The Optimal Compositions of Sex Pheromones for Monitoring Major Lepidopteran Pests of Food Crops in Korea (식량작물 주요 나비목 해충의 성페로몬 조성 연구 현황)

  • In this review, the compositions of the sex pheromones of 22 lepidopteran species were analyzed and briefly arranged by comparing the research results conducted in different locations worldwide. These compositions can be used as monitoring tools for pests of rice, maize, leguminous crops, perilla, and sesame in Korea. The sex pheromone compounds and those compositions of four species were successfully identified within Korean populations. The optimal compositions of sex pheromones of three other species were selected through behavioral studies using synthetic chemicals of the known pheromone compounds in laboratories and fields, without chemical identification processes in the sex pheromone compounds of Korean populations. For a few species, the roles of the minor components of the sex pheromone volatiles should be re-assessed for use in the development of optimal monitoring lures.

  • In order to elucidate and characterize the signal transduction pathway(s) whereby yeast cells respond to mating pheromone, we have isolated mutants which are able to conjugate in the absence of the alpha-factor receptor. Sixty-one suppressors of a ste2-deletion mutation which also confer a ts conditional "start" arrest phenotypw have been subjected to genetic analysis. The mutants could be assigned to three complementation groups designated CDC70, CDC72 and CDC73, which are unlinked to each other as well as to the previously identified start genes. Quantitation of mating ability of the cdc70, cdc72 and cdc73 mutations in a ste2-deletion background gives levels ranging from 0.1% to 0.3% of wild type, depending on the allele and the gene. The results indicate that the signals from mating pheromone might be mediated by the CDC70, CDC72 and CDC73 products. products.

  • When the membrane protein fraction of mating type a cells of heterobasidiomycetous yeast R. toruloides was phosphorylated in vitro, two phosphorylated proteins of 72Kd and 57Kd were detected on SDS-polyacryamide gel. The phosphorylation reaction was inhibited by rhodotorucine A(Rh. A) which is a sexual pheromone secreted by mating type A cells. The inhibition of phosphorylation by Rh. A was dependent on $Ca^{2+}$, and independent on $Mg^{2+}$ or calmodulin. When adding trigger peptidase(TPase) inhibitor, antipain, no inhibition of phosphory was observed. Also, by adding the trysin-digested product of Rh. A, the phosphorylation was inhibited as the action of Rh. A. From these results, it is expected that the inhibition of membrane protein phosphorylation should be caused by the digested product of Rh. A with TPase.

A Practical Synthesis of (Z)-7-Eicosen-11-one and (Z)-7-Nonadecen-11-one, the Pheromone of Peach Fruit Moth, and Its Biological Activity Test

  • A new practical method is described for the synthesis of (Z)-7-eicosen-11-one (1) and (Z)-7-nonadecen-11-one (2), the active principles of the sex pheromone of the peach fruit moth. Lithium anion of 1-octyne (from acetylene and 1-bromohexane) was alkylated with 3-bromo-1-propanol tetrahydropyranyl ether and deprotected to afford 4-undecyn-1-ol, which was oxidized with pyridinium chlorochromate to 4-undecyn-1-al. 4-Undecyn-1-al was treated with 1-nonylmagnesiumbromide or 1-octylmagnesiumbromide to yield 7-eicosyn-11-ol and 7-nonadecyn-11-ol. Jones oxidation, followed by Lindlar catalytic hydrogenation afforded the target compounds. Biological activity of the synthetic pheromones as attractants for males of the peach fruit moth was tested in the field using polyethylene capsules as containers. For each vial containing about 3.0 mg of the synthetic pheromone, the number of trapped moths were counted.

  • (Z)-13-Eicosen-10-one, an active component of the sex pheromone of the peach fruit moth was synthesized from 4-oxo-tridecan-1-al and heptylidenetriphenylphosphonium ylide by Wittig reaction. The key intermediate, 4-oxo-tridecan-1-al, was synthesized by three different methods. The biological activity test of the synthetic pheromone as attractant for the male peach fruit moth was tested at several districts in Korea.

  • (Z)-13-Octadecen-1-yl acetate, the pheromone mimic of the Rice Leaf Folder Moth, Cnaphalocrosis medinalis, was synthesized from 1,13-tridecanediol in three steps. Monoacetylation of 1,13-tridecanediol followed by PCC oxidation gave 13-acetoxytridecan-1-al. Wittig olefination of the 13-acetoxytridecan-1-al with pentylidenetriphenylphosphonium ylide afforded (Z)-13-octadecen-1-yl acetate, the pheromone mimic of the Rice Leaf Folder.

