Development of New Natural Antioxidants for Cosmeceuticals (천연물 유래 항산화 기능성 화장품 신소재 개발)

  • New antioxidative substances for cosmeceuticals were screened from natural resources such as microbial metabolites, mushrooms, and medicinal plants. Four antioxidants were isolated from the fungal metabolite of Eupenicillium shearii and their structures were determined to be new phenolic compounds. The compounds were designated as melanocins A, B, C, and D. Melanocins $A{\sim}D$ exhibited free radical scavenging activity on DPPH and superoxide with $EC_{50}$ values of $21{\sim}94\;and\;7{\sim}84{\mu}M$, respectively, which were stronger activity than those of ${\alpha}-tocopherol$ and BHA. Melanocin A showed anti-wrinkle effects on the UV-irrated hairless mouse skin. A novel hispidin antioxidative compound designated as inoscavin A was isolated from the fruiting body of the mushroom, Inonotus xeranticus. Inoscavin A scavenged superoxide radical with $EC_{50}$ values of $0.03{\mu}g/mL$, and inhibited rat liver microsomal lipid peroxidation with $EC_{50}$ values of $0.3{\mu}g/mL$. Benzastatins $A{\sim}G$, the novel antioxidants isolated from the culture of Streptomyces nitrosporeus showed potent lipid peroxidation inhibitory activity with $EC_{50}$ values of $3{\sim}30{\mu}M$. A cyclopentene compound with strong hypopigmentary effect was isolated from the fungal metabolite of Penicillium sp. and identifed as terrein. Terrein significantly reduced melanin levels in a melanomacyte cell line, Mel-Ab. It showed 10 times stronger activity than kojic acid, but exhibited no cytotoxic effect even in $100{\mu}M$. It was suggested that terrein reduced melanin synthesis by reducing tyrosinase production by MITF down-regulation.

Screening of Antioxidants and Cosmeceuticals from Natural Plant Resources in Jeju Island (제주 자생식물로부터 항산화 및 화장품 기능성 소재 탐색)

  • Fifty-four natural plants from Jeju Island, Korea were extracted by 70% methanol. The extracts containing the highest total phenolic contents (TPC) (>250 mg gallic acid equivalents/g dry sample) were obtained from Ostrya japonica, Geranium thunbergii, Malus sieboldii, Ardisia japonica, and Agrimonia pilosa. DPPH inhibition activity was greatest in Ardisia crenata at 94.1%. A high correlation was observed between DPPH inhibition activity and TPC ($R^2=0.87$). Tyrosinase inhibition activities of more than 85% were obtained from the extracts of Persicaria filiformis, Rhus javanica, Alnus firma, and Myrica rubra. On the other hand, the P. filiformis and M. rubra extracts each showed more than 90% XOD inhibition activity. The five natural plants with the highest biological activities were also extracted by pressurized liquid (PLE, 100% methanol, 13.6 MPa, $40^{\circ}C$). The DPPH and tyrosinase inhibition activities were almost the same in both the 70% methanol and PLE extracts. The $IC_{50}$ for tyrosinase and elastase inhibition activities in PLE were 802 and 88 ppm in A. japonica, and 959 and 66 ppm in M. rubra, respectively.

Physicochemical and Antioxidant Properties of Broccoli Sprouts Cultivated in the Plant Factory System (식물공장 시스템에서 재배한 브로콜리 새싹의 건조방법에 따른 이화학 및 항산화 특성 연구)

