Effect of Storage Temperature, Washing, and Cooking on Postharvest-treated Pesticide Residues in Polished Rice (쌀의 저장온도, 세척 및 취반이 일부 농약잔류의 제거에 미치는 효과)

  • Effect of storage temperature, washing, and cooking on postharvest-treated pesticide residues in polished rice was investigated. After being treated with each 500 mg/kg of captan, carbaryl, phenthoate, fenthion, fenitrothion, chlorpyriphos-methyl, pirimiphos-methyl, the polished rice was stored for 8 weeks at 4 and 3$0^{\circ}C$, respectively. The penetation rate of carbaryl was the highest as 27.5% and the others, 13 to 18%. The half lives of pesticide residues were estimated as 30 to 230 weeks at 4$^{\circ}C$, but 1 to 12 weeks at 3$0^{\circ}C$. The residues were reduced faster at 3$0^{\circ}C$ than at 4$^{\circ}C$. The half lives of pesticide residues by water washing were estimated as 0.7 to 4.6 trials and the removed with the washing trials. The residues of captan and carbaryl in cooked rice were removed 100 and 98%, respectively, comparing to initial residues concentration in treated rice but those of other 5 pesticides were removed 80%.

  • Water quality contamination issues are of critical concern to human health, whilst pesticide release generated from irrigated land should be considered for protecting natural habitats and human health. This paper suggests new method for evaluation and analysis using the GIS technique based on integrated spatial modeling framework. The pesticide use on irrigated land is a subset of the larger spectrum of industrial chemicals used in modern society. The behavior of a pesticide is affected by the natural affinity of the chemical for one of four environmental compartments; solid matter, liquid, gaseous form, and biota. However, the major movements are a physical transport over the ground surface by rainfall-runoff and irrigation-runoff. The irrigated water carries out with the transporting sediments and makes contaminated water by pesticide. This paper focuses on risk impact identification and assessment using GIS technique. Also, generated data on pesticide residues on farmland and surface water through GIS simulation will be reflected to environmental research programs. Finally, this study indicates that GIS application is a beneficial tool for spatial pesticide impact analysis as well as environmental risk assessment.

Study on the Development of Analytical Method (Multi-Pesticide Residue Method) for Organophosphate Pesticides (유기인제 농약 분석 방법 (Multi-Pesticide Residue Method) 개발에 관한 연구)

  • Organophosphate pesticides were extracted with 70% acetone and then transferred to dichloromethane. Extracts were applied to open-column chromatography with florisil. The florial extract was analyzed by gas chromatography with nitrogen-phosphorus detector(GC/NPD). Recoveries of the 18 organophosphate pesticides were ranged from 88.7% to 100. 0% for the narrow-bore capillary GC(Ultra-21. The minimum detectable level of this analytical method was 0.019 - 0.035 mg/kg. Sample throughput(extraction, open-column chro- matography, and GC analysts) was decreased considerably (8h per sample).

  • Much of the increase in agricultural productivity over the past half century has been due to the control of the pests with synthetic pesticides. The use of these pesticides has caused environmental problems and public health concern. The guidelines of maximum residue levels of pesticides in agricultural products has been well documented but more careful monitoring of their residues is required. Pyrethorid class pesticides are dominant in modern agricultural industry but public health concerns have been recently considered. The major route of pesticide exposure is the diet and with improved surveillance of pyrethorid residues in agricultural products their exposure should be controlled and minimized. In suitable products with reduced matrix effects such as agricultural products, aqueous samples, fruits and vegetables the use of immunoassays for pyrethorid residue monitoring could satisfy this requirement. Immunoassays have several advantages, namely they are highly sensitive, selective and cost-effective and enable large-scale sample handling and analysis in the laboratory.

  • The use pattern and survey data on the residue levels of organochlorine insecticides in Korea were summarized. On the basis of available data on food consumption and residue levels, the daily intake of the pesticide chemicals by average Korean adults were calculated to give $13.77\;{\mu}g$ of total BHC, $2.45\;{\mu}g$ of total DDT and $4.06\;{\mu}g$ of heptachlor and its epoxide. These intake levels were compared with the acceptable daily intake proposed by FAO/WHO and values obtained in other countries. It was then proposed to undertake a total diet study on pesticide chemicals for the sake of food safety assurrance and reasonable regulation of pesticides for food production in this country.

