Comparative analysis of paeoniflorin, albiflorin and phenolic compound contents as bioactive components of peony was performed by Reverse Phase-High Performance Liquid Chromatography (RPHPLC) using the four- year-old peony which were different plant parts and pretreatment, such as removing or unremoving the cork layer of peony root before drying. The contents of paeoniflorin, albiflorin, (+)-taxifolin $3-O-{\beta}-D-glucopyranoside$, (+)-catechin and (-)-epicatechin were the highest in rhizome part, but those of gallic acid and benzoic acid in the leaves were higher than other parts. The contents of albiflorin, gallic acid, benzoic acid and (-)-epicatechin in the cork layer were higher than in those of the core, but the contents of paeoniflorin, (+)-taxifolin $3-O-{\beta}-D-glucopyranoside$ and (+)-catechin in the core were higher than those in the cork layer. In general, the rhizome part of peony root has been used only propagation purpose, but this part contained high contents of bioactive component. Therefore, it is needed that medicinal application of rhizome part in peony root was firmly investigated. Also, In the use of peony root for medicinal purpose, the use of peony root with cork layer can be efficient way on the practical use of useful components and the reduction of labor for removing the cork layer.
Onion vinegar has an undesirable flavor and taste that results from alcohol and acetic acid production from fermentation. In this study, we have used onion vinegar to develop an onion vinegar beverage with better sensory quality. The objective of this study was to determine the optimum blending ratio by using response surface methods to produce an onion vinegar beverage containing Yuza (Citrus junos Sieb ex Tanaka). The optimal formula for a fermented onion beverage was determined using a central composite design by the response surface methodology. The independent variables were obtained by regression analysis of the reaction surface of brown sugar, apple extracts and Yuza extracts. The optimum mixing ratio for onion vinegar:water:brown sugar:apple extracts:Yuza extracts was 6.0:77.6:4.9:9.2:2.3 (w/w). The actual overall acceptance was 7.08 under optimum conditions, which was close to the maximum predicted value of 6.96. The concentration of phenolic compounds, total flavonoids, and quercetin present in the onion vinegar beverage was 14.8 mg/100 g, 2.6 mg/100 g and 1.4 mg/100 g, respectively. The onion vinegar beverage showed antimicrobial activity against Staphylococcus aureus, Escherichia coli, Salmonella typhimurium and Enterobacter aerogenes. It also showed antioxidant effects, with a DPPH radical inhibition rate of 18.2% and superoxide dismutase (SOD)-like activity of 11.5%. In conclusion, the onion vinegar beverage described here seems to have nutritional value and potential biological activity.
This experiment has conducted to evaluate whether single injection of red ginseng extract including 50% ethanol extract, crude saponin, and lipid soluble fraction can induce oxidative burst of mouse peritoneal macrophages with use of fluorescence spectrophotometer. To optimize conditions of fluorescent spectrophotometry, concentrations of DCFH-DA(2', 7' -dichlorofluorescin diacetate) was 1.6 ${\mu}{\textrm}{m}$ and control oxidative burst by Zymosan A and PMA(phorbol myristate acetate) were 100$\mu\textrm{g}$, 250ng, respectively. Though in vitro macrophages failed to induce increment of H2O2 production, but 50% ethanol extract group induced significant enhancement of H2O2 production when zymosan A triggered oxidative burst. On the other hand, lipid soluble fraction enhanced significantly H2O2 production than that of control group. These findings consisted with the other reports which showed ginsenosides inhibited nitric oxide production and lipid soluble fraction activated colony stimulating factor(granulocyte - monocyte) activity in bone marrow stem cells. As is well known, lipid soluble fraction contains phenol compound, polyacetylene compound and alkaloids. Further study would unravel which component of it can induce H2O2 production of macrophages. We purified polyphenols from persimmon leaf and tested their biological activity. The 60% acetone extract was lyophilized and applied to test enzyme inhibition of glucosyltransferase and tyrosinase. GTase was 82.4% inhibited at $1.8{\times}10^{-1}$ mg/ml and tyrosinase 21.7% inhibited at 0.8 mg/ml. The acetone extract was fractionated into F-1, 2, 3, 4, 5 by Sephadex Q-50 gel filtration and the fraction-1 and 2 showed higher enzyme inhibition activity than the other fractions. To the Proteinase K treatment and autoclaving of the two fractions had no effect on the enzyme activity, but these results suggested that active fraction was not protein but phenol ring completed compounds. By Sephadex LH-20, MCI-gel and Bondapak $C_{18}$ column chromatographies, compouds 1, 2, 3 and 4 from F-1 fraction, compounds 5 and 6 from F-2 fraction and compounds 7 , 8 from F-3 fraction were purified and re-crystallized. The purified compounds was assumed to be condensed tannins of frame flavan-3-ol frame on the basis of color reagent reaction and to be a mixture of monomer, dimer and trimer according to TLC analysis.