  • Recently, an interest in functional foods has been increasing. It was recommended placing a short definition. Therefore, we performed research on the chemical functions and antioxidant ability of broccoli. This research is vital for preparing the most favorable conditions and environment for highly-functional broccoli. Broccoli produced after applying sprouting and light sources were used for research. The chemical properties of the broccoli, including composition, free sugar, citric acid, mineral and vitamin (A, C, E) content, were analyzed. In addition, the ability of broccoli compounds to reduce total phenolic compounds, SOD-liked activity, EDA (electron donating ability), and hydroxyl radicals were inspected. Total analysis relied on the SAS (statistical analysis system). Broccoli sprouts produced through plant factory system's photosynthesis, treated under different light sources, had superior amounts of crude protein, crude fat, and crude ash, compared to normal sprouts under fluorescent light. Is it a facility or does it refer to the inner metabolism of the cell? Broccoli sprouts under red light had superior amounts of glucose, fructose, malic acid, and oxalic acid, while broccoli sprouts under turquoise light had superior amounts of citric acid. Broccoli sprouts under white light had superior amounts of various minerals, such as potassium, magnesium, and sodium. In terms of antioxidant activity, data from the plant factory system shows an increase in EDA antioxidants (1.63 mg/mL, 30.82%). Sprouts applied with turquoise light had superior amounts of hydroxyl radical scavenging (65.62%), and sprouts applied with white light had superior amounts of activated SOD-like activity (52.69%). Research on dehydrated broccoli sprouts showed that sprouts dehydrated with cold air had superior amount of malic, citric, oxalic acid compared to sprouts dehydrated with hot air. In terms of vitamin levels, sprouts dehydrated with cold air had five times the normal amount of vitamin A and E, whereas sprouts dehydrated with hot air had higher amounts of vitamin C. Dehydration at low temperature also produced a higher amount of activated antioxidants (1.6 mg/mL of activated antioxidant ability, 63.04% of SOD-like activity, and 67.76% of hydroxyl radical scavenging). Our results show that antioxidant ability can vary by the type of photosynthesis and temperature level in which the sprouts are dehydrated. Therefore, thorough foundational data is required to product the most functional broccoli.

Determination of Antioxidants Contents in Various Plants Used as Tea Materials (다류소재 식물류 중의 황산화물질 함량 분석)

  • Forty plants used as tea materials were chosen for determining the contents of total phenolics, selenium (Se), ${\beta}-carotene$, ${\alpha}-tocopherol$ and ascorbate. Total phenolics and ascorbate contents were analyzed colorimetrically. The Se contents were measured by hydride-atomic absorption spectrometry. The contents of ${\beta}-carotene$ and ${\alpha}-tocopherol$ were simultaneously determined by high performance liquid chromatography using separate detectors, UV for ${\beta}-carotene$ and FL for ${\alpha}-tocopherol$ analyses. The contents of these antioxidants were as follows (per 100 g dry plant); Contents of total phenolics in green tea leaf, black tea leaf, oolong tea leaf and instant coffee were about 7 g and the Se contents in corni fructus and arrowroot were found to be about $4{\mu}g$, which were the highest among all plants used. Contents of ${\beta}-carotene$ in eucommiae cortex, persimmon leaf and green tea leaf were 8587, 6222 and $3652\;{\mu}g$ respectively. The persimon leaf contained the highest ${\alpha}-tocopherol$ content (33 mg) and then followed by eucommiae cortex (26 mg), green tea leaf (16 mg) and black tea leaf (13 mg) in order. Ascorbate contents were found to be high in green tea leaf (199 mg) and black tea leaf (117 mg).

PHENOLOXIDASE AND ANTIOXIDANT IN KOREAN GINSENG (고려인삼에 있어서의 페놀 산화효소의 항산화물질)