Development of Multi-Residue Methods for Carbamate Pesticides by the Enzyme Inhibition Test (효소 저해법을 이용한 Carbamate계 농약의 다성분 잔류분석법 개발)

  • This study was carried out with the detection for multiresidue of the carbamate pesticide such as carbaryl and cabofuran by enzyme-inhibition method. The check time for determination of acetylcholinesterase(AChE) activity was selected at 60 sec. The AChE activity in chicken brain determined by the Ellman's method was $162{\mu}$mol/min/g protein. $I_{50}$ for AChE by carbamate pesticide with wet kit was 0.169mg/L of carbaryl and 0.089mg/L of cabofuran, respectively. The incubation time for enzyme kit with substrate kit was 30min for determination of AChE activity. Enzyme kit with substrate kit was stable at $4^{\circ}C\;and\;25^{\circ}C$ for 5 days. Limit detection concentration of carbaryl with dry kit for AChE was 0.05mg/L. The dry kit such as wet kit applied Enzyme-Inhibition(EI) method with AChE was confirmed the multi residue method to detect the carbamate pesticides.

Residual characteristics of tolclofos-methyl treated by seed dressing in ginseng (인삼 중 종자분의 처리 tolclofos-methyl의 잔류 특성)

  • This study was carried out to evaluate residual characteristics of tolclofos-methyl in ginseng and elucidate the reason for its high detection rate from fresh ginseng selling at markets. Seeds of ginseng were sowed after seed dressing with tolclofos-methyl and after a year of growth, the young seedlings were transplanted to field. They were then harvested annually until three-years of growth and the pesticide residue was analyzed in them. LOD and LOQ of the pesticide were 0.001 and 0.003 mg/kg, respectively. Recovery test was carried out to validate the analytical method for tolclofos-methyl in ginseng. The ginseng seedlings were fortified with the test pesticide at the level of LOQ, ten times of LOQ and maximum residue concentration of tolclofos-methyl. Its recovery ranged from 77.37 to 100.16%. Residual concentration of tolclofos-methyl in ginseng seedlings just before transplanting and two-year-old ginseng were from 7.58 to 8.05 and from 6.46 to 6.79 mg/kg, respectively. In case of three-year-old ginseng, it was found to be from 4.18 to 4.35 mg/kg. As a result of annual pesticide residue analysis, concentration of the pesticide was found to decrease time-coursely in ginseng. This may be due to decomposition and increasing of fresh weight of the ginseng during the cultivation periods of three years.

Developing a Web-based System for Computing Pre-Harvest Residue Limits (PHRLs)

  • This study describes the development of a web-based system that collects all data generated in the research conducted to set pre-harvest residue limits (PHRLs) for agricultural product safety control. These data, including concentrations of pesticide residues, limit of detection, limit of quantitation, recoveries, weather charts, and growth rates, are incorporated into a database, a regression analysis of the data is performed using statistical techniques, and the PHRL for an agricultural product is automatically computed. The development and establishment of this system increased the efficiency and improved the reliability of the research in this area by standardizing the data and maintaining its accuracy without temporal or spatial limitations. The system permits automatic computation of the PHRL and a quick review of the goodness of fit of the regression model. By building and analyzing a database, it also allows data accumulated over the last 10 years to be utilized.

Establishment of Analytical Method for Residues of Ethychlozate, a Plant Growth Regulator, in Brown Rice, Mandarin, Pepper, Potato, and Soybean Using HPLC/FLD

  • BACKGROUND: Ethychlozate (ECZ) is a plant growth regulator of synthetic auxin for agricultural commodities (ACs). Accurate and sensitive method to determine ECZ in diverse ACs on global official purpose is required to legal residue regulation. As the current official method is confined to the limited type of crops with poor validation, this study was conducted to improve and extend the ECZ method using high-performance liquid chromatography (HPLC) in all the registered crops with method verification. METHODS AND RESULTS: ECZ and its acidic metabolite (ECZA) were both extracted from acidified samples with acetone and briefly purified by dichloromethane partition. ECZ was hydrolyzed to form ECZA and the combined ECZA was finally purified by ion-associated partition including hexane-washing. The instrumental quantitation was performed using HPLC/ FLD under ion-suppression of ECZA with no interference by sample co-extractives. The average recoveries of intra- and inter-day experiment ranged from 82.0 to 105.2% and 81.7 to 102.8%, respectively. The repeatability and reproducibility for intra- and inter-day measurements expressed as a relative standard deviation was less than 8.7% and 7.4%, respectively. CONCLUSION: Established analytical method for ECZ residue in ACs was applicable to the nation-wide pesticide residues monitoring program with the acceptable level of sensitivity, repeatability and reproducibility.