Lactobacillus helveticus YM-1and Streptococcus lactis Ml$_3$ were inoculated together in reconstituted non-fat skim milk medium, and then their proteolytic activity and stimulatory compound for acid production were investigated. Significant difference between Lactobacillus helveticus YM-1 and Streptococcus lactis Ml$_3$was observed in the proteolytic activities. The proteolytic activity of Lactobacillus helveticus YM-1 and Streptococcus lactis Ml$_3$ was 105 $\mu\textrm{g}$/$m{\ell}$ and 30 $\mu\textrm{g}$/$m{\ell}$ when converted the amounts of hydrolysates of milk protein determined by Folin Ciocaleau phenol method into their tyrosine equivalent Stimulatory compounds in cell-free filtrate of Lactobacillus helveticus YM-1were identified as peptide with a molecular weight of approximately 4, 300 for the acid production by Streptococcus lactis Ml$_3$. Some kinds of amino acids, such as histidine, lysine, arginine and glutamic acid, were rich in acid hydrolysates of peptide. Among amino acids, histidine, glutamic acid and phenylalanine stimulated acid production, on the contrary isoleucine inhibited.
Terpene-modified phenolic resins were used to improve the tires wet traction related to the driving safety and also rolling resistance related to fuel efficiency. In this work, alpha-pinene, beta pinene, and delta limonene resins, which constitute different basic structures of terpene-modified phenolic resins, were individually added to the tread compounds of tires and their physical properties were compared with those of the alkyl phenol resin compounds. Alkyl phenolic resins showed no significant difference in tangent delta from terpene-modified phenolic resins at 0 ℃, which is related to wet traction, but showed higher tangent delta at 80 ℃, which is related to rolling resistance, indicating smaller fuel efficiency improvement effects. Among the terpene-modified phenolic resins, beta pinene one showed improved wet traction and fuel efficiency compared to those of other resins. Delta limonene resin showed the best wet traction improvement effect, and alkyl phenolic resins showed relatively high tensile strength and abrasion property. All terpene-modified resins exhibited better rolling resistance than those of alkyl phenolic ones so that they can be said to have better fuel efficiency improvement effects and also to improve other properties compared to those of blanks. Terpene-modified phenolic resins could be used when mixing tire compounds referring to the properties of the phenolic resins revealed in this work, which could result in preparing compounds with improved wet traction and rolling resistance.