  • Enzymatic browning is considered desirable in tea and tobacco processing but undesirable in many fruits processing at the present time. It is necessary to understand the nature of the enzyme, phenoloxidase, in order to control browning reactions, and extend its effects to formation of browning products as antioxidants in ginseng. Ginseng exhibits antioxidant activity when incorporated with turkey dark meat patties. The activity in red ginseng showed about two times stronger than white ginseng. One of the phenolic antioxidants from fresh, white and reprocessed white ginseng was identified as phenol 2.6 Bis(1.1 dimethyl ethyl) 4-methyl among several unknown compounds by GC/mass spectrometer. In red ginseng, no phenol 2.6 Bis (1.1 dimethyl ethyl) 4-methyl was detected, the compound may be polymerized by phenoloxidase and form some higher molecular compounds which may possess high antioxidant activity. Phenoloxidase isozymes in fresh Korean ginseng (panax ginseng C.A. Meyer) were extracted with phosphate buffer at pH 7.3. The isozymes were purified through ammonium sulfate fractionation, dialysis and chromatography on a DEAE-cellulose column. Two groups of phenoloxidase were shown to be present, one in the floating agglomerated group and the other in the precipitate. group from the 0.85 saturation ammonium sulfate. The DEAE-cellulose column chromatography, the phenoloxidase isozyme present in the precipitate appears as the first peak (I), and that in the agglomerate in the second peak (II). Isozyme I showed higher activity with catechin and catechal, and isozyme II showed higher activity with p-cresol. The isozyme showed two optimum pH activity one at pH 4.5 and the other at 8.5 with catechin as substrate. Korean ginseng phenoloxidase has high heat stability. When heated at $75^{\circ}C$ for 2 hours, its activity remained $90\%\;and\;80\%$ on phenoloxidase I and II respectively. Phenoloxidase I was most active on (+) catechin followed by p-cresal, catechol and epicatechin. Phenoloxidase II was most active on p-cresal followed by (+) catechin, catechol, p-coumanic acid and epicatechin. Sodium bisulfite, sodium cyanide, ascorbic acid glutachion in the oxidized form, sodium diethyl dithiocarbomate and ethylendiamine tetra acetate (EDTA) acted as inhibitors. Red ginseng color development was initiated by phenoloxidase and finished by a followed sun drying process. The antiaging activity of ginseng may be initiated by the antioxidant in the ginseng.

Isolation and Identification of Antioxidants from Makgeolli (막걸리로부터 항산화 화합물의 단리.정제 및 동정)

  • The present study was carried out to isolate and identify of antioxidants from makgeolli. Makgeolli (3 L) was filtered and the residue was extracted by MeOH. The combined filtrates and MeOH extracts were successively solventfractionated by n-hexane, EtOAc, and BuOH. In the antioxidative activity against DPPH and $ABTS^+$ radicals of each fraction obtained after solvent-fractionation, EtOAc and BuOH layers showed higher activities than other fractions. Therefore, the two layers were respectively purified by column chromatography and HPLC. The isolated compounds were subjected to NMR and MS analyses and identified as 4-hydroxybenzaldehyde (1, 2.0 mg), 2-(4-hydroxyphenyl)ethanol (2, tyrosol, 15.3 mg), trans- and cis-ferulic acids (3 and 4, 1.2 mg), 1H-indole-3-ethanol (5, tryptophol, 3.4 mg), dimethyl succinate (6, 14.9 mg), succinic acid (7, 7.4 mg), and mono-methyl succinate (8, 7.8 mg). The presence of 1-5 in makgeolli have never before been reported.

Screening for Antioxidant Effects of Aerial Part Extracts Obtained from Sixteen Compositae Species (국화과 식물 16종 지상부 추출물의 항산화효과 탐색)

  • The attempts to develop natural antioxidants have been made with aerial part of 16 Compositae species by analyzing their phenolic compound contents, scavenging activities on 1,l-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radicals, ferrous ion chelating effects and inhibition effects on peroxidation of linoleic acids. Total polyphenol and flavonoid contents were highest in Matricaria recutica. Scavenging activity on DPPH and ABTS radicals were highest with extracts of Echinacea angustifolia and Serratula coronata var. insularis f. insularis, respectively. Aerial palt extracts of all species showed lower DPPH scavenging activity than ascorbic acid and 2,6-Di-tert-butyl-4-methylphenol (BHT). But Serratula coronata var. insularis f. insularis demonstrated higher ABTS scavenging activity than ascorbic acid and BHT. In Hieracium pilosella, Echinacea angustifolia, Matricaria reculica extracts showed higher ABTS scavenging activity than BHT. Ferrous ion chelating effects was highest with Matricaria recutica extract, but the effects were much lower than ethylenediaminetetraacetic acid (EDTA). The inhibition activity in lipid peroxidation of linoleic acids was highest in Eupatorium japonicum with 90.06% inhibition 4 days after reaction and 40.52% after 24 days. This demonstrated higher inhibition activity and longer lasting than BHT. Aerial part of Matricaria recutica for extraction source, rather than flower, has higher potential for antiox.idant material. In conclusion, development of natural antioxidants in Compositae is possible by studying antioxidant activity of each species.

