Carbohydrate Metabolism During Germination of Ginkgo (Ginkgo biloba L.) Seed (은행나무 종자의 발아에서 탄수화물 대사)

  • Ginkgo (Ginkgo biloba L.) seeds were analyzed to determine the level of soluble sugars and insoluble starch during germination. Also the activities of the hydrolytic enzymes such as amylase, invertase and phosphatase were compared. As amylase activity was sharply increased, significant decline of starch was observed in the female gametophyte and increase of soluble sugars occurred concurrently. Invertase activity was gradually increased in cotyledon and radicle, while it was very low in dry seeds. In addition, phosphatase activity was variable only in radicle, and acid phosphatase showed higher activity than alkaline phosphatase.hatase.

  • 조사해역에서의 Phosphatase에 의한 최대분해속도(Vmax)는 $0.26\;{\sim}\;7.09{\mu}M/L/hr$ 로서, 식물플랑크톤 bloom을 보였던 6월27일 조사 시에 높게 나타났으며 수온이 하강하고 클로로필 농도가 낮았던 9월13일 조사결과에서 가장 낮게 나타났다. 식물플랑크톤에 의한 유기 인산염 분해를 알아보기 위해 Specific activity를 구하였다. Phosphatase activity와 Chlorophyll. a의 변화는 유사한 변화를 보여주고 있으며, 이는 식물플랑크톤 이 Phosphatase의 분비에 큰 기여를 하는 것으로 나타났다.

Screening of Marine Microbial Extracts for Tyrosine Phosphatase 1B Inhibitors

  • Protein tyrosine phosphatase 1B (PTP1B) acts as a negative regulator of insulin signaling, and selective inhibition of PTP1B has served as a potential drug target for the treatment of type 2 diabetes. As part of our searching for PTP1B inhibitors from natural products, the extracts of marine microorganisms were screened for the inhibitory effects on the activity of protein tyrosine phosphatase 1B (PTP1B). Among the tested 304 extracts, 29 extracts exhibited inhibition rate ranging 40.1 - 83.6 % against PTP1B at the concentration level of $30{\mu}g/mL$.

Inhibitory Effect of Schizandrin on Toxicity of Paraquat (Paraquat 독성에 대한 Schizandrin의 억제효과)

  • Paraquat is a useful nonselective herbicide widely used throughout the world. However, accidental or intentional ingestion of the paraquat cause fetal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of DDB(dimethyl-4,4'dimethoxy-5,6,5',6'-dimethylene dioxyphenyl-2,2'-dicarboxylate) on paraquat toxicity. DDB (100mg/kg) was administered orally to SD rats lhr after paraquat(50mg/kg) injection. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups sGPT, sGOT, BUN, creatinine, MDA and alkaline phosphatase levels in blood and MDA, glucose-6-phosphatase activity in tissues were elevated by 2 to 5 times of normal values. However in schizandrin treated groups, sGPT, sGOT, MDA and alkaline phosphatase activity in blood and MDA and glucose-6-phosphatase activity were significantly decreased to notmal levels but not in biochemical parameters of nephrotoxicity, BUN and creatinine levels. Therefore, we concluded that schizandrin can be used as an antidote of pulmono, hepatotoxicity of paraquat.

  • Eelctron microscopic cytochemical methods reveal that acid phosphatase activity appears exclusively in vacuoles surrounding established symbiotes. However, copius amounts of acid phosphatase reaction product are visible between and around some of the degenerating symbiotes in the amebae after treatment of chloramphenicol. It is thought that bacteriostasis by chloramphenicol has served to lost the symbiotic interference to intracellular digestion by the ameba and possibly phodphatase enxymes have been implicated in phagocytosis and intracellular digestion of the symbiotic bacteria.

  • Pyrantal pamoate's anthelmintic activity is due to its action as a neuromuscular blocking agent. It is generally well tolerated. Transient rises in SGOT levels have been reported in the drug-treated patients. Decreased levels of serum alkaline phosphatase post treatment were found in yound dogs. The present study was performed to investigate the possible toxic effects of pyrantal pamoate in pregnant mice progenies. The drug was given in different doses to these mothers in the first, second and third trimester. Serum alkaline phosphatase, SGOT and SGPT of one or two month old offspring were monitored. SGOT levels showed an increase in some doses in one and two month old offspring where alkaline phosphatase showed a decrease in some doses in one and two month old offspring. The latter effect may be due to osteoblastic alkaline phosphatase inhibition. The effect on SGOT levels, however, was difficult to explain, but may be due to a toxic effect on liver cells or cardiac muscles.

Screening of Medicinal Herbs for Inhibitory Activity against Protein Tyrosine Phosphatase 1B (생약의 Protein Tyrosine Phosphatase 1B 저해활성 검색)

  • Protein tyrosine phosphatase 1B (PTP1B) is predicted to be therapeutic target in treatment of type 2 diabetes and obesity. Thus, in order to search for PTP1B inhibitors, we screened the inhibitory activity of PTP1B in the water extracts of 84 medicinal herbs. Among them, the extracts of Pini Folium, Magnoliae Cortex, Artemisiae asiaticae Herba, Schizonepetae Herba, Menthae Herba, Mume Fructus, Cimicifugae Rhizoma, and Amomi Cardamomi Fructus showed relatively significant (58-68%) inhibitory activity against PTP1B. Especially, the methylene chloride fraction of the methanol extract of Menthae Herba (81% inhibition at 30 ${\mu}g$/ml) showed more potent inhibitory activity against PTP1B than others.