Reductive dechlorination of chlorophenols by nickel coated iron was investigated to understand the feasibility of using Ni/Fe for the in situ remediation of contaminated groundwater. Zero valent iron (ZVI) was amended with Ni(II) ions to form bimetal (Ni/Fe). Dechlorination of 4-chlorophenol and formation of intermediates was studied using Ni/Fe. Effects of initial contaminant concentration, bimetal loading, presence of humic acid, and solution chemistry were also evaluated. Experimental results showed that Ni/Fe bimetal was so effective that more than 95% of 4-CP degradation was achieved within 240 minutes. Pseudo first-order rate constant for the dechlorination reaction was well correlated with bimetal loading. Humic acid competed for the reactive sites on the nickel coated iron with chlorophenols, lowering the dechlorination efficiency. No significant changes in solution pH were observed in the dechlorination of chlorophenols with Ni/Fe in the absence of buffer, indicating that reactivity of bimetal (Ni/Fe) could be prolonged. Phenol was found as a dechlorination intermediate of the conversion of 4-chlorophenol compound by Ni/Fe.
A board was manufactured for each resin and sawdust addition using the chaff made by carbonizing the chaff charcoal, an agricultural by-product that emerge during the rice pounding process, and sawdust. And effects of the additions of resin and sawdust on coefficients of dynamic and static modulus of elasticity, modulus of rupture, as well as the relationship between the dynamic modulus of elasticity, statis modulus of elasticity, and modulus of rupture were investigated. As phenol resin addition of chaff charcoal-sawdust compound board increases to 10~25%, the bending performance has increased. This suggests that resin addition largely effects the bending performance. Although the bending performance was gradually increased with the increase in sawdust addition, since the coefficients of determination (R2) between the sawdust addition with the coefficients of dynamic, static modulus of elasticity, and modulus of rupture were 0.4012, 0.0809, and 0.1971, respectively. Thus, it showed a relatively lower correlation, and the effect of sawdust on bending performance was small. Since a high correlation was confirmed between dynamic and static modulus of elasticity, and modulus of rupture of chaff charcoal-sawdust compound board, it was confirmed that prediction of static modulus of elasticity and modulus of rupture can be made in a nondestructive way from the dynamic modulus of elasticity.
To collect the basic data for the use of major softwood and hardwood vinegar, the chemical compositions were analized. In softwood vinegar the phenolic compounds derieved from lignin and extractives are vanillin, guaiacol, 3-methylphenol, acetoguaiacone, 1,2-benzenediol, ethylguaiacol, pyrocatechol. The compound derived from carbohydrates is 2-hydroxy-3-methyl-2-cyclopenten-1-one. The isolated compounds are the most in the phenolic fraction, and in the vinegar of Pinus koraiensis. In hardwood vingar 2-methoxyphenol, 2,6-djmethoxyphenol, 1-(4-hydroxy-3-methoxyphenyl) -ethanone, 3-methoxy-4-hydroxybenzylalcohol, 1-(4-hydroxy-3,5-dimethoxy-phenyl)-ethanone are the compounds derived from lignin and extractives. 2-hydroxy-3-methyl-2,4-cyclopen-1-one, methyl-4-t-butyl-2-furoate are the major compounds derived from the decomposition of carbohydrates. Many different kinds of compounds are analized in the neutral, phenolic and acidic fraction of hardwood vinegar The amount and kinds of compounds in hardwood vinegar are more higher than those of softwood than in softwood.
To analyze chemical compositions of softwood vinegar prepared with continuous carbonized kiln, the chemical compositions were analyzed by GC-MS spectrometry. The results were summarized as follow : 1. The amounts of methylalcohol and acetic acid and of vinegar were 0.12% and 0.8% respectively, and acidity was 0.85 2. Perfume components of vinegar were frufual, 5-meayl-2-furancarboxyaldehyde, 2,3-pentanedione, 2-butanol, 2,3-dihydrofuran, 1-(2-furanyl)-etanone, benzaldehyde, 2-furan carboxyaldehyde and acetic acid. 3 Vinegar prepared from softwood, so that murk amount of guaiacyl compound and phenol derivetives are produced from lignin and extractives was analγzed. 4 The yield of 4-methyl-di-tert-butylphenol was the highest in the nutural and carbonyl and acetic acid in the acid fractions, 3-ethylpentane in the basic fraction, and guaiacol in the phenolic fraction.