Mineral Content and Antioxidants Activity of Portulaca oleracea (쇠비름의 무기물 함량 및 항산화 활성)

  • We investigated the mineral content and antioxidant activity of Portulaca oleracea for biological properties. Total mineral content was 6025.80 mg/100 g, and potassium was the highest at 3846.99 mg/100 g. Water and 80% ethanol extract yields were 14.84% and 24.93%, respectively. The contents of phenolic compounds and flavonoids of ethanol extract were 58.16 mg/g and 20.08 mg/g, and were significantly higher than that of its water extract (49.09 mg/g and 14.98 mg/g, respecitvely). Antioxidant activities and nitrite scavenging activity was significantly elevated in a dose-dependent manner, and that of ethanol extract was higher than that of the water extract. ${\alpha}$-Glucosidase inhibition activity of ethanol extract was significantly higher than the water extract as well. We suggest that the biological properties of Portulaca oleracea are due to its mineral and phenolic contents.

Comparision of SOD Activity and Phenolic Compound Contents in Various Korean Medicinal Plants (한국산 약용식물로부터의 SOD활성 및 Phenolic Compounds 함량 비교)

  • 82 species of Korean medicinal plants were measured to their SOD (superoxide dismutase) activity, total phenol and individual phenol compounds. The SOD activity of medicinal plants was varied from 3.67% to 48.3%, and especially, 4 species including Polygala tenuifolia Willd. showed activity over 40% higher than other species. The 16 individual phenol compounds concentrations have a severe variation in medicinal plants. Codonopsis lanceolata is the highest concentration $(12,176\;{\mu}g/g)$, and 10 species including Glycyrrhiza uralensis Fisch were showed to concentrations more than $1,000\;{\mu}g/g$. And also, salicylic acid was generally found in many plants, but trans-cinnamic acid, naringenin and kampferol was little found in plants. The total phenol by Folin-Dennis methods showed a respectively variation $(3{\sim}249.731\;{\mu}g/g)$ in 82 medicinal plants, and 7 species including Astragalus membranaceus showed low total phenol concentrations less than $10\;{\mu}g/g$. In conclusion, we expect that selecting medicinal plants with high biological activity might develop to chemicals related to antioxidants, as well as these become most popular substance at many industrial materials, and suggest to more researchs in future.

Bioactive Materials and Antioxidant Properties of Fermented Rice-bran Extract (쌀겨발효추출물의 이화학적 특성 및 항산화 작용)

  • This study suggests that fermented rice bran extract contains natural antioxidants. The contents of bioactive materials (e.g., polyphenolic compounds and flavonoids), antioxidative properties (DPPH (α,α'- diphenyl-β-picrylhydrazyl) free radical scavenging activity, Fe reducing, Cu reducing power, peroxidation of linoleic acid and rat hepatocyte microsome) were tested by in vitro experimental models using fermented rice bran (FRB) extract. The concentrations of phenolic compound and flavonoid were 19.92 mg/g and 11.56 mg/g, respectively. In oxidation in vitro models using DPPH free radical scavenging activity, (free radical scavenging activity 69.8%) Fe reducing power and Cu reducing power (effect of dose-dependent manner), Fe2+/ascorbate induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods (inhibition activity 81%), and autooxidation of rat hepatic microsomes membrane (lipid peroxidation inhibition activity 38%), antioxidative activities were stronger in FRB extract than FRS (Fermented Rice and Soybean, positive control) extract and, these effects were dose-dependent manner. From these results, FRB extract was shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. Overall, these results may provide the basic data to understand the antioxidative properties of fermented rice bran for development of functional foods.