  • Two major sex pheromone components (Z-8-dodecenyl acetate and E-8-dodecenyl acetate) are known in the peach fruit moth, Grapholita molesta. From a putative biosynthetic pathway of these sex pheromone components, delta 10 desaturase ($10{\Delta}$ DES) has been proposed to play a crucial role in synthesizing a species-specific stereoisomer of the double bond. However, its molecular identity was not known. This study determined a putative desaturase (Gm-comp1575) as a $10{\Delta}$ DES candidate from G. molesta transcriptome constructed from the sex pheromone gland. Its open reading frame encodes 370 amino acid sequence with a predicted molecular weight at 43.2 kDa and isoelectric point at 8.77. It was predicted to have four transmembrane domains and six glycosylation sites at N-terminal or cytosolic domains. A phylogenetic analysis with its predicted amino acid sequence indicated that Gm-comp1575 is closely related with known $10{\Delta}$ DES genes of other insects. Gm-comp1575 transcript was detected in female adults at sex pheromone gland and other abdominal tissues. RNA interference of Gm-comp1575 significantly reduced attractiveness of virgin females in apple orchard compared to control females. These results suggest that Gm-comp1575 is associated with sex pheromone biosynthesis of G. molesta.

  • Electroantennography, wind tunnel observation, and field trapping experiments were carried out to investigate the biological activity of synthetic sex pheromone in the onental tobacco budworm. Heli-couerpa assulta. Two major sex pheromone components of H. assulta, Z9-16' Ald and Z11-16: Ald. elicited a big EAG response in male, but not in female Their mIXture ratios did not give much influence on EAG size Fema]e H assulta showed a great EAG response only to its host plant e extract. EAG size also increased with the amount of mixture from 001 to 10 [lg but rather decreased w when the amount was 1 00 $\mug$. H. assulta always revealed a series of stereotyped behavior in a wind tunnel. The behavioral response was different when the males were stimulated with the sex pheromone containing some minor components, 16: AId and Z9.16: Ac, or being different in mixing ratios of the two major components. The best ratio of the sex phemmone components for a attracting H assulta male adults was 20-25: 1 between Z9-16: Aid and Z11-16: Ald in net house a and red pepper field experiments in Korea When the lure contamed Z9-16: OH, attracting power rapidly decreased. Flight of Matsucoccus thunbergianae Males in Response to Synthetic Pheromone Placed at Various Heights above Ground and the Wind Speed (합성(合成)페로몬의 지상(地上)높이별(別) 위치(位置) 및 풍속(風速)에 따른 솔껍질깍지벌레 수컷의 비행(飛行))

  • Matsucoccus thunbergianae is a major insect pest of Pinus thunbergiana in southern Korean peninsula. To study the flight behavior of M. thunbergianae males responding to the synthetic pheromone, five sticky traps were placed on a bamboo pole at various heights, between 0.1m and 2.0m above ground. A bait impregnated with the synthetic pheromone was placed at 0.1m, 1.0m or 2.0m above ground and the number of male catches on each trap was counted. In an open area, numbers of males caught per trap were not different between heights when the bait was placed at 2m or 1m above ground ; when the bait was placed at 0.1m height, male flight was aggregated near the ground. In a forest with low crown closure, trap catches on five traps on the same bamboo pole were not different one another when the bait was placed at 2m height, but most males were flying near the bait when it was placed at 1m height. In a dense pine forest, most males were flying around the bait regardless of the bait position. In all three places, most males were caught on the trap near the ground when the baits were placed 0.1m above ground. Thus, for monitoring the frontal zone of infestation of the scale, placing the pheromone trap near the ground was considered the most efficient. When the males perceived pheromone, they tended to fly in the air with low wind speed.

Seasonal Occurrence Trends of Hemipteran Bug Pests Monitored by Mercury Light and Aggregation Pheromone Traps in Sweet Persimmon Orchards (수은유아등과 집합페로몬 트랩에 의한 단감원 노린재류의 발생소장)

  • The seasonal occurrence trends of the brown-winged green bug, Plautia stali, and the brown malmorated stink bug, Halyomorpha halys, were monitored in sweet persimmon orchards in southern region of Korea using light traps (in 2000 and 2001), and aggregation pheromone traps (methyl (E, E, Z)-2 ,4, 6-decatrienoate)) of P. stali (in 2001). Light trap data showed that H. halys started to occur from the end of June, and reached its peak in early or mid August, while P. stali was mostly attracted to the traps from mid July to late August without any distinct attraction peak. Both species did not occur after September in the persimmon orchards studied. The attraction patterns of both species to aggregation pheromone traps were different from those observed in light traps. Both species were attracted to the pheromone traps from mid May to late August. The peak occurrence of H. halys could not be detected due to low catches. However, the period of peak attraction for P. stali was from late June to late August depending on the geographical locations. The aggregation pheromone traps of P. stali attracted more number of P stali than H. hails, but the light traps showed a reverse pattern. In both species, more females were attracted to the aggregation pheromone traps than males. The difference of attraction patterns between 2 types of traps was discussed in relation to content of soluble tannin in persimmon fruits.
