and protein has been studied. The fat body glycogen significantly increased in all the groups treated with 2,4-D whereas there is no significantly change in fat body glycogen in all the groups treated with NDA. However, the haemolymph trehalose was significantly increased in all 274-D and NOA treated groups except in the groups treated with 400 and 600 $\mu$g/ml 2,4-D where the increase was not significant.The fat body protein did not show any significant change in all groups treated with 2,4-D and NOA except in the groups treated with 200 $\mu$g/ml 2,4-D where the fat body protein was significantly increased. The total lipids, phospholipids and neutral lipids of the fat body increased significantly in all the groups treated with 2,4-D and NOA when compared with those of the corresponding parameters of the carrier control.

  • The effect of topical application with 5,10 and 15 $\mu$g/ ml androstenedione and methyltesterone to the fifth stadium larvae of the si1kworm,B. mori on fat body glycogen, protein, tota1 lipids, phospholipids, neutral lipids and haemolymph trehalose and protein has been studied. The fat body glycogen increased significantly in all the treated groups except in the 15$\mu$g/ml androstenedione treated group. However the fat body glyco- gen increased significantly in 10 ${\mu}g$/ml methyltesterone treated group. The androstenedione treated groups showed increased haemolymph trehalose but the increase was not significant. However the haemolymph trehalose increased significantly in all the methyltestosterone treated groups. The fat body protein decreased in all the androstenedione and methyltesterone treated groups except 15 ${\mu}g$/ml methyltesterone where the decrease in fat body protein was significant. Whereas that of haemolymph protein significantly decreased in all the groups except in the group treated with 5 ${\mu}g$/ml androstenedione where the decrease was not significant. The total lipids, phospholipids and neutral lipids of the fat body decreased significantly in all the groups treated with androstenedione and methyltestosterone when compared with those of carrier control.

  • Frog legs were stored before and after 30, 60, 90, 120 days at $-30^{\circ}C$ and $-70^{\circ}C$. The amounts of triglycerides, glycolipide and phospholipids by TLC and composition of fatty acids by GC were shown in this experiment, there fore, I have got these following results. 1. The amount of PL in frog legs consisted of 80% of the total lipids before storage. As the days of storage was longer, the amount of TG was increased, while PL and GL were decreased. 2. TG and ES & HC were composed of 66% to 77% of the neutral lipids composition before storage. As the days of storage was longer, the amount of MG, DG. FFA and ES & HC were increased, while FS and TG were decreased. 3. PE and PC were composed of 78% to 81% of the phospholipids composition before storage. As the days of storage was longer, the amount of FA and PS were increased, while PC and PE were decreased. 4. The composition of fatty acids in neutral lipids of frozen frog legs at $-30^{\circ}C$, 120 days was studied, Capric acid. Stearic acid, Behnic acid. Oleic acid and Linoleic acid were increased as compared with initial neutral lipids.

Gene Targeting of the Acyl-CoA Synthetase Specific to Arachidonate

  • The synthesis of acyl-CoA catalyzed by acyl-CoA synthetase (ACS, EC 6.2.1.3) from fatty acid, ATP, and CoA is a crucial reaction in mammalian fatty acid metabolism. In arachidonate metabolism, acyl-CoA synthetase(ACS) plays a key role in the esterification of free arachidonate into membrane phospholipids. Following its release by the action of calcium dependent phospholipase, free arachidonate is believed to be rapidly converted to arachidonoyl-CoA and reesterified into phospholipids in order to prevent excessive synthesis of eicosanoids. In previous studies, we have characterized five ACSs (designated as ACS1-5) with different tissue distribution. ACS1, ACS2, and ACS5 are similar in structure and fatty acid preference, and completely different from ACS3 and ACS4. The latter are arachidonate-preferring enzymes closely related in structure but expressed in different tissues: ACS3 mRNA is highly expressed in the brain and the mRNA for ACS4 is expressed in steroidogenic tissues including adrenal gland, ovary, and testis. To learn more about the potential function of ACS4 in arachidonate metabolism, we have produced knock-out mice for ACS4 gene. ACS4+/- females become pregnant less frequently and produce small litters with extremely low transmission of the disrupted alleles. Striking morphological changes including extremely enlarged uterine filled with numerous proliferative cysts of various size were detected in ACS4+/- females. Furthermore, marked accumulation of prostaglandins were seen in the uterus of heterozygous females. These results indicate that ACS4 is critical for the uterine arachidonate metabolism and heterozygous disruption of its gene lead to impaired pregnancy.

Hair Growth Promoting Potential of Phospholipids Purified from Porcine Lung Tissues

  • BP201, porcine lung tissue-derived phospholipids, consists of phosphatidylcholine as a major phospholipid species. BP201 promoted hair growth after application onto the shaved backs of BALB/c and C3H mice. Its effect was enhanced when applied together with minoxidil (MNX) in C3H mice. When the tissue specimens prepared from the shaved skins of BP201-treated and control mice were microscopically examined, the total numbers of hair follicles in both anagen and telogen phases of BP201-treated mice were significantly higher than those of control mice. The numbers of hair follicles in the anagen phase of BP201-treated mice were also higher than those of control mice. In combination with MNX, BP201 further increased the total number of hair follicles, but did not alter the percentage of hair follicles in the anagenic phase. BP201 also increased the proliferation of human hair follicle dermal papilla cells. Collectively, BP201 possesses hair growth promoting potential, which would suggest its use singly or in combination for hair growth products.

Changes in Fatty Acid Composition of Phospholipids from Human Milk in the Course of the Lactation (수유기간의 경과에 따른 인유 인 지방질의 지방산 조성의 변화)

  • Changes in fatty acid composition of phospholipids from human milk during the course of lactation were determined in milk from Korean lactating mothers giving birth at term. The levels of 8 : 0 increased significantly from colostral to transitional stages with a further small decrease at mature stage. The proportions of 10 : 0, 12 : 0 and 14 : 0 increased markedly from colostrum to transitional to mature milk. The w6-derived long-chain polyunsaturated fatty acids, 20 : 2w5, 20 : 4w6 and 22 : 4w5, were decreased with time of lactation. The levels of 24 : 1w9 were significantly higher in transitional milk than in colostrum and then significantly decreased to lowers than the colostral levels at mature milk.
